Dynamic electrostimulation consists of the application of local or global electrostimulation together with physical exercise. This study aimed to investigate the immediate effects of a dynamic electrostimulation session on the thickness of the abdominal musculature, inter-rectus distance, heart rate, blood pressure, and body temperature, and to identify possible differences in its form of application. A total of 120 healthy participants were divided into three groups: the whole-body electrostimulation group, the local electrostimulation group, and the control group without electrical stimulation. All groups performed a single session with the same dynamic exercise protocol. Muscle thickness and inter-rectus distance were evaluated ultrasonographically using the Rehabilitative Ultrasound Imaging technique both at rest and in muscle contraction (the active straight leg raise test) to find the post-intervention differences. The results showed significant differences in immediate post-intervention heart rate, with a smaller increase in the local electrostimulation group compared to the control and whole-body electrostimulation groups. No significant differences were identified between the groups after the interventions in the rest of the variables analyzed. Therefore, a local application, with the same effects as a global application on the abdominal musculature, has fewer contraindications, which makes its use more advisable, especially in populations with cardiorespiratory disorders, for which more research is needed.

Vitamin D is an important human hormone, known primarily to be involved in the intestinal absorption of calcium and phosphate, but it is also involved in various nonskeletal processes (molecular, cellular, immune, and neuronal). One of the main health problems nowadays is the vitamin D deficiency of the human population due to lack of sun exposure, with estimates of one billion people worldwide with vitamin D deficiency, and the consequent need for clinical intervention (i.e., prescription of pharmacological vitamin D supplements). An alternative to reduce vitamin D deficiency is to produce good dietary sources of it, a scenario in which the yeast Saccharomyces cerevisiae seems to be a promising alternative. This review focuses on the potential use of yeast as a biological platform to produce vitamin D, summarizing both the biological aspects of vitamin D (synthesis, ecology and evolution, metabolism, and bioequivalence) and the work done to produce it in yeast (both for vitamin D2 and for vitamin D3 ), highlighting existing challenges and potential solutions.

Environmental stimuli attack the skin daily resulting in the generation of reactive oxygen species (ROS) and inflammation. One pathway that regulates oxidative stress in skin involves Protein Phosphatase 2A (PP2A), a phosphatase which has been previously linked to Alzheimer's Disease and aging. Oxidative stress decreases PP2A methylation in normal human dermal fibroblasts (NHDFs). Thus, we hypothesize agents that increase PP2A methylation and activity will promote skin health and combat aging. To discover novel inhibitors of PP2A demethylation activity, we screened a library of 32 natural botanical extracts. We discovered Grape Seed Extract (GSE), which has previously been reported to have several benefits for skin, to be the most potent PP2A demethylating extract. Via several fractionation and extraction steps we developed a novel grape seed extract called Activated Grape Seed Extract (AGSE), which is enriched for PP2A activating flavonoids that increase potency in preventing PP2A demethylation when compared to commercial GSE. We then determined that 1% AGSE and 1% commercial GSE exhibit distinct gene expression profiles when topically applied to a 3D human skin model. To begin to characterize AGSE's activity, we investigated its antioxidant potential and demonstrate it reduces ROS levels in NHDFs and cell-free assays equal to or better than Vitamin C and E. Moreover, AGSE shows anti-inflammatory properties, dose-dependently inhibiting UVA, UVB and chemical-induced inflammation. These results demonstrate AGSE is a novel, multi-functional extract that modulates methylation levels of PP2A and supports the hypothesis of PP2A as a master regulator for oxidative stress signaling and aging in skin.

