RESUMEN

Myocarditis has been reported in male F344 rats given a diet containing hinokitiol (HT). A subchronic toxicity study was here performed to re-evaluate toxic effects of HT in both sexes of F344 rats with dietary administration at concentrations of 0%, 0.02%, 0.07% and 0.2% for 13 weeks. Significant reduction of body weight gain was noted in 0.2% males and 0.07% and above females. Significant decrease in RBC counts, hemoglobin and hematocrit was detected in 0.07% and 0.2% females. Significant increase in MCV was observed in 0.07% and above males and 0.2% females. In the rats given 0.07% and 0.2%, significant increase in total protein and albumin were detected in males, and in total cholesterol in females. Significant increases in total cholesterol, urea nitrogen and creatinine were also detected in the 0.2% males. Significant increase in relative liver weights was detected in the 0.07% and above males and females. Absolute and relative heart weights were significantly decreased in the 0.07% and above males. Based on the above findings the no-observed-adverse-effect level (NOAEL) of HT for both male and female rats was estimated to be 0.02%, translating into 12.7 and 14.8 mg/kg b.w./day, respectively. Myocarditis was not evident in the present study.

A subchronic toxicity study of soybean extract was performed in F344 rats with dietary administration at concentrations of 0%, 1.25%, 2.5% and 5% for 13 weeks. No mortality or abnormal clinical signs in any group were observed. Body weight gains were decreased with a tendency for reduction of feed intake in the 1.25% and above female and 5% male groups. In males, absolute and relative liver weights were increased in the 1.25% and above groups. In females relative kidney weights were increased in the 1.25% and above groups. Other significant changes such as decreased RBC and hematocrit and increased urea nitrogen were detected in the 2.5% and/or 5% groups. On histopathological observation, atrophy of the ventral prostate was observed in all animals in the 5% male group. Mucification and atrophy of the vaginal epithelium and increased atretic follicles in ovaries were noted in 2.5% and 5% female rats. Based on the above findings the lowest-observed-adverse-effect level for male and female rats was estimated to be 1.25% (707.2 and 751.8 mg/kg b.w./day, respectively).

Green tea catechins (GTC), polyphenols extracted from the stalks and leaves of Camellia sinensis, are found in the different types of tea beverages and as antioxidant additives to many foods, snacks, fats and fatty oils. As a part of their safety assessment, subchronic toxicity was investigated in male and female F344 rats with dietary administration at concentrations of 0 (control), 0.3%, 1.25% and 5.0% for 90 days. The average daily intakes of GTC in each group were 180, 764 and 3525mg/kg body weight/day, respectively for males, and 189, 820 and 3542mg/kg body weight/day, respectively for females. No mortality or obvious clinical signs were observed throughout the experimental period but body weights were reduced from week 1 to the end of the experiment in 5.0% males. In serum biochemistry, alanine transaminase and alkaline phosphatase in 5.0% males and females and aspartate transaminase in 5.0% females were increased, together with the relative liver weights in both sexes receiving 5.0%. Although decreases were evident for total cholesterol in 0.3-5.0% males and triglycerides in 1.25% and 5.0% males and 5.0% females, these changes were not considered to be adverse. Hematology and histopathological observation revealed no GTC-related toxicological changes. Based on above findings, the no observed adverse effect level (NOAEL) of GTC was estimated to be 1.25% (764mg/kg body weight/day for males and 820mg/kg body weight/day for females).

