RESUMEN

Elaeagnus angustifolia (EA) mediated green chemistry route was used for the biofabrication of NiONPs without the provision of additional surfactants and capping agents. The formation of NiONPs was confirmed using advanced different characterization techniques such as Scanning electron microscopy, UV, Fourier transmission-infrared, RAMAN, and energy dispersal spectroscopic and dynamic light scattering techniques. Further, different biological activities of EA-NiONPs were studied. Antibacterial activities were performed using five different bacterial strains using disc-diffusion assays and have shown significant results as compared to standard Oxytetracycline discs. Further, NiONPs exhibited excellent antifungal performance against different pathogenic fungal strains. The biocompatibility test was performed using human RBCs, which further confirmed that NiONPs are more biocompatible at the concentration of 7.51-31.25 µg/mL. The antioxidant activities of NiONPs were investigated using DPPH free radical scavenging assay. The NiONPs were demonstrated to have much better antioxidant potentials in terms of % DPPH scavenging (93.5%) and total antioxidant capacity (81%). Anticancer activity was also performed using HUH7 and HEP-G2 cancer cell lines and has shown significant potential with IC50 values of 18.45 µg/mL and 14.84 µg/mL, respectively. Further, the NiONPs were evaluated against Lesihmania tropica parasites and have shown strong antileishmanial potentials. The EA-NiONPs also showed excellent enzyme inhibition activities; protein kinase (19.4 mm) and alpha-amylase (51%). In conclusion, NiONPs have shown significant results against different biological assays. In the future, we suggest various in vivo activities for EA-NiONPs using different animal models to further unveil the biological and biomedical potentials.

The remediation in plant enzymatic activities in Cd-contaminated soil was monitored through tea waste. Tea is an extensively used beverage worldwide with the release of a high quantity of tea waste utilized in the growing condition of Helianthus annuus on Cd metal contaminated soil. The study was a plan for the natural environmental condition in the greenhouse. For this purpose, four sets of plants were cultivated in triplicate and marked as (i) control, (ii) Cd stress plants, (iii) dry tea waste and Cd stress, and (iv) fresh tea waste and Cd stress. The improved efficiency of biochemical reactions in plants under Cd stress with tea waste treatment was the consequence of blocking Cd movement in the soil through adsorption on tea waste, showing that the tea waste effectively controls the mobility of Cd from the soil to the roots of the plants. Scan electron microscopy (SEM) validates the recovery of the leaves of the plants. The remediation of plant growth and enzyme activities such as amylase, peroxidase, nitrate reductase (NR), and nitrite reductase (NiR) under Cd metal-contaminated soil through tea waste was investigated. The source of tea waste in contaminated soil resulted in the recovery of the photosynthetic process and an improvement in amylase, NR, NiR, and peroxidase activities, thereby resulting in the recovery of pigments coupled with an increase in the biomass of the plants. It was suggested that tea waste acts as a good biosorbent of Cd and energy provider to the plants for normal enzyme activity under Cd stress and may be used by farmers in the future for safe and healthy crops as a cost-effective technology.

Information on reproductive biology and pollination ecology studies of threatened plants are essential to develop strategies for their sustainable utilization and effective conservation. As such, these studies were conducted on Rheum webbianum, a high-value "vulnerable" medicinal herb of the north-western Himalaya. This species presents a unique mode of reproductive behavior through the involvement of different floral events, including the movement of reproductive organs. The plants survive extremely cold conditions through underground perennating rhizomes that sprout into juvenile shoots with the onset of the favorable climatic conditions. The peduncle arises from the axils of the radical leaves, bearing a globular collection of densely arranged hermaphrodite flowers with temporally separated male and female phases; the male phase precedes the female phase (protandry). Anther dehiscence and stigma receptivity is post-anthesis. Anthers dehisce longitudinally along margins, liberating a large mass of spherical and tricolpate pollen with spinulose exine. Pollen viability decreased to < 10% on day 9. Pistil is tristylous, with each style terminating into a fan-shaped stigma lobe. The pollen receptive surface of each stigmatic lobe remains incurved at an angle of 360° and shows upward movement after anthesis, forming a funnel-like structure at an angle of 180° with respect to the ovary. Pollination syndrome is ambophilous. Spontaneous autogamy or geitonogamy to a certain extent is achieved in this species due to the arrangement of flowers in the inflorescence and overlapping of male and female reproductive phases among them. Incurved stigmatic lobes and outward movement of stamens too facilitate outcrossing. Pollen/ovule ratio estimates, results of pollination experiments on breeding behavior, outcrossing, and self-compatibility indices demonstrated that plants are self-compatible and cross-fertile.

