Peony root is an important herbal drug used as an antispasmodic analgesic. To evaluate peony roots with different botanical origins, producing areas, and post-harvest processing, 1H NMR-based metabolomics analysis was employed. Five types of monoterpenoids, including albiflorin (4), paeoniflorin (6), and sulfonated paeoniflorin (25), and six other compounds, including 1,2,3,4,6-penta-O-galloyl-ß-D-glucose (18), benzoic acid (21), gallic acid (22), and sucrose (26) were detected in the extracts of peony root samples. Among them, compounds 4, 6, 18, and total monoterpenoids including 21 were quantified by quantitative 1H NMR (qHNMR). Compound 25 was detected in 1H NMR spectra of sulfur-fumigated white peony root (WPR) extracts indicating that 1H NMR was a fast and effective method for identifying sulfur-fumigated WPR. The content of 26, the main factor affecting extract yield, increased significantly in peony root after low-temperature storage for one month, whereas that in WPR did not increase due to the boiling treatment after harvesting. We investigated the impact of preprocessing methods to such analysis for NMR data from commercial samples, resulting that the data matrix transformed from qHNMR spectra and normalized to internal standard were optimum for multivariate analysis. The multivariate analysis demonstrated that among commercial samples derived from P. lactiflora, peony root samples in Japanese market (PR) had high contents of 18 and 22, and red peony root (RPR) samples had high content of monoterpenoids represented by 6; and among RPR samples, those derived from P. veitchii showed higher contents of 18 and 22 than those from P. lactiflora. The 1H NMR-based metabolomics method coupled with qHNMR was useful for evaluation of peony root and would be applicable for other crude drugs.

Panax ginseng, as a kind of rare and valuable Chinese materia medica with the largest global trade volume, has been widely applied in many fields, such as medicine, food, health care, and production of daily chemical products. It is widely used in Asia, Europe, and America. However, its global trade and standardization present different features and an uneven development in different countries or regions. As the main country for its production and consumption, Panax ginseng in China is characterized by its large cultivation area and high total yield and is mainly sold as a raw material or primary processed product. By contrast, Panax ginseng produced in South Korea is mainly sold in manufactured products. Besides, European countries, as another consumption market of Panax ginseng, pay more attention to the research and development of its products. Although Panax ginseng has been widely recorded in various national pharmacopoeias and regional standards, the current standards of Panax ginseng differ in quantity, composition, and distribution, and the existing standards cannot be enough to meet the demands of its global trade. Based on the above issues, we systemically summarized and analyzed the status and features of Panax ginseng standardization and put forward suggestions on the development needs of international standardization of Panax ginseng to guarantee its quality and safety, regulate the order of its global trade, and resolve trade disputes, thereby promoting the high-quality development of the Panax ginseng industry.

The Lycium genus, perennial herbs of the Solanaceae family, has been an important source of medicines and nutrient supplements for thousands of years in China, where seven species and three varieties are cultivated. Among these, Lycium barbarum L. and Lycium chinense Mill., two "superfoods", together with Lycium ruthenicum Murr, have been extensively commercialized and studied for their health-related properties. The dried ripe fruits of the genus Lycium are well recognized as functional foods for the management of various ailments including waist and knee pain, tinnitus, impotence, spermatorrhea, blood deficiency and weak eyes since ancient times. Phytochemical studies have reported numerous chemical components in the Lycium genus, categorized as polysaccharides, carotenoids, polyphenols, phenolic acids, flavonoids, alkaloids and fatty acids, and its therapeutic roles in antioxidation, immunomodulation, antitumor treatment, hepatoprotection and neuroprotection have been further confirmed by modern pharmacological studies. As a multi-functional food, the quality control of Lycium fruits has also attracted attention internationally. Despite its popularity in research, limited systematic and comprehensive information has been provided on the Lycium genus. Therefore, herein, we provide an up-to-date review of the distribution, botanical features, phytochemistry, pharmacology and quality control of the Lycium genus in China, which will provide evidence for further in-depth exploration and comprehensive utilization of Lycium, especially its fruits and active ingredients in the healthcare field.

