RESUMEN

This study was conducted to investigate the effects of providing oxygenated and hydrogenated water on the growth performance, blood biochemical parameters, and immunoglobulin concentrations and antioxidant enzyme activity of broiler chickens. In our investigation, 144 Ross × Ross broiler chicks were randomly allotted to three different treatment groups with four replicates (treatment × replicate × bird = 3 × 4 × 12). All chicks were given one of the following types of water for five weeks: tap water (CON), hydrogenated water (HNW), and oxygenated water (ONW). ONW supplementation increased the final body weight and weight gain and also improved both feed intake and feed conversion of broiler chickens as compared to those of CON broiler chickens (P < 0.05). The abdominal fat and its ratio to the final body weight showed that fat accumulation in the broiler chicken abdomen was reduced when broiler chickens drank only ONW for five weeks (P < 0.05). ONW supplementation improved blood parameters, including triacylglyceride, total cholesterol, and low-density lipoprotein-cholesterol. Additionally, in accordance with a globulin increase in broiler chickens, both IgG and IgM generation were significantly enhanced when ONW was supplied to broiler chickens (P < 0.05) but only a numerical advance was observed in the HNW group (P > 0.05). Both oxygenated and hydrogenated water supplementation significantly improved the antioxidant effects (P < 0.05), and it seems that superoxide dismutase refinement was completed due to oxygen and/or hydrogen enhancement of drinking water. These results indicate that oxygen enhancement of drinking water may be recommended to improve growth performance by increasing immunoglobulins mainly IgG and IgM.

The aim of the present study was to evaluate the protective effect of aqueous extract from Platycodon grandiflorum (BC703) on bile duct ligation (BDL)-induced hepatic fibrosis in rats. BDL rats were divided into three groups, which orally received distilled water or BC703 (10 or 50 mg/kg/day) for consecutive 28 days. Antifibrotic effects of BC703 on BDL-induced hepatic fibrosis in rats were estimated by assessing serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), blood urea nitrogen (BUN), transforming growth factor-beta 1 (TGF-ß1) and hepatic levels of malondialdehyde (MDA), glutathione (GSH), total superoxide dismutase (SOD) and nitric oxide (NO). The biochemical observations were supplemented by histopathological examination of liver samples stained with hematoxylin and eosin and Masson's trichrome stain. ALT, AST, TBIL and BUN were elevated in the group treated with BDL alone than in the sham-operated group. These elevations were significantly decreased by BC703 treatment. Hepatic GSH and SOD levels, depressed by BDL, were also increased in the BC703 group. In addition, increases in hepatic MDA and NO levels in the BDL-induced cholestasis were attenuated by BC703 treatment. Furthermore, BC703 treatment significantly reduced the serum level of fibrogenic cytokine, TGF-ß1. Histopathological studies further substantiated the protective effect of BC703 on BDL-induced hepatic fibrosis in rat. BC703 may have beneficial effects not only on hepatic fibrosis by cholestasis but also on hepatic fibrosis development in patients with chronic hepatic disease.

The effects of sequentially combined carbon (SCC) using a symbiotic relationship of methanol and acetic acid on biological nutrient removal were investigated in both the continuous bench scale process consisting of an anoxic, an aerobic and a final settling tank and intensive batch tests. Compared to the use of respective sole carbon sources, methanol and acetic acid, the use of SCC showed superior removal efficiency of nitrogen (98.3%) and phosphorus (approximately 100%). Furthermore, the use of SCC enhanced simultaneous denitrification and phosphorus uptake by denitrifying phosphorus removal bacteria (DPB), resulting in the highest specific denitrification rate (SDNR) of 0.252 g NO3-N/g VSS/d achieved from the first anoxic zone with methanol of 30 mg COD/I. From batch tests performed under carbon limited anoxic conditions, 1 g of nitrate was used by DPB for P-uptake of 1.19 g. According to this result, 0.205 g NO3-N/g VSS/d was accomplished by normal denitrifiers using methanol, and 0.047 g NO3-N/g VSS/d was achieved by DPB. This research also demonstrated that the increase of poly-beta-hydroxybutyrate (PHB) stored by phosphorus accumulating organisms (PAOs) could be of importance in improving aerobic denitrification. The use of SCC produced the highest P-release in the anoxic zone, indicating the amount of PHB would be higher compared to the use of other sole carbons. Therefore, the SCC could be a very effective carbon source for the enhancement of aerobic denitrification as well.

RESUMEN

The sequentially combined carbon (SCC) of methanol and acetic acid was used for the biological nutrient removal (BNR). Its BNR performance was compared with methanol or acetic acid as a sole carbon substrate. Compared to the sole carbon substrate, the use of SCC demonstrated the highest overall TIN removal of 98.3% at a COD ratio of 30 mg COD/l of methanol/50 mg CDO/l of acetic acid. Furthermore, denitrification was more enhanced when methanol was used as one of the SCC, rather than as a sole carbon source. Complete phosphorus removal was accomplished with a non-detectable o-P concentration when SCC was added. This research also showed that aerobic denitrifiers appear to prefer acetic acid to methanol, and the amount of poly-beta-hydroxybutyrate (PHB) stored by P accumulating organisms (PAOs) using acetic acid in the anoxic zone could be another important factor in improving the aerobic denitrification. The SCC was a very favorable carbon source for the aerobic denitrification since acetic acid was utilized more efficiently for P-release in accordance with increase of PHB stored in the cell of PAOs by removing nitrogen first using methanol.

To define the role of caspase-3 in H2O2-induced apoptosis, we introduced caspase-3 cDNA into MCF-7 breast carcinoma cells that otherwise lack caspase-3 expression. H2O2 treatment induced DNA fragmentation and nuclear condensation in the caspase-3-expressing cells, but not in the caspase-3-deficient cells. This indicated that caspase-3 is essential for nuclear events. However, H2O2 induced an externalization of membrane phosphatidylserine (PS) and cell death regardless of caspase-3 expression. These events were not suppressed by Ac-DEVD-CHO and Z-VAD-fmk, which inhibit DEVD-specific caspases and a broad spectrum of caspases, respectively. In Jurkat T cells, these inhibitors abolished H2O2-induced PS relocalization, but not cell death. Therefore, caspases appear to be dispensable for lethality by H2O2, but required for PS redistribution in a cell-type-specific manner.

The purpose of this 85-day study was to investigate the long-term effects of histidine depletion on nitrogen utilization in young adult men. A low nitrogen (6.3 g/day), low histidine (10 mg/day) amino acid diet was fed to seven men for 8 weeks. Mean nitrogen balance became negative at the end of the 8-week period. Free histidine in postabsorptive plasma and 24-hour urine decreased significantly during the first 2 weeks of the depletion and remained low and constant for the remaining 6 weeks. Hemoglobin concentration decreased somewhat, and serum iron concentration increased significantly during histidine depletion. Lean body mass, urinary N'-methylhistidine and total creatinine did not change significantly. On addition of histidine to the low histidine diet for 2 weeks, nitrogen retention became positive, plasma and urinary histidine returned to initial values, serum iron fell, and hemoglobin concentration slowly increased. These parameters remained unchanged in two control men fed the same diet supplemented with histidine (1.05 g/day) for 8 weeks. The results suggest that histidine is indispensable for young men consuming a low nitrogen diet.
