RESUMEN

The increase of urbanization level has led to the rapid increase of impervious surface area (ISA). The aim of this work is to clarify the relationship between the ISA and water quality and lay a foundation for the improvement and protection of the water quality in the basin. Taking the Fuxian Lake Basin in Yunnan Province as an example, based on the Landsat ETM+ remote sensing image and the Gram-Schmidt (GS) image fusion algorithm, the four-terminal model and the linear spectral mixture model (LSMM) were used to extract the impervious surface of the watershed from 2006 to 2015. And statistical methods were used to distinguish its relationship with water quality. The results show that the four-terminal model and the linear spectral mixture model can effectively extract the impervious surface information of the Fuxian Lake Basin. The average root mean square error (RMS) of the image decomposition results from 2006 to 2015 was less than 0.02. In the past 10 years, the ISA has changed significantly in the Fuxian Lake Basin. The ISA showed an overall upward trend from 2006 to 2015. It increased from 24.73 km2 in 2006 to 35.14 km2 in 2015, an increase of 10.81 km2. From the value anomaly, the ISA in 2006 and 2009 is lower than the multiyear average, and those in the other years are higher than the multiyear average. The percentage of ISA in the basin was significantly positively correlated with Chemical Oxygen Demand-Mn (CODMn) and total phosphorus (TP) (r is 0.772, 0.763), and the correlation in the flooding season was greater than that in the dry season. The ISA threshold for water quality deterioration is around 10% in the Fuxian Lake Basin. Reducing ISA coverage, controlling ISA to less than 10%, and preventing nonpoint source pollution during flooding season will be the best measures to effectively improve the water quality environment in the basin.

Pregnancy is an oxidative stress and immune challenge for the mother. Fish oil is rich in EPA and DHA, which can partly inhibit aspects of inflammation and restore antioxidant capacity. In the present study, we investigated the effect of maternal diet with fish oil during the late gestation period on oxidative stress and inflammatory response in sows and their progenies. Twelve second-parity sows were allocated equally into two groups. Sows were fed either the soybean oil diet (SD) or soybean oil+fish oil diet (FD) during the gestation period. The plasma of sows, cord blood, and new-born piglets were collected. Full-term placentas and livers of new-born piglets were also sampled. The activities of glutathione peroxidase (GSH-Px) and total superoxide dismutase (T-SOD) in the plasma of sows on farrowing day were higher, and the concentrations of interleukin-6 (IL-6) and prostaglandin-endoperoxide synthase 2 (PGE2) in the plasma of sows on farrowing day and interleukin-1ß (IL-1ß) in the plasma of cord blood were lower in the FD group than those in the SD group (P < 0.05). The FD downregulated the expression of SOD, IL-1ß, IL-6, and transforming growth factor-ß-activated kinase 1 binding protein 1 (TAB1) mRNA but upregulated the expression of lipoxygenase enzyme 5 (ALOX5) and interleukin-10 (IL-10) mRNA in placentas (P < 0.05). The FD downregulated the protein expression level of p-JNK/JNK in placentas (P < 0.05). In the livers of new-born piglets, the FD upregulated the expression of ALOX5 (P < 0.05) and G-protein-coupled receptor 120 (GPR120) (P < 0.05) mRNA. Our results suggest that the maternal diet with fish oil might alleviate oxidative stress in sows on farrowing day and modulate inflammatory response in full-term placentas by inhibiting the JNK signal pathway. Moreover, the maternal diet with fish oil might partly regulate the neonatal immune response of their progenies.