Medicinal plants or Chinese materia medica (CMM) are now attracting worldwide attention as they have increasingly prominent advantages over chemical drugs in disease treatment and healthcare. Since the 1990s, World Health Organization (WHO) and International Organization for Standardization established the Technical Committee of Traditional Chinese Medicine (ISO/TC 249) have carried out the development of quality standards on medicinal plants or CMMs respectively, and a considerable number of monographs and international standards have been published. Since the two international organizations adhere to different principles, the standards they develop naturally have different emphasis. Driven by market demand and international trade, ISO mainly takes quality, efficacy and safety into consideration when developing standards, while WHO pays more attention to clinical practice, quality control and medication guidance. Up to now, there is a lack of comparative analysis on the records, background, principles, basic content, and main requirements of quality standards on medicinal plants or CMMs respectively published by WHO and ISO. Therefore, based on international standards of CMM developed by ISO/TC 249 platform and WHO Monographs on Selected Medicinal Plants, this paper systematically compares the purposes, selected principles, standard-developing process, basic content, and main quality requirements to summarize their similarities and differences, and find their merits, aiming to serve as a reference to the development of international standards for CMMs that helps them go global.

ETHNOPHARMACOLOGICAL RELEVANCE: Pinellia ternata tuber (PTT), the dried tuber of Pinellia ternata (Thunb.) Breit., has a long history of use in traditional Chinese medicine for drying dampness, resolving phlegm, down-bearing counterflow to check vomiting and dissipating masses. Modern pharmacology studies have revealed that PTT has diverse pharmacological effects such as antitussive and expectorant, anti-emetic, anti-tumor, and anti-inflammatory effect, etc. AIM OF THE REVIEW: This review aims to provide a critical and comprehensive evaluation on ethnopharmacological uses, chemical constituents, pharmacological and toxicological effects, analytical methods and quality control of PTT, which would provide scientific evidence for exploring future therapeutic, and formulating quality and safety criteria of PTT. MATERIALS AND METHODS: Pertinent information was systematically collected from several electronic scientific databases including Web of Science, Science Direct, PubMed, Elsevier, Wiley Online Library and China national knowledge infrastructure (CNKI), as well as the classic Chinese medical books. RESULTS: PTT is reported to be widely used traditionally for the treatment of cough, vomiting, infection, and inflammatory diseases in many southeast Asian countries. Phytochemical studies have revealed the presence of a total of 233 compounds belonging to alkaloids, nucleosides, organic acids, polysaccharides, volatile oils, amino acids, proteins, starches, etc. The extracts and components of PTT have possessed diverse pharmacological activities, such as antitussive, antiemetic, antitumor, antibacterial, and sedative-hypnotic activities. Raw P. ternata tuber (RPTT) with a pungent taste causes acrid irritation of the oral and laryngopharynx mucosa when taken by mistake, while its toxicity and side effects of RPTT can be dramatically reduced with proper processing. Three kinds of processed P. ternata tuber with different processing methods are available and traded in market, as well as applied in clinical treatments. Additionally, although raw or processed PTT have been recorded in several mainstream pharmacopoeias such as Chinese Pharmacopoeia, Japanese Pharmacopoeia, and Korean Pharmacopoeia, the quality items and requirements varies a lot. Therefore, a unified international standard of raw and processed PTT is urgent need to be done. CONCLUSIONS: The ethnopharmacological, phytochemical, pharmacological and toxicological and quality evaluation of PTT were highlighted in this review, which provides potential reference information to future investigate and commercially explore for pharmaceutical applications. Nevertheless, an efficient method for chemical profiling is still unavailable to find potent bioactive markers for quality control, and then comprehensive pharmacological effects and mechanisms and toxicological evaluation of PTT require further detailed research to ensure their quality and safety.