In an era of anthropocene, threatened and endemic species with small population sizes and habitat specialists experience a greater global conservation concern in view of being at higher risk of extinction. Predicting and plotting appropriate potential habitats for such species is a rational method for monitoring and restoring their dwindling populations in expected territories. Ecological niche modelling (ENM) coalesces species existence sites with environmental raster layers to construct models that describe possible distributions of plant species. The present study is aimed to study the potential distribution and cultivation hotspots for reintroducing the high value, vulnerable medicinal herb (Rheum webbianum) in the Union territories of Jammu and Kashmir and Ladakh using population attributes and ecological niche modelling approach. Sixty-three populations inventoried from twenty-eight areas display a significant change in the phytosociological attributes on account of various anthropogenic threats. The current potential habitats coincide with actual distribution records and the mean value of Area Under Curve (AUC) was 0.98 and the line of predicted omission was almost adjacent to omission in training samples, thus validating a robustness of the model. The potential habitat suitability map based on the current climatic conditions predicted a total of 103760 km2 as suitable area for the growth of Rheum webbianum. Under the future climatic conditions, there is a significant reduction in the habitat suitability ranging from -78531.34 Km2 (RCP 4.5 for 2050) to -77325.81 (RCP 8.5 for 2070). Furthermore, there is a slight increase in the suitable habitats under future climatic conditions, ranging from +21.99 Km2 under RCP 8.5 (2050) to +3.14 Km2 under RCP 4.5 (2070). The Jackknife tests indicated Precipitation of Driest Month (BIO14) as the most contributing climatic variable in governing the distribution of R. webbianum. Therefore, scientifically sound management strategies are urgently needed to save whatever populations are left in-situ to protect this species from getting extinct. Present results can be used by conservationists for mitigating the biodiversity decline and exploring undocumented populations of R. webbianum on one hand and by policymakers in implementing the policy of conservation of species with specific habitat requirements by launching species recovery programmes in future on the other.

Due to their versatile applications, ZnONPs have been formulated by several approaches, including green chemistry methods. In the current study, convenient and economically viable ZnONPs were produced using Elaeagnus angustifolia (EA) leaf extracts. The phytochemicals from E. angustifolia L. are believed to serve as a non-toxic source of reducing and stabilizing agents. The physical and chemical properties of ZnONPs were investigated employing varying analytical techniques (UV, XRD, FT-IR, EDX, SEM, TEM, DLS and Raman). Strong UV-Vis absorption at 399 nm was observed for green ZnONPs. TEM, SEM and XRD analyses determined the nanoscale size, morphology and crystalline structure of ZnONPs, respectively. The ZnONPs were substantiated by evaluation using HepG2 (IC50: 21.7 µg mL-1) and HUH7 (IC50: 29.8 µg mL-1) cancer cell lines and displayed potential anticancer activities. The MTT cytotoxicity assay was conducted using Leishmania tropica "KWH23" (promastigotes: IC50, 24.9 µg mL-1; and amastigotes: IC50, 32.83 µg mL-1). ZnONPs exhibited excellent antimicrobial potencies against five different bacterial and fungal species via the disc-diffusion method, and their MIC values were calculated. ZnONPs were found to be biocompatible using human erythrocytes and macrophages. Free radical scavenging tests revealed excellent antioxidant activities. Enzyme inhibition assays were performed and revealed excellent potential. These findings suggested that EA@ZnONPs have potential applications and could be used as a promising candidate for clinical development.

The purpose of this study was to evaluate etiology and pregnancy outcome of recurrent miscarriage women. The enrolled patients (280) were evaluated for Triiodothyronine, Thyroxine, Thyroid stimulating hormone, prolactin, chromosomal analysis, Haemoglobin A1C, blood sugar, Magnetic resonance imaging, 3D-ultrasound, auto-antibodies profile (antiphospholipid antibodies, anticardiolipin antibodies, lupus anticoagulant, antinuclear antibodies, anti-thyroid antibodies and ß2 glycoprotein1), torch profile (Toxoplasmo gondii, rubella, cytomegalo virus and herpes simplex virus), blood vitamin D3 levels, psychological factors, Body mass index and thrombotic factors (protein S and C deficiency, Prothrombin G20210A mutation, anti-thrombin III, Factor V Leiden and Methylenetetrahydrofolate reductase mutation), uterosalpingography (hysteronsalpingography) and hysteroscopy. The therapeutic regimens either singly or combined were employed for the treatment of recurrent miscarriage patients on the basis of etiology (single or multiple) and include intravenous immunoglobulin, low molecular weight heparin, low dose aspirin, levothyroxine, progesterone, folic acid, human chorionic gonadotrophin, vitamin D3, psychotherapy, genetic counselling. However, patients with idiopathic recurrent miscarriage were treated with progesterone supplementation, anticoagulation and/or immune modulatory agents. The incidence of primary recurrent miscarriage was highest and most of the women experienced recurrent miscarriage during first trimester. Endocrinological disorders (39%) were found as the major pathological factor for recurrent miscarriage. Other factors include uterine abnormalities (5.7%), vitamin D3 deficiency (3.5%), psychological factors (3.2%) infection (3.6%), autoimmune abnormalities (1.8%) and protein S deficiency (1.8%). However, 40% cases were idiopathic. The overall live birth rate achieved after the management of recurrent miscarriage patients was 75.7%. Enocrinopathy was the major cause of recurrent miscarriage. The overall live birth rate achieved was 75.7% with highest pregnancy outcome in secondary recurrent miscarriage patients after the management.

