RESUMEN

Exposure to high doses of acetaminophen is the most common cause of drug induced liver injury. We investigated the protective effects of Hedera helix extract against acetaminophen induced oxidative stress and hepatotoxicity using a mouse model. We used two control groups: group A was given 0.9% NaCl, group B was an acetaminophen control that was given a single injection of 600 mg/kg acetaminophen. T1-T4 groups were pretreated orally with different doses of H. helix extract. The mice were sacrificed and blood samples were collected to estimate the levels of glutathione peroxidase (GPx), malondialdehyde (MDA), superoxide dismutase (SOD) and total bilirubin. Liver samples also were used for histopathological studies. We found that acetaminophen significantly increased the levels of serum ALP, ALT, AST and MDA, and also significantly reduced the antioxidant factors, CAT, GPX and SOD. H. helix extract treatment produced a significant reduction in the levels of ALP, ALT, AST and MDA in serum and restored the levels of CAT, GPX and SOD to control levels. The histopathological findings were consistent with the biochemical findings. H. helix extract exhibited antioxidant and hepatoprotective effects against acetaminophen induced liver damage.

Overcrowding stress is common in the poultry industry. Chickens exposed to long-term stressful situations are characterised by welfare impairment and immunosuppression. 1. The present study evaluated the effects of a blend of essential oils (EOB; cinnamaldehyde and thymol) and stocking density on the performance, gut microflora, meat quality and physiological stress markers of broilers. 2. One-day-old Ross 308 male broiler chickens (n = 360) were allocated to 4 experimental groups from d 22 to 42. Each treatment had 6 replicates of 15 chicks. Two groups were subjected to a high stocking density (HSD) of 20 birds/m2 and the other two groups were kept at a low stocking density (LSD) of 10 birds/m2. 3. The results of this study indicate that overcrowding stress decreased growth performance parameters, blood immunoglobulin (Ig)G and heterophil:lymphocyte (H:L) ratio but increased IgA and IgM levels. HSD reduced water-loss rate and pH decline at 45 min post mortem in the breast muscle. 4. Essential oils supplementation elevated H:L ratio but decreased breast meat redness and pH24. 5. Significant interactions between EOB and stocking density were observed for corticosterone (CS) level and mRNA levels of heat shock protein 70 (HSP70) in brain and heart. Although HSD increased CS and HSP70 when compared to LSD, the effects of the former were inconsistent with EOB supplemented diets. 6. In conclusion, dietary EOB supplementation could improve some of the biomarkers associated with overcrowding stress in broiler chickens.

Vitamin K2 (VK2), acts as an electron carrier in mitochondria and thereby effects reactive oxygen species (ROS) and ATP production. This study evaluates role of VK2 on in vitro developmental competency and cryo-survival of pre-implantation ovine embryos. Initially the optimal and beneficial concentration of VK2 on compaction and blastocyst formation rates was defined (0.1 µM). Subsequently, it was shown that 0.1 µM VK2, at blastocyst stage, reduces H2O2 production, increase the expression of mitochondrial related gene and improved embryos quality. We further assessed presence VK2 supplementation before and/or after vitrification of in vitro derived blastocysts. Our results reveal that presence of VK2 before and after vitrification improves rates of blastocysts re-expansion (88.19± 3.37% vs 73.68± 1.86%, P < 0.05) and hatching (49.55± 4.37% vs 32.7± 3.32%) compared to control group. These observation were consistent with reduction in H2O2 production and improved in expression of mitochondrial related genes. However, VK2 before or after vitrification, not only had no positive effect on these two parameters, but also significantly reduced these parameters. Therefore, in concordance with pervious report in bovine, we show that VK2 supplementation post genomic activation (Day 3-7) improved developmental competency of ovine in vitro derived embryos. We also showed that presence of VK2 after vitrification improves the cryo-survival of ovine embryos.

Cognitive training is an emergent approach to improve cognitive functions in various neurodevelopmental and neurodegenerative diseases. However, current training programs can be relatively lengthy, making adherence potentially difficult for patients with cognitive difficulties. Previous studies suggest that providing individuals with real-time feedback about the level of brain activity (neurofeedback) can potentially help them learn to control the activation of specific brain regions. In the present study, we developed a novel task-based neurofeedback training paradigm that benefits from the effects of neurofeedback in parallel with computerized training. We focused on executive function training given its core involvement in various developmental and neurodegenerative diseases. Near-infrared spectroscopy (NIRS) was employed for providing neurofeedback by measuring changes in oxygenated hemoglobin in the prefrontal cortex. Of the twenty healthy adult participants, ten received real neurofeedback (NFB) on prefrontal activity during cognitive training, and ten were presented with sham feedback (SHAM). Compared with SHAM, the NFB group showed significantly improved executive function performance including measures of working memory after four sessions of training (100min total). The NFB group also showed significantly reduced training-related brain activity in the executive function network including right middle frontal and inferior frontal regions compared with SHAM. Our data suggest that providing neurofeedback along with cognitive training can enhance executive function after a relatively short period of training. Similar designs could potentially be used for patient populations with known neuropathology, potentially helping them to boost/recover the activity in the affected brain regions.