ABSTRACT: Bottled tea beverages (BTB) are popular for the health benefits and convenience. Because chlorpyrifos (CP) is commonly used as a biomarker for exposure, as well as a pesticide in the field, it is important to determine the dynamics of CP dissipation in BTB to better perform risk assessments. This study focused on the dynamic behavior of CP for 22 days by fortifying bottled green tea, dark tea, and oolong tea beverages with the parent chemical and analyzing the degradation products. Photoinduction was used to generate the two transient intermediates: the reactive oxygen species from H2O2 and the triplet excited state of CP from the parent chemical in water were designed to observe the effects of (-)-epigallocatechin-3-gallate (EGCG) on the dissipation and transformation of CP. The results indicated that the CP degraded in BTB and the main products were detected. The half-life values of CP illustrated that EGCG increased the dissipation of CP by combination with CP and inhibited the generation of CP-oxon by scavenging the emerged oxidant, the reactive oxygen species, and interfering with the transformation of the triplet excited state of CP. This work suggests EGCG could play various roles in the dissipation and transformation of CP. Thus, a comprehensive identification of CP degradation should be performed when assessing the exposure risk in drinking BTB.

Lycium fruits have a high content of phenolics as bioactive constituents with various pharmacological effects, but there is a lack of comparative analysis and chemical profiling of phenolics in Lycium fruit varieties. An ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MSE) combined with chemometrics was developed to characterize the phenolics in fruits from four Lycium species, including Lycium barbarum L. (LBL), L. chinense Mill. (LCM), L. barbarum var. auranticarpum (LBA) and L. ruthenicun Murr. (LRM). 63 phenolics were identified according to reported tandem mass fragmentation patterns and the UNIFI scientific informatics platform. Nine phenolics (5, 18, 20, 29, 31, 37, 41, 43, 60) were common and predominant components among four types of Lycium fruit. The partial least squares discriminant analysis (PLS-DA) and the orthogonal partial least squares discriminant analysis (OPLS-DA) were analyzed on the basis of a matrix created from 653 sets of data, and 20 Lycium fruits were classified into four groups. Further analysis identified that phenolics profiles were characteristic for each Lycium species, and five markers (13, 29, 31, 35, 99) could be utilized for fruit identification. Subsequently, inhibitory activity against 5α-reductase of phenolic extracts of Lycium fruits showed that LBL extract was the relative better effective, followed by LCM, whereas LBA and LRM showed no activity, which might be associated with the high contents of marker compounds (29, 31, 35, 43, 71, 99) in LBL. These findings will provide guidance for the development of Lycium phenolics with beneficial properties for the prevention and treatment of Benign prostatic hyperplasia (BPH).

LianhuaQingwen capsule, prepared from an herbal combination, is officially recommended as treatment for COVID-19 in China. Of the serial pharmacokinetic investigations we designed to facilitate identifying LianhuaQingwen compounds that are likely to be therapeutically important, the current investigation focused on the component Glycyrrhiza uralensis roots (Gancao). Besides its function in COVID-19 treatment, Gancao is able to induce pseudoaldosteronism by inhibiting renal 11ß-HSD2. Systemic and colon-luminal exposure to Gancao compounds were characterized in volunteers receiving LianhuaQingwen and by in vitro metabolism studies. Access of Gancao compounds to 11ß-HSD2 was characterized using human/rat, in vitro transport, and plasma protein binding studies, while 11ß-HSD2 inhibition was assessed using human kidney microsomes. LianhuaQingwen contained a total of 41 Gancao constituents (0.01-8.56 µmol/day). Although glycyrrhizin (1), licorice saponin G2 (2), and liquiritin/liquiritin apioside (21/22) were the major Gancao constituents in LianhuaQingwen, their poor intestinal absorption and access to colonic microbiota resulted in significant levels of their respective deglycosylated metabolites glycyrrhetic acid (8), 24-hydroxyglycyrrhetic acid (M2D; a new Gancao metabolite), and liquiritigenin (27) in human plasma and feces after dosing. These circulating metabolites were glucuronized/sulfated in the liver and then excreted into bile. Hepatic oxidation of 8 also yielded M2D. Circulating 8 and M2D, having good membrane permeability, could access (via passive tubular reabsorption) and inhibit renal 11ß-HSD2. Collectively, 1 and 2 were metabolically activated to the pseudoaldosterogenic compounds 8 and M2D. This investigation, together with such investigations of other components, has implications for precisely defining therapeutic benefit of LianhuaQingwen and conditions for its safe use.