BACKGROUND: Chia seeds have gained importance as it is the highest known plant source of omega-3 (ω3) polyunsaturated fatty acids. Specifically, chia seeds possess ω3 α-linolenic acid (ALA) and ω6 linoleic acid (LA), together known as Vitamin F, which play an important role in maintaining skin function. Protein phosphatase 2A (PP2A) is a master regulatory protein that plays a critical role in skin barrier function and its activity is modulated by natural lipids. AIMS: Obtain a chia seed extract (HYVIA™) with significant higher levels of Vitamin F, determine in vitro PP2A activity and skin hydration markers compared to other commercial chia seed extracts (CCSEs), and evaluate the potential skin hydration benefits clinically in human subjects. METHODS: A PP2A demethylation assay was utilized to assess PP2A activity. In vitro studies utilizing normal human epidermal keratinocytes (NHEKs) were treated with HYVIA™ and gene expression of hydration markers (AQP3, HAS2) were measured by quantitative PCR (qPCR). A 16-subject clinical trial was performed with 0.1% HYVIA™ formulated in a cream and applied topically to assess its skin moisturizing potential. RESULTS: We demonstrate here that HYVIA™, ALA, and LA inhibit PP2A demethylation, boosting PP2A activity, while most other CCSEs do not. Unlike other CCSEs, HYVIA™ increases keratinocyte hydration factors aquaporin-3 and hyaluronic acid synthase-2 in vitro. Clinical assessment of 0.1% HYVIA™ cream shows that HYVIA™ improves skin hydration. CONCLUSIONS: HYVIA™ is a novel chia seed extract, enriched for Vitamin F, that modulates PP2A activity and clinically improves skin hydration and barrier function.

OBJECTIVES: To describe the case of a patient with infection due to a KPC-producing Klebsiella pneumoniae (K. pneumoniae) isolate developing ceftazidime-avibactam resistance with restored carbapenem susceptibility during ceftazidime-avibactam therapy. To review the clinical/microbiological cure and survival rates using carbapenems in other similar case reports and case series. PATIENTS AND METHODS: A patient with an intra-abdominal infection due to K. pneumoniae producing the KPC-48 variant (L169P-A172T) (resistant to ceftazidime/avibactam and susceptible to carbapenems) who was treated with imipenem-cilastatin in combination with tigecycline and gentamicin. The literature was reviewed in order to summarise the in vivo (clinical/microbiological cure and survival rate) use of carbapenems in this emerging scenario. RESULTS: The patient was successfully treated with the indicated regimen. In other reported cases (mostly with pneumonia) all-cause mortality was 50% and clinical cure was 62.5%. Meropenem-vaborbactam has been successful used in an additional case. CONCLUSIONS: A carbapenem-based regimen of combination therapy seems to be an option for treating patients infected with K. pneumoniae resistant to ceftazidime/avibactam and susceptible to carbapenems, at least when the risk of mortality is low.

RESUMEN

Se realizó un estudio descriptivo y transversal de 56 pacientes con diabetes mellitus, a quienes se le diagnosticó pie diabético, pertenecientes al área de salud del Policlínico Universitario Josué País García de Santiago de Cuba, desde enero hasta diciembre del 2016, a fin de determinar el costo-beneficio del tratamiento con Heberprot-P®. La aplicación de dicho tratamiento resultó beneficiosa, pues se logró la curación de todos los pacientes. Se concluyó que, a pesar del costo elevado que representa la aplicación del novedoso tratamiento, constituyó un beneficio significativo para los afectados, puesto que se evitó la aplicación de técnicas quirúrgicas costosas y las amputaciones; además, favoreció una mejor calidad de vida, se extendió la vida socialmente útil, disminuyeron los problemas desde el punto de vista social y una alta erogación de gastos en asistencia social(AU)


A descriptive and cross-sectional study of 56 patients with diabetes mellitus to whom diabetic foot was diagnosed, belonging to the health area of Josué País García University Polyclinic in Santiago de Cuba was carried out from January to December, 2016, in order to determine the cost-benefit of the treatment with Heberprot-P®. This treatment was beneficial, because the cure of all patients was achieved. It is concluded that, in spite of the high cost that represents the administration of the novel treatment, it constituted a significant benefit for those affected, since the use of expensive surgical techniques and the amputations were avoided; also, it favored a better life quality, the socially useful life was expanded, the problems decreased from the social point of view as well as the high expenditure of expenses in social care(AU)

