RESUMEN

There are 200-500 species of Potentilla(Rosaceae) worldwide, among which 90 species are widely distributed in China and have a long history of ethnic medicinal use. According to our statistics, a total of 367 compounds have been isolated and identified from plants of this genus, including terpenoids, flavonoids, phenolic acids, tannins, and phenylpropanoids. The medicinal materials made from these plants mainly have antioxidative, blood sugar-lowering, anti-inflammatory, anti-tumor, cardiovascular system-protecting, neuroprotective, and hepatoprotective activities. This study systematically reviews the research progress on chemical constituents and pharmacological activities of Potentilla plants to provide a basis for further research and clinical application.

RESUMEN

Objective:To observe the clinical efficacy of Xiao Qinglongtang on chronic heart failure with cold phlegm in lung, and explore its mechanism of action. Method:A total of 87 patients with definite chronic heart failure were divided into observation group and control group by random number table method.The two groups received routine western medicine at the same time. Forty-two cases in observation group were treated with Xiao Qinglongtang based on western medicine, and 45 cases in control group received the same dose of placebo. Both groups were treated for 4 weeks, and then their cardiac function, traditional Chinese medicine (TCM) syndrome score and efficacy were compared before and after treatment. Left ventricular ejection fraction (LVEF) and N-terminal pro-brain natriuretic peptide (NT-proBNP) were measured in both groups before and after treatment. The changes of standard deviation of NN intervals (SDNN), square root of the mean squared differences of successive NN intervals (RMSSD), high frequency (HF) and low frequency (LF) that reflect autonomic nerve function indexes in heart rate variability (HRV) after treatment were compared between two groups. The changes of inflammatory indicators such as interleukin-6 (IL-6) and highly sensitive C reaction protein (hs-CRP) were detected. Result:After treatment, the total effective rate for cardiac function in observation group was higher than that in control group (P<0.05). The TCM symptom scores were improved after treatment in both groups (P<0.05), and the total effective rate in observation group was higher than that in control group (P<0.01). After treatment, LVEF levels significantly increased (P<0.01) and NT-proBNP levels significantly decreased (P<0.01) in both groups, and the effect in observation group was more obvious (P<0.01). After treatment, SDNN, RMSD, HF and LF indicators in HRV were all higher than those before treatment in both groups (P<0.01), and the improvement in observation group was more significant than that in control group (P<0.01). The levels of IL-6 and hs-CRP decreased after treatment in both groups (P<0.01), and the level of observation group was significantly lower than that of control group (P<0.01). Conclusion:Xiao Qinglongtang has certain clinical efficacy in treating chronic heart failure with cold phlegm in lung as it can improve the clinical symptoms of patients, regulate autonomic nervous balance, and inhibit inflammatory factors, providing new clinical ideas to treat chronic heart failure in TCM.

Objective:To investigate the effects of electroacupuncture on blood-brain barrier to promote intake of drugs into brain of SAMP8 mice. Methods:The male SAMP8 mice (30-week-old) were randomly divided into model group (n = 7), drug group (n = 7), acupuncture group (n = 7) and combined group (n = 7). Other SAMR1 mice were as control group (n = 7). The acupuncture group and the combined group accepted electroacupuncture at Baihui (DU20) and Yintang (EX-HN3), the drug group and the combined group accepted Donepezil, for four weeks. They were observed hippocampus tight junction (TJ) under transmission electron microscope. The activities of acetylcholinesterase (AChE) in hippocampus were detected with immunohistochemistry. The mRNA expression of ZO-1, Claudin-5 and Occludin was detected with real-time quantitative polymerase chain reaction. Results:TJ and basement membrane in the control group, the acupuncture group and the combined group were better than those in the model group and the drug group. AChE was the most in the combined group, and then the drug group and the control group, the acupuncture group and the model group (P < 0.001). The expression of mRNA of ZO-1, Claudin-5 and Occludin was more in the acupuncture group and the combined group than in the drug group (P < 0.01). Conclusion:Electroacupuncture could ameliorate blood-brain barrier disruption and promote drug to enter the brain in SAMP8 mice, which may relate to the adjustion of ZO-1, Claudin-5 and Occludin.