OBJECTIVE: The root of Helicteres angustifolia L. (Sterculiaceae) has been used as tea to treat diabetics effectively by local people in Laos. However, no scientific evidence is available for this ethnomedicinal usage. This study was undertaken to explore the hypoglycemic effect of aqueous extract from Helicteres angustifolia root. PATIENTS AND METHODS: The effect of aqueous extract from Helicteres angustifolia root on glucose consumption in C2C12 myotubes was investigated at a dose of 25, 50 and 100 µg/mL, respectively. The alpha-glucosidase inhibitory activity of the extract was evaluated using rat intestinal maltase and sucrose. Moreover, oral sucrose tolerance test (OSTT) in normal and streptozotocin induced diabetic rats was performed. Finally, their cytotoxicity in C2C12 cells and acute oral toxicity in rats was analyzed. RESULTS: Aqueous root extract of Helicteres angustifolia significantly enhanced glucose consumption in C2C12 myotubes. The extract also significantly inhibited rat intestinal maltase (IC50 = 1.44 ± 0.24 mg/mL) and sucrase activity (IC50 = 0.54 ± 0.12 mg/mL), respectively. The OSTT results showed that the extract significantly suppressed the increase of blood glucose levels in normal and diabetic rats. The extract was also proven to have low acute toxicity (LD50 > 5 g/kg) and low cytotoxicity in C2C12 cells (IC50 > 0.4 mg/mL). CONCLUSIONS: The findings from this study indicate that aqueous root extract of Helicteres angustifolia possesses significant alpha-glucosidase inhibitory activity and moderate enhanced glucose consumption activity, while with low cytotoxic and acute toxicity.

OBJECTIVES: To study the changes of alcohol content and pharmacokinetic parameter in rats after taking Huoxiang Zhengqi liquid. METHODS: The rats were randomly divided into three groups and given with white alcohol at the dose of 3.0 mL/kg, low-dose and high-dose Chinese medicine liquor, respectively. The blood was collected before administration and 5 min, 10 min, 15 min, 30 min, 1 h, 2 h, 3 h, 4 h, 5 h, 6 h and 8 h after administration by cutting rats' tails. The concentrations of alcohol in blood were detected by headspace-gas chromatography method. The main pharmacokinetic parameters were calculated by DAS 2.0, and then analyzed by SPSS 17.0. RESULTS: The difference of maximum blood concentrations between high-dose Chinese medicine alcohol group and white alcohol group was statistically significant (P<0.05). There was no significant difference in other pharmacokinetic parameters among three groups (P>0.05). CONCLUSIONS: The Chinese herbal medicinal ingredients in the Huoxiang Zhengqi liquid has no effect on the metabolism and elimination of ethanol in rats. The research provides useful reference for the qualitative assessment and processing of traffic accident cases involved in Huoxiang Zhengqi liquid and the studies related to drug-interaction.

BACKGROUND AND AIM: The effect of soy isoflavones on blood pressure is controversial. The objective of this study was to evaluate the effect of dietary soy isoflavones on blood pressure. METHODS AND RESULTS: Trials were searched in PubMed, the Cochrane Library, Embase and references cited in related reviews and studies. A total of eleven trials were reviewed. Meta-analysis results showed a mean decrease of 2.5 mm Hg (95% CIs, - 5.35 to 0.34 mm Hg; P = 0.08) for systolic blood pressure and 1.5 mm Hg (95% CIs, - 3.09 to 0.17 mm Hg; P = 0.08) for diastolic blood pressure in the soy isoflavones-treated group compared to placebo. Meta-regression and subgroup analyses indicated that blood pressure status was a significant predictor of heterogeneity for the effect of soy isoflavones on blood pressure. Subgroup analysis of hypertensive subjects revealed that a greater blood pressure reduction was identified in the soy isoflavone-treated group compared to placebo (5 trials; SBP: - 5.94, 95% CIs [- 10.55, - 1.34] mm Hg, P = 0.01; DBP: - 3.35, 95% CIs [- 6.52, - 0.19] mm Hg, P = 0.04). In contrast, treatment with soy isoflavones did not lead to a significant reduction in blood pressure in normotensive subjects (6 trials; SBP: 0.29, 95% CIs [- 2.39, 2.97] mm Hg, P = 0.83; DBP: - 0.43, 95% CIs [- 1.66, 0.81] mm Hg, P = 0.50). CONCLUSION: Soy isoflavones had an effect of lowering blood pressure in hypertensive subjects, but not in normotensive subjects. Larger trials need to be carried out to confirm the present findings.

