This study was conducted to investigate the impact of supplementing blue and red light on the biomass yield, metal uptake, contaminant purification, and the alleviation of leaching risks by Noccaea caerulescens, a well-known hyperaccumulator of Cd and Zn. As previously reported for the closely related Thlaspi arvense, N. caerulescens retarded the leaching of Cd and Zn but aggravated the leaching of Pb and Cu, because the species mobilized all metals in soil but only extracted Cd and Zn. Monochromic red light reduced the leaching of Pb and Cu by 13.8% and 1.3%, respectively, but simultaneously weakened Cd phytoremediation by reducing shoot biomass. Our results demonstrated that a small proportion of blue light (10%) could eliminate the negative effect of monochromatic red light on plant shoot growth. However, root biomass decreased by 14.3%, 26.2%, 21.4%, and 61.9% as the percentage of blue light increased from 10 to 100%. Noccaea caerulescens generated the most biomass and accumulated the highest metal concentrations, except for Pb, when the ratio of red to blue light was 1:1. In addition, leachate volume was significantly reduced under the 10% and 50% blue light treatments compared to other light treatments. Therefore, light supplementation with a suitable proportion of blue light can enhance metal purification by N. caerulescens and alleviate potential leaching risk during phytoremediation.

OBJECTIVE: To investigate the biological mechanisms underlying the effect of the Chinese herbal medicine Oxalis corniculata on human prostate cancer PC-3 cells. METHODS: Through in vitro experiment, we treated human prostate cancer PC-3 cells with different concentrations of Oxalis corniculata, assessed the viability of the cells by MTT assay, examined their apoptosis by flow cytometry, evaluated their migration and invasiveness by Transwell assay, and determined the expressions of the proteins p65, p-p65, IκBα and p-IκBα in the NF-κB pathway using protein imprinting technology. RESULTS: Compared with the blank control, Oxalis corniculata significantly inhibited the proliferation and induced the apoptosis of the PC-3 cells (P< 0.05), suppressed their migration and invasiveness in a dose-dependent manner (P< 0.05), and upregulated the expression of IκBα and downregulated those of p-p65 and p-IκBα in the NF-κB pathway (P< 0.05). CONCLUSION: Oxalis corniculata can inhibit the proliferation, migration and invasiveness and induce the apoptosis of human prostate cancer PC cells, which may be attributed to its abilities of inhibiting the expressions of p-p65 and p-IκBα and regulating the activity of the NF-κB pathway.

Electrocoagulation (EC) using iron (Fe), zinc (Zn) and aluminum (Al) electrodes was comparatively applied in the treatment of selenium (Se) in flour production (FP) wastewater. It was indicated that EC treatment with Fe anode obtained highest removal efficiency (79.1%) for Se in the 90 min treatment in the comparative study, which could be attributed to the superior adsorption capacity of in-situ generated iron flocs. Removal of Se resulted from electrodeposition and adsorption to in-situ generated flocs in EC treatment, and the operational conditions significantly influenced the Se removal performance in this work. The results showed the acidic condition and higher current density favored EC treatment on Se removal, EC removed up to 97.8% of Se at pH 4 under 15 mA cm-2, whereas it obtained 83.5% and 50.4% of removal efficiency at pH 7 and 10, respectively. There was competitive adsorption in the process of selenium removal, as the in-situ generated flocs effectively removed 35.6% of humic acid-like (HA-like) substance in FP wastewater after 90 min treatment. The FTIR results showed that HA-like substance mainly contained the protein water hydrogen bond, carboxylate COO antisymmetric stretching and other functional groups. Through the analysis of existence of Se in flocs and wastewater, it can be found that approximately 2.8%-3.92% of Se was removed by electrodeposition process. This study illustrated the Se removal mechanism and provided constructive suggestion for food manufacturing to the metal removal and utilization of advanced treatment.