Taurine bulls are highly susceptible to heat stress, leading to increased oxidative stress (OS) and impaired sperm viability. Polyunsaturated fatty acids (PUFAs) supplementation can be an alternative to improve semen quality, which also results in more sperm susceptibility to lipid peroxidation. Moreover, this deleterious effect can be exacerbated in animals affected by heat stress. Vitamin E is a key antioxidant that counteracts lipid peroxidation of sperm membrane caused by OS. Thus, combining PUFAs with vitamin E may improve sperm quality. In this context, this study aimed to evaluate the effect of interaction between PUFAs and vitamin E on sperm quality in Bos taurus bulls under testicular heat stress. Sixteen taurine bulls under testicular heat stress were randomly assigned in four groups: Control, Vitamin E, PUFA, and PUFA + Vitamin E. All groups lasted for 60 days. Samples were cryopreserved/thawed and analyzed for motility variables (CASA), membrane and acrosome integrity, mitochondrial activity, susceptibility to oxidative stress, DNA integrity, and sperm-binding capacity. Results showed that vitamin E had a beneficial effect on some sperm characteristics, whereas PUFA supplementation had an adverse effect when the two treatments were evaluated separately. Finally, the association between PUFAs and vitamin E did not improve sperm quality.

Soluble forms of oligomeric beta-amyloid (Aß) are thought to play a central role in Alzheimer's disease (AD). Transgenic manipulation of methylation of the serine/threonine protein phosphatase, PP2A, was recently shown to alter the sensitivity of mice to AD-related impairments resulting from acute exposure to elevated levels of Aß. In addition, eicosanoyl-5-hydroxytryptamide (EHT), a naturally occurring component from coffee beans that modulates PP2A methylation, was shown to confer therapeutic benefits in rodent models of AD and Parkinson's disease. Here, we tested the hypothesis that EHT protects animals from the pathological effects of exposure to elevated levels of soluble oligomeric Aß. We treated mice with EHT-containing food at two different doses and assessed the sensitivity of these animals to Aß-induced behavioral and electrophysiological impairments. We found that EHT administration protected animals from Aß-induced cognitive impairments in both a radial-arm water maze and contextual fear conditioning task. We also found that both chronic and acute EHT administration prevented Aß-induced impairments in long-term potentiation. These data add to the accumulating evidence suggesting that interventions with pharmacological agents, such as EHT, that target PP2A activity may be therapeutically beneficial for AD and other neurological conditions.

Introducción y objetivos: la incidencia de enfermedad inflamatoria intestinal (EII) en España ha sido clásicamente inferior a la incidencia de los países del norte de Europa. Estudios epidemiológicos recientes muestran que la brecha está disminuyendo. Este estudio estima la incidencia de EII en Málaga, una ciudad en el sur de España, y relaciona dichas cifras con los datos de nuestro entorno. Material y métodos: este es un estudio prospectivo que fue diseñado para recoger los nuevos casos diagnosticados durante los años 2007-2008, y para realizar el seguimiento de estos pacientes. La incidencia se expresa como el número de pacientes por cada 100.000 habitantes por año. La distribución de la población del estudio colaborativo europeo se utilizó para normalizar la tasa de incidencia. Resultados: la incidencia bruta de la EII en Málaga es 9/105, la incidencia estandarizada es de 12,3/105 (9,7-15,6). Conclusiones: estos datos son similares a los de nuestro medio ambiente, aunque se observa una mayor incidencia para la enfermedad de Crohn (EC) que para la colitis ulcerosa (CU). Las características clínicas y evolución de nuestro paciente no presentan grandes diferencias con respecto a lo descrito en otras poblaciones (AU)


