RESUMEN

Previous clinical studies have shown that anisodamine could improve no-reflow phenomenon and prevent reperfusion arrhythmias, but whether this protective effect is related to the antagonism of the M-type cholinergic receptor or other potential mechanisms is uncertain. The aim of the present study was to investigate the role of the mitochondrial ATP-sensitive potassium channel (mitoK ATP ) in cardioprotective effect of anisodamine against ischemia/reperfusion injury. Anisodamine and 5- hydroxydecanoic acid were used to explore the relationship between anisodamine and mitoK ATP . Using a Langendorff isolated heart ischemia/reperfusion injury model, hemodynamic parameters and reperfusion ventricular arrhythmia were evaluated; in addition, changes in myocardial infarct size, cTnI from coronary effluent and myocardial ultrastructure, as well as ATP, MDA and SOD in myocardial tissues, were detected. In the hypoxia/reoxygenation injury model of neonatal rat cardiomyocyte, cTnI release in the culture medium and levels of ATP, MDA and SOD in cardiomyocytes and mitochondrial membrane potential, were analyzed. Overall, anisodamine could significantly improve the hemodynamic indexes of isolated rat heart injured by ischemia/reperfusion, reduce the occurrence of ventricular reperfusion arrhythmia and myocardial infarction area, and improve the ultrastructural damage of myocardium and mitochondria. The in vitro results demonstrated that anisodamine could improve mitochondrial energy metabolism, reduce oxidative stress and stabilize mitochondrial membrane potential. The cardioprotective effects were significantly inhibited by 5-hydroxydecanoic acid. In conclusion, this study suggests that the opening of mitoK ATP could play an important role in the protective effect of anisodamine against myocardial ischemia/reperfusion injury.

An oilfield reservoir over long-term operation may have different petrophysical information, which has a significant impact on oilfield maintenance and finance. Successful oilfield enhanced oil recovery benefits a lot from identifying and analyzing the variations of the critical properties after long-term waterflooding treatments. Since the inspection wells drilled within different development periods contain the core samples that have the petrophysical information at that period, it is necessary to collect and test the samples from different periods to investigate the overall tendency of the petrophysical properties. The samples from four inspection wells, which were drilled in four stages since the very beginning of development, were subjected to in-laboratory core analysis methods to illustrate the variation of some critical parameters in the reservoir. The permeability and porosity variation are revealed clearly by the experimental results. The migration and dissolution of clay minerals play a crucial role in the variation of petrophysical information and pore structure. To quantify the variations above, we applied the multiple linear regression model into our investigation. The dependent variable and all of the predictors in the model come from the experimental results. The quantitative results show the closed correlation between different parameters in the formation. With the development stage moving forward, the weight coefficients for different predictors have multiple trends. The experimental and statistical approach provides a novel understanding of the reservoir properties with the effect of waterflooding treatment.

