RESUMEN

OBJECTIVE@#To compare the clinical effect of the combined treatment of acupuncture, moxibustion, Chinese herbal medicine and western medication and simple western medication on polycystic ovary syndrome (PCOS) of kidney deficiency and blood stagnation pattern and explore the effect on endometrial receptivity and the expression of serum homeobox gene A10 (HOXA10).@*METHODS@#A total of 60 patients with PCOS of kidney deficiency and blood stagnation pattern were randomized into a combined treatment group and a western medication group, 30 cases in each one. In the western medication group, on the fifth day of menstruation, clomiphene citrate tablets were taken orally, 50 mg each time, once daily, consecutively for 5 days. On the day when the follicle diameter was ≥ 18 mm, chorionic gonadotrophin for muscular injection, a dose of 10 000 U was given. Before sleep, the aspirin enteric-coated tablets were taken orally, 50 mg (except during menstruation). In the combined treatment group, on the base of the treatment as the western medication group, acupuncture and moxibustion were adopted and the Chinese herbal for tonifying the kidney and activating blood circulation was taken orally. The acupoints were Guanyuan (CV 4), Qihai (CV 6), Zusanli (ST 36), Sanyinjiao (SP 6), Zigong (EX-CA 1), etc. Acupuncture was remained for 30 min each time, once every two days and discontinued during menstruation. Chinese herbal was given from the 3rd day of menstruation till the onset of the next menstruation, one dose each day. After consecutive treatment for 3 menstrual cycles in the two groups, the real-time polymerase chain reaction (RT-PCR) method was adopted to determine the expression of serum HOXA10 before and after treatment in the patients of the two groups. The endometrial thickness at ovulatory phase, uterine arterial flow 7 days after ovulation [including uterine arterial pulsatility index (PI), resistance index (RI), peak systolic velocity (PSV)/end diastolic velocity (EDV), meaning S/D], pregnancy rate and the score of Chinese medicine symptoms before and after treatment were compared in the patients between the two groups.@*RESULTS@#① After treatment, the expression of serum HOXA10 was higher than that before treatment in the patients of the two groups (@*CONCLUSION@#The combined treatment with acupuncture, moxibustion and medication effectively improves endometrial receptivity and uterine arterial flow in the patients with PCOS of kidney deficiency and blood stagnation pattern and increases pregnancy rate. The therapeutic effect is better than the simple western medication and its mechanism is probably related to the regulation of serum HOXA10 expression.

RESUMEN

OBJECTIVE: To observe the protective effect of atorvastatin-induced increase of EPC-MVs on myocardial cells in ST-segment elevation myocardial infarction (STEMI) patients, and to investigate its potential mechanism. METHODS: Totally 168 STEMI patients was collected from our hospital during Feb. 2015-Feb. 2018, and then divided into group A (88 cases) and group B (94 cases) according to the dose of atorvastatin. All patients received percutaneous coronary intervention, and then given Bivaleridine for injection, Clopidogrel bisulfate tablets and Atorvastatin calcium tablets. Group A was given Atorvastatin calcium tablets 20 mg, once a day. Group B was given Atorvastatin calcium tablets 20 mg, twice a day. A treatment course lasted for 30 d, and two groups were treated for 3 courses at least. The levels of blood lipid (TC, LDL-C, HDL-C) (before treatment and 30th, 60th, 90th day after treatment) and the number of EPCs positive cells (30th, 60th day after treatment) were observed in 2 groups. The expression of miRNA of EPC-MVs (60th day after treatment) was detected, and the expression difference of miRNA were validated. Target gene and KEGG pathway enrichment of miRNA with most significant expression difference were analyzed, and the effects of it on the proliferation of cardiac HCM-a cells were evaluated. The occurrence of ADR was recorded in 2 groups. RESULTS: Totally 8 patients withdrew from the study in group A, and 6 patients in group B. There was no statistical significance in the levels of TC, LDL-C and HDL-C or the number of EPCs positive cells in peripheral blood between 2 groups before treatment or 30th day after treatment (P>0.05). After treatment, the level of HDL-C in 2 group (60th and 90th day after treatment) and the number of EPCs positive cells in peripheral blood in group B (60th day after treatment) were increased significantly, and group B was significantly higher or more than group A at the same time point (P<0.05). Microarray analysis showed that compared with group A, 16 miRNAs expressed more than 1.5 times differentially in EPC-MVs of group B, 7 of which were up-regulated and 9 down-regulated. Top five differentially expressed genes were hsa-miR-126 (up-regulated), hsa-miR-1275 (up-regulated), hsa-miR-7704 (down-regulated), hsa-miR-105-5p (down-regulated), and hsa-miR-3180 (down-regulated). Fluorescence quantitative polymerase chain reaction results showed that compared with group A, relative expression of hsa-miR-126 and hsa-miR-1275 in group B were increased significantly; and relative expression of hsa-miR-7704, hsa-miR-105-5p and hsa-miR-3108 were decreased significantly (P<0.05). The expression difference of hsa-miR-126 was the most significant, and its target genes included Ang-1, PDGF, p38 MAPK, Smad2/3, HIF-1, TGF-β, etc. The signaling pathways involved in regulation mainly included angiogenesis signaling pathway, chronic myelogenous leukemia related pathway, renal epithelial cell carcinoma related pathway and so on. CCK-8 test showed that the optical density (OD) of cells in hsa-miR-126 specific interfering substance group was decreased significantly, and the OD value of cells in simulated substance group was increased significantly, compared with blank group (P<0.05). There was no statistical significance in the incidence of ADR as diarrhea, nausea and vomiting, rash, etc. (P>0.05). CONCLUSIONS: Different doses of atorvastatin can regulate the level of HDL-C, and large dose of atorvastatin can increase the number of EPCs significantly, but dose not influence the safety of drug use. This effect may be associated with up-regulating the expression of hsa-miR-126 in EPC-MVs so as to promoting the proliferation of myocardial cells.

