RESUMEN

OBJECTIVES: To investigate the mechanism of electroacupuncture (EA) at "Neiguan" (PC6) in impro-ving myocardial electrical remodeling in rats with acute myocardial infarction (AMI) by enhancing transient outward potassium current. METHODS: A total of 30 male SD rats were randomly divided into control, model and EA groups, with 10 rats in each group. The AMI model was established by subcutaneous injection with isoprenaline (ISO, 85 mg/kg). EA was applied to left PC6 for 20 min, once daily for 5 days. Electrocardiogram (ECG) was recorded after treatment. TTC staining was used to observe myocardial necrosis. HE staining was used to observe the pathological morphology of myocardial tissue and measure the cross-sectional area of myocardium. Potassium ion-related genes in myocardial tissue were detected by RNA sequencing. The mRNA and protein expressions of Kchip2 and Kv4.2 in myocardial tissue were detected by real-time fluorescence quantitative PCR and Western blot, respectively. RESULTS: Compared with the control group, cardiomyocyte cross-sectional area in the model group was significantly increased (P<0.01), the ST segment was significantly elevated (P<0.01), and QT, QTc, QTd and QTcd were all significantly increased (P<0.05, P<0.01). After EA treatment, cardiomyocyte cross-sectional area was significantly decreased (P<0.01), the ST segment was significantly reduced (P<0.01), and the QT, QTc, QTcd and QTd were significantly decreased (P<0.01, P<0.05). RNA sequencing results showed that a total of 20 potassium ion-related genes co-expressed by the 3 groups were identified. Among them, Kchip2 expression was up-regulated most notablely in the EA group. Compared with the control group, the mRNA and protein expressions of Kchip2 and Kv4.2 in the myocardial tissue of the model group were significantly decreased (P<0.01, P<0.05), while those were increased in the EA group (P<0.01, P<0.05). CONCLUSIONS: EA may improve myocardial electrical remodeling in rats with myocardial infarction, which may be related to its functions in up-regulating the expressions of Kchip2 and Kv4.2.

RESUMEN

OBJECTIVE: The study explores the effects of electroacupuncture (EA) at the governing vessel (GV) on proteomic changes in the hippocampus of rats with cognitive impairment. METHODS: Healthy male rats were randomly divided into 3 groups: sham, model and EA. Cognitive impairment was induced by left middle cerebral artery occlusion in the model and EA groups. Rats in the EA group were treated with EA at Shenting (GV24) and Baihui (GV20) for 7 d. Neurological deficit was scored using the Longa scale, the learning and memory ability was detected using the Morris water maze (MWM) test, and the proteomic profiling in the hippocampus was analyzed using protein-labeling technology based on the isobaric tag for relative and absolute quantitation (iTRAQ). The Western blot (WB) analysis was used to detect the proteins and validate the results of iTRAQ. RESULTS: Compared with the model group, the neurological deficit score was significantly reduced, and the escape latency in the MWM test was significantly shortened, while the number of platform crossings increased in the EA group. A total of 2872 proteins were identified by iTRAQ. Differentially expressed proteins (DEPs) were identified between different groups: 92 proteins were upregulated and 103 were downregulated in the model group compared with the sham group, while 142 proteins were upregulated and 126 were downregulated in the EA group compared with the model group. Most of the DEPs were involved in oxidative phosphorylation, glycolipid metabolism and synaptic transmission. Furthermore, we also verified 4 DEPs using WB technology. Although the WB results were not exactly the same as the iTRAQ results, the expression trends of the DEPs were consistent. The upregulation of heat-shock protein ß1 (Hspb1) was the highest in the EA group compared to the model group. CONCLUSION: EA can effect proteomic changes in the hippocampus of rats with cognitive impairment. Hspb1 may be involved in the molecular mechanism by which acupuncture improves cognitive impairment.