BACKGROUND: Total knee arthroplasty (TKA) is an optimal option for patients with middle-to-end-stage knee osteoarthritis. However, the management of postoperative acute pain remains inefficient. Transcutaneous electrical acupoint stimulation (TEAS) is a nonpharmacological method to manage postoperative acute pain. Different frequencies of TEAS have been tested using varying parameters, but the optimal analgesic frequency remains controversial. The aim of this study was to explore the optimal analgesic frequency of TEAS for treating acute pain after the primary unilateral TKA. METHODS/DESIGN: This is a double-blind, randomized controlled trial. A total of 156 patients are randomly assigned to: G1, 5 Hz TEAS; G2, 100 Hz TEAS; G3, mixed TEAS (alternative use of daily 5 Hz and 100 Hz TEAS) and G4, placebo TEAS. In the G1, G2 and G3 groups, TEAS is conducted at acupoints SP9 and GB34 of the leg that was operated on (at a wave of continuous, balanced and asymmetrical biphasic square, with a pulse width of 200 µs, and a strong but comfortable current) for 30 min prior to a 30-min rehabilitation session per day for 2 weeks. In G4 group, TEAS is delivered at a strong but comfortable current for 30 s, then the current is gradually decreased to none over the next 15 s. The primary outcomes are measured before surgery, at baseline (POD 3, before TEAS intervention), week 1 and 2 after TEAS intervention with the Numeric Pain Rating Scale and The American Knee Society Score. The secondary outcomes include: (1) Active range of motion of the knee that was operated on; (2) Surface electromyography of both quadriceps; (3) Modified 30-s sit to stand test; (4) Additional usage of analgesia; and (5) SF-36. The additional outcomes include: (1) Patients' satisfaction rate; (2) Patient's expectation rate; and (3) Incidence of analgesia-related side effects. To test the blinding of participants and assessors, they are asked to guess whether the subjects received active or placebo TEAS within 5 min after the latest intervention. The safety and financial cost of TEAS are assessed. DISCUSSION: Mixed TEAS has more favorable effect on acute pain control than the placebo or 5 Hz or 100 Hz TEAS. OBJECTIVE: Glioma-associated oncogene homolog 1 (Gli1) in Hedgehog signal pathway regulates Cyclin D1 expression, cell cycle or proliferation modulation. Esophageal cancer patients had significantly elevated Gli1 expression, which is related with survival and prognosis. It has been demonstrated that the level of miR-150 was decreased in esophageal cancer patients compared to normal control. As a complementary relationship exists between miR-150 and 3'-UTR of Gli1, this study investigated if miR-150 played a role in regulating Gli1 expression, and proliferation or cell cycle of esophageal cancer cells. PATIENTS AND METHODS: Esophageal squamous cell carcinoma (ESCC) patients from our hospital were recruited to collect tumor and adjacent tissues for miR-150 and Gli1 expression. Esophageal carcinoma cell line EC9706 and normal esophageal epithelial cell line HEEC were compared for expression of miR-150, Gli1 and Cyclin D1. Dual luciferase reporter gene assay examined the targeted relationship between miR-150 and 3'-UTR of Gli1. In vitro cultured EC9706 cells were treated with miR-150 mimic, si-Gli1 or the combination of miR-150 mimic and si-Gli1, respectively, to check their gene expression, cell cycle and proliferation. RESULTS: ESCC tissues had significantly higher Gli1 expression and lower miR-150 expression. EC9706 cell also had higher Gli1 expression than that in HEEC, whilst miR-150 was down-regulated. Via targeting 3'-UTR of Gli1 gene, miR-150 inhibited its expression. Transfection of miR-150 mimic, si-Gli1 or the combination of miR-150 mimic and si-Gli1, respectively, remarkably decreased expression of Gli1 and Cyclin D1 expression in EC9706 cells, whose cell cycle arresting at G0/G1 phase was enhanced with weakened proliferation. CONCLUSIONS: MiR-150 can induce G0/G1 cell cycle arresting and weaken proliferation of esophageal carcinoma cells via targeted inhibition on Gli1 and downstream expression of Cyclin D1.

Heortia vitessoides has been a serious defoliating pest of Aquilaria sinensis forests in recent years.The adults displayed strong tropism to the frequency trembler grid lamps and the nectar source plants.The favorite nectar source plants of H.vitessoides adults as the trap plants and the frequency trembler grid lamps in the integrated management of H.vitessoides were studied in the adult eclosion period through both the laboratory and field.The results showed that Kuhnia rosmarnifolia and Santalum album plants showed strong attraction to the H.vitessoides adults, with significant differences among the different nectar source plants.K.rosmarnifolia and S.album as trap plants with board type of planting area to total planting area of 5%-10%, and the frequency trembler grid lamps trapped significantly more adults of H.vitessoides. These results suggested that the frequency trembler grid lamps and trap plants could play an important role in the integrated management of the pest H.vitessoides of A.sinensis.