Background and objectives: The incidence of inflammatory bowel disease (IBD) in Spain has been traditionally lower than in Northern European countries. Recent epidemiological studies have found that these differences are diminishing. This study estimates the incidence of IBD in Málaga (Spain), a city in Southern Spain and relates its results to those found in our neighboring countries. Material and methods: This was a prospective study designed to collect new cases diagnosed during the period from 2007-2008 and follow up these patients. Incidence is expressed as number of patients per 100,000 population per year. The population distribution found in the European Collaborative Study was used to standardize incidence rates. Results: The gross incidence rate of IBD in Málaga is 9/105, the standardized incidence rate is 12.3/105 (9.7-15.6). Conclusions: These data are similar to those found in our surroundings, although a higher incidence rate for Crohn's disease (CD) as compared to ulcerative colitis (UC) was found. The clinical characteristics and outcomes of our patients do not differ significantly from those described for other populations (AU)

OBJECTIVE: In the present study we explored the psychometric properties of three widely used questionnaires to assess the subjective effects of hallucinogens: the Hallucinogen Rating Scale (HRS), the Mystical Experience Questionnaire (MEQ), and the Addiction Research Center Inventory (ARCI). METHODS: These three questionnaires were administered to a sample of 158 subjects (100 men) after taking ayahuasca, a hallucinogen whose main active component is N,N-dimethyltryptamine (DMT). A confirmatory factorial study was conducted to check the adjustment of previous data obtained via theoretical proposals. When this was not possible, we used an exploratory factor analysis without restrictions, based on tetrachoric and polychoric matrices and correlations. RESULTS: Our results sparsely match the theoretical proposals of the authors, perhaps because previous studies have not always employed psychometric methods appropriate to the data obtained. However, these data should be considered preliminary, pending larger samples to confirm or reject the proposed structures obtained. CONCLUSIONS: It is crucial that instruments of sufficiently precise measurement are utilized to make sense of the information obtained in the study of the subjective effects of psychedelic drugs. A tales efectos se analizan algunos aspectos relacionados con la introducción de la magia como expresión artística en la relación psicólogo-paciente pediátrico, lo cual favorece una mejor adherencia terapéutica y mayor motivación por el tratamiento psicológico(AU)

With the continuous scaling of resistive random access memory (RRAM) devices, in-depth understanding of the physical mechanism and the material issues, particularly by directly studying integrated cells, become more and more important to further improve the device performances. In this work, HfO2-based integrated 1-transistor-1-resistor (1T1R) RRAM devices were processed in a standard 0.25 µm complementary-metal-oxide-semiconductor (CMOS) process line, using a batch atomic layer deposition (ALD) tool, which is particularly designed for mass production. We demonstrate a systematic study on TiN/Ti/HfO2/TiN/Si RRAM devices to correlate key material factors (nano-crystallites and carbon impurities) with the filament type resistive switching (RS) behaviours. The augmentation of the nano-crystallites density in the film increases the forming voltage of devices and its variation. Carbon residues in HfO2 films turn out to be an even more significant factor strongly impacting the RS behaviour. A relatively higher deposition temperature of 300 °C dramatically reduces the residual carbon concentration, thus leading to enhanced RS performances of devices, including lower power consumption, better endurance and higher reliability. Such thorough understanding on physical mechanism of RS and the correlation between material and device performances will facilitate the realization of high density and reliable embedded RRAM devices with low power consumption.

