Nutritionists have been discussing whether the dietary supplementation of cyst(e)ine is required as a part of the dietary methionine (Met) in the total sulfur amino acid (TSAA) requirement to achieve optimum performance in broilers. Part of Met is converted to cysteine (Cys) to meet the Cys requirement, especially for feather growth. The TSAA requirement has been determined by using graded levels of free Met in the diet, without supplementation of free cyst(e)ine. It has also been argued that the Met to Cys ratio (Met : Cys) changes with age and even with different Met sources. The objective of this study was to evaluate the two sources of Met, while determining the proportion of Met and Cys in total dietary TSAA that optimize the performance of broilers. A performance assay was carried out in a factorial arrangement (5 × 2) using 1080 broilers from 42 to 56 days of age fed diets having different dietary proportions of Met and Cys (44 : 56, 46 : 54, 48 : 52, 50 : 50 or 52 : 48) while maintaining the same dietary TSAA in the diets. Two synthetic Met sources (dl-Met or l-Met) were used for each of the diets with different dietary Met : Cys ratios. Twenty-one broilers of the same age were fed the diets 44 : 56, 48 : 52 and 52 : 48 by supplementing the diet with L-(15N) Met or L-(15N2) Cystine to study the metabolism of TSAA. No differences were observed between Met sources for feed intake, BW gain and feed conversion ratio (FCR; P > 0.05); however, FCR was numerically improved at 50 : 50 Met : Cys. Regarding TSAA utilization, the conversion of Met to Cys increased with increase in Met : Cys ratios, but the concentration of Met intermediates decreased. Broiler chickens responded to different dietary proportions of sulfur amino acids by altering their sulfur amino acid metabolism, and diets containing 50 : 50 Met : Cys is recommended for broilers of age 42 to 56 days.

PURPOSE: Differentiated thyroid cancer (DTC) patients with an unresectable primary tumor cannot benefit from curative surgery, and radioiodine treatment for locoregional and distant disease is not possible with the thyroid gland still in place. Due to local invasion, these patients cannot be included in clinical trials, so that treatment options are limited. The aim of this study was to describe the characteristics and the prognosis of patients with these locally unresectable DTC. PATIENTS AND METHODS: A retrospective and multicentric analysis of consecutive cases of unresectable DTC diagnosed between 2000 and 2015 was performed. RESULTS: The study population consisted in 22 patients, 13 females (59%); median age: 77 years (range: 52-91). Thyroid tumors were papillary in six, follicular in seven, Hürthle cell in one and poorly differentiated in eight patients. Patients were treated with external beam radiation therapy (EBRT) (57%), locoregional therapy of distant metastases (41%), cytotoxic chemotherapy (38%) and tyrosine kinase inhibitors (TKIs) (33%). TKI treatment resulted in median disease control duration of 7 months with a grade 3-4 toxicity rate of 44%. Only one patient had a total thyroidectomy after neo-adjuvant EBRT. The 1, 3 and 5-year cumulative survival rate was 81%, 27.7% and 21.5%, respectively. The cause of death was DTC in 11 cases (local progression in 7), and to other causes in 7 cases; no patient died from treatment toxicity. CONCLUSIONS: Clinical trials and approved treatments are lacking for unresectable DTC. TKI treatment may allow prolonged disease control with acceptable toxicity.

RESUMEN

Sarcopenia is a major component of the frailty syndrome, both being considered as strong predictors of morbidity, disability, and death in older people. In this review, we explore the definitions of sarcopenia and frailty and summarize the current knowledge on their relationship with oxidative stress and the possible therapeutic interventions to prevent or treat them, including exercise-based interventions and multimodal strategies. We highlight the relevance of the impairment of the nervous system and of the anabolic response (protein synthesis) in muscle aging leading to frailty and sarcopenia. We also discuss the importance of malnutrition and physical inactivity in these geriatric syndromes. Finally, we propose multimodal interventions, including exercise programs and nutritional supplementation, as the strategies to prevent and treat both sarcopenia and frailty.