RESUMEN

Background: Osteosarcoma (OS) is a common type of malignant bone tumor in adolescents with high risk of metastasis. However, the clinical management still remains unsatisfactory. Traditional Chinese medicine (TCM) has been widely considered as an alternative treatment, and their extracts have proved to possess great potential for drug discovery. Baicalein (BA), the active pharmaceutical ingredient of rhizoma coptidis, was proved to have anti-tumor properties in OS, but the mechanism remains poorly understood. Methods: The potential anti-cancer effects on cell growth, cell cycle, apoptosis and migration were examined in OS cells. Moreover, the lncRNA-Neighboring Enhancer of FOXA2 (lncRNA-NEF) and Wnt/ß-catenin signaling were detected by qPCR and Western blotting assays. The in vivo effect of GA on tumor growth was investigated using a xenograft mice model. Results: In the present study, BA was found to significantly suppress tumor growth in vitro and in vivo. And it was also found to inhibit the invasion and metastasis as well. As for the mechanism investigation, lncRNA-NEF was obviously upregulated by BA in OS cells, and thus induced the inactivation of Wnt/ß-catenin signaling. Moreover, lncRNA-NEF knockdown partially reversed the BA-induced anti-cancer activities; and successfully compensated the suppressive effect on Wnt/ß-catenin signaling. We therefore suggested that BA induced the inactivation of Wnt/ß-catenin signaling through promoting lncRNA-NEF expression. Conclusions: In conclude, our results demonstrated that BA suppressed tumor growth and metastasis in vitro and in vivo through an lncRNA-NEF driven Wnt/ß-catenin regulatory axis, in which lncRNA-NEF was upregulated by BA, and thus induced the inactivation of Wnt/ß-catenin signaling. The Translational potential of this article: The findings derived from this study validates the anti-cancer activity of BA in OS and provides a novel underlying mechanism, which suggest that BA may be a potential candidate to develop the effective drug for OS patients.

RESUMEN

OBJECTIVE: To evaluate the effects of interactive dynamic scalp acupuncture (IDSA), simple combination therapy (SCT), and traditional scalp acupuncture (TSA) on motor function and gait of the lower limbs in post-stroke hemiplegia patients. METHODS: A total of 231 patients with post-stroke hemiplegia was randomly divided into IDSA (78 cases), SCT (78 cases), and TSA (75 cases) groups by a random number table. Scalp acupuncture (SA) and lower-limb robot training (LLRT) were both performed in the IDSA and SCT groups. The patients in the TSA group underwent SA and did not receive LLRT. The treatment was administered once daily and 6 times weekly for 8 continuous weeks, each session lasted for 30 min. The primary outcome measures included Fugl-Meyer assessment of the lower extremity (FMA-LE), berg balance scale (BBS), modified barthel index (MBI), and 6-min walking test (6MWT). The secondary outcome measures included stride frequency (SF), stride length (SL), stride width (SW), affected side foot angle (ASFA), passive range of motion (PROM) of the affected hip (PROM-H), knee (PROM-K) and ankle (PROM-A) joints. The patients were evaluated before treatment, at 1- and 2-month treatment, and 1-, and 2-month follow-up visits, respectively. Adverse events during 2-month treatment were observed. RESULTS: Nineteen patients withdrew from the trial, with 8 in the IDSA and 5 in the SCT groups, 6 in the TSA group. The FMA-LE, BBS, 6MWT and MBI scores in the IDSA group were significantly increased after 8-week treatment and 2 follow-up visits compared with the SCT and TSA groups (P<0.05 or P<0.01). Compared with pre-treatment, the grade distribution of BBS and MBI scores in the 3 groups were significantly improved at 1, 2-month treatment and 2 follow-up visits (P<0.05 or P<0.01). The SF, PROM-H, PROM-K and PROM-A in the IDSA group was significantly increased compared with the SCT and TSA groups after 8-week of treatment (P<0.05 or P<0.01). Compared with the SCT group, ASFA of the IDSA group was significantly reduced after 8-week of treatment (P<0.05). SF, SL, PROM-K and PROM-A were significantly increased at the 2nd follow-up visit whereas the ASFA was significantly reduced in the IDSA group compared with the SCT groups at 1st follow-up visit (P<0.05 or P<0.01). The SF was significantly increased in the SCT group compared with the TSA group after 8-week treatment (P<0.05). Compared with the TSA group, PROM-K, PROM-A were significantly increased at the 2nd follow-up visit (P<0.05). CONCLUSIONS: The effects of IDSA on lower-limb motor function and walking ability of post-stroke patients were superior to SCT and TSA. The SCT was comparable to TSA treatment, and appeared to be superior in improving the motion range of the lower extremities. (Registration No. ChiCTR1900027206).