RESUMEN

A rapid, sensitive, and selective liquid chromatography-tandem mass spectrometry was developed for the simultaneous determination of lycorine and galanthamine, two major constituents in Lycoris radiata extract, in rat plasma. Liquid-liquid extraction with ethyl ether was carried out using diphenhydramine as the internal standard. The two bioactive alkaloids were separated on a Zorbax SB-C18 reserved-phase column (150 mm × 4.6 mm, i.d., 5 μm) by gradient elution using a mobile phase consisting of methanol with 0.1% formic acid (A) and water with 0.1% formic acid (B) at a flow rate of 0.6 mL/min. All analytes showed good linearity over a wide concentration range (r (2)>0.99) and the lower limit of quantification was 3.00 ng/mL for each analyte. The average extraction recovery of the analytes from rat plasma was more than 82.15%, and the intra-day and inter-day accuracy and precision of the assay were less than 12.6%. The validated method was successfully applied to monitoring the concentrations and pharmacokinetic studies of two Amaryllidaceous alkaloids in rat plasma after an oral administration of Lycoris radiata extract.

RESUMEN

OBJECTIVETo study the antagonistic effect of 3'-daidzein sulfonate sodium (DSS) on benign prostatic hyperplasia (BPH) and its possible mechanism.

METHODSForty healthy mice were randomly divided into five groups: a normal control group without any treatment, a model group of BPH treated by subcutaneous injection of testosterone propionate, a positive control group with the BPH procedure treated by Qianliekang, a 20 mg/(kg x d) DSS group with the BPH procedure and a 40 mg/(kg x d) DSS group with the BPH procedure. After 12 days of the above treatments, the mice were sacrificed for measurement of the prostate glandular wet weight, the index of prostate gland (PI), the morphological changes of prostate gland by light microscopy and the contents of testosterone and estradiol in the serum.

RESULTSThe prostate wet weight and PI decreased dose-dependently after DSS treatment for 12 days compared with the BPH model group (P < 0.05 or P < 0.01). The hyperplastic epithelioglandular papilla waned and even disappeared in the DSS treated groups under the light microscope, the epithelial cells became cubical or flat. The effect of DSS at 40 mg/(kg x d) was similar to that of the positive anti-BPH drug Qianliekang. DSS reduced the serum testosterone, estradiol contents and the T/E2 ratio (P < 0.05 or P < 0.01).

CONCLUSIONDSS has significant antagonistic effect on BPH induced by testosterone propionate in mice, which may involve its regulatory action on the sex hormone balance.

RESUMEN

OBJECTIVETo investigate the effect of ginkgo biloba extract (ginaton) preconditioning on discordant cardiac xenografts from guinea pig to rat, and explore its mechanism.