BACKGROUND: Propionibacterium acnes is a major contributing factor to the inflammatory component of acne. The interaction of P. acnes with keratinocytes leads to an innate immune response via activation of toll-like receptors (TLR2, TLR4) resulting in the production and secretion of pro-inflammatory mediators. SIG1273, an isoprenylcysteine small molecule modulates inflammatory signaling pathways and kills P. acnes. SIG1273 represents a novel cosmetic functional ingredient that provides relief from blemishes in acne prone skin. OBJECTIVE: To assess the keratinocyte response and microbial growth of SIG1273 in vitro and evaluate the tolerability of SIG1273 gel applied topically in acne prone subjects. METHODS: For in vitro studies, human keratinocytes were exposed in culture to live P. acnes and peptidoglycan (PGN) to induce IL-8 production. P. acnes were cultured to determine minimal inhibitory concentration and minimal bactericidal concentration values. A total of 30 subjects were randomized in a double-blind controlled trial receiving 3% SIG1273 gel or vehicle for 6 weeks. Evaluation included inflammatory lesions, noninflammatory lesions, microcomedones, Sebutape scores, and P. acnes counts. RESULTS: In vitro studies demonstrate SIG1273 inhibits P. acnes-induced IL-8 production and inhibits P. acnes growth. SIG1273 gel was well tolerated with no signs of stinging, redness, or itching. Furthermore, improvement in some aspects of acne was observed in subjects applying SIG1273 gel, including inflammatory lesions, microcomedone counts and Sebutape scores. Facial scrubs taken to measure P. acnes colony-forming units showed those applying SIG1273 gel had ~1.0 Log 10 colony reduction over the length of the study, a statistically significantly improvement when compared with vehicle. No significant effects above vehicle were observed for noninflammatory lesions. CONCLUSIONS: SIG1273 represents a novel cosmetic functional ingredient that provides a safe dual modulating benefit to individuals with acne prone skin by reducing P. acnes counts and reducing inflammation.

Os ácidos graxos poli-insaturados garantem fluidez à membrana plasmática do espermatozoide. No entanto, as duplas ligações presentes, os tornam mais susceptíveis aos efeitos nocivos da peroxidação lipídica. A adição do antioxidante Glutationa reduzida (GSH) poderia conferir proteção aos espermatozoides de ovinos submetidos à congelação contra os graves danos causados pelo estresse oxidativo. O objetivo do presente experimento foi avaliar se a GSH protege os espermatozoides ovinos criopreservados contra danos causados pelo estresse oxidativo. Foram colhidos ejaculados de quatro carneiros adultos. As análises convencionais foram: concentração, motilidade, vigor e morfologia. As análises funcionais foram: integridade de membrana, integridade acrossomal, integridade de cromatina e atividade mitocondrial. O sêmen foi criopreservado utilizando o diluidor Tris-gema-critrato, suplementado com GSH (controle, 1, 5 e 10 mM). As amostras foram submetidas ao protocolo de peroxidação lipídica induzida e subsequente quantificação das substâncias reativas ao ácido tiobarbitúrico (TBARS). As análises estatísticas foram realizadas utilizando o Sistema SAS para Windows. Não houve efeito do tratamento com GSH sobre as variáveis avaliadas pelos testes convencionais. A GSH diminuiu a proporção de acrossomas íntegros. Amostras tratadas com 5mM de GSH apresentaram menor percentual de células com membranas íntegras quando comparadas às amostras controle e aquelas tratadas com 10 mM; o percentual de células sem atividade mitocondrial foi influenciado pela GSH e também não houve efeito nas TBARS. As amostras do grupo controle foram mais susceptíveis à denaturação da cromatina. Em conclusão, a adição do antioxidante Glutationa reduzida confere proteção ao DNA e à atividade mitocondrial de espermatozoides de ovinos.


The high susceptibility of sperm to the oxidative stress occurs especially due to high content of poly-unsaturated fatty acids (PUFAs) in its plasma membrane. The PUFAs provide the necessary fluidity to the plasma membrane. However double bonds present in those fatty acids are more susceptible to oxidative stress. Studies in human indicate that cryopreservation may lead to damages to the sperm due to oxidative stress. This study aimed to verify if the antioxidant glutathione (GSH) may protect ovine cryopreserved sperm against damages caused by oxidative stress. Semen samples of four rams were cryopreserved using Tris-egg yolk extender supplemented with different concentrations of reduced glutathione (control, 1, 5 and 10 mM). After thawing, samples were evaluated using conventional (motility and vigor) and functional tests (membrane integrity and mitochondrial activity). Aliquots of each thawed sample were submitted to protocol of induced lipid peroxidation using ascorbate (20 mM) and ferrous sulphate (4 mM), with further measurement of tiobarbituric acid reactive substances (TBARS), index of oxidative stress. No effect of GSH was observed on variables assessed by conventional tests. GSH decreased the proportion of intact acrosomes. Samples treated with 5 mM GSH showed lower percentage of intact membrane cells when compared to control samples and those treated with 10 mM. The percentage of cells with mitochondrial activity was affected by GSH, but no effect on TBARS. Samples from control group were more susceptible to denaturation of chromatin. In conclusion, the addition of Glutathione (GSH) offers protection to DNA and mitochondrial activity of ovine sperm.