In this study, we characterized the effects of 25-hydroxyvitamin D3 (25-OH D3) and manipulated dietary cation-anion difference (DCAD) on the performance, urine pH, serum constituents, carcass traits, tissue residual vitamin D and its metabolites, beef tenderness, and mRNA and protein concentrations of Ca-dependent proteinases in LM using 24 cull native Korean cows. The cows were divided into 3 groups of 8: control, 25-OH D3 supplemented (25-OH D3), and manipulated DCAD plus 25-OH D3 supplemented (DCAD+25-OH D3). Cows receiving 25-OH D3 or DCAD+25-OH D3 were dosed with 125 mg of 25-OH D3 6 d before slaughter. The manipulated DCAD (-10 mEq/100 g of DM) diet was fed from 20 to 6 d (14 d) before slaughter. The DCAD+25-OH D3 treatment decreased urine pH and increased serum Ca concentrations. Although the vitamin D concentrations in LM, liver, and kidney were not affected by 25-OH D3 or DCAD+25-OH D3, muscle tissue 25-OH D3 concentrations were increased by both regimens. Serum 25-OH D3 concentrations were increased by 25-OH D3 supplementation, and the increase was even greater for DCAD+25-OH D3. The same pattern was observed for serum 1,25- (OH)2 D3. However, the LM concentration of 1,25-(OH)2 D3 was less for DCAD+25-OH D3 than for control. Although Ca concentrations of LM increased numerically in response to 25-OH D3 supplementation, no statistical differences in Warner-Bratzler shear force or sensory traits of LM were detected. The LM of cows receiving 25-OH D3 with or without manipulated DCAD had greater concentrations of mu-calpain and m-calpain mRNA, whereas the reverse was observed for calpastatin mRNA. Expression of mu-calpain protein was increased relative to control by DCAD+25-OH D3. The amount of 25-OH D3 and manipulated DCAD administered to cull native Korean cows was insufficient to improve tenderness of beef by increasing muscle Ca concentration. However, DCAD+25-OH D3 induced greater expressions of mu-calpain protein as well as mRNA.

To reduce the residual organic matter and phosphorus contained in secondary effluent, a biofiltration system combined with electrocoagulation using bipolar iron electrodes was evaluated as a supplementary treatment to existing small-community sewage treatment. Based on the results of batch tests, bipolar electrocoagulation (BEC) was found to be more effective on phosphorus removal than monopolar electrocoagulation (MEC) but energy consumption was less in monopolar electrocoagulation. Optimum conditions of BEC to treat the secondary effluent were current density 15 A/m2, electrode spacing 1 cm and pH < 8. The removals of COD(Cr) and phosphorus by biofiltration system without BEC were 69.1% and 9.6%, respectively. However, biofiltration system combined with BEC showed 76.6-83.7% and 70.7-93.0% removal for COD(Cr) and phosphorus respectively. Extraordinary increase in phosphorus could be achieved by introducing electrocoagulation to biofiltration, and BEC/biofiltration system was evaluated to be applicable to existing small-community sewage treatment plants as a supplementary process.

Our interest in improving the efficiency of targeted scission reagents has prompted us to study the influence of ring substituents on the nuclease activity of 1,10-phenanthroline-copper conjugated to oligonucleotides and DNA-binding proteins. Since methyl substitution at all but the 2 and 9 positions enhances the copper-dependent chemical nuclease activity of 1,10-phenanthroline, we have compared the activity of conjugates prepared from 5-(aminomethyl)-1,10-phenanthroline (MOP) to those of conjugates prepared from 5-amino-1,10-phenanthroline (amino-OP). Tethering MOP derivatives to the Escherichia coli Fis protein enhances DNA scission several-fold at the weaker cleavage sites initially observed with conjugates prepared from amino-OP. However, scission efficiency is not increased at the stronger cleavage sites, or when scission is targeted to single-stranded DNA by a complementary oligonucleotide. These results are consistent with a change in the rate-determining step for cleavage associated with the differential accessibility of the DNA-bound coordination complex to solvent and reductant. Although the free bis cuprous complex of 2,9-dimethyl-1,10-phenanthroline (neocuproine) is redox-inactive, an oligonucleotide tethered to neocuproine through C5 of the phenanthroline ring efficiently cleaves a complementary DNA sequence. These results establish that the nucleolytic species in targeted scission is the 1:1 cuprous complex and suggest that the oxidative reaction proceeds through a copper-oxo intermediate rather than a metal-coordinated peroxy species. However, substituents at the 2 and 9 positions of the ligand will often hinder close approach of the phenanthroline-copper moiety to the oxidatively sensitive ribose as shown by the preference of the oligonucleotide-targeted chimera for cleavage of single-stranded regions and the failure of neocuproine-DNA-binding protein chimeras and a C2-tethered chimera to cleave DNA.