RESUMEN

OBJECTIVE: To compare the clinical effects of interactive dynamic scalp acupuncture (IDSA), simple combination therapy (SCT), and traditional scalp acupuncture (TSA) on cognitive function, depression and anxiety in patients with post-stroke cognitive impairment. METHODS: A total of 660 patients with post-stroke cognitive impairment who were admitted to 3 hospitals in Shenzhen City between May 2017 and May 2020 were recruited and randomly assigned to the IDSA (218 cases), SCT (222 cases) and TSA groups (220 cases) according to a random number table. All the patients received conventional drug therapy for cerebral stroke and exercise rehabilitation training. Scalp acupuncture and computer-based cognitive training (CBCT) were performed simultaneously in the IDSA group, but separately in the morning and in the afternoon in the SCT group. The patients in the TSA group underwent scalp acupuncture only. The course of treatment was 8 weeks. Before treatment (M0), 1 (M1) and 2 months (M2) after treatment, as well as follow-up at 1 (M3) and 2 months (M4), the cognitive function of patients was assessed using the Mini-Mental State Examination (MMSE) and Montreal Cognitive Assessment Scale (MoCA) Scales; depression, anxiety, sleep quality, and self-care ability of patients were assessed using Hamilton Depression Rating Scale (HAMD), Hamilton Anxiety Rating Scale (HAMA), Pittsburgh Sleep Quality Index (PSQI), and Modified Barthel Index (MBI), respectively. During this trial, all adverse events (AEs) were accurately recorded. RESULTS: There were no significant differences in the MMSE, MoCA, HAMD, HAMA, PSQI, and MBI scores among the 3 groups at M0 (all P>0.05). In the IDSA group, the MMSE, MoCA and MBI scores from M2 to M4 were significantly higher than those in the SCT and TSA groups, while the HAMD, HAMA and PSQI scores were significantly reduced (all P<0.01). The changes of all above scores (M2-M0, M4-M0) were significantly superior to those in the SCT and TSA groups (all P<0.01, except M4-M0 of HAMD). At M2, the severity of MMSE, HAMD, HAMA, PSQI and MBI in the IDSA group was significantly lower than that in the SCT and TSA groups (all P<0.01). There was no serious AE during this trial. CONCLUSIONS: IDSA can not only significantly improve cognitive function, but also reduce depression, anxiety, which finally improves the patient's self-care ability. The effect of IDSA was significantly better than SCT and TSA. (Trial registration No. ChiCTR1900027206).

RESUMEN

OBJECTIVE: To compare the efficacy of scalp acupuncture combined with lower-limb intelligent feedback training and lower-limb intelligent feedback training alone for lower-limb motor dysfunction after stroke. METHODS: A total of 154 patients with lower-limb motor dysfunction after stroke were randomly divided into an observation group (76 cases, 6 cases dropped off) and a control group (78 cases, 8 cases dropped off). The patients in both groups were treated with conventional medication and exercise rehabilitation training. In addition, the patients in the observation group were treated with scalp acupuncture combined with lower-limb intelligent feedback training. The scalp acupuncture was given at upper 1/5 of the anterior oblique line of parietal temporal area and upper 1/5 of the posterior oblique line of parietal temporal area. The patients in the control group were treated with lower-limb intelligent feedback training alone. All the treatment was given once a day, 6 days a week, totaling for 8 weeks. The affected-side lower-limb Brunnstrom stage and modified Ashworth scale (MAS) grade, 6-minute walk test (6MWT), Berg balance scale (BBS) score and modified Barthel index (MBI) score were evaluated before and after treatment in the two groups. The plantar pressure was measured by gait function evaluation system. RESULTS: Compared before treatment, the Brunnstrom stage in the two groups was improved after treatment (P<0.01); the MAS grade in the observation group was improved after treatment (P<0.01); the Brunnstrom stage and MAS grade in the observation group were superior to those in the control group (P<0.01, P<0.05). After treatment, the 6MWT, BBS and MBI scores in the two groups were increased (P<0.05), and those in the observation group were higher than those in the control group (P<0.05). After treatment, the touchdown area of health-side hind foot, affected-side front-hind foot and bilateral full foot in the observation group was increased (P<0.05), and the touchdown area of affected-side front-hind foot and full foot in the observation group was larger than that in the control group (P<0.05). The weight-bearing ratio of health-side forefoot and full foot in the observation group was decreased after treatment (P<0.05), and the weight-bearing ratio of affected-side forefoot, hind foot and full foot was increased after treatment (P<0.05). The weight-bearing ratio of health-side forefoot and full foot in the observation group was lower than that in the control group (P<0.05), and the weight-bearing ratio of health-side hind foot, affected-side forefoot and affected-side full foot in the observation group was higher than that in the control group (P<0.05). CONCLUSION: The scalp acupuncture combined with lower-limb intelligent feedback training could reduce the muscle tension of lower limbs, promote the separation movement mode of lower limbs, improve the plantar pressure distribution, and improve the balance ability and walking ability in stroke patients, and the curative effect is better than lower-limb intelligent feedback training alone.