Objective:To explore the effect and mechanism of Tuina on denervated skeletal muscle atrophy. Methods:A total of 77 male Sprague-Dawley rats were randomly divided into sham group (n = 7), model group (n = 35) and Tuina group (n = 35). The latter two groups were established skeletal muscle atrophy model by exposing and cutting off the common tibial nerve of rats. One day after modeling, the lower limbs of the surgical side received Tuina in Tuina group. Separately, the surgical side of gastrocnemius muscle were sampled on the 0th, 7th, 14th, 21st and 28thday after modeling, and measured the wet mass ratio. The cross-sectional area and diameter of muscle fiber were measured after HE staining. The mRNA expression of autophagy-realated factor Beclin-1, vacuolar protein sorting (Vps34) and microtubule-associated protein light chain 3 (LC3) were tested with reverse transcription real-time quantitative polymerase chain reaction. Results:There was no statistical difference in the ratio of gastrocnemius wet weight, the cross-sectional area and diameter of muscle fiber, and the mRNA expression of Beclin-1, Vps34 and LC3 among three groups on the 0th day (F < 1.321, P > 0.05). Compared with the sham group, the ratio of gastrocnemius wet weight, the cross-sectional area and diameter of muscle fiber decreased at different time points in the model group and Tuina group (P < 0.05), the ratio of gastrocnemius wet weight was higher, and the cross-sectional area and diameter of muscle fiber were bigger, both except on the 21st day, in Tuina group than in the model group (P < 0.05). Compared with the sham group, the mRNA expression of Beclin-1, Vps34 and LC3 increased at different points in the model group than in Tuina group (P < 0.05), and all the mRNA expression was higher, except on the 14th day, in Tuina group than in the model group (P < 0.05). The ratio of gastrocnemius wet weight, the cross-sectional area and diameter of muscle fiber showed a trend of progressive decrease with time in the model group and Tuina group (P < 0.05). The mRNA expression of Beclin-1 and Vps34 increased (P < 0.05), and the mRNA expression of LC3 increased in the model group 21 days after intervention (P < 0.05). The mRNA expression of Beclin-1, Vps34 and LC3 increased first and then decreased, except the mRNA expression on the 14th day in Tuina group (P < 0.05). Conclusion:Tuina may promote the activation of autophagy by up-regulating the expression of autophagy-realated factor Beclin-1, Vps34 and LC3, remove the damaged organelles and proteins, provide certain synthetic substrate and energy for muscle fiber regeneration, thereby reduce the loss of degree of denervated skeletal muscle atrophy.

@#Objective To investigate the effect of p53 protein on the nerve function of rats with acute cerebral ischemia-reperfusion by electro-acupuncture pretreatment. Methods A total of 72 male Sprague-Dawley rats were randomly divided into sham group(n=24),model group(n=24) and electroacupuncture pretreatment group(n=24).Each group was equally divided into two hours and 72 hours subgroups. The neurological behavior was assessed with modified Neurological Severity Score (mNSS). HE staining and TUNEL assay were used to observe the injury and cell apoptosis in ischemic brain,and Western blot-ting was used to detect the expression of p-p53(ser392),p53 and microtubule associated protein 1 light chain 3 (LC3)II in ischemic area. Results Compared with the model group,the mNSS scores,the injury of brain tissue,the number of TUNEL-positive cells, and the level of p-p53 and LC3II decreased (P<0.05) in the electroacupuncture pretreatment group.Two hours after reperfusion,there was no significant difference in the level of p53 between the model group and the electroacupuncture pretreatment group(P>0.05),however,72 hours after reperfusion,the level of p53 decreased in the electroacupuncture pretreatment group(P<0.05). Conclusion Electroacupuncture pretreatment can improve neurological deficit in cerebral ischemia-reperfusion injury rats,which may be related to the involvement of p53 protein in autophagy and apoptosis.

@#Objective To explore the effects and mechanism of electroacupuncture(EA)on denervation-induced atrophy. Methods A total of 21 male Sprague-Dawley rats were divided into sham group(n=7),model group(n=7)and EA group (n=7).The latter two groups were cut off their right sciatic nerve.Since one day after modeling,EA group accept-ed electroacupuncture at right Zusanli(ST36)and Huantiao(GB30)for eight weeks.Then,the gastrocnemius of all the rats were obtained,and measured the wet mass ratio.Cross-sectional area(CSA)and fiber diameter were measured after HE staining. The expression of autophagy-related gene ULK1, Atg13, Beclin1, Atg14, Atg7, Atg12,Atg5 and Atg16L1 were tested with reverse transcription real-time quantitative polymerase chain reaction. Results Compared with the sham group,the wet mass ratio,CSA and fiber diameter of gastrocnemius were lower signifi-cantly in the model group and EA group(P<0.001),and they were more in EA group than in the model group(P<0.05).Compared with the sham group,the mRNA expression of ULK1,Atg13,Beclin1,Atg14,Atg7,Atg12,Atg5 and Atg16L1 was more significantly in the model group (P<0.001), and they decreased in EA group compared with those of the model group(P<0.05). Conclusion Electroacupuncture can inhibit the overexpression of autophagy-related gene in denervated rats,which may steady skeletal muscle cells to delay atrophy.

