RESUMEN

The natural product Honokiol exhibits robust antitumor activity against a range of cancers, and it has also received approval to undergo phase I clinical trial testing. We confrmed that honokiol can promote the apoptotic death of tumor cells through cell experiments. Then siRNA constructs specific for PIAS3, PIAS3 overexpression plasmid and the mutation of the STAT3 Tyr705 residue were used to confirm the mechanism of Honokiol-induced apoptosis. Finally, we confrmed that honokiol can promote PIAS3 upregulation, in turn suppressing STAT3 Tyr705 phosphorylation through the in vivo and in vitro experiments. Honokiol was ultimately found to reduce tumor cell viability by promoting apoptosis through a mechanism dependent on the ability of Honokiol to promote PIAS3 upregulation and the selective inhibition of p-STAT3 (Tyr705) without affecting p-STAT3 (Ser727) or p-STAT1 (Tyr701) levels. PIAS3 knockdown and overexpression in tumor cells altered STAT3 activation and associated DNA binding activity through the control of Tyr705 phosphorylation via PIAS3-STAT3 complex formation, ultimately shaping Honokiol-induced tumor cell apoptosis. Honokiol was also confirmed to significantly prolong the survival of mice bearing xenograft tumors in a PIAS3-dependent fashion. Together, these findings highlight a novel pathway through which Honokiol can promote PIAS3 upregulation, in turn suppressing STAT3 Tyr705 phosphorylation and promoting the apoptotic death of tumor cells.

Phycoerythrin (PE) is a natural protein-pigment complex with a strong pink color, but it is sensitive to thermal and light variations. In this study, PE was extracted from Porphyra haitanensis in a yield of 0.2% (w/w). The phycoerythrin hydrolysates (PEH) (3-10 kDa) were prepared by enzymatic hydrolysis of PE with bromelain (8000 U/g) at 47 °C for 30 min, with a degree of hydrolysis (DH) of 11.57 ± 0.39% and a color degradation rate of 7.98 ± 0.39%. The physicochemical properties of PEH were evaluated. The UV and fluorescence spectra indicated that bromelain changed the microenvironment around phycoerythrobilin (PEB). The infrared spectrum revealed that the bromelain hydrolysis increased the α-helix content of PEH. The scanning electron microscope showed that bromelain destroyed the dense and smooth structure of PE, resulting in irregular porous structures. The radical scavenging activities of DPPH and ABTS of PEH were increased relative to that of PE (p < 0.05). The thermal (50-80 °C)-, UV (0.5-3 h)-, visible light irradiation (2-8 h)-, and metal ion exposing stabilities of PEH were significantly improved (p < 0.05). This study provides a potential scheme for overcoming the sensitivity of PE to thermal and light variations and facilitates PEH as a natural colorant ingredient in food and pigment applications.

Four Chinese herbs from the Citrus genus, namely Aurantii Fructus Immaturus (Zhishi), Aurantii Fructus (Zhiqiao), Citri Reticulatae Pericarpium Viride (Qingpi) and Citri Reticulatae Pericarpium (Chenpi), are widely used for treating various cardiovascular and gastrointestinal diseases. Many ingredients have already been identified from these herbs, and their various bioactivities provide some interpretations for the pharmacological functions of these herbs. However, the complex functions of these herbs imply undisclosed cholinergic activity. To discover some ingredients with cholinergic activity and further clarify possible reasons for the complex pharmacological functions presented by these herbs, depending on the extended structure-activity relationships of cholinergic and anti-cholinergic agents, a simple method was established here for quickly discovering possible choline analogs using a specific TLC method, and then stachydrine and choline were first identified from these Citrus herb decoctions based on their NMR and HRMS data. After this, two TLC scanning (TLCS) methods were first established for the quantitative analyses of stachydrine and choline, and the contents of the two ingredients and synephrine in 39 samples were determined using the valid TLCS and HPLC methods, respectively. The results showed that the contents of stachydrine (3.04‱) were 2.4 times greater than those of synephrine (1.25‱) in Zhiqiao and about one-third to two-thirds of those of Zhishi, Qingpi and Chenpi. Simultaneously, the contents of stachydrine, choline and synephrine in these herbs present similar decreasing trends with the delay of harvest time; e.g., those of stachydrine decrease from 5.16‱ (Zhishi) to 3.04‱ (Zhike) and from 1.98‱ (Qingpi) to 1.68‱ (Chenpi). Differently, the contents of synephrine decrease the fastest, while those of stachydrine decrease the slowest. Based on these results, compared with the pharmacological activities and pharmacokinetics reported for stachydrine and synephrine, it is indicated that stachydrine can be considered as a bioactive equilibrist for synephrine, especially in the cardio-cerebrovascular protection from these citrus herbs. Additionally, the results confirmed that stachydrine plays an important role in the pharmacological functions of these citrus herbs, especially in dual-directionally regulating the uterus, and in various beneficial effects on the cardio-cerebrovascular system, kidneys and liver.