ETHNOPHARMACOLOGICAL RELEVANCE: Gnaphalium affine D. Don is an important Traditional Chinese herbal Medicine (TCM) used to treat hyperuricemia, asthma, rheumatic arthritis, antitussive, expectorant and cardiovascular in folk medicine because of anti-inflammatory and anti-oxidant activity. The aim of this study was to investigate the potential beneficial effect of G. affine extract (GAE) on hydrogen peroxide (H2O2)-induced apoptosis and explore the possible underlying mechanism in cardiomyocyte. MATERIALS AND METHODS: The ingredients of GAE were isolated and tentatively identified by HPLC-ESI-Q-Qribatrip-MS/MS. The cardioprotective and anti-oxidant effects of GAE were evaluated in the experimental model with H2O2 induced apoptosis in H9c2 cells. H9c2 cells were pretreated for 3 h with or without GAE or with GAE plus PX866 (PI3K inhibitor), then exposed to H2O2 for 6 h, H9c2 cells viability were detected by CCK8 kit, the content of intracellular reactive oxygen species (ROS) and malondialdehyde (MDA) and intracellular superoxide dismutase (SOD) activity were measured by the commercial biochemical kits, western blotting, immunohistochemical (IHC), immunofluorescence (IF) and reverse transcription-polymerase chain reaction (RT-PCR) assays were performed to evaluate the proteins and mRNA expression, propidium iodide (PI) staining was adopted to indicate H9c2 cells apoptosis. RESULTS: Firstly, seventeen polyphenols and flavonoids compounds with the characteristics of anti-inflammatory and anti-oxidant in GAE were tentatively identified by HPLC-ESI-Q-Qribatrip-MS/MS. In the experimental model, GAE not only significantly improved cells viability, but also showed anti-oxidant effects through improving SOD activity, up-regulating nuclear factor E2-related factor 2 (Nrf2), and decreasing intracellular concentration of ROS and MDA and the proteins expression of p47phox, p67phox and gp91phox. On the other hand, GAE revealed anti-apoptotic effect through up-regulating the expression of B-cell lymphoma-2 (Bcl-2), down-regulating Bcl2-associated X (BAX) and cleaved-caspase 3. Furthermore, GAE significantly facilitated phosphorylation of AKT and glycogen synthase kinase-3 beta (GSK-3ß) but not AMPK, while the effects were blocked by PX866 (PI3K inhibitor). CONCLUSIONS: Our data suggested that GAE showed strong anti-oxidant effect to ameliorate oxidative stress and attenuate apoptosis induced by H2O2 in H9c2 cells by targeting PI3K/AKT/GSK-3ß signaling pathway.

Flowback water generated during shale gas extraction in Pennsylvania is mostly reused for hydraulic fracturing operation. Abandoned mine drainage (AMD), one of the most widespread threats to water quality in Pennsylvania, can potentially serve as a make-up water source to enable flowback water reuse. This study demonstrated co-treatment of flowback water and AMD produced in northeastern Pennsylvania in a pilot-scale system consisting of rapid mix reactor, flocculation tank and sedimentation tank. Sulfate concentration in the finished water can be controlled at a desired level (i.e., below 100 mg/L) by adjusting the ratio of flowback water and AMD in the influent. Fe3+ contained in the AMD can serve as a coagulant to enhance the removal of suspended solids, during which Fe2+ is co-precipitated and the total iron is reduced to a desirable level. Solid waste generated in this process (i.e., barite) will incorporate over 99% of radium present in the flowback water, which offers the possibility to control the fate of naturally occurring radioactive materials (NORM) brought to the surface by unconventional gas extraction. Sludge recirculation in the treatment process can be used to increase the size of barite particles formed by mixing flowback water and AMD to meet specifications for use as a weighting agent in drilling fluid. This alternative management approach for NORM can be used to offset the treatment cost and promote flowback water reuse, reduce environmental impacts of AMD and reduce pressure on fresh water sources.

Acremoniumterricola milleretal mycelium (AMM) exerts numerous protective effects on organs, and has been used in Chinese herb prescriptions to treat refractory diseases. The aim of this study was to investigate the effects of AMM on immunological hepatic fibrosis induced by porcine serum (PS) in rats. Male Sprague Dawley rats were administered 0.5 ml sterile PS by intraperitoneal injections twice a week for 18 weeks. AMM (175, 350 or 700 mg/kg) and colchicine (0.1 mg/kg) were administered intragastrically each day until the rats were sacrificed. PS administration resulted in marked hepatic fibrosis, as assessed by increased oxidative stress and hepatic collagen content, as well as α­smooth muscle actin (α­SMA) expression. AMM significantly reduced liver damage and fibrosis. In addition, AMM decreased the elevation in hydroxyproline, hyaluronic acid, laminin and procollagen type III; increased the activity of superoxide dismutase and glutathione peroxidase; decreased α­SMA expression; and eliminated hepatic collagen deposits. Furthermore, AMM inhibited Smad2/3 phosphorylation and Smad7 expression. These results indicate that AMM is able to reduce oxidative stress, inhibit collagen synthesis and block the transforming growth factor­ß/Smad signaling pathway in a dose­dependent manner.