BACKGROUND: Low serum magnesium (MgS) is a known risk factor for cardiovascular and mineral bone disease. In renal transplant recipients (RTRs), low MgS levels have been related to higher glomerular filtration rates (GFR) and with calcineurin inhibitors, particularly tacrolimus. We aimed to evaluate MgS in renal transplant recipients with over 1 year of follow-up to establish related risk factors and the impact of the use of cyclosporine versus tacrolimus. METHODS: Cross-sectional study of 94 RTRs with more than 12 months of follow-up. Hypomagnesemia was defined as serum magnesium level <1.5 mg/dL. RESULTS: Hypomagnesemia was found in 5.3% of patients. MgS showed a negative correlation with creatinine clearance. A positive correlation between MgS with urinary magnesium and phosphorus was found. Cyclosporine versus tacrolimus analysis did not show a significant difference regarding MgS when considering all the population and the subgroup of patients with GFR >45 mL/min/1.73 m2. On the subgroup with GFR <45 mL/min/1.73 m2, those on tacrolimus had lower MgS than those on cyclosporine, but those same patients presented with significantly different GFR, higher in the tacrolimus subgroup. CONCLUSIONS: Hypomagnesemia has a low prevalence in RTRs with more than 1 year of follow-up. MgS levels evidenced a strong correlation with GFR. A significant difference on MgS levels between patients on tacrolimus and cyclosporine was found only when considering GFR <45 mL/min/1.73 m2, in which patients on tacrolimus had significantly higher GFR than patients on cyclosporine, which may explain these results.

Altered tissue fatty acid (FA) composition may affect mechanisms involved in the control of energy homeostasis, including central insulin actions. In rats fed either standard chow or a lard-enriched chow (high in saturated/low in polyunsaturated FA, HS-LP) for eight weeks, we examined the FA composition of blood, hypothalamus, liver, and retroperitoneal, epididymal and mesenteric adipose tissues. Insulin-induced hypophagia and hypothalamic signaling were evaluated after intracerebroventricular insulin injection. HS-LP feeding increased saturated FA content in adipose tissues and serum while it decreased polyunsaturated FA content of adipose tissues, serum, and liver. Hypothalamic C20:5n-3 and C20:3n-6 contents increased while monounsaturated FA content decreased. HS-LP rats showed hyperglycemia, impaired insulin-induced hypophagia and hypothalamic insulin signaling. The results showed that, upon HS-LP feeding, peripheral tissues underwent potentially deleterious alterations in their FA composition, whist the hypothalamus was relatively preserved. However, hypothalamic insulin signaling and hypophagia were drastically impaired. These findings suggest that impairment of hypothalamic insulin actions by HS-LP feeding was not related to tissue FA composition.

RESUMO O presente trabalho teve como objetivo avaliar a ação antifúngica dos extratos de Allamanda blanchetti A.DC. e Momordica charantia L. no controle de Colletotrichumgloesporioides e qualidade pós-colheita em frutos de mamoeiro da cultivar Sunrise solo. Os experimentos foram conduzidos nos Laboratório de Fitopatologia e Laboratório de Produtos de Origem Animal, CCA-UFPB, Campus.II, Areia-PB. Os frutos foram colhidos no estádio 1 de maturação, em seguida, sanitizados com hipoclorito de sódio a 1% por três minutos e pulverizados com os extratos etanólicos de A. blanchetti e M. charantia nas concentrações: 0, 10, 100, 500 e 1000 µg.mL-1. Os frutos foram distribuídos em bandejas de polipropileno expandido em temperatura ambiente (23±2 °C) e umidade relativa em torno de 70%. Após doze dias de armazenamento, os frutos foram avaliados quanto à incidência natural de C. gloesporioides, severidade, atividade da enzima peroxidase, sólidos solúveis, acidez titulável, relação SS/AT, perda de massa e aparência externa dos frutos. Foi realizada análise de regressão polinomial testando modelos até o terceiro grau para as concentrações dos extratos utilizando análise de variância pelo teste F enquanto as médias foram comparadas pelo teste de Tukey a 5% de probabilidade, através do programa SAS® 9.2. Não se observou diferença estatística quando avaliou-se a incidência natural de C. gloesporioides em frutos de mamoeiro utilizando os extratos de A. blanchetti e M. charantia. Já na avaliação da severidade da antracnose, verificou-se diferença para o tratamento com o extrato de A. blanchetti, que apresentou as menores escalas de notas em relação ao tratamento com a M. charantia. Observou-se atividade da peroxidase (POD) em frutos de mamoeiro nos extratos de A. blanchetti e M. charantia. Os frutos tratados com A. blanchetti apresentaram teor de acidez, com máximo de 0,12 g de ácido cítrico.100 g-1 e no de M. charantiaapresentou aumento para 0,15 g de ácido cítrico.100 g-1. A relação SS/AT diminuiu com o aumento das concentrações dos dois extratos. Os frutos tratados com A. blanchetti apresentaram maior perda de massa durante todo o armazenamento, sendo maior que 10% aos nove dias. Já a perda de massa dos frutos tratados com M. charantia apresentou comportamento linear, sendo superior a 12% aos 12 dias de armazenamento. A cor evoluiu de 1 (verde) para 6 (casca alaranjada) e a aparência dos frutos diminuiu com o decorrer do tempo de 5 (excelente) para 3 (regular) em todas concentrações dos extratos de A. blanchetti e M. charantia. Os extratos de A. blanchetti e M. charantia não reduziram a incidência natural, porém, só o extrato de A. blanchetti foi responsável por reduzir a severidade da antracnose. Ocorreu a atividade da enzima peroxidase em todos os tratamentos avaliados e conservaram a qualidade pós-colheita em frutos até os nove dias de armazenamento.