Juzen-taiho-to is an immunostimulatory herbal formulation that is clinically used in East Asia for cancer patients undergoing chemotherapy and radiation. The formulation stimulates various leukocytes, including T, B, and NK cells and macrophages. Although Juzen-taiho-to is known to contain numerous compounds with various pharmacological activities, it is not clear which compounds are responsible for the stimulation of individual cell types. Here, we conducted what we call "biomarker-guided screening" to purify compounds responsible for the macrophages stimulatory activity. To this end, gene expression was analyzed by a DNA array for macrophages treated with Juzen-taiho-to and DMSO (vehicle control), which identified intercellular adhesion molecule 1 as a biomarker of macrophage stimulation by Juzen-taiho-to. A quantitative reverse transcription polymerase chain reaction assay of intercellular adhesion molecule 1 was then used to guide the purification of active compounds. The screening resulted in the purification of a glycolipid mixture, containing ß-glucosylceramides. The glycolipid mixture potently stimulated intercellular adhesion molecule 1 expression in primary dendritic cells as well as in primary CD14+ (macrophages) cells. The identification of this glycolipid mixture opens up an opportunity for further studies to understand how plant-derived glycolipids stimulate macrophages and dendritic cells in a safe and effective manner as demonstrated by Juzen-taiho-to.

OBJECTIVE: The aim of the present study was to screen the Metarhizium strains with high virulence against the larvae of Dorysthenes hydropicus, a serious pest of Citrus grandis. METHOD: Thirty six strains of Metarhiziums were isolated from the soil of C. grandis GAP base and collected from other institutions, and the pathogenicity of these strains against 1st instar larvae of D. hydropicus was detected at concentration of 1 x 10(8) conidia/g. The high violence strains against D. hydropicus were cultivated in sabouraud dextrose yeast medium at first, then transfer to rice grain. And the sporulations of these violent strains against D. hydropicus were detected. RESULT: Twenty-eight strains showed virulence against D. hydropicus by preliminary study, and 7 strains of them were collected for further study, 6 of the 7 showed high virulence, the highest cadaver rate was higher than 74%. The conidia production of strain 1 and strain 4 were 2.35 +/- 0. 25 (1 x 10(9) conidia/g), 2.21 +/- 0.27 (1 x 10(9) conidia/g), respectively, showed significantly higher than other strains. CONCLUSION: Strain 1 and strain 4 of the 36 Metarhiziums strains showed high virulence against D. hydropicus, and the highest sporulation ability, so they have a best application prospect.

RESUMEN

A type of entomopathogenic fungus of soil in Citrus grandis 'tomentosa' production base was isolated and identified with morphological and molecular biological methods, including pathogenesis, spore characteristic and ITS sequence analysis were conducted. The results showed that eighteen entomopathogenic fungi strains were isolated from the Tenebrio molitor infected in the soil samples, which were identified as Metarhizium anisopliae var. anisopliae. Based on results above, we concluded that there was quantity of Metarhizium resources in this area. These provided the useful information for controlling some pests of C. grandis by using these strains of fungus.

To analyze naringin, naringenin and its metabolites in rat urine and feces after intragastric administration of alcohol extract of Exocarpium Citri Grandis, healthy SD rats were fed with alcohol extract of Exocarpium Citri Grandis for 3 days. On the last day, 0-24 h feces and 0-4 h, 4-8 h, 8-24 h urine were collected and analyzed by UPLC-Q-TOF/MS. The post-acquisition data were processed using Metabolynx The result is that naringin and its 6 metabolites, naringenin and its 4 metabolites were detected in the urine of rat. Meanwhile, naringin and its 3 metabolites, naringenin and its 2 metabolites were detected in the feces of rat.

OBJECTIVE: To analyze naringin, naringenin and its metabolites in rat plasma after intragastric administration of exocarpium Citri grandis alcohol extract. METHOD: Rat blood samples were collected 1.0 hour after oral administration of 50 g x kg(-1) exocarpium Citri grandis alcohol extract and analyzed by UPLC-Q-TOF with MS(E) function. The post-acquisition data were processed using Metabolynx. RESULT: Naringin (M1), naringenin (M2), naringin-5-O-glucuronide (M3), naringin-4-O-glucuronide (M4), glucuronide conjugate of naringenin (M5), naringin-4-O-sulfate (M6), methylated conjugate of hydroxylated naringenin (M7), glucuronide and sulfate conjugate of naringenin (M8), glucuronide conjugate of hydroxylated naringenin (M9) in rat plasma were detected. M3, M4, M6 were first reported as the metabolites of naringin. M7, M9 were first reported as the metabolites of naringenin. CONCLUSION: The results indicated that naringin, naringenin can be metabolited as the forms of glucuronidation, sulfation and naringenin can also be metabolited as the forms of methylation with hydroxylation and glucuronidation with hydroxylation in vivo after administration.