RESUMEN

OBJECTIVE: To observe the effect of acupuncture plus hyperbaric oxygen (HBO) on cerebral blood perfusion in patients with delayed encephalopathy after carbon monoxide poisoning (DEACMP). METHODS: Twenty-eight patients with DEACMP were randomly divided into acupuncture group (n=14 cases) and control group (n=14 cases). Patients of the acupuncture group were treated by using "Xingnao Kaiqiao" needling technique (specific set of acupuncture points as Yintang ï¼»EX-HN3ï¼½, Shangxing ï¼»GV23ï¼½, Baihui ï¼»GV20ï¼½, Sishenchong ï¼»EX-HN1ï¼½, Fengchi ï¼»GB20ï¼½, Taichong ï¼»LR3ï¼½, etc., and strong stimulation) combined with HBO in an air pressurized tank, and those of the control group treated by simple HBO. The treatment was conducted once a day, 5 days a week for 6 weeks. All the patients underwent head routine magnetic resonance imaging (MRI) and intravoxel incoherent motion imaging(IVIM) scan before and after the treatment. The values of pseudo-diffusion coefficient D (D*) and perfusion fraction (f) of the bilateral semi-oval centers were measured by using MITK software package to calculate the ave-rage fD* value (f×D*). The Barthel index (BI) score was used to assess the patients' daily living ability, and the correlation between parameter values was evaluated by Pearson method. RESULTS: After the treatment, the values of f, fD* and BI scores were significantly increased in both acupuncture group and control groups (P<0.05). The values of the 3 indexes were significantly higher in the acupuncture group than those in the control group (P<0.05). The values of f and fD* were positively correlated with BI score (P<0.05), with the correlation coefficients being 0.822, 0.636 and 0.601, respectively between the fD* and f, BI and f, and BI and fD*. CONCLUSION: Acupuncture combined with HBO can significantly improve the low-perfusion of bilate-ral semi-oval center and daily living ability in patients with DEACMP, being significantly superior to simple HBO therapy in the curative effect.

RESUMEN

OBJECTIVE: To explore the effect of electroacupuncture (EA) on learning-memory ability and its correlation with miR-664-3p levels in the hippocampus in cerebral ischemia-reperfusion injury (CI/RI) rats. METHODS: A total of 27 male SD rats were randomly divided into sham operation, CI/RI model and EA groups (n=9 in each group). The CI/RI model was established by occlusion of the middle cerebral artery (MACO) according to the modified Longa methods. EA was applied to "Baihui" (GV20) and "Shenting" (GV24) for 20 min, once a day for 7 days. The Morris water maze tests (place navigation tasks and spatial probe trials) were used to evaluate the rats' learning-memory ability. After EA intervention, the hippocampal tissue was collected to measure the mRNA and miRNA expression profiles by using high-throughput next generation sequencing. Bioinformatics analyses including Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed to screen the most abundant differentially-expressed miRNAs and mRNAs, particularly the target genes of miR-664-3p. Their biological functional categories including the key biochemical metabolic pathways and signaling pathways involved in CI and EA effect in the left hippocampus tissue were analyzed. RESULTS: The average escape latency of place navigation tests was significantly longer in the CI/RI model group than in the sham operation group (P<0.01), and obviously shorter in the EA group than in the model group (P<0.01); and the times of escape-platform quadrant crossing of the spatial probe trials were considerably fewer in the CI/RI model group than in the sham operation group (P<0.05), and markedly more in the CI/RI EA group than in the model group (P<0.05). Combined analysis of miRNA and mRNA showed that the differentially-expressed target genes of miR-664-3p were significantly up-regulated after CI/RI relevant to the sham operation rats (P<0.05) and notably down-regulated in the EA group relevant to the CI/RI model group (P<0.05). The GO analysis displayed that the target genes of miR-664-3p were significantly enriched in the category of cell transportation activity, ion binding, and transmembrane transporter activity, the production of precursor metabolites and energy, transportation, signal transduction, localization, etc. KEGG analysis showed that the target genes of miR-664-3p in this study were mainly enriched in the thyroid hormone pathway, Hippo signaling pathway, MAPK signal transduction pathway,cAMP signaling pathway, neurotrophic factor signal transduction pathway, etc. CONCLUSION: EA of GV20 and GV24 can improve the learning-memory ability of CI/RI rats, which is closely related to its effect in down-regulating the levels of hippocampal target genes of miR-664-3p involving cellular transmembrane transportation, cell to environment interaction,cell-cell communication, etc.