The essential oil composition of Peumus boldus and Drimys winterii was analyzed by means of capillary GC-FID and GC-MS. More than 96% of the total oil components (43 and 54 compounds, respectively) were identified, with ascaridole (51.17 ± 9.51), p-cymene (16.31 ± 2.52) and 1,8-cineole (14.45 ± 2.99) as the main compounds in P. boldus and g-eudesmol (21.65 ± 0.41), followed of elemol (12.03 ± 0.34) and terpinen-4-ol (11.56 ± 1.06) in D. winterii. The herbicidal activity was tested against Amaranthus hybridus and Portulaca oleracea. P. boldus essential oil was the most phytotoxic against both weeds, inhibiting seed germination and seedling growth at all concentrations assayed (0.125-1 µL/mL). D. winterii essential oil did not show any effect on A. hybridus germination and only affected P. oleracea germination at the highest concentration. The results suggest the possible use of the essential oil from P. boldus as a natural herbicide.

INTRODUCTION: Epilepsy is considered a health problem. 85% of patients are satisfactorily controlled with antiepileptic drugs (AEDs) and 15% have refractory seizures. The combination of omega 3 and omega 6 fatty acids (EquazenTM) can contribute to changes in ionic currents and to stabilization of neurotransmitter's function resulting in cell membrane equilibrium. These changes lead to potential seizure control. OBJECTIVE: To compare efficacy and safety in the treatment of refractory epilepsy with a supplement combining EPA, DHA (omega 3 fatty acids) and GLA (omega 6 fatty acids). METHODS: We reviewed patients from the Neurology Department of the Children's Hospital of Mexico who have refractory epilepsy, evaluating clinical characteristics of seizures, number of seizures and AEDs. 792 mg of EPA, DHA and GLA per day (6 capsules of EquazenTM) were administrated for four weeks in order to assess the frequency of seizures as well as tolerability and probable side effects. RESULTS: The study was conducted with a total of 13 patients with follow-up four weeks after the start of supplementation. We obtained a satisfactory clinical response with ≥ 80% decrease in the daily number of seizures in more than 60% of patient. The mean of number of seizures over all patients was reduced significantly from 26.61±37.2 to 5.92 daily. In addition a significant improvement in the neurocognitive capacity was observedin all patients. CONCLUSION: The co-adjuvant supplementation with EquazenTM may result in a reduction of the number of seizures in refractory epilepsy having in addition significant impact on neurocognitive aspects. To enhance the quality of life of epileptic patient supplementation with a specific combination of EPA, DHA and GLA should be reconsidered.


INTRODUÇÃO: epilepsia é considerada um problema de saúde em que 85% dos pacientes são satisfatoriamente controlados com drogas antiepilépticas (DAE) e 15% tem crises refratárias. A combinação de ácidos graxos ômega 3 e ômega 6 (EquazenTM) pode contribui em mudanças em correntes iônicas e estabilização de neurotransmissores, resultando em equilíbrio da membrana celular. Estas mudanças levam a potencial controle de crises. OBJETIVO: comparar a eficácia e segurança suplementos combinando EPA, DHA (ácidos graxos ômega 3) e GLA (ácidos graxos ômega 6) no tratamento da epilepsia refrataria. MÉTODOS: crianças com epilepsia refratária do Departamento de Neurologia do Hospital Infantil do México tiveram avaliadas as características de suas crises, frequência e numero de DAEs. EPA, DHA e GLA (6 capsulas de Equazem - 729mg) foram administrados em um período de 4 semanas, sendo acessados tolerabilidade, efeitos colaterais e frequência de crises. RESULTADOS: 13 pacientes foram avaliados por 4 semanas após o início da suplementação. Um controle clínico satisfatório (redução de 80% ou mais das crises) foi obtido em 60% dos pacientes (26.61±37.2 crises para 5.92 crises ao dia), além de significativa melhora em sua capacidade neurocognitiva. CONCLUSÃO: a suplementação de Equazen pode determinar redução no número de crises em epilepsia refratária e determinar melhora em parâmetros neurocognitivos. O uso da combinação EPA, DHA, GLA deveria ser reconsiderado neste cenário.