ABSTRACT This study aimed to evaluate the antifungal effect of the extracts Allamandablanchetti A.DC. and Momordica charantia L. in the control of Colletotrichum gloesporioides in papaya fruits of the Sunrise Solo cultivation, as well as their post-harvest quality. The trials were conducted in the Laboratory of Plant Pathology and Animal Products Laboratory, CCA-UFPB, Campus II, Areia-PB. The fruits were harvested at maturity stage 1, then sanitized with sodium hypochlorite 1% for three minutes and sprayed with ethanol extracts of A. blanchetti and M.charantia at concentrations of 0, 10, 100, 500 and 1000 μg.mL-1.The fruits were distributed in polypropylene trays expanded at room temperature (23±2 °C) and relative humidity around 70%. After twelve days of storage, the fruits were evaluated for natural incidence of C. gloesporioides, severity, peroxidase enzyme activity, soluble solids, titratable acidity, SS/TA ratio, weight loss and external appearance of the fruit. Polynomial regression testing models to the third degree were employed for the extracts of concentrations, using analysis of variance by F test. The averages were compared by Tukey test at 5% probability, through the SAS® 9.2 program. There was no statistical difference observed when we evaluated the natural incidence of C. gloesporioides in the papaya fruit, using extracts from A. blanchetti and M. charantia. In the assessment of the severity of the anthracnose, there was no statistical difference for the treatment with the extract of A. blanchetti, which had smaller scales of notes when compared with the treatment using the M. charantia. It was observed peroxidase activity (POD) in the papaya fruits treated with the extracts of A. blanchetti and M. charantia. The fruits treated with A. blanchetti presented a maximum dose of acidity of 0.12 g g-1 of citric acid.100 g-1. In the fruits treated with M. charantia this increase was of 0.15g of citric acid.100 g-1. The SS/TA ratio decreased with increasing concentrations of the two extracts. The fruits treated with A. blanchettipresented higher weight loss of approximately 10%, since the M. charantia was over 12%. The color evolved from 1 (green) to 6 (Orange Peel) and the appearance of the fruit got worse according to the time, from 5 (excellent) to 3 (regular) in all the concentrations of the extracts of A. blanchetti and M. charantia. The A. blanchetti and M. charantia extracts did not reduce the natural incidence, however, only the A. blanchetti extract was able to decrease the severity of the anthracnose. The peroxidase enzyme presented activity in all treatments and kept the postharvest quality of the fruits in the nine days of storage.

This study evaluated the expression of insulin-like growth factor I (IGF-I), growth hormone receptor (GHR), and uncoupling protein (UCP) mRNA in muscle and liver of quails that were in thermal comfort or exposed to heat stress and that were fed diets with or without methionine supplementation. Meat quails were fed a diet that either met the nutritional demands for methionine (MS) or did not meet this demand (methionine-deficient diet, MD). The animals were either kept at a thermal comfort temperature (25°C) or exposed to heat stress (38°C for 24 h starting on the 6th day). RNA was extracted from liver and breast muscle, and cDNA was synthesized and amplified using quantitative reverse transcription-polymerase chain reaction. Animals that were fed the MS diet and remained at the thermal comfort temperature exhibited increased IGF-I mRNA expression in the liver (0.56 AU). The GHR mRNA expression in the liver and muscle was influenced by both the study variables. Animals receiving the MS diet showed higher GHR expression, while increased expression was observed in animals at the thermal comfort temperature. The UCP mRNA expression in the muscle was influenced by both methionine supplementation and heat stress. Higher expression was observed in animals that received the MD diet (2.29 vs 3.77 AU) and in animals kept in thermal comfort. Our results suggest that heat stress negatively affects the expression of growth-related genes and that methionine supplementation is necessary to appropriately maintain the levels of IGF-I, GHR, and UCP transcripts for animal metabolism.

