ETHNOPHARMACOLOGICAL RELEVANCE: Eucommia ulmoides Oliv., a Chinese medicinal herb called "Duzhong" from the bark of Eucommia ulmoides Oliv., has been shown to possess significant protective effects in Parkinson's disease (PD). However, the molecular mechanism remains unclear. AIM OF THE STUDY: In this study, we explored the anti-neuroinflammatory mechanisms of Duzhong on the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mouse model to elucidate the traditional medical theories with modern pharmacological methods and to provide a reference for further clarifying its mechanisms of action. MATERIALS AND METHODS: The representative components in Duzhong extract were identified by UPLC-Q-TOF/MS. Male C57BL/6J mice were intraperitoneally injected with MPTP to establish an in vivo PD model. The pole, rotarod, and grip strength tests were performed to evaluate the motor coordination ability of the PD mice. HPLC-ECD was used to detect the striatal levels of dopamine (DA), 3,4- dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA). The expression of tyrosine hydroxylase (TH) was studied by immunohistochemistry (IHC) and Western blot assays. ELISA and Q-PCR were used examined the levels of proinflammatory cytokines in the serum and midbrain, respectively. Whole-transcriptome analysis of the midbrain was performed to explore the therapeutic effect of Duzhong on PD mice, and Q-PCR was then used to validate the differential gene expression changes in the PD mice treated with Duzhong. RESULTS: Ten compounds were identified from Duzhong extract. Duzhong significantly alleviated the behavioral impairments and dopaminergic neuron degeneration of PD mice, and inhibited the expression of proinflammatory cytokines. Whole-transcriptome analysis revealed nine oppositely regulated genes, and the Fosl2 gene was consistent with the trend of observed by RNA-seq. Furthermore, Duzhong downregulated mRNA expression of p38 and JNK, which are key upstream genes of Fosl2. CONCLUSIONS: Duzhong has promising therapeutic potential in PD mice, and its molecular mechanism is mediated by downregulating p38/JNK-Fosl2 gene expression to alleviate neuroinflammation.

OBJECTIVE: To evaluate the effect of decoction prepared with Yang-activating and stasis-eliminating medicinals from Traditional Chinese Medicine (TCM) on the intestinal mucosal permeability in rats with ulcerative colitis induced by dextran sulfate sodium (DSS). METHODS: Totally 55 male Wistar rats (body weight of 170-190 g) were randomly divided into the blank group (n = 10) and the model duplication group (n = 45). The blank group was not intervened, while the other was modeled with 5% dextran sulfate sodium by gavaging in a dosage of 4 mL per day to induce ulcerative colitis, a total of 7 days. Then, the model rats were divided into model blank group, mesalazine group and TCM group, and each group was consisted of 15 rats. They were given retention enema 10 min with normal saline, mesalazine enema (0.036 g/mL), and Yang-activating and stasis-eliminating decoction [0.54 g/mL of a decoction boiled by Puhuang (Pollen Typhae), Xiebai (Bulbus Allii Macrostemonis) and Wulingzhi (Faeces Trogopteri)] for 10 days respectively. Afterwards, all of the rats were evaluated by disease activity index (DAI), histological changes of distal colon, expression of occludin protein and ultrastructure of intestinal epithelial cells. Furthermore, ratio of lactulose to mannitol (L/M) discharged in urine was evaluated. RESULTS: Comparing the results between TCM and model control groups, scores of DAI and histological lesions decreased significantly (P = 0.000 < 0.01), ultrastructures of intestinal epithelial cells and tight junctions were more complete. The expression of occludin protein (P = 0.001 < 0.01) increased while the L/M value decreased significantly (P = 0.000 < 0.01) in TCM group. There was no statistical difference between the TCM and mesalazine groups in results of each item (P > 0.05). CONCLUSION: The decoction prepared with Yang-activating and stasis-eliminating TCM medicianls can restore intestinal mucosal epithelial cells and tight junctions the model rats with ulcerative colitis; it can reduce histological lesions and protect the permeability of intestinal mucosa barrier in the rats as well.

Chemical investigation of the whole plants of Valeriana hardwickii has led to the isolation of 11 flavones and 2 monoterpe- noids by using various chromatographic techniques including column chromatography on silica gel and Sephadex LH-20, preparative TLC, and preparative HPLC. Their structures were identified by spectroscopic data analysis as syzalterin (1), 6-methylapigenin (2), 5-hydroxy-7,4'-dimethoxyflavone (3), genkwanin (4), acacetin (5), apigenin (6), quercetin (7), tricin (8), (-)-farrerol (9), sosakuranetin (10), 5,3',4'-trihydroxy-7-methoxyflavanone (11), (-)-bornyl ferulate ( 12) , and (-)-bornyl caffeate ( 13). All compounds were isolated from this plant for the first time, while compounds 1, 9-13 were obtained from this genus for the first time.

OBJECTIVE: Discussion on assessment and intervention models to promote the rational use of medicines of Chinese medicine injection effect. METHOD: Using systematic prescription assessment and intervention work mode, formed expert group guide established assessment standard, developed the prescription audit specification, and extracted all Chinese medicine injection prescription of outpatient 2010 first quarter (in front of intervention) and 2011 first quarter (behind intervention), respectively for 2 543 and 3 122. RESULT: The percent of the non-indication of medication in front of intervention outpatient fell from 3.44% to behind intervention of 2.66% (P<0.05), the percent of contraindicated medication in front of intervention outpatient fell from 2. 14% to behind intervention of 1.33% (P<0.05), the incidence of adverse reactions in front of intervention outpatient fell from 1.01% to behind intervention of 0.29% (P<0.05). CONCLUSION: The modes of prescription assessment and intervention have obvious results for the promotion of rational use of traditional Chinese medicine injections.

Calcium peroxide is applied to sediment remediation. Based on laboratory-simulated test, trend of soluble reactive phosphorus (SRP) concentration in overlying was measured after two means of calcium peroxide treatment, mixing and capping. Transformation among different phosphorus fractions in the sediments was studied to analyze the distinguished effect. The results show that SRP concentration in overlying water presents different trend after mixing and capping of calcium peroxide. SRP concentration of the former decreased at first and increased subsequently, while that of the latter kept decreasing and average release velocity of SRP was 6.077 mg/(m2 x d) and -0.879 mg/(m2 x d) respectively. The transformation of phosphorus fractions in the sediments shows that diverse change of iron-bound phosphate are deemed to cause the effect under different adding options of calcium peroxide.
