RESUMEN

Background: Primary ovarian insufficiency (POI) is a common gynecological disease with serious ramifications including low pregnancy rate and low estrogen symptoms. Traditional Chinese medicine is regarded as an effective treatment for POI. However, the therapeutic mechanism of it is unclear. Methods: In this study, a mouse model of primary ovarian insufficiency was established by intraperitoneal injection of cyclophosphamide (CTX) and He's Yang Chao Recipe (HSYC) concentrate was used for intragastric administration. Serum hormone levels (Anti-Müllerian Hormone, Estradiol, Progesterone, Luteinizing Hormone and Follicle Stimulating Hormone) and Oxidative Stress (OS) related products, superoxide dismutase (SOD), GSH-Px, and malondialdehyde (MDA) were measured by enzyme-linked immunosorbent assay. Pathological changes in ovarian tissue were evaluated by hematoxylin and eosin staining, and flow cytometry was used to determine reactive oxygen species content and mitochondrial membrane potential levels in granulosa cells. Mitochondrial distribution and morphology were investigated using immunofluorescence staining. The level of mitophagy was evaluated by LC3 immunofluorescence staining and autophagosome counts using electron microscopy. Western blotting and qPCR were used to detect the expression of proteins and genes related to mitophagy and the NLRP3 inflammasome. Results: After HSYC treatment, the ovarian damage was milder than in the CTX group. Compared with the CTX group; SOD, GSH-Px, and the total antioxidant capacity were significantly increased, while MDA and ROS were decreased in the HSYC treatment groups. Furthermore, mitochondrial distribution and membrane potential levels were improved after HSYC treatment compared to the CTX group. After the HSYC treatment, the LC3 fluorescent intensity and autophagosome counts were decreased. Similarly, mitophagy related markers PINK1, Parkin, LC3, and Beclin1 were decreased, while p62 was significantly increased, compared with the CTX groups. The mRNA and protein expression of NLRP3 inflammasome, NLRP3, caspase-1, GSDMD, IL-18, and IL-1ß were significantly decreased in the HSYC treatment groups. Conclusion: This is the first study in molecular mechanisms underlying HSYC against granulosa cell injury in POI. HSYC protects ovaries from CTX-induced ovarian damage and oxidative stress. HSYC enhanced ovarian function in mice with primary ovarian insufficiency by inhibiting PINK1-Parkin mitophagy and NLRP3 inflammasome activation.

Panax notoginseng, a traditional Chinese medicine, exerts beneficial effect on diabetic kidney disease (DKD), but its mechanism is not well clarified. In this study we investigated the effects of ginsenoside Rb1 (Rb1), the main active ingredients of Panax notoginseng, in alleviating podocyte injury in diabetic nephropathy and the underlying mechanisms. In cultured mouse podocyte cells, Rb1 (10 µM) significantly inhibited high glucose-induced cell apoptosis and mitochondrial injury. Furthermore, Rb1 treatment reversed high glucose-induced increases in Cyto c, Caspase 9 and mitochondrial regulatory protein NOX4, but did not affect the upregulated expression of aldose reductase (AR). Molecular docking analysis revealed that Rb1 could combine with AR and inhibited its activity. We compared the effects of Rb1 with eparestat, a known aldose reductase inhibitor, in high glucose-treated podocytes, and found that both alleviated high glucose-induced cell apoptosis and mitochondrial damage, and Rb1 was more effective in inhibiting apoptosis. In AR-overexpressing podocytes, Rb1 (10 µM) inhibited AR-mediated ROS overproduction and protected against high glucose-induced mitochondrial injury. In streptozotocin-induced DKD mice, administration of Rb1 (40 mg·kg-1·d-1, ig, for 7 weeks) significantly mitigated diabetic-induced glomerular injuries, such as glomerular hypertrophy and mesangial matrix expansion, and reduced the expression of apoptotic proteins. Collectively, Rb1 combines with AR to alleviate high glucose-induced podocyte apoptosis and mitochondrial damage, and effectively mitigates the progression of diabetic kidney disease.