Objetivo: Estudiar los resultados de aplicación de la versión española del TCI-R en una muestra de sujetos que inician tratamiento por consumo problemático de sustancias. Explorar las relaciones entre las puntuaciones del TCI-R y las escalas de trastornos de la personalidad del MCMI-II. Material y método: Se administran el TCI-R y el MCM-II a una muestra de 316 sujetos que inician tratamiento por abuso/dependencia de heroína,cocaína, alcohol o cannabis, en un centro público de tratamiento ambulatorio del Ayuntamiento de Madrid. Resultados: La versión española del TCI-R ofrece adecuadas propiedades psicométricas. Existe una amplia concordancia entre los instrumentos de Cloninger y Millon en la caracterización de los trastornos de personalidad. Bajas puntuaciones en Autodirección y Cooperatividad, junto con elevada Autotrascendencia, sugieren existencia de un trastorno de personalidad. Las diversas combinaciones de los cuatro rasgos temperamentales informan del tipo de trastorno presente. Conclusiones: La versión española del TCI-R es un instrumento fiable y válido para el estudio de los trastornos de personalidad en sujetos adictos. Ambos instrumentos, TCI-R y MCMI-II, presentan indudable utilidad para una caracterización dimensional de los trastornos del Eje II


Objective. The results of the administration of the Spanish version of the TCI-R were examined in a sample of subjects undergoing treatment for substance addiction. The relationships between the TCI-R scores and the MCMI-II personality disorders scales were explored. Material and method: The TCI-R and MCMI-ii were administered to a sample of 316 subjects who had begun treatment for heroin, cocaine, alcohol or cannabis abuse/dependency in a public outpatient treatment centre in Madrid in Spain. Results: the Spanish version of the TCI-R provides suitable psychometric properties. There is a broad concordance between the Cloninger and Millon instruments on the characterisation of personality disorders. Low scores in self-directedness and cooperativeness, together with high self-transcendence, suggest the existence of a personality disorder. The different combinations of the four temperatmental traits provide information on the specific disorder. Conclusions: the Spanish version of the TCI-R appears to be a reliable and valid instrument for the study of personality disorders in addicts. Both instruments, the TCI-R and the MCMI-II, offer undoubted utility inthe dimensional characterisation of Axis II disorders

Bacterial chemotaxis receptors are posttranslationally modified by carboxyl methylation of specific glutamate residues within their cytoplasmic domains. This highly regulated, reversible modification counterbalances the signaling effects of ligand binding and contributes to adaptation. On the basis of the crystal structure of the gamma-glutamyl methyltransferase CheR, we have postulated that positively charged residues in helix alpha2 in the N-terminal domain of the enzyme may be complementary to the negatively charged methylation region of the methyltransferase substrates, the bacterial chemotaxis receptors. Several altered CheR proteins, in which positively charged arginine or lysine residues were substituted with alanines, were constructed and assayed for their methylation activities toward wild-type receptor and a series of receptor variants containing different glutamates available for methylation. One of the CheR mutant proteins (Arg53Ala) showed significantly lower activity toward all receptor constructs, suggesting that Arg53 may play a general role in catalysis of methyl transfer. The rest of the mutant proteins exhibited different patterns of relative methylation rates toward different receptor substrates, indicating specificity, probably through interaction of CheR with the receptor at sites distal to the specific site of methylation. The findings imply complementarity between positively charged residues of the alpha2 helix of CheR and the negatively charged glutamates of the receptor. It is likely that this complementarity is involved in discriminating different methylation states of the receptors.