RESUMEN

Acupuncture is widely used in the treatment of ischemic diseases of the central nervous system in different clinical stages and has achieved a good clinical effect. The current research showed that acupuncture can improve cerebral blood flow perfusion via increasing blood flow volume, blood flow velocity, the levels of vasomotor substances and reduction of blood viscosity and erythrocyte aggregation index. In recent years, many studies focused on the pathophysiological mechanism of acupuncture in improving cerebral ischemia via triggering the cholinergic vasodilatation, up-regulation of expression of vascular-related proteins and genes, attenuation of inflammatory reaction, etc. Currently, the methods for evaluating the effect of acupuncture are mostly noninvasive functional magnetic resonance imaging in healthy subjects. Future studies should include united selection of acupoints and acupuncture needle manipulations, more reasonable combination of different acupoints, united outcome evaluative standards, better repeatability, employment of big data, etc.

RESUMEN

OBJECTIVE: To explore the specificity of the relevant points along the governor vessel in chronic gastritis through the synchronous observation of the tenderness reaction and tenderness threshold on the back along the governor vessel in the volunteers with chronic gastritis and make the comparison with healthy volunteers. METHODS: A chronic gastritis group and a healthy volunteer group were prepared, 30 cases in each one. Using synchronous comparison, the tenderness reaction at the point inferior to each of the spinous processes from T1 to L3 along the governor vessel was observed. The mechanical pressure pain threshold was determined. RESULTS: Regarding the tenderness reaction at the point inferior to each spinous process on the back along the governor vessel, the occurrence rate was different significantly between the chronic gastritis group and the healthy volunteer group (P<0.01). The tenderness threshold at the point inferior to each spinous process from T1 to L3 along the governor vessel in the chronic gastritis group was lower than the healthy volunteer group, the tenderness thresholds from T4 to T9 and T12 were different significantly between the two groups (P<0.01, P<0.05). The tenderness reaction at the point inferior to each spinous process on the back along the governor vessel presented the significant negative correlation with the tenderness threshold in the chronic gastritis group (P<0.01), and the correlation was not displayed in the healthy volunteer group (P>0.05). CONCLUSION: The point inferior to the spoinous process of T4 on the governor vessel, Shendao (GV 11, T5) and Lingtai (GV 10, T6) are the points with the high specificity in chronic gastritis, which provides the reference to clinical treatment with acupuncture and moxibustion. The occurrence rate of high tenderness reaction and the low tenderness threshold may be the important manifestation of the point specificity on the back along the governor vessel in chronic gastritis.