A recombinant yEGFP gene yeast strain necessary for the routine screening of estrogen activity in the chemical product of the environment was described. Two plasmids, one containing human estrogen receptor (hER) cDNA fused to the GPD gene promoter and another yEGFP inserted under the control of ERE, were constructed. The use of hER cDNA and the yEGFP reporter in the yeast cell sensor resulted in estrogenic chemical product-dependent light emission of yEGFP without additions owing its advantages: a simple and reagent-free measurement of GFP, and a non-toxic protein characteristic. The time needed for the optimal induction of light emission was 4h. The maximal fold induction of 8.80 over uninduced levels at the concentration of 10(-5)M of bisphenol A and 0.1 nM sensitivities for four different estrogenic chemicals tested were obtained. Five different chemicals which could not bind to hER did not cause an induction of yEGFP. This bioassay can be performed completely in 96-well plates. Thus, this test system can be used as a rapid screening system for the surveillance of estrogenic chemical products in the environment. The yEGFP assay is sensitive, reproducible, and cheap, which makes it highly suitable to be used as a high throughput system.

OBJECTIVE: To discover new chemopreventive agents, a drug screening cell model based on reporter gene and antioxidative response element (ARE) has been established. METHOD: Four repeas of ARE DNA binding conserved sequences were synthesized and cloned into a GFP expression vector. This construct was stably transfected into HepG2 cells in vitro. The cell model was tested with known chemopreventive agents and the effects of resveratrol, protocatechuic aldehyde, ursolic acid and oleanolic acid at different concentration (0, 12.5, 25, 50, 100, 200 micromol x L(-1)) were observed by determining reporter gene GFP activity. RESULT: The induce level of GFP was regulated by ARE and the dose-dependence in a certain range was observed. The induce level of GFP by resveratrol was significantly increased. CONCLUSION: The method can be used to screening of chemopreventive agents from Chinese traditional medicine by measurement of luminescent value of expressed GFP in wells of microtiter plate.

OBJECTIVE: To study the biological characteristics of Dorysthenes hydropicus in the farm of Cirtus grandis, and offer scientific evidence for prevention and controlling of D. hydropicus. METHOD: Indoor-rearing and light trap were applied to study the biological characteristics, development course and harmful effect of D. hydropicus. RESULT: D. hydropicus reproduces one generation in 1-2 year in Guangdong province, and overwinters in the form of larvae. Its imago comes out of the earth mainly in late May after mature. The body length has great individual diversity normally ranged from 25-60 cm, It also shows strong phototaxy. One lamp can trap more than 2 000 of them per night. Female imago has a large egg load with the maximum amount of 543. The eggs hatching is in depth of 1-3 cm soil. The dominant hatching period of egg is from late June to early July, and hatchability is over 85%. The living space of larva ranges from 15-60 cm in soil. D. hydropicus has caused serious harm and lead to thousands of Cirtus grandis trees death every year. CONCLUSION: Dorysthenes hydropicus showed serious threat to the growth of Cirtus grandis and should be prevented and controlled.

OBJECTIVE: To study the content of phytoestrogen in dissimilarity herbs. METHOD: The activity of phytoestrogen in heat-clearing drugs, drugs for relieving exterior syndrome, diuretic, anastaltics, tonics and astringents were detected based on the recombinant yeast cell (W303-1A/hER-ERE-Lac Z). The estrogenic activity in traditional Chinese materia medica were assayed quantitatively by determining the expression of beta-galactosidase. RESULT: The phytoestrogen concentration (6.35 x 10(-3) nmol x g(-1) E2 equivalent) in heat-clearing drugs was the highest while that in anastaltic and tonic drugs was the lowest, which was less than the detected limit. CONCLUSION: Compared with the other traditional Chinese materia medica, the content of phytoestrogen, which can bind to estrogen receptor, in giant knotweed rhizome, forsythia suspense, ash bark, baical skullcap root and ophiopogonis tuber were higher.