Objective To learn the adverse drug reaction (ADR) of traditional Chinese medicine injection (TCMI) in Dongfang Hospital of Beijing University of Chinese Medicine (BUCM),to master the characteristics of the occurrences of ADR,and to provide basis for safe and rational clinical drug use.Methods Locating all the reported ADR cases by Dongfang Hospital of BUCM to national ADR monitoring system from June 2011 to May 2016,and make retrospective analysis of them.Results Eighteen kinds of TCMI that caused the total 135 ADR cases,50 out of all the 135 instances were male patients,whilst the rest 85 instances were female patients;The main kinds of TCMI that caused ADR problems were TCMI for relieving heat and toxic material and TCMI for promoting blood circulation to remove blood stasis;TCMI was used jointly with antibiotics and biological product etc.;And the circumstances of ultra-dose and improper infusion allocation existed.The clinical manifestation included many skin lesion cases.All the patients suffered from ADR were cure.Conclusion Prudent and rational use of drugs,standardized clinical use procedure,and clinical re-evaluation are key aspects to lower the incidence rate ofADR of TCMI and to safe and rational use of drugs.

<p><b>OBJECTIVE</b>To observe the differences in timely effectiveness of immediate analgesia on acute migraine between electroacupuncture (EA) at shaoyang meridian points and non-meridian points.</p><p><b>METHODS</b>The randomized controlled trial method was adopted. One hundred and ten cases of acute migraine were randomized into two groups. 55 cases in each one. In the observation group, EA was applied to Fengchi (GB 20), Waiguan (TE 5), etc. In the control group. EA was applied to non-meridian points. Only one treatment was given, and the nee dies were retained for 30 min in the two groups. Separately. at 10 time points, named before acupuncture, in 5 min, 10 min. 20 min and 30 min (at the moment of needle removal), 1 h, 2 h, 4 h, 6 h and 8 h after acupuncture, the pain intensity numerical rating scale (PI-NRS) was adopted to evaluate the pain severity and the adverse reac tion was recorded.</p><p><b>RESULTS</b>(1) The effective rate of imnmediate analgesia was 87. 3% (48/55) in the observation group. significantly higher than 52.7% (29/55) in the control group (P<0.01). (2) NRS of each group was re duced at each time point, from 5 min to 8 h after acupuncture (all P<0.01). (3) NRS score at 30 min after acu puncture in the control group was obviously lower than that in the observation group (P<0.05), but the scores in 2 h, 4 h, 6 h and 8 h after acupuncture in the observation group were lower apparently than those in the control group (all P<0. 01). The differences in the scores in 5 min, 10 min, 20 min and 1 h after acupuncture were not significant (all P>0. 05). (4) For the reducing amplitude of NRS score at the same time point after acupuncture, the results in 20 min, 30 min and 1 h in the control group were higher apparently than those in the observation group (all P<0. 05). But, in 2 h, 4 h, 6 h and 8 h, the results in the observation group were higher apparently than those in the control group (all F<0.05). The differences in 5 min and 10 min after acupuncture were not significant (both P>0. 05). (5) In 24 h after acupuncture, the recurrence rate of migraine was 12. 7% (7/55) in the observation group, which was lower obviously than 34. 5% (19/55) in the control group (P<0. 05). The adverse reaction was not found in the two groups.</p><p><b>CONCLUSION</b>EA at the shaoyang meridian points achieves the definite immediate analgesia effect on acute migraine and presents the time effectiveness of analgesia.</p>