RESUMEN

OBJECTIVE: To observe changes of skin temperature along the Governor Vessel (GV) after moxibustion stimulation. METHODS: A total of 36 healthy volunteer subjects were recruited in the present study. The skin temperature of Mingmen (GV 4), Dazhui (GV 14), Shenzhu (GV 12), Zhiyang (GV 9), Zhongshu (GV 7), Yaoyangguan (GV 3) regions and their bilateral control sites (1.5 cun lateral to GV 4) was detected by using a Thermo Tracer TH 9100. RESULTS: Following moxibustion stimulation of GV 4 region, a marked infrared radiant track along the central part of the back covering the GV was observed in the healthy volunteer subjects, and the skin temperature levels of both GV 4 and its bilateral control points were significantly increased (P < 0.001). The skin temperature of GV 4 was remarkably higher than those of both left and right control points (P < 0.001). After moxibustion stimulation, the average skin temperature levels of GV 9, GV 7, GV 4 and GV 3 were all up-regulated in comparison with pre-moxibustion (P < 0.01). CONCLUSION: Moxibustion stimulation of Mingmen (GV 4) can induce an infrared radiant track along the central part of the back covering the GV and raise skin temperature of acupoint areas of the GV.

RESUMEN

OBJECTIVE: To observe the effect of electroacupuncture (EA) stimulation at Zhongwan (CV 12) on the skin microcirculatory blood flow along the Conception Vessel (CV) in yin-deficiency constitution volunteers. METHODS: A total of 16 yin-deficiency constitution volunteers (9 woman and 6 men) from 200 students of Fujian University of Traditional Chinese Medicine finished a body constitution questionnaire and showed a yin-deficiency constitution. Skin microcirculatory blood perfusion units (MBPU) at Danzhong (CV 17), Xiawan (CV 10) , and Qihai (CV 6) of the CV were measured by using a Laser Doppler Flowmetry (Peri Flux 5000) in normal constitution, before and after EA stimulation of Zhongwan (CV 12) for 20 min. RESULTS: Under normal condition, the cutaneous MBPU values were significantly lower in the Qihai (CV 6) than in both Danzhong (CV 17) and Xiawan (CV 10) (P < 0.01). After EA stimulation of CV 12 for 20 min, the MBPU values were significantly decreased in the CV 17 and CV 6 acupoint regions compared with pre-stimulation (P < 0.05). No significant changes of skin MBPU were found in Xiawan (CV 10) after EA stimulation of CV 12 (P > 0.05). CONCLUSION: EA at Zhongwan (CV 12) can reduce the skin MBPU values of CV 6 and CV 17 of the Conception Vessel in yin-deficiency constitution volunteers.

RESUMEN

The Akt/mTORC1 pathway plays a central role in the activation of Warburg effect in cancer. Here, we present for the first time that halofuginone (HF) treatment inhibits colorectal cancer (CRC) growth both in vitro and in vivo through regulation of Akt/mTORC1 signaling pathway. Halofuginone treatment of human CRC cells inhibited cell proliferation, induced the generation of reactive oxygen species and apoptosis. As expected, reduced level of NADPH was also observed, at least in part due to inactivation of glucose-6-phosphate dehydrogenase in pentose phosphate pathway upon HF treatment. Given these findings, we further investigated metabolic regulation of HF through Akt/mTORC1-mediated aerobic glycolysis and found that HF downregulated Akt/mTORC1 signaling pathway. Moreover, metabolomics delineated the slower rates in both glycolytic flux and glucose-derived tricarboxylic acid cycle flux. Meanwhile, both glucose transporter GLUT1 and hexokinase-2 in glycolysis were suppressed in CRC cells upon HF treatment, to support our notion that HF regulates Akt/mTORC1 signaling pathway to dampen glucose uptake and glycolysis in CRC cells. Furthermore, HF retarded tumor growth in nude mice inoculated with HCT116 cells, showing the anticancer activity of HF through metabolic regulation of Akt/mTORC1 in CRC.

RESUMEN

OBJECTIVE: To observe the effect of different-frequency-electroacupuncture (EA) and moxibustion stimulation on rabbits' arrhythmias. METHODS: Newzealand rabbits were randomly divided into arrhythmia model (n = 18), EA-3 Hz (n = 19), EA-20 Hz (n = 19), EA-100 Hz (n = 18) and moxibustion (n = 18) groups. The arrhythmia model was induced by intravenous injection of 25 µg/kg aconitine. EA (3 Hz, 20 Hz or 100 Hz, 3.5-4 V) or moxibustion was applied to bilateral "Neiguan" (PC 6) for 20 min and electrocardiogram (ECG) of the standard limb lead II was recorded by using a Biosignal Processing Instrument. The concentration of serum hypersensitive C reactive protein (hs-CRP) content was detected by using ELISA method. RESULTS: Following EA intervention, the duration of aconitine-induced arrhythmia was significantly shorter in both EA-20 Hz and EA-100 Hz groups than in the model group (P < 0.05, P < 0.01), and the serum hs-CRP level in the EA-20 Hz group was significantly lower than that of the model group ( P < 0.05). No significant changes were found in the duration of arrhythmia in both EA-3 Hz and moxibustion groups, and in the serum hs-CRP level in the EA-3 Hz, EA-100.Hz and the moxibustion groups (P > 0. 05). CONCLUSION: EA stimulation of PC 6 at 20 Hz and 100 Hz has an anti-arrhythmic effect in aconitine-induced arrhythmia rabbits.

