RESUMEN

An experiment on the use of farmyard manure and biofertilizer along with application of chemical phosphorus was conducted to assess the impact of differential doses of phosphorus, farmyard manure and consortium biofertilizer application on the development, yield and phosphorus uptake during the year 2018 and 2019. The impact of different treatments was recorded on the plant height, dry matter partition, yield and yield attributes, phosphorus uptake and soil phosphorus availability using standard methods. The data revealed significant improvement in yield, yield attributes, phosphorus uptake and soil phosphorus availability. The integration of farmyard manure and biofertilizer with 60 kg ha-1 SSP (single superphosphate) has improved the black gram yield by 7.4% and 3.28% respectively over the use of 60 SSP alone. The phosphorus uptake in black gram with application of Farm yard manure and biofertilizer along with 60 kg ha-1 SSP has improved the uptake by 7.18% and 2.51% respectively over the use of 60 kg ha-1 SSP alone. The results highlight the need of integrated application of farm yard manure, biofertilizer for sustainable production of black gram in the region.

Cadmium (Cd) metal extraction through efficient plant roots has attracted much attention as this methodology is environment-friendly and cost-effective. Brassica species are well known for their tolerance towards high Cd concentration in contaminated soils. The tolerance ability may vary among species; hence the assessment of this variability is mandatory for selecting Brassica species. For this purpose, a greenhouse pot experiment was carried out using three Brassica species (Brassica juncea L., Brassica campestris L., and Brassica napus L.). To evaluate the effect of chelating agent ethylenediamine tetraacetic acid (EDTA) on Cd uptake, EDTA (0, 1, and 2 g kg-1 soil) was supplemented along with Cd (0, 5, 10, 20, 40, and 80 mg kg-1 soil). Among different species, B. juncea possessed the highest root dry biomass and lowest root Cd concentration in untreated soil. Overall root dry biomass of all tested Brassica species reduced on increasing Cd and EDTA levels. The trend was appeared to be related to an increase in root Cd concentration on the supplementation of EDTA that formed a complex with the target metal contaminate and resulted in vacuolar sequestration. Roots of B. juncea showed maximum Cd accumulation and highest values at Cd and EDTA levels up to 20 mg kg-1 and 1 g kg-1 soil due to the combined effect of root biomass and Cd concentration in roots. Thus, present findings inferred that Cd and EDTA supplementation might prove as a feasible strategy to improve remediation of Cd-polluted soil using B. juncea as an efficient Cd accumulator.

RESUMEN

Three triblock copolymers based on the poly(lactide) or poly(lactide-co-glycolide) and poly(ethylene glycol) or poly(ethylene oxide) blocks were synthesized and characterized. The weight average molecular weight and number average molecular weight were determined by gel permeation chromatography and proton nuclear magnetic resonance spectroscopy, respectively. Fourier transform infrared spectroscopy was used to determine the completion of synthesis of polymers. Thermoreversible sol-gel transition temperature and concentration were determined by an inverted tube method. Two formulations each of three synthesized polymers containing 5% (w/v) of lysozyme or bromelain but differing in polymer concentrations (20-30%, w/v) were prepared and studied for in vitro release of the incorporated protein. In vitro biocompatibility of the delivery systems was studied by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) cell viability assay. Biological activities of lysozyme and bromelain were determined by enzyme activity assays. Critical gelling concentrations were found in the range of 20-30% (w/v). In vitro biocompatibility study showed that all the formulations were biocompatible. Increasing the polymer concentration led to a decrease in burst release and extended the in vitro release of proteins. Furthermore, biological activities of lysozyme and bromelain in released samples were found to be significantly (p<0.05) greater in comparison to the control. Thus, the above thermosensitive polymers were able to deliver proteins in biologically active forms at a controlled rate for 2-8 weeks.

The effect of chemical enhancers (e.g., fatty acids, limonene, depilatory lotions) and iontophoresis was investigated on the in vitro permeability of leuprolide acetate through porcine epidermis. Franz diffusion cells and Scepter iontophoretic power source were used for the percutaneous absorption studies. Anodal iontophoresis was performed at 0.2 mA/cm2 current density. Fatty acids used were palmitic (C16:0), palmitoleic (C16:1), stearic (C18:0), oleic (C18:1), linoleic (C18:2), and linolenic (C18:3) acids. The passive and iontophoretic flux were significantly (p < 0.05) greater through fatty acids-treated porcine epidermis in comparison to the control (untreated epidermis) for leuprolide acetate. The passive and iontophoretic permeability of leuprolide acetate increased with increasing number of cis double bonds. Among the fatty acids tested, linolenic acid (C18:3) exhibited the maximum permeability of leuprolide acetate during passive (51.42 x 10(-4) cm/hr) and iontophoretic (318.98 x 10(-4) cm/hr) transport. The passive and iontophoretic flux of leuprolide acetate were significantly (p < 0.05) greater through the limonene and depilatory lotion treated epidermis in comparison to their respective control. In conclusion, iontophoresis in combination with chemical enhancers synergistically increased (p < 0.05) the in vitro permeability of leuprolide acetate through porcine epidermis.

Using lysozyme as a model protein, this study investigated protein stability, protein--polymer interaction in different release media and their influence on protein release profile and in vitro--in vivo correlation. Lysozyme was microencapsulated into PLGA 50:50 by a double emulsion--solvent extraction/evaporation method. Protein stability, protein--PLGA adsorption and protein in vitro release were studied in various test media. Differential scanning calorimetry analysis showed lysozyme to be most conformationally stable in pH 4.0 acetate buffer with highest T(m) at 77.2 degree C and DeltaH(cal) 83.1 kcal/mol. Lysozyme exhibited good stability in pH 2.5 glycine buffer with T(m) at 63.8 degree C and DeltaH(cal) 69.9 kcal/mol. In pH 7.4 phosphate-buffered saline (PBS), lysozyme showed a trend toward aggregation when the temperature was elevated. When PLGA polymer was incubated with lysozyme in the various buffers, adsorption was found to occur in PBS only. The adsorption severely limited the amount of lysozyme available for release from microspheres, resulting in slow and incomplete release in PBS. In contrast, the release of the microspheres in acetate and glycine buffers was complete within 40 and 70 days, respectively. Radiolabeled lysozyme blood levels in rats from the microspheres correlated qualitatively well with in vitro release in glycine buffer as a release medium. This study suggests that protein stability and adsorption are critical factors controlling protein release kinetics and in vitro--in vivo correlation of PLGA microspheres.
