RESUMEN

Generally, in gymnosperms, pollination and fertilization events are temporally separated and the developmental processes leading the switch from ovule integument into seed coat are still unknown. The single ovule integument of Ginkgo biloba acquires the typical characteristics of the seed coat long before the fertilization event. In this study, we investigated whether pollination triggers the transformation of the ovule integument into the seed coat. Transcriptomics and metabolomics analyses performed on ovules just prior and after pollination lead to the identification of changes occurring in Ginkgo ovules during this specific time. A morphological atlas describing the developmental stages of ovule development is presented. The metabolic pathways involved in the lignin biosynthesis and in the production of fatty acids are activated upon pollination, suggesting that the ovule integument starts its differentiation into a seed coat before the fertilization. Omics analyses allowed an accurate description of the main changes that occur in Ginkgo ovules during the pollination time frame, suggesting the crucial role of the pollen arrival on the progression of ovule development.

KEY MESSAGE: BbrizGID1 is expressed in the nucellus of apomictic Brachiaria brizantha, previous to aposporous initial differentiation. AtGID1a overexpression triggers differentiation of Arabidopsis thaliana MMC-like cells, suggesting its involvement in ovule development. GIBBERELLIN-INSENSITIVE DWARF1 (GID1) is a gibberellin receptor previously identified in plants and associated with reproductive development, including ovule formation. In this work, we characterized the Brachiaria brizantha GID1 gene (BbrizGID1). BbrizGID1 showed up to 92% similarity to GID1-like gibberellin receptors of other plants of the Poaceae family and around 58% to GID1-like gibberellin receptors of Arabidopsis thaliana. BbrizGID1 was more expressed in ovaries at megasporogenesis than in ovaries at megagametogenesis of both sexual and apomictic plants. In ovules, BbrizGID1 transcripts were detected in the megaspore mother cell (MMC) of sexual and apomictic B. brizantha. Only in the apomictic plants, expression was also observed in the surrounding nucellar cells, a region in which aposporous initial cells differentiate to form the aposporic embryo sac. AtGID1a ectopic expression in Arabidopsis determines the formation of MMC-like cells in the nucellus, close to the MMC, that did not own MMC identity. Our results suggest that GID1 might be involved in the proper differentiation of a single MMC during ovule development and provide valuable information on the role of GID1 in sexual and apomictic reproduction.

Previous ERP studies have shown that musicians detect a pitch change in spoken sentences better than non-musicians in both native (French, Schön et al., 2004) and unfamiliar (Portuguese, Marques et al., 2007) language. The aim of the present study was to further investigate differences between musicians and non-musicians in processing pitch changes in spoken sentences. To study the effects of familiarity of intonational contour and of the presence of meaningful context, behavioral and electrophysiological data from Italian musicians and non-musicians were compared in a pitch incongruity detection task using sentences in the native (Italian) and foreign (French) language and in jabberwocky (meaningless sentences formed by pseudowords). Moreover, to examine whether these differences depend on enhanced auditory sensitivity to pitch, the frequency discrimination threshold (FDT) for tones was obtained using a psychophysical procedure. Musicians were more accurate than non-musicians in detecting small pitch changes in all languages showing a smaller response bias, as well as much lower FDTs than non-musicians. The ERP data revealed shorter latencies of a late positivity over parietal sites in musicians than in non-musicians for weak and strong incongruities. Overall results confirmed musicians' advantage in detection of subtle pitch changes not only with tones but also with speech sentences in both native and unfamiliar languages. Such effect appears to emerge from more efficient pitch analysis trained by musical experience.

This study aimed at investigating the effects of acoustic distance and of speaker variability on the pre-attentive and attentive perception of French vowels by French adult speakers. The electroencephalogram (EEG) was recorded while participants watched a silent movie (Passive condition) and discriminated deviant vowels (Active condition). The auditory sequence included 4 French vowels, /u/ (standard) and /o/, /y/ and /ø/ as deviants, produced by 3 different speakers. As the vowel /o/ is closer to /u/ than the other deviants in acoustic distance, we predicted smaller mismatch negativity (MMN) and smaller N1 component, as well as higher error rate and longer reaction times. Results were in line with these predictions. Moreover, the MMN was elicited by all deviant vowels independently of speaker variability. By contrast, the Vowel by Speaker interaction was significant in the Active listening condition thereby showing that subtle within-category differences are processed at the attentive level. These results suggest that while vowels are categorized pre-attentively according to phonemic representations and independently of speaker variability, participants are sensitive to between-speaker differences when they focus attention on vowel processing.

Initiation of transcription mediated by RNA polymerase II requires a number of transcription factors among which TFIID is the major core promoter recognition factor. TFIID is composed of highly conserved factors which include the TATA-binding protein (TBP) and about 14 TBP-associated factors (TAFs). Recently, the complete Arabidopsis TAF family has been identified. To obtain functional information about Arabidopsis TAFs, we analyzed a T-DNA insertion mutant for AtTAF6. Segregation analysis showed that plants homozygous for the mutant allele were never found, indicating that inhibition of the AtTAF6 function is lethal. Genetic experiments also revealed that the male gametophyte was affected by the attaf6 mutation since significant reduced transmission of the mutant allele through the male gametophyte was observed. Detailed histological and morphological analysis showed that the T-DNA insertion in AtTAF6 specifically affects pollen tube growth, indicating that the transcriptional regulation of only a specific subset of genes is controlled by this basal transcription factor.