RESUMEN

BACKGROUND: Electroacupuncture (EA) is a traditional Chinese medicine treatment guided by meridian theory. As it gradually gains more worldwide acceptance, a clarification of its mechanisms is extremely urgent. We observed variations in transcutaneous oxygen pressure/carbon dioxide pressure (tcpO2/tcpCO2) and microcirculation blood perfusion units (BPU) along the pericardium meridian, and cardiac function during EA at Neiguan (PC6) to explore variations in energy metabolism and its relationship with visceral function adjustments during EA. METHODS: Twenty-two healthy volunteers participated in this study. Three channel laser Doppler flowmetry and tcpO2/tcpCO2 detection systems were used to detect tcpO2/tcpCO2 and microcirculation BPU along the pericardium meridian. A hemodynamic monitor was used to detect cardiac function. RESULTS: In the normal state, the microcirculatory BPU along the pericardium meridian were significantly higher than that of their bilateral corresponding control points (p < 0.05). During EA at PC6, the values of the microcirculatory BPU along the pericardium meridian did not vary, and few increased. In the normal state, the values of tcpO2 along the pericardium meridian were significantly higher than those of their bilateral corresponding control points (p < 0.05). In addition, the values of tcpCO2 along the pericardium meridian were lower than those of their bilateral corresponding control points. In comparison with the normal state, EA could decrease tcpO2 along the meridian significantly (p < 0.05) and increase tcpCO2. During EA at PC6 in healthy volunteers treated by artificial acute mild hypoxia, cardiac output and cardiac index (p < 0.05) decreased and systemic vascular resistance increased significantly (p < 0.05). CONCLUSIONS: In the normal state, the values of microcirculatory BPU and tcpO2 along the pericardium meridian were both higher than those of their bilateral corresponding control points. Energy metabolism was vigorous along the meridian. During EA, the decrease in oxygen partial pressure along the pericardium meridian might be a result of strengthened energy metabolism of associated tissue and increased oxygen consumption. The variations in energy metabolism along the pericardium meridian during the course of EA had a close relationship with visceral function adjustments. RESUMEN

OBJECTIVE: To observe the difference of the microcirculatory blood perfusion between acupoints and non-acupoints of the Stomach Meridian and its bilateral control points and to analyze the effect of electroacupuncture (EA) on subcutaneous microcirculatory perfusion in healthy volunteers. METHODS: A total of 21 healthy adult volunteers were recruited in the present study. Subcutaneous microcirculatory blood perfusion units (BPU) of Shangjuxu (ST 37), Liangqiu (ST 34), Yinshi (ST 33), Futu (ST 32) and non-acupoint (the midpoint between ST 33 and ST 32) of the Stomach Meridian and their bilateral on-meridian points were measured by using Laser Doppler Flowmetry (LDF, PeriFlux 5000). EA (0.5 Hz,duration 0.2 ms, 3-4 V) was applied to Zusanli (ST 36) for 10 min. The subcutaneous microcirculatory perfusion was detected under room temperature of (28 +/- 1) degree C and humidity of about 60% after inserting a single-use syringe needle into the subcutaneous tissues (about 1 cm in depth). RESULTS: In comparison with the bilateral control points, the subcutaneous microcirculatory BPU values of Shangjuxu (ST 37), Liangqiu (ST 34), Yinshi (ST 33), Futu (ST 32) and non-acupoint regions of the Stomach Meridian were significantly higher (P < 0.01). In comparison with pre-EA, the cutaneous microcirculatory BPU was increased mildly after EA of ST 36 (P > 0.05). Following EA for 5 to 10 min, the waveforms of LDF of the Stomach Meridian got regular and smooth. CONCLUSION: The microcirculatory blood perfusion of acupoints and non-acupoint of the Stomach Meridian in the deeper subcutaneous tissues are significantly higher than that of their bilateral corresponding control points, suggesting that subcutaneous microvascular blood flow may be the heat source for forming infrared radiation trace along the meridian in the human body observed in our past studies.