BACKGROUND: Garcinia subelliptica (Fukugi in Japanese) is an evergreen tropical tree, first identified in Batanes, the Philippines, which has been planted as a homestead windbreak and in coastal forests extensively on the Ryukyu Archipelago, Japan. This article focuses on the traditional uses and cultural values of Fukugi trees and provides ethnobotanical information for the conservation scheme of this important tree species. METHODS: A combination of ecological and ethnobotanical approaches was applied in this study. Extensive field surveys were conducted to collect dimensions of relatively large trees, and in-depth interviews with the village leaders and knowledgeable persons were conducted to collect ethnobotanical data. RESULTS: Fukugi trees have been primarily planted as homestead or farmland windbreaks. Timber was harvested during difficult times, for example, after WWII, and used for recreational purposes for children or farmers. The fruits were also eaten on some remote islands. Old-growth Fukugi trees are widely found in sacred sites, within cities, and as symbolic trees. The older generations respect Fukugi trees; however, the cultural significance valued by older generation seems to be lacking in the younger generation. We argue that Fukugi is a cultural keystone species in Okinawa, which underpins Ryukyu culture and has transformed islands into a pleasant land, a unique place, and shared identity for the community. CONCLUSIONS: Fukugi windbreaks provide significant ecosystem services, such as biodiversity in the forest, reducing soil erosion, and spiritual and cultural values. A combination of biophysical environment, as well as tradition and custom, has played an essential role in tree species selection for windbreaks. The positive impacts that anthropogenic activities have had on the sustainability of woody species, namely, the active utilization of tree species, may have enabled the species to sustain. Strategies for protecting old-growth Fukugi trees, in addition to restoration of damaged trees, are needed to improve the sustainable management of Fukugi trees in Okinawa.

PURPOSE: To examine radiofrequency-induced tissue heating around intracranial aneurysm clips during a 7 Tesla (T) head MR examination. METHODS: Radiofrequency (RF), temperature simulations, and RF measurements were employed to investigate the effects of polarization and clip length on the electric field (E-field) and temperature. Heating in body models was studied using both a conservative approach and realistic exposure scenarios. RESULTS: Worst-case orientation was found for clips aligned parallel to the E-field polarization. Absolute tissue temperature remained below International Electrotechnical Commission regulatory limits for 44 of 50 clinical scenarios. No significant effect on heating was determined for clip lengths below 18.8 mm, and worst-case heating was found for clip length 51.5 mm. The conservative approach led to a maximum permissible E-field of 72 V/m corresponding to B1+ of 1.2 µT, and an accepted power of 4.6 W for the considered RF head coil instead of 38.5 W without clip. CONCLUSION: Safe scanning conditions with respect to RF-induced heating can be applied depending on the information about the clip gained during screening interviews. However, force and torque measurements in the MR system shall be conducted to give a final statement on the MR safety of aneurysm clips at 7T. OBJECTIVE: To observe the clinical efficacy of acupuncture combined with massage on infiltrative exophthalmos so as to provide a more effective treatment for infiltrative exophthalmos in hyperthyroidism. METHODS: Forty-five cases of infiltrative exophthalmos were randomly divided into an acupuncture massage group (25 cases) and a western medicine group (20 cases). In acupuncture massage group, acupuncture was applied mainly at Jingming (BL 1), Qiuhou (EX-HN 7), Chengqi (ST 1) and Shangming (Extra); and massage was applied in combination on the back of the neck and the local points around the orbit. In western medicine group, Dexamethasone and Methotrexate were used for intravenous infusion, and Prednisone was for oral administration. The exophthalmos extent and clinical efficacy were compared between two groups. RESULTS: Before and after the treatment, the exophthalmos extents were (20.27 +/- 1.22) mm and (18.11 +/- 0.87) mm in acupuncture massage group and were (20.34 +/- 1.20) mm and (19.47 +/- 1.11) mm in western medicine group separately, indicating remarkable improvement in two groups (both P < 0.01). The improvement in acupuncture massage group was superior remarkably to that in western medicine group (P < 0.01). The total effective rate (83.3%, 40/48) in acupuncture massage group was superior to that (53.8%, 21/39) in western medicine group, and with less adverse reaction. CONCLUSION: Acupuncture combined with acupoint massage is an advanced therapy and has definite efficacy on infiltrative exophthalmos, which is superior to the conventional treatment in western medicine.