Chinese materia medica (CMM) is indispensable component of Traditional Chinese Medicine (TCM) therapy. With the widespread of TCM around the world, the quality control and safe use of CMM become a major concern. This paper introduces the role of ISO standards for industrial development and current development status of CMM standards in ISO/TC 249. Through the comparison of similarities and differences between CMM standards in ISO/TC 249 and pharmacopoeias of main stakeholders, this paper suggests strengthening standard formulation in the following areas to provide more appropriate documents to facilitate the international trade and promote the industrial development of CMM: (1) Develop standards to fill the blanks among the whole industry chain of CMM; (2) Develop standards for new forms of CMM and services; (3) Develop specification and grade standards for CMM with large quantity and high value.

Numerous natural preparations in traditional Chinese medicine are prepared as decoctions. Processing factors (PFs) comparing the levels of pesticide residues in decoctions to those in the corresponding unprocessed products should be considered in exposure assessments. Thus, this study determined the residue levels of six pesticides (chlorpyrifos, phoxim, imidacloprid, thiamethoxam, fenpropathrin, and emamectin benzoate), as well as 3,5,6-trichloropyridinol, the primary metabolite of chlorpyrifos, and clothianidin, the main metabolite of thiamethoxam in Baishao, Paeoniae radix lactiflora (Fam. Ranunculaceae). The results showed that significant time-response effects were present for the release of pesticides from P. radix. The PFs calculated were < 1, indicating a significant reduction in pesticide residues after TCM processing. The water solubility and partition coefficient values of the pesticides may have played a basic role in the dissipation of the residues during the TCM decocting process. A risk assessment based on the hazard quotient with PFs revealed that exposure to pesticide residues in P. radix was far below the levels that might pose a health risk. In conclusion, the results presented here are of theoretical and practical value for the safety evaluation of TCMs.

Huangqin Decoction(HQD) is a classic prescription for treating dysentery in the Treatise on Cold Damage and now is mainly used for the treatment of ulcerative colitis(UC). Since there are no requirements on specific Paeonia species, both Paeoniae Radix Alba(white peony root, WPR) and Paeoniae Radix Rubra(red peony root, RPR) are clinically used in HQD now. Although the two types of peony roots are close in origin and similar in primary components, the medicinal properties and efficacies are different. Furthermore, the systematic comparative analysis on the efficacy differences in treating UC of HQD with the roots of multi-originated peony has been seldom reported. This study compared and evaluated the pharmacological effects of HQD prepared from the roots of multi-originated peony, including WPR, RPR-l(derived from P. lactiflora), and RPR-v(derived from P. veitchii) based on the mouse model of UC induced by dextran sodium sulfate(DSS) by animal behaviors, pathological section(colon), and cytokine expression(IL-1ß and IL-6), aiming to provide evidence for the identification of the original resource of peony root in HQD. The results indicated that all HQD samples prepared from WPR, RPR-l, and RPR-v could improve the symptoms of UC. Compared with the HQD-WPR, HQD-RPR-l and HQD-RPR-v were significantly different in weight loss, colon length, and disease activity index(DAI) score, but there was no significant difference between HQD-RPR-l and HQD-RPR-v. Moreover, HQD-RPR-v exhibited the most significant improvement in the pathological morphology of colonic tissue and mucosal defects. According to the previous comparative analysis of chemical profiling and content distribution of HQD prepared from the roots of multi-originated peony, RPR-v in HQD was potent in protecting against UC, which was presumedly attributed to a large number of monoterpene glycosides and galloyl glucoses. This study provided a scientific basis for the determination of peony root in HQD and its clinical medication.