<p><b>OBJECTIVE</b>To observe the clinical analgesic efficacy and the relation between clinical analgesic effect and duration of effect of two acupuncture methods of Longhu Jiaozhan (fight of dragon and tiger, an acupuncture reinforcing and reducing manipulation characterized with nine counterclockwise and six clockwise rotations) and even manipulations.</p><p><b>METHODS</b>Sixty-two patients with primary dysmenorrhea were randomly divided into an observation group (32 cases) and a control group (30 cases). The observation group was applied with Longhu Jiaozhan manipulation, while the control group was applied with even manipulation. Acupuncture treatment was given on both of the groups since onset of the pain. The score of the visual analogue scale (VAS) of the 2 groups was observed at different times. And 8 VAS values were recorded at the point right before acupuncture, needle remaining of 5 min, 10 min, 20 min and 30 min as well as and 30 min, 60 min, 120 min after needle withdrawal. Comparison was made on differences between the 2 groups.</p><p><b>RESULTS</b>Comparison with the same group before acupuncture showed that the VAS difference of the time from needling remaining of 5 min to 120 min after acupuncture were all with statistic significance (all P<0. 01). The score of VAS of needle remaining for 20 min and 30 min of the observation group was without significant difference (P>0. 05). The score of VAS value of needle remaining for 20 min and 30 min of the control group was with significant difference (P<0. 01). Comparison of the VAS scores before the treatment and the scores of the 2nd menstrual cycle were found with significant difference (P<0. 01). The VAS score of 2nd menstrual cycle was (28. 73 +/- 16. 15) in the observation group, which was better than (46. 93+/-12. 18) in the control group (P<0. 001). Comparison of the VAS score of the two groups at 5 min r emaining of the needle was without statistic significance between two groups (P>0. 05). However, the VAS impairment magnitude difference at that moment was with statistic significance (P<0.01). From the time of needle remaining for 10 min, 20 min, 30 min until 120 min after needling, the differences of the VAS scores and impairment range were all with statistic significance (all P<0. 01). The effects of the two acupuncture techniques could both be maintained up to 2 hours after needling.</p><p><b>CONCLUSION</b>Both methods of acupuncture have immediate and long-term analgesic effect in a certain degree on primary dysmenorrhea. However, compared with the control group, the advantage of analgesic effect in the observation group is significantly superior. 20 min needling remaining can reach the best analgesic effect.</p>

ETHNOPHARMACOLOGICAL RELEVANCE: Miaoyao Fanggan Sachets (MFS) has long been used as a folk medicine for the prevention of influenza in Southeast of Guizhou Province, China. However, the precise immunological mechanisms by which MFS confers protection have not been defined. STUDY AIM: To explore the effects of MFS on innate immune system responses using a cold stress-induced immune impairment model as a means of examining MFS-mediated influenza prevention. We investigated the effects of MFS on toll-like receptor 2 and 4 (TLR2/4) gene and protein expression levels and on the percentage of NKp46(+) cells present in serum. No overt toxicity was observed following continuous administration of MFS at high doses. METHODS: Kunming male mice (n=40) were randomly divided into 4 groups consisting of the continuous inhalation Sachet group, Yu-Ping-Feng powder (YPF-P) gavage positive control group, discontinuous inhalation MFS group and untreated controls. After 4 weeks, mice were sacrificed and lungs harvested. The expression of toll-like receptors 2 and 4 (TLR2/4) gene and protein levels was assessed using real-time polymerase chain reaction (RT-PCR) and Western blot analyses, respectively. An additional 60 Kunming mice were randomly divided into 6 groups comprised of a blank control group, continuous MFS inhalation group, an immune-compromised continuous MFS inhalation group, an immuno-compromised group, an immune-compromised MFS discontinuous inhalation group and an immune-compromised positive control group. Immune suppression was induced by cold stress (4 °C/4 h daily for 3 days) and mice were treated with MFS or YPF-P before cold stress treatments. Immuno-compromised mice were treated with MFS continuously or intermittently, or treated with YPF-P. Blood samples were collected and examined for natural killer cells based on positive NKp46 staining. The isorhamnetin associated with MFS-induced immune modulation was obtained from 'wo ga le' which is considered to be a major component of MFS, and analyzed by HPLC. RESULTS: Mice continuously inhaling MFS showed a moderate increase in TLR2/4 mRNA and protein levels compared to mice in the control and discontinuous inhalation groups. MFS significantly increased the TLR2/4 expression in a dose-dependent manner. Furthermore, there was also a slightly significant increase in the number of NKp46(+) cells in the continuous inhalation group compared to controls and discontinuous inhalation group. Pretreatment with MFS partially prevented cold stress-induced inhibition of NKp46(+) cells. HPLC analysis of the 'wo ga le' associated with immune function identified the major component to be isorhamnetin. CONCLUSIONS: Taken together, these data suggested that MFS significantly enhanced TLR2/4 expression levels and the number of NKp46(+) cells in mice and moderately affected innate immune responses associated with protection against influenza, suggesting that isorhamnetin in the MFS enhanced innate immune potency. The use of MFS for the prevention of various respiratory tract infections can be attributed to its antimicrobial properties. In a pilot study, a large quantity (40 g) was administered over a prolonged period did not produce apparent toxicity.