A new dicoumarinoid glycoside, named giraldoid A (1), has been isolated from Daphne giraldii Nitsche. The structure of 1 was determined as 7-O-beta-glucosyl-8-(7-hydroxy-2H-1-benzopyran-2-one-8-)yl-2H-1-benzopyran-2-one on the basis of chemical reactions and spectroscopic methods.

RESUMEN

Two new megastigmane glycosides, eriojaposides A (1) and B (2), and a new acylated triterpenoid (3) were isolated along with nine known compounds from a leaf extract of Eriobotrya japonica. The structures of 1--3 were characterized as (6R,9R)-3-oxo-alpha-ionyl-9-O-beta-xylopyranosyl-(1' '-->6')-beta-glucopyranoside, (6R,9R)-3-oxo-alpha-ionyl-9-O-alpha-rhamnopyranosyl-(1' '-->6')-beta-glucopyranoside, and 3 alpha-trans-feruloyloxy-2 alpha-hydroxyurs-12-en-28-oic acid, respectively, on the basis of spectral and chemical evidence.

AIM: To study the effect of ligustrum fruit on spermatogenesis and blood gonadal hormones in diabetic rats. METHODS: Experimental diabetes was induced in male Wistar rats with streptozotocin. Ligustrum fruit extract was given by gastric gavage at a dose of crude drug 30 g x kg(-1) x d(-1) for 110 days. The serum gonadadotropic hormones and testosterone were determined on d 60 and testicular histology examined on d 110. RESULTS: In the control diabetic rats, the seminiferous tubules were dilated and the spermatogenic cells irregularly arranged. Spermatogenesis was arrested with the number of spermatids highly reduced and spermatozoa not observed. In the treated rats, all types of spermatogenic cells were practically normal. The serum luteinizing hormone (LH), follicle-stimulating hormone (FSH) and testosterone levels were higher in the treated than in the control rats, but the difference was insignificant. CONCLUSION: In experimental diabetic rats, ligustrum fruit extract protects the damaging effect of experimental diabetes on spermatogenesis.

RESUMEN

Five new 11(15-->1)-abeo-taxane diterpenoids, taxuyunnanines K-O (1-5), were isolated from an ethanol extract of the bark of Taxus yunnanensis, and their structures were determined using MS and NMR techniques. Compounds 1/2 and 4/5 are rearranged taxane diterpenoids possessing an opened oxetane ring moiety at C4(20). Compounds 4/5 are rearranged taxoids lacking an oxygenated functionality at C-4.

Three new flavonoid glycosides, together with 15 known flavonoids, have been isolated from the leaves of Eriobotrya japonica, and characterized as (2S)- and (2R)-naringenin 8-C-alpha-L-rhamnopyranosyl-(1-->2)-beta-D-glucopyranosides, and cinchonain Id 7-O-beta-D-glucopyranoside, respectively, based on spectral analyses including two dimensional (2D) NMR techniques. Higher proanthocyanidin fraction in the water-soluble portion of the extract was characterized as a procyanidin oligomer mixture mainly composed of undecameric procyanidin. These polyphenols have also been assessed for cytotoxic activity against two human oral tumor (human squamous cell carcinoma and human salivary gland tumor) cell lines. Selective cytotoxicity of the procyanidin oligomer between tumor and normal gingival fibroblast cells, and its possible mechanism, were also described.