PURPOSE: Traditional Chinese medicine (TCM) has fully engaged and played an essential role in the prevention and treatment of Coronavirus Disease 2019 (COVID-19). This study compares relevant standards on high-frequent Chinese Materia Medicia (CMM) used in this pandemic aiming at reaching a global consensus and ensuring the use of Chinese medicines safely. METHODS: 141 representative Chinese formulas and Chinese Patent Medicines from the National Protocol and the most of Provincial Protocols for controlling COVID-19 in China have been collected to statistical analyze the composition and characteristics of CMM. Among them, the domestic and international standards of 47 varieties with the frequency usage over 10 times were selected to compare their quality requirements in the mainstream pharmacopoeias and international standards. RESULTS: The quality requirements of used CMM for fighting COVID-19 on the terms of overall quality control, marker compounds, and safety indicators showed different patterns in these mainstream pharmacopoeias and international standards. The uniformed and scientific quality standards of CMM were urgently needed to promote global acceptation and trade. CONCLUSIONS: These findings will provide evidence for building unified quality and safety standards that can adapt to the characteristics of CMM and promote international trade, and also will be stated that it is of the highest priority for ISO/TC 249 to formulate high-quality standards that consolidate international consensus to ensure quality and safety of the urgently needed CMM.

Clarification of the quality and biological effect equivalence of traditional Chinese medicines containing multi-origin species is essential to improve their current quality standards, and also is the core problem to clarify the origins of single herbs with multi-species in Chinese formulas that will guarantee their clinical application. Huangqin decoction is the typical one of multi-origin formulas frequently used in traditional Chinese medicine and Kampo medicine. An ultra high performance liquid chromatography with electrospray ionization-tandem mass spectrometry was developed for chemical profiling and marker quantification of Huangqin decoction prepared with two different original types of peony root, white and red peony root. Forty-seven main peaks in chemical profiling of Huangqin decoction prepared with white and red peony root were identified: nine were from peony root, 20 from baical skullcap root, 17 from licorice root, and one from jujubae fruit. The markers characteristics of the respective types of peony root in Huangqin decoction differ from that in single herbs, especially in terms of monoterpenoids and hydrolysable tannins. Subsequently, 17 representative markers in Huangqin decoction prepared with three types of peony root and their chemical characteristics and content distribution were carried out.

A simple, specific and selective quantitative analysis of multi-components by single marker(QAMS) method for simultaneous determination of anthraquinones and anthraquinone glycosides in Polygonum multiflorum was developed. Four main anthraquinones and its glycosides, emodin, emodin-8-O-ß-D-glucoside, physcion and physcion-8-O-ß-D-glucoside were selected as analytes to evaluate the quality of P. multiflorum. Emodin was used as the internal standard, and the relative correction factors(RCFs) between emodin and the other three anthraquinones were calculated. Comparison of the contents of the four components in 30 batches of P. multiflorum from different regions and 12 batches decoction pieces from different manufacturers by QAMS and external standard method(ESM) showed that there was no significant difference between QAMS and ESM for quantification of the four main components by using relative error results, and the QAMS method was accurate and reliable, and had a good repeatability. In addition, compared with the results calculated by the difference method between total anthraquinone and free anthraquinone in the content determination of P. multiflorum in Chinese Pharmacopoeia, the results of direct determination combined anthraquinone by QAMS were very close to that by measured the external standard method. Therefore, simultaneous quantification of four main anthraquinones by using QAMS is suitable to evaluate the quality of P. multiflorum. Then the optimized assay method of the combined anthraquinone contents showed simple and feasible, which could be replaced and improved the quantification method of the combined anthraquinone in the current Chinese Pharmacopeia.

Endotoxic shock exhibits a considerably high mortality risk. It is defined as a systemic inflammatory response syndrome caused by a microbial infection. Radix Isatidis has anti-inflammatory, antiviral, and antipyretic effects and is used worldwide. This study investigated the antiendotoxin sepsis effects of an aqueous R. Isatidis extract (RIE) and explored the possible pharmacological molecular mechanisms. Male C57BL/6J mice were intravenously injected with 15 mg/kg lipopolysaccharide (LPS) to induce endotoxic shock. The results demonstrated that the survival rate of mice pretreated with RIE increased, and LPS-induced liver and lung damage were reduced by inhibiting inflammation. For elucidating detailed molecular mechanisms, we focused on LPS-induced transcription factors: nuclear factor-κB (NF-κB) and interferon regulatory factor 3 (IRF3). Our results demonstrated that the protective effects of RIE were strongly dependent on IRF3-induced interferon-ß, not on NF-κB-induced tumor necrosis factor-α and interleukin-1ß. In addition, RIE suppressed the phosphorylation of IRF3, not NF-κB. In conclusion, this study revealed the antiendotoxic properties of RIE on LPS-induced sepsis and provided mechanistic evidence for the beneficial effects of RIE.