Previously, we have shown that electroacupuncture (EA) in rats decreased eosinophil infiltration into the pulmonary tissue (PT) and in the bronchoalveolar lavage (BAL) in an experimental model of asthma. Th2 cytokines, leukotriene B4 (LTB4) and nitric oxide (NO) are involved in the asthma inflammatory process. The aim of this study was to verify the effects of EA on these asthma mediators. Male Wistar rats were divided into control (C), immobilized (I), sham acupuncture (SA), and acupuncture (A) groups. All rats were sensitized, and EA treatment using clinical acupuncture points was started 24h after antigen priming. EA was done every other day for 2weeks. Subsequently, animals were challenged by inhalation and sacrificed 24h later. At this time, the BAL and lungs were collected and used to analyze cytokine production, LTB4 and NO. The EA increased IL-1 and IFN-gamma and decreased IL-4, IL-10, NO and LTB4 in the BAL and PT compared to the C and SA groups. The presence of eosinophils in the BAL negatively correlated with IL-1 and IFN-gamma production and positively correlated with IL-4 and IL-10 production. Our results show that the beneficial anti-inflammatory action of EA on asthma is related to the balance of the Thl/Th2 response and the reduction of LTB4 and NO. These results suggest that EA therapy could be an important complementary treatment for asthma.

OBJECTIVE: To investigate the colon's development in rats subjected to protein energy malnutrition followed by supplementation with rice bran. MATERIALS AND METHODS: Weaned Wistar male rats (21 days old), weight (40-50 g) were divided into two groups: diet with 17% protein (C; control group) or an aproteic diet (A; aproteic group), for 12 days. After this, 50% of the rats from each group were sacrificed. The remaining rats were further distributed in the three groups for a recovery (21 days): control (C) continued to receive the control diet whereas the aproteic group (A) received either a control diet (AC) or a control diet supplemented with 5% of rice bran (ARB). RESULTS: The A group showed alterations in the colon and cecum, excreted dry feces mass and fecal nitrogen, compared with C rats. In the proximal colon of A rats, the external muscularis and the width of the colon wall were higher whereas in the distal colon they were lower than C. After the recovery period, the relative cecum mass, colon mass and colon length of the recovered groups (AC and ARB) were higher than in the C group. Dry feces and fecal nitrogen excreted of the rats from recovered groups were lower than C group. Colon length of the AC group was lower than in the C group. Only the crypt's depth from ARB group was higher than in the C group. CONCLUSION: Control diet supplemented with 5% rice bran, reestablished the large intestine of aproteic rats. The recovery in the ARB group was even better than in the AC rats.

The aim of the present study was to evaluate the effect of diets rich in n-6 and n-3 fatty acids on adipose tissue metabolism. Starting at weaning, male Wistar rats were fed ad libitum, for 8 weeks with one of the following diets: C, rat chow; S, rat chow containing 15 % (w/w) soyabean oil; F, rat chow containing 15 % (w/w) fish oil; SF, rat chow containing 15 % (w/w) soyabean and fish oil (5:1, w/w). Casein was added to the fat diets to achieve the same 20 % (w/w) protein content as in the control chow. Food intake and body weight were measured weekly. The rats were killed by decapitation and the retroperitoneal (RET) and epididymal (EPI) white adipose tissues were removed and weighed. Tissue lipid and protein content, in vivo lipogenesis rate, uptake of diet-derived lipids, in vitro lipolytic rate, adipocyte area, lipoprotein lipase, ATP citrate lyase, and malic enzyme activities were evaluated. Carcass lipid and protein contents were also measured. Energy intake was reduced while carcass lipid content was increased in the three fat-fed groups. However, carcass protein and body weight gains were elevated only with diets F and SF. Lipolysis rate was diminished by diets F and SF, while the uptake of diet-derived lipids was elevated by the diet S in both RET and EPI tissues. These metabolic alterations may have contributed to the increase in in vivo lipogenesis rate in the presence of decreased ATP citrate lyase and malic enzyme activities induced by the three lipid diets. These results indicate that enrichment of the diet with polyunsaturated fatty acids causes changes in adipose tissue metabolism that favour fat deposition. Different metabolic pathways were preferentially affected by each type of fatty acid used.

To examine the relationship between the type of dietary fat on liver and adipose tissue metabolism, in this study, fresh weight, protein, DNA, lipid content, and rate of lipogenesis from 3H2O of liver and retroperitoneal (RET) and epididymal (EPI) adipose tissues were assessed in rats fed polyunsaturated omega-6 (PUFA), saturated medium chain (SMC) or saturated long chain (SLC) fatty acid rich chows and control chow (CC). The results obtained indicate that fatty acid rich diets decreased liver fresh weight, protein and DNA content. However, they increased lipid content in liver and carcasses without changing the weight of RET and EPI. The rate of lipogenesis in vivo was increased by SMC and PUFA in liver and RET. These results indicate that not only the lipid content of diet but also the type of fatty acid caused metabolic alterations both in liver and white adipose tissues.