Decreasing the side effects of chemotherapy in testis has been the subjects of many studies. In this study, the protective effects of Zingiber officinale extract on rat testis were investigated after chemotherapy with cyclophosphamide. Histological and biochemical parameters were compared in cyclophosphamide-treated rats with or without ginger extract intake. Wistar male rats were randomly divided into four groups each 10. The control group received a single injection of 1 ml isotonic saline intraperitoneally. The Cyclophosphamide (CP) group received a single dose of cyclophosphamide (100 mg kg(-1) BW) intraperitoneally. CP + 300 and CP + 600 groups received orally 300 or 600 mg of ginger extract, respectively, for a period of 6 weeks after cyclophosphamide injection. The morphologic and histological structure of the testis was compared in different groups of the rats. Also, factors like malondialdehyde, reactive oxygen species, total antioxidant capacity and testosterone level were assessed in blood serum as well. Our results showed that although ginger extract could not change testis weight, malondialdehyde (MDA) and ROS, but antioxidant and testosterone levels in serum were increased significantly. Also, an obvious improved histological change was seen in CP + 300 and CP + 600 groups in comparison with CP group. These protective effects of ginger on rat testis after cyclophosphamide treatment could be attributed to the higher serum level of antioxidants.

The present study aims to investigate if prematuration culture (PMC) of ovine oocytes in the presence of a phosphodiesterase type 3 (PDE3) inhibitor cilostamide can improve the shortcomings of conventional in vitro maturation (IVM) system. Therefore, a two-step culture system consisting of 22 hours culture in the presence of 1, 10, and 20 µM cilostamide (PMC medium), followed by 22 hours culture in maturation medium, was designed. The effect of cilostamide on gap junction communications and nuclear status was studied. The variables assessed were chromosome organization, spindle pattern, polar body extrusion, and embryonic development. According to the results, inhibition of PDE3 could not permanently block nuclear maturation in ovine oocytes but it delayed the process of nuclear maturation. Elevation of intra-oocyte cAMP concentration could inhibit cumulus cells expansion and maintain gap junction communications between oocyte and cumulus cells. Deletion of cilostamide and refreshing maturation medium after 22 hours culture revealed that cumulus cells were completely expanded. The inhibitory effect induced by 1 µM cilostamide was reversible, and it increased the number of mature oocytes with aligned chromosomes and normal spindle. However, the inhibitory effects of 10 and 20 µM cilostamide was not fully reversible and was associated with deleterious effects on chromosome organization and spindle pattern. Investigation of embryonic development via parthenogenetic activation and in vitro fertilization revealed that the blastocyst rate of oocytes that were prematured with 1 µM cilostamide was not significantly different from oocytes that underwent conventional IVM but it was significantly reduced in oocytes that were prematured with 10 and 20 µM cilostamide. Our results provide the evidence that reduced cAMP via PDE3 is not the only mechanism that controls the progress of nuclear maturation in sheep oocytes, and that alternative or additional mechanisms may also exist.

Spirulina spp. and its processing products are employed in agriculture, food industry, pharmaceutics, perfumery and medicine. Spirulina has several pharmacological activities such as antimicrobial (including antiviral and antibacterial), anticancer, metalloprotective (prevention of heavy-metal poisoning against Cd, Pb, Fe, Hg), as well as immunostimulant and antioxidant effects due to its rich content of protein, polysaccharide, lipid, essential amino and fatty acids, dietary minerals and vitamins. This article serves as an overview, introducing the basic biochemical composition of this algae and moves to its medical applications. For each application the basic description of disease, mechanism of damage, particular content of Spirulina spp. for treatment, in vivo and/or in vitro usage, factors associated with therapeutic role, problems encountered and advantages are given.