METHODSCervical cardiac transplantation model was established in the rats, which were divided into 4 groups Group 1 (cobra venom factor ( CVF) pretreatment, n = 10]; Group 2 (CVF + ginaton, n = 5) ; Group 3 Ccyclosporine (CsA); Group 4 (CVF + CsA + ginaton, n = 8]. The survival time and histopathology after xenograft were observed and expressions of intercellular adhesion molecule-1 (ICAM-1) heme oxygenase-1 (HO-1) CD68 and CD57 were detected.

RESULTSPathologic manifestion of grafts showed changes of acute vascular rejection (AVR) in all groups. The mean survival time after car diac xenograft was 41 hrs in Group 1, 68 hrs in Group 2, 55 hrs in Group 3 and 74 hrs in Group 4. Expression of intercellular adhesion molecule-1 (ICAM-1 ) decreased after ginaton preconditioning (P < 0. 05). CD68 and CD57 expressions were down-regulated, HO-1 expression was up-regulated, as well as the apoptotic index (Al) reduced significantly after ginaton with cyclosporine A preconditioning.

CONCLUSIONGinaton preconditioning can prolong the survival time after discordant xenograft, and significantly alleviate pathological lesion from acute xenograft vascular rejection combined with cyclosporine A.

RESUMEN

OBJECTIVETo observe the clinical effect of Yuantong Capsule (YTC) in treating vascular dementia (VD).

METHODSEighty-three patients of VD were randomized on ratio of 2:1 into two groups, the 54 patients in the treated group were treated with YTC orally administered, 3 times a day, 1 capsule in each time. The remaining 29 patients in the control group were treated with Hydergine orally, 3 times a day, 2 mg in each time. The therapeutic course for both groups was 2 months.

RESULTSThe therapeutic effect in the treated group was significantly better than that in the control group, significant difference (P < 0.05 or P < 0.01) was shown in comparison of the two groups in terms of the mini-mental state examination (MMSE) and activity of daily living (ADL) test, symptoms scoring, total effective rate, and laboratory indexes findings.

CONCLUSIONThe therapeutic effect of YTC in treating VD was obvious.

RESUMEN

OBJECTIVETo observe the effect of Kangxin capsule (KXC) on homocysteine (HCY) and beta-amyloid protein (beta-Ap) in patients with vascular dementia.

METHODSSixty-three patients with vascular dementia were randomly divided into two groups, the 33 patients in the treated group and the 30 in the control group. Basal treatment were given to both groups, besides, KXC was given to the treated group and Hydergine was given to the control group. The levels of plasma HCY, beta-Ap as well as the changes of mini mental state examination (MMSE), activity of daily living (ADL) and TCM Syndrome scoring were observed before and after treatment.

RESULTSKXC could significantly decrease the levels of HCY and beta-Ap (P < 0.01), rise the MMSE scores, reduce ADL scores and ameliorate TCM Syndrome.

CONCLUSIONKXC can effectively regulate the levels of HCY, beta-Ap in vascular dementia patients.

RESUMEN

OBJECTIVETo explore the effect of Tripterygium wilfordii polyglycoside (TWP) on level of serum interleukin 2 (IL-2), and tumor necrosis factor alpha (TNF-alpha) in patients of acute anterior uveitis (AAU).

METHODSPatients of AAU were randomly divided into two groups. The treated group (n = 50) was mainly treated with TWP and the control group (n = 50) treated with bimolani. The level of IL-2 and TNF-alpha before and after treatment were determined and compared between the two groups and also compared between the treated group and normal group consisted of 50 healthy subjects.

RESULTSLevels of IL-2 and TNF-alpha in the treated group before treatment were 1.31 +/- 0.27 micrograms/L and 1.20 +/- 0.65 micrograms/L, and after treatment 1.19 +/- 0.27 micrograms/L and 0.96 +/- 0.54 microgram/L, those in the control group were 1.31 +/- 0.26 micrograms/L and 1.22 +/- 0.66 micrograms/L before treatment and 1.20 +/- 0.27 micrograms/L and 0.98 +/- 0.51 microgram/L after treatment respectively. Comparisons of the two parameters before and after treatment in both groups showed significant difference (P < 0.05 in TWP group and P < 0.01 in bimolani group). The two parameters in both treated groups were all higher than those in the normal group before treatment (P < 0.05), but showed insignificant difference after treatment (P > 0.05).

CONCLUSIONAbnormal changes of IL-2 and TNF-alpha exist in AAU patients, TWP could suppress both parameters markedly therefore has a reliable effect in treatment of AAU.