<p><b>OBJECTIVE</b>To study the clinical efficacy and mechanisms of Youdujing (YDJ) preparation in treating the cervical high-risk human papilloma virus (HR-HPV) infection.</p><p><b>METHODS</b>Totally HR-HPV infection 70 patients were assigned to the treatment group and the control group using random single blind method, 35 cases in each group. YDJ external lotion and YDJ cream were applied to patients in the treatment group, while normal saline was applied for those in the control group. HR-HPV DNA detection was performed by the end of the 1st menstruation after 3 menstrual cycles. The cervical biopsy and cervical smear were performed using vaginoscope before and after treatment. The mRNA expression of human telomerase reverse transcriptase (hTERT) was detected from fresh tissues of the cervical lesion.</p><p><b>RESULTS</b>The total effective rate was 96.6% in the treatment group, higher than that of the control group (70.00%, P<0.01). Before treatment there was no statistical difference in the pathological results of the cervix and the mRNA expression of hTERT between the two groups (P>0.05). The mRNA expression of hTERT decreased in the two groups after treatment, showing statistical difference (P<0.01). Better effects on the pathological results of the cervix and the mRNA expression of hTERT were obtained in the treatment group after treatment (P<0.05).</p><p><b>CONCLUSION</b>YDJ played a role in clearing HR-HPV infection and reversing the cervical precancerous changes possibly through down-regulating the mRNA expression of hTERT and lowering the telomerase activation.</p>

<p><b>OBJECTIVE</b>To observe the effect on intervention of sub-health with pestle needle (Chuzhen).</p><p><b>METHOD</b>Randomized controlled trail was adopted for this research. One hundred and fifty-three cases were randomly divided into two groups of a Chuzhen group (79 cases) and a massage group (74 cases). Acupoint of Bazhen (Baihui Bazhen, Shendao Bazhen, Zhiyang Bazhen, Mingmen Bazhen, Yaoyangguan Bazhen), Hechelu on the head, the neck and the lumbar area were adopted in Chuzhen group. While regular whole-body massage was applied in the massage group. The human sub-health score, the cornell medical index (CMI) and thermal texture maps system (TTM) technology of the two groups before and after the intervention were observed.</p><p><b>RESULTS</b>1) After treatment, sub-health condition score, the CMI score, the M-R score and the TTM index were all increased in both groups (all P<0.01) 2) Comparison of D-value of the two groups before and after the intervention: the level of the sub-health score, the total score of CMI, and the index of sleep, pressure, Governor Vessel, Hukou (first web), blood lipid, viscosity of blood, microcirculation of TTM index of the Chuzhen group changed more obvious (all P<0.01), but there was no statistic significances in the M-R score and blood sugar of the TTM (both P>0.05). 3) The sub-health condition score in Chuzhen group was higher than that in the massage group (P<0.01).</p><p><b>CONCLUSION</b>Chuzhen therapy has definite effect on intervention of sub-health, which is better than regular general massage.</p>

<p><b>OBJECTIVE</b>To compare the curative effects of simple obesity of spleen deficiency and dampness excess treated by hour-prescription of points and routine acupuncture therapy.</p><p><b>METHODS</b>Sixty cases were randomly divided into an observation group and a control group; Taibai (SP 3), Gongsun (SP 4), Sanyinjiao (SP 6), Yinlingquan (SP 9), Fujie (SP 14) and Daheng (SP 15) were applied in both groups. Hour-prescription of points (the acupoints of Spleen Meridian were selected at the period of the day from 10:04 a.m. to 12:04 a.m. when the Spleen Meridian is most energetic at Chengdu) was applied in observation group, and routine acupuncture was applied in control group (acupoints weren't selected at 10:04 a.m. to 12:04 a.m.). Once a day, and 10 days of treatment made one session, totally 3 sessions were required.</p><p><b>RESULTS</b>The total effective rate was 86.2% (25/29) in observation group, and 75.9% (22/29) in control group. The total effect in observation group was superior to that in control group (P < 0.05). The observation group was better than the control group in reduction of weight, body mass index, obesity degree, awaist circumference and the ratio between waist and hip with significant differences between two groups (P < 0.01, P < 0.05).</p><p><b>CONCLUSION</b>The therapeutic effect of obesity of spleen deficiency and dampness excess treated by hour-prescription of points therapy is superior to that by routine acupuncture therapy.</p>