Five pairs of alkaloid enantiomers (1a/1b-5a/5b) were obtained from Isatis indigotica (I. indigotica) roots. Among them, 1a/1b, 2a/2b and 3a/3b were determined as three pairs of new alkaloid enantiomers. Their structures were elucidated by physicochemical properties and spectroscopic methods. The absolute configurations were deduced by comparison of their experimental circular dichroism (CD) and calculated electronic circular dichroism (ECD) spectra, as well as by single-crystal X-ray crystallography using anomalous scattering of Cu Kα radiation. Alkaloids 1a and 1b possess an unpresented carbon skeleton and their putative biosynthetic pathways are discussed. Moreover, all of the alkaloids were tested for their nitric oxide (NO) inhibitory effects in RAW 264.7 cells, and 4a and 4b showed inhibitory effects with IC50 values of 76.97 µM and 65.88 µM, respectively.

Seventeen lignans (1-17) were obtained from the roots of Isatis indigotica (I. indigotica). Among them, isatindigosesquilignans A-C (1-3) were deduced as three undescribed sesquilignans, which possess unusual carbon skeleton of aryltetralin unit connected with a C6-C3 moiety via a five-membered ring of C-3-C-8″-C-7″-O-C-4. Isatindigosesquilignans B and C were determined as the first examples of its glycosides from a natural source and a plausible biosynthetic pathway was proposed. Moreover, all of the isolated lignans were assayed regarding their inhibitory effects on nitric oxide (NO) production in RAW 264.7 cells and compounds 1, 2 and 7 showed inhibitory effects with IC50 values ranging from 19.46 µM to 64.82 µM.

BACKGROUND: Botanical pesticides increasingly play important roles in the control of agricultural pests. In this study, the aphidicidal effect of moso bamboo (Phyllostachys pubescens) extract against mustard aphid was confirmed, the main active compounds identified, and aphidicidal mechanism of the most active compound established. RESULTS: When the treatment concentration was 10.0 g L-1 , the corrected mortality of bamboo leaf extract (BE) was 53.22 ± 5.20% and the petroleum ether component of bamboo leaf extract (PE) reached 82.76 ± 4.50%, which also showed a synergistic effect with imidacloprid. Four flavonoids were identified as the main active components in the BE via activity tracking and phytochemical method. Isoorientin had an LC50 of 313.22 mg L-1 , and affected the activities of acetylcholinesterase and peroxidase significantly, revealing the possible aphidicidal mechanism. When the treatment of 11.1% PE·imidacloprid was 200 mL, the control effect was 99.07%, which was better than that observed with 10% of imidacloprid or 0.5% of matrine. CONCLUSIONS: These data provide a better understanding of the aphidicidal activity and aphidicidal mechanism of moso bamboo leaf extract and the most active compound, isoorientin. This will help in developing a more effective botanical aphicide. © 2019 Society of Chemical Industry.

The safety of Traditional Chinese Medicine (TCM) is of concern worldwide. Herein, Paeoniae Radix Alba, Chaenomelis Fructus and Moutan Cortex, representing three medicinal components, were subjected to toxicological analysis to investigate possible pesticide contamination. Exposure using a point estimate model identified 47 residues that were simultaneously validated by the QuEChERS-UPLC-MS/MS method, which is sufficiently reliable for measuring residue concentrations. Of the 313 samples tested, 94.57% contained pesticide residues, with concentrations ranging from 0.10 to 1199.84 µg kg-1, of which >83.17% contained 4-15 different residues. Carbendazim was the most frequently detected pesticide (>85%), and procymidone, pendimethalin and phoxim were also abundant (median concentration = 15.33-623.12 µg kg-1). Risk assessment based on the hazard quotient/hazard index (HQ/HI) approach revealed that exposure to pesticide residues in all three TCMs (95th percentile) were far below levels that might pose a health risk. However, insecticides contributed to cumulative exposure, especially phoxim, and worryingly, several banned pesticides were detected. The results are of theoretical and practical value for evaluating the safety TCMs, and could improve their quality and safety.