The present study was conducted to investigate the effects of Cuscuta reflexa extract (CRE) on the activities of germination enzymes, seed germination vigor, biomass production, physio-biochemical attributes, and seed yield of water-stressed wheat plants. Different levels of CRE (0, 10, 20, 30, 40, and 50%), including water soaking, were used as seed priming. Water stress negatively affected the seed germination, germination enzyme activities, growth, yield, and different physio-biochemical attributes of wheat plants. Low doses of CRE (10, 20, and 30%) ameliorated the adverse effects of water stress on seed germination attributes, and activities of germination enzymes, but negative impacts were recorded at higher doses (40 and 50%) of CRE. Water-stressed wheat plants grown from seeds pre-treated with low doses of CRE also showed better growth and yield as compared with non-treated ones, and that was associated with an improvement in water relations, photosynthetic pigments, nutrient acquisition, reduced lipid peroxidation, and better antioxidative defense mechanisms. The maximum increase in seed yield was 14.77 and 12.32%, found in plants grown from seeds treated with 20% and 10% CRE, respectively. In conclusion, it is suggested that using low doses of CRE as seed priming can contribute to better wheat yield under water stress, especially in semi-arid and arid areas.

This study aimed to describe the protective efficacy of Moringa oleifera ethanolic extract (MOEE) against the impact of cobalt chloride (CoCl2) exposure on the rat's kidney. Fifty male rats were assigned to five equal groups: a control group, a MOEE-administered group (400 mg/kg body weight (bw), daily via gastric tube), a CoCl2-intoxicated group (300 mg/L, daily in drinking water), a protective group, and a therapeutic co-administered group that received MOEE prior to or following and concurrently with CoCl2, respectively. The antioxidant status indices (superoxide dismutase (SOD), catalase (CAT), and reduced glutathione (GSH)), oxidative stress markers (hydrogen peroxide (H2O2), 8-hydroxy-2-deoxyguanosine (8-OHdG), and malondialdehyde (MDA)), and inflammatory response markers (nitric oxide (NO), tumor necrosis factor (TNF-α), myeloperoxidase (MPO), and C-reactive protein (CRP)) were evaluated. The expression profiles of pro-inflammatory cytokines (nuclear factor-kappa B (NF-kB) and interleukin-6 (IL-6)) were also measured by real-time quantitative polymerase chain reaction (qRT-PCR). The results showed that CoCl2 exposure was associated with significant elevations of oxidative stress and inflammatory indices with reductions in the endogenous tissue antioxidants' concentrations. Moreover, CoCl2 enhanced the activity of the NF-κB inflammatory-signaling pathway that plays a role in the associated inflammation of the kidney. MOEE ameliorated CoCl2-induced renal oxidative damage and inflammatory injury with the suppression of the mRNA expression pattern of pro-inflammatory cytokine-encoding genes. MOEE is more effective when it is administered with CoCl2 exposure as a prophylactic regimen. In conclusion, MOEE administration exhibited protective effects in counteracting CoCl2-induced renal injury in rats.

Piroxicam (PM) is an oxicam-NSAID commonly recommended for various pain and associated inflammatory disorders. However, it is reported to have a gastric and hepato-renal toxic effect. Therefore, the current research was planned to investigate the possible mechanisms behind the mitigating action of the coenzyme (CoQ10), a natural, free radical scavenger, against PM tissue injury. Rats were assigned to five equal groups; Control, CoQ10 (10 mg/kg, orally), PM (7 mg/kg, i.p.), CoQ + PM L, and CoQ + PM H group. After 28 days, PM provoked severe gastric ulceration and marked liver and kidney damage indicated by an elevated gastric ulcer index and considerable alteration in liver and kidney biochemical tests. The toxic effects might be attributed to mitochondrial dysfunction and excess generation of reactive oxygen species (ROS), as indicated by enhanced malondialdehyde (MDA) levels along with decreased reduced-glutathione (GSH) levels and catalase (CAT) activity. Apoptotic cell death also was demonstrated by increased regulation of activated caspase-3 in the stomach, liver, and kidney tissues. Interestingly, external supplementation of CoQ10 attenuated the PM-inflicted deleterious oxidative harm and apoptosis. This ameliorative action was ascribed to the free radical scavenging activity of CoQ10.