AIM: To study the anti-inflammatory effects of total saponins of Panax notoginseng (PnS). METHODS: Rat air-pouch acute inflammatory model was established with s.c. carrageenan (Car, 25 mg.kg-1). The protein content in exudate was measured. Micro-acid titration assay and radioimmunoassay (RIA) were applied respectively to investigate effects of PnS on phospholipase A2 (PLA2) activity and dinoprostone (Din) content in exudate. Fura-2 fluorescence technique was used to determine the intracellular free calcium concentration in neutrophils (Neu-[Ca2+]i). RESULTS: At 12 h, PnS 60-240 mg.kg-1 i.p. reduced Neu counts, protein content [(7.7 +/- 1.3) to (4.4 +/- 1.4) g.L-1], and Din content [(1619 +/- 391) to (883 +/- 268) ng.L-1]; inhibited the PLA2 activity in exudate [(248 +/- 42) to (157 +/- 35) kU.L-1] in a dose-dependent manner. PnS 60, 120, and 240 mg.kg-1 lowered the level of Neu-[Ca2+]i with the inhibitory rate of 9.1%, 33.2%, and 39.4%, respectively. CONCLUSION: PnS has an obvious anti-inflammatory effect and its mechanisms are related to the inhibition of the Neu-[Ca2+]i level and PLA2 activity, and reduction of Din content.

Dulcitol was isolated chemically from Celastrus obiculatus Thumb and determined by HPLC. Effects of dulcitol were examined on collagen-induced arthritis (CIA) in DBA/1J mice. From 6 weeks after the first immunization with bovine type II collagen, dulcitol (100 mg/kg body weight/day) was administered orally to immunized mice for 9 weeks. Clinical score of CIA was improved significantly by dulcitol intervention compared with the non-treated CIA mice. Radiographic score of phalangeal destruction was also improved by dulcitol treatment. These findings suggest that dulcitol may play a role in regulation of some inflammatory responses in the present arthritis model. Significant reduction of percentage of CD4+ and CD8+ T cell subsets in the spleen leucocytes of CIA mice was observed by flow cytometry. Almost normal level of CD4+ and CD8+ T cells was observed in dulcitol-treated groups, suggesting T cell-modifying effect of dulcitol in CIA. Weight of spleen was larger in CIA mice and it was not affected by dulcitol. Anti-collagen antibody titer was increased in CIA mice, and it was not affected by dulcitol, either. Improvement of the changes of CD4+ and CD8+ T cells in the spleen by dulcitol may suggest its modulatory effect on cellular immunity.

Dentatorubral and pallidoluylsian atrophy (DRPLA) is a progressive neurological disorder characterized by neuronal degeneration, especially in the cerebellar dentate nucleus. DRPLA is caused by an unstable expansion of a CAG trinucleotide repeat coding for glutamine in a gene of unknown function, termed atrophin-1, located on chromosome 12. To gain additional understanding of atrophin-1, we have isolated a second member of the atrophin-1 gene family by screening rat cDNA libraries. The 1006-amino-acid product of this gene, which we have termed rat atrophin related protein(rARP), does not contain a glutamine repeat, but it does contain two regions of alternating acidic and basic amino residues similar to those found in atrophin-1. rARP is widely expressed as both a 7.4- and a 9.4-kb message, with enrichment in cerebellum and testis. Like atrophin-1, the rARP in vitro translation product migrates more slowly on SDS-polyacrylamide gel electrophoresis than predicted by molecular weight. We conclude that, at least in the rat, polyglutamine is not an essential feature of the atrophin family of genes.

1. The effects of a formula of traditional Chinese medicine, TBL-1, on collagen-induced arthritis (CIA) were investigated in DBA/1J mice. 2. From 4 weeks after the first immunization with bovine type II collagen (CII), TBL-1 or indomethacin was administered orally for 13 weeks. 3. Clinical scores of CIA were decreased by both TBL-1 and indomethacin intervention compared with the control CII-immunized group. 4. Radiographic scores of phalangeal destruction were markedly improved by TBL-1 intervention (P < 0.001), but indomethacin failed. 5. The suppressive effects of TBL-1, but not indomethacin, were manifested in reduced serum anti-CII antibody titres (P < 0.001). 6. These findings suggest that TBL-1 may play a role in regulating some immune responses in the present arthritis model.