This study was conducted to examine the effects of saffron extract on preventing D-galactose and NaNO2 induced memory impairment and improving learning and memory deficits in amnestic mice. In this study, the learning and memory functions in ovariectomized mice were examined by the one way passive and active avoidance tests. In active avoidance test, training in amnestic treated (AT) and amnestic prophylaxis (AP) groups, was improved so that there was a significant difference between them and the amnestic control (AC) group. In passive avoidance test, animal's step through latency, as an index for learning, in all test groups was significantly greater than control group. Total time spent in dark room (DS), which opposes the memory retention ability, in AC was significantly greater than AT group at 1 and 2 hours after full training, while there was not any significant difference between this index in AP and AT as compared with normal control (NC) group. Our findings indicate that saffron hydro-alcoholic extract prevents and improves amnesia induced by D-galactose and NaNO2 in mice.

Substitution of egg yolk with soybean lecithin may reduce hygienic risks in extenders. Though a few studies have been performed on the effect of soybean lecithin in bull, to date evaluation of ram semen in vitro fertility after cryopreservation with use of soybean lecithin has not been studied. This study assessed the effect of 1% or 2% (wt/vol) soybean lecithin (L1 or L2) or 15% or 20% (vol/vol) egg yolk (E15 or E20) supplemented with 5% or 7% glycerol (G5 or G7) in a Tris-based medium for cryopreservation of ram (Oviss arries) semen. Although no significant difference was observed in pattern of capacitation, the best results in terms of sperm motility, viability postthaw, and cleavage rates were observed with L1G7 (51.9+/-4.8%, 48.1+/-3.5%, and 79.6+/-3.9%, respectively) and E20G7 (51.8+/-2.9%, 46.7+/-4.0%, and 72.9+/-6.4%, respectively). Our results also showed that 1% lecithin and 20% egg yolk was superior to 2% lecithin and 15% egg yolk. In terms of cleavage rate, 7% glycerol was superior to 5% glycerol. No significant difference was obtained between groups in terms of blastocysts rate per cleaved embryo. Therefore, we concluded that the optimal concentration of lecithin and egg yolk is 1% and 20%, respectively, along with 7% glycerol. In addition, our results suggest that lecithin can be used as a substitute for egg yolk.

The aim of this study was to evaluate the feeding value and also the effect of different supplementary energy on increasing in vitro DM digestibility of date-palms leaves obtained in pruning. A randomized complete design with different energy supplementary with 3 replicates was conducted. Harvested date-palm leaves were air-dried, ground through a 5 mm screen, then palm leaves (L) ensiled with different levels of urea (U) and other supplementary energy. Combination, mixture was packed tightly in 21 (10 L) plastic containers. The containers were opened after a fermentation of 60 days. Treatments consisting: L (Palm leaves)+5% U (urea) (T1), L+5% U+5% BP (Beet pulp) (T2), L+5% U+10% BP (T3), L+5% U+5% M (Molasses) (T4), L+5% U+10% M (T5), L+5% U+5% CP (Citrus pulp) (T6) and L+%5 U+10% CP (T7). The results of this experiment showed that supplementation of date-palm leaves with energy supplements significantly (p < 0.05) increased IVDMD and T5 had higher digestibility than those of the other treatments. ADF content of silage was not affected by dietary treatments. However, NDF content was significantly (p < 0.05) affected by treatments and T2 and T6 had higher NDF content compared with the other treatments. Supplementation of palm leaves with energy supplementary had no significant effect (p > 0.05) on Crude Protein (CP) content of silages. However, these supplements had a significant effect (p < 0.05) on calcium and phosphorus levels of silage materials and T7 had higher Ca and P content compared with the other treatments. In conclusion, supplementation of date-palm leaves with energy supplements can improve IVDMD and feeding value of this feed source.

AIMS AND OBJECTIVES: The aim of this study is to find easier way of home care while obviating the colostomy before single stage pull through operation. MATERIALS AND METHODS: From August 2005 to December 2006, eight cases of neonatal Hirschsprung disease were treated. Mean age 4.5 (2-6) day/old with absent anorectal inhibitory reflex, rectosigmiod disease in Barium enema, positive Acetylcholine esterase (Ache) staining, good response to rectal washout. They underwent botulinium toxin injection (5 unit /kg/quadrant) in four quadrant intrasphincteric. They were followed until pull through operation in 8-10 weeks post injection. RESULTS: Four of 8 (50%) cases only needed rectal washout for three to five days post injection until pull through operation, two had decrease in number of rectal washouts /day and the remaining two underwent colostomy five days post injection because of no response. CONCLUSION: Botulinium toxin injection can help in palliative care in patients with Hirschsprung disease who are waiting for colostomy or definitive pullthrough. It gives an option of eaiser home care for these patients.