RESUMEN

OBJECTIVE: To observe the effect of electroacupuncture (EA) of different acupoints on electroretinogram (ERG) and cerebral visual evoked potentials (VEP) in healthy subjects so as to evaluate the relative specificity of the function of acupoints. METHODS: A total of 33 healthy adult volunteers were observed in the present study. The subject was asked to lie on a testing bed for a while, two disk electrodes were respectively fixed to the suborbital region and the occiput (the crossing point, 5 cm apart from the right median line of the head and 5 cm above the ear), and the reference electrode was fixed to the medial side of the earlobe for recording ERG and cerebral VEP separately. The testing room was asked to keep faint light during the whole recording process. A flash stimulation (frequency 1 Hz) was applied to the subject's eye (with a distance of about 50 cm between the light source and the tested eye, and the other eye was covered by an eyepatch). EA (0.5 Hz, 0.2 ms and 3-4 V) was applied to Guangming (GB 37), Neiguan (PC 6) and Zusanli (ST 36) for 30 min respectively in different testing days (with the interval being 3 days at least). ERG and cerebral VEP were recorded by using Polygraphy (RM-6200). RESULTS: Following EA of Guangming (GB 37), Neiguan (PC 6) and Zusanli (ST 36),the changing values (increase and decrease) of the amplitude of b waves of ERG and P100 waves of cerebral VEP were (19.68 +/- 12.61)% and (12.84 +/- 14.08)%, (6.09 +/- 14.06)% and (5.61 +/- 8.01)%, (2.39 +/- 3.52)% and (5.43 +/- 8.22)%, respectively, while those of b waves of ERG and P100 waves of cerebral VEP of GB 37 were significantly bigger than those of PC 6 and ST 36 (P < 0.01). CONCLUSION: EA of Guangming (GB 37)can significantly change the amplitude of both ERG and cerebral VEP in volunteer subjects, which is markedly bigger than those of EA of Zusanli (ST 36) and Neiguan (PC 6), suggesting a relative specificity of the effect of EA of different acupoints.

RESUMEN

OBJECTIVE: To observe the difference of the microcirculation blood perfusion volume between the Governor Vessel (GV) running course and its bilateral control points in healthy volunteers. METHODS: A total of 53 healthy volunteers were recruited in the present study. Cutaneous and muscular microvascular blood perfusion units (BPU) of Mingmen (GV 4), Jizhong (GV 6), Zhiyang (GV 9), Shenzhu (GV 12) and non-acupoint (at the 12th thoracic vertebra level) of GV and their bilateral non-meridian points (1 cm lateral to the above mentioned GV-acupoints and non-acupoint on both sides) were measured by using a Laser Doppler Flowmetry (PeriFlux 5000). RESULTS: 1) In comparison with bilateral control points, the cutaneous microvascular BPU values of Mingmen (GV 4), Jizhong (GV 6), Zhiyang (GV 9), Shenzhu (GV 12) and non-acupoint of GV Meridian were significantly higher (P < 0.01). 2) Compared with bilateral control points, BPU values of GV4, GV6, GV9, GV12 and non-acupoint of GV Meridian 1 cm, 1.5 cm and 2 cm beneath the skin were significantly higher (P < 0.05). No significant differences were found between the bilateral corresponding control points of GV4, GV6, GV9, GV12 and non-acupoint of GV in cutaneous BPU values, and between bilateral control points and GV4 in BPU values of deep (2.5 cm beneath the skin) tissues (P > 0.05). CONCLUSION: Blood flow perfusion volumes in the skin and deeper tissues along GV are significantly higher than those of their bilateral corresponding control points, indicating that cutaneous microvascular blood flow and shallower muscular blood flow may be the heat source for forming infrared radiation trace along meridian (IRRTM) in our past studies.
