RESUMEN

Tacca leontopetaloides (T. leontopetaloides) contain a number of active compounds such as flavonoids, tannins, phenolics, steroids, and alkaloids. The active compounds from plants have been shown to reduce the risk of cardiovascular disease by lowering cholesterol levels by inhibiting the enzyme 3-hydroxy-3-methylglutaryl-coenzym A (HMG-CoA) reductase activity. This study aims to investigate the potential active compounds in the ethanolic extract of Tacca tubers (T. leontopetaloides) from the Banyak Islands, Aceh Singkil Regency, Aceh Province both in vitro and in silico. Tacca tubers contain secondary metabolites including flavonoids, phenolics, tannins, steroids and saponins, according to phytochemical screening. In vitro investigation of ethanolic extract of Tacca tuber revealed inhibitory activity of HMG Co-A reductase with an IC50 value of 4.92 ppm. Based on the in silico study, active compound from the extract, namely Stigmasterol with the highest binding affinities with HMG Co-A reductase (-7.2 kcal/mol). As a comparison, the inhibition of HMG Co-A reductase activity by simvastatin with an IC50 4.62 ppm and binding affinity -8.0 Kcal/mol. Our findings suggest that the ethanolic extract of Tacca tuber (T. leontopetaloides) from Banyak Islands, Aceh Province has the potential to inhibit the activity of HMG Co-A reductase.

Neem (Azadirachta indica A. Juss) is one of the tropical plants found in Indonesia that has been used to prevent and treat various diseases. This study aimed to investigate the effect of the ethanol extract of neem leaves on the concentration of aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, and creatinine in male rats. Twenty-four male Wistar rats were randomly divided into four groups (T0, T1, T2, and T3) with 6 rats in each group. T0 is the control group, and T1, T2, and T3 are the treatment groups that were administered 100, 200, and 300 mg/kg body weight of neem leaf ethanolic extracts for 48 days, respectively. On day 49, blood samples were collected to measure the concentration of AST, ALT, creatinine, and urea followed by an evaluation of liver and kidney histology. The results showed that the ethanolic extract of neem leaves did not affect the concentration of AST, ALT, and creatinine, The ethanol leaves reduced extract on the urea concentration, no abnormal changes were observed in the liver and kidney organs. In the future, it is required to carry out a comprehensive safety evaluation of the neem leaf ethanol extract for herbal medicines.

Investigations of antibacterial, antioxidant, and anti-type 2 diabetes mellitus activities have been carried out on Garcinia macrophylla Mart. plant extract fractions. An isolate from a fraction of ethyl acetate extract was characterized with spectroscopic data. A new biflavonoid compound was found to have a skeleton of 5,7,4',5″,7″,3‴,4‴-heptahydroxyflavanone[3-6″] flavones which was named macrophylloflavone (1). The compound was evaluated for its antibacterial activity against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 25923 with cephazolin as a positive control, antioxidant assay against 2,2 diphenyl-1-picrylhydrazyl (DPPH) with ascorbic acid as the positive control, and anti-type 2 diabetes mellitus treatment with metformin as a positive control. The biflavonoid compound exhibited a good inhibition for bacteria and free radical DPPH. Furthermore, biflavonoid compound treatment on the diabetic rats suggested its ability to decrease the blood glucose level. This study provided evidence that the plant has antibacterial, antioxidant, and antidiabetic properties.