The Inositol 1,4,5-trisphosphate receptor (IP3R) is an intracellular calcium channel involved in coupling cell membrane receptors to calcium signal transduction pathways within the cell. We have cloned a cDNA for the human type 1 inositol 1,4,5-trisphosphate receptor. The sequence contains the S2 splice site which appears to be the region most divergent between rat and human. We now report an additional alternatively spliced region in the coupling domain, that is 9 amino acids long, which we term S3. Alternatively spliced forms are found in both human and rat. PCR analysis of brain and peripheral tissues from human and rat shows both transcripts of the type 1 inositol 1,4,5-trisphosphate receptor in all tissues. The long form predominates in most brain regions (except the cerebellum) while the short form predominates in peripheral tissues. The sequence of the longer form in human appears to create an additional consensus protein kinase C phosphorylation site.

RESUMEN

Expansion mutation is the cause of eight neuropsychiatric disorders. Thus far each disease is the result of expansion of a C-G rich trinucleotide repeat that is polymorphic for length in the general population. We now report the identification of seven novel cDNA clones with CCA or equivalent trinucleotide repeats obtained by screening a human frontal cortex cDNA library. The repeat lengths of two clones, CCA11 (linked to D20S101, expressed in human brain as a 3.2 kb message) and CCA38 (linked to D5S404), are highly polymorphic in a normal human population. CCA54, mapped to chromosome 19, appears to correspond to a portion of the human gene encoding the alpha 1 subunit of a P-type calcium channel. Expansion mutations at these loci should be considered as possible candidates in evaluating the genetic etiologies of diseases linked to chromosomes 5, 19, and 20.

Dentatorubral pallidoluysian atrophy (DRPLA) is a rare, progressive, fatal neuropsychiatric disorder similar to Huntington's disease, caused by an expansion of a CAG trinucleotide repeat encoding glutamine. We have cloned the cDNA of the rat homologue of this gene. The cDNA contains a 3549 base pair open reading frame that is 88.2% identical to the human cDNA, with a predicted amino acid sequence that is 93.6% identical to the human sequence. The consecutive glutamine repeat is only five residues in length (normal range in human: 7-35 glutamines) and is followed by a polymorphic region of alternating glutamine and proline residues (QQQQQPQPQPQPQQ). The sequence also includes a polymorphic proline repeat, a serine repeat, and a region of alternating acidic and basic residues. Northern analysis and in situ hybridization indicate that the gene is widely expressed as a 4.5 kb mRNA, with a neuronal distribution in the brain. The widespread expression of this gene is consistent with the possibility that DRPLA, like other glutamine repeat diseases, is a result of an abnormality at the protein level.

Lao-guan-cao has long been used in Chinese traditional medicine. It is used as an antirheumatic and circulation promoting drug for the treatment of acute and chronic rheumatalgia; also used as a detoxicant for enteritis and bacillary dysentery. The dried aerial parts of Erodium stephanianum Willd. and Geranium wilfordii Maxim. (family Geraniaceae) have been specified in the Chinese Pharmacopoeia (1990 ed.) According to the literature and the authors' market investigations, the commercial drug Lao-guan-cao chiefly consists of the aerial parts of Erodium stephanianum Willd. Also those rather popularly used in several provinces are the aerial parts of Geranium nepalense Sweet and G. sibiricum L. In this paper, the morphological and histological characters of the crude drugs derived from E. stephanianum Willd., G.nepalense Sweet and G.sibiricum L. are described. It is found that the following characters are useful for distinguishing the drugs derived from the above mentioned three species. (1) The shape and size of leaf; (2) the number and arrangement of the vascular bundles of stem; (3) the number of the vascular bundles in midrib and layers of palisade cells of leaf; (4) the presence or absence of stomata in upper epidermis; (5) the characters of cells containing crystals in sepal; (6) the characters of epidermis cells, cells containing crystals and fibers of pericarp; (7) the types of hairs and glandular hairs on epidermis of stem, leaf, sepal and pericarp; (8) the characters of anticlinal walls of epidermal cells of seed coat.