Maize (Zea mays L.) is one of the most important food crops in the world. Drought is currently the most important abiotic factor affecting maize yield. L-arginine has emerged as a nontoxic plant growth regulator that enhances the tolerance of plants to drought. An experiment was conducted to examine the role of L-arginine in alleviating the inhibitory effects of drought on the photosynthetic capacity and activities of antioxidant enzymes when the plants were subjected to drought stress. The results showed that the biomass of maize seedlings decreased significantly under a 20% polyethylene glycol-simulated water deficit compared with the control treatment. However, the exogenous application of L-arginine alleviated the inhibition of maize growth induced by drought stress. Further analysis of the photosynthetic parameters showed that L-arginine partially restored the chloroplasts' structure under drought stress and increased the contents of chlorophyll, the performance index on an adsorption basis, and Fv/Fm by 151.3%, 105.5%, and 37.1%, respectively. Supplementation with L-arginine also reduced the oxidative damage caused by hydrogen peroxide, malondialdehyde, and superoxide ions by 27.2%, 10.0%, and 31.9%, respectively. Accordingly, the activities of ascorbate peroxidase, catalase, glutathione S-transferase, glutathione reductase, peroxidase, and superoxide dismutase increased by 11.6%, 108.5%, 104.4%, 181.1%, 18.3%, and 46.1%, respectively, under drought. Thus, these findings suggest that L-arginine can improve the drought resistance of maize seedlings by upregulating their rate of photosynthesis and their antioxidant capacity.

The Ganoderma species in Polyporales are ecologically and economically relevant wood decayers used in traditional medicine, but their genomic traits are still poorly documented. In the present study, we carried out a phylogenomic and comparative genomic analyses to better understand the genetic blueprint of this fungal lineage. We investigated seven Ganoderma genomes, including three new genomes, G. australe, G. leucocontextum, and G. lingzhi. The size of the newly sequenced genomes ranged from 60.34 to 84.27 Mb and they encoded 15,007 to 20,460 genes. A total of 58 species, including 40 white-rot fungi, 11 brown-rot fungi, four ectomycorrhizal fungi, one endophyte fungus, and two pathogens in Basidiomycota, were used for phylogenomic analyses based on 143 single-copy genes. It confirmed that Ganoderma species belong to the core polyporoid clade. Comparing to the other selected species, the genomes of the Ganoderma species encoded a larger set of genes involved in terpene metabolism and coding for secreted proteins (CAZymes, lipases, proteases and SSPs). Of note, G. australe has the largest genome size with no obvious genome wide duplication, but showed transposable elements (TEs) expansion and the largest set of terpene gene clusters, suggesting a high ability to produce terpenoids for medicinal treatment. G. australe also encoded the largest set of proteins containing domains for cytochrome P450s, heterokaryon incompatibility and major facilitator families. Besides, the size of G. australe secretome is the largest, including CAZymes (AA9, GH18, A01A), proteases G01, and lipases GGGX, which may enhance the catabolism of cell wall carbohydrates, proteins, and fats during hosts colonization. The current genomic resource will be used to develop further biotechnology and medicinal applications, together with ecological studies of the Ganoderma species.

Salts reduction/substitution have gained a lot interest from food industry since the US Food and Drug Administration (FDA) has issued a draft guidance for salt reduction. However how changes of salts in food formulation could influence lipid oxidation is still not fully understood. Using oil-in-water emulsions stabilized by a natural emulsifier - lecithin at pH 7.0 as a model system, this study evaluated how salts affect the physical parameters of the emulsion, the chelating activity of lecithin and thus the lipid oxidation of these emulsions. Results showed that salts increased the particle size, the negative charge of the oil droplets, and the amount of iron chelated by lecithin. Lipid oxidation lag phases were shortened by addition of salts, by 1 day and 2 days for lipid hydroperoxides and thiobarbituric acid reactive substances measurements respectively. These results provide some new insights on the mechanisms of how salts could affect the lipid oxidation of food emulsions.

A total of 14 types of sludge from household sewage, mixture of domestic and industrial wastewater, and industrial and oil wastewater treatment plants were selected to evaluate the effectiveness and adaptability of thermal hydrolysis pre-treatment. Organic solubilization, dewatering improvement, volume reduction, high-strength filtrate biodegradation, and dewatered sludge incineration were investigated using bench and pilot thermal hydrolysis experiments (170 °C/60 min). Results showed that sludge types significantly affected the treatment effects. Organic content has a primary influence on thermal effects. The relationship between suspended solid (SS) solubilization and raw sludge organic content was linear with an R(2) of 0.73. The relationship between raw sludge organic content and treated sludge dewatering was linear with an R(2) of 0.86 and 0.65 for pilot and bench pre-treatments, respectively. Household and oil sludge possessed incineration possibilities with high heat value. Industrial and oil sludge filtrate was unsuitable for digestion to recover bioenergy.

The contributions of various plasma species to the removal of low-concentration formaldehyde (HCHO) in air by DC corona discharge plasma in the presence and absence of downstream MnO(x)/Al(2)O(3) catalyst were systematically investigated in this study. Experimental results show that HCHO can be removed not only by short-living active species in the discharge zone, but also by long-living species except O(3) downstream the plasma reactor. O(3) on its own is incapable of removing HCHO in the gas phase but when combined with the MnO(x)/Al(2)O(3) catalyst, considerable HCHO conversion is seen, well explaining the greatly enhanced HCHO removal by combining plasma with catalysis. The plasma-catalysis hybrid process where HCHO is introduced through the discharge zone and then the catalyst bed exhibits the highest energy efficiency concerning HCHO conversion, due to the best use of plasma-generated active species in a two-stage HCHO destruction process. Moreover, the presence of downstream MnO(x)/Al(2)O(3) catalyst significantly reduced the emission of discharge byproducts (O(3)) and organic intermediates (HCOOH).

In this study, solubilization of PAHs from a manufactured gas plant (MGP) soil and two artificially spiked soils using fatty acid methyl esters (FAME) was investigated. PAH removals from both the MGP and the spiked soils by FAME, methanol, soybean oil, hydroxypropyl-beta-cyclodextrin, Triton X-100, and Tween 80 were compared. The effect of FAME:MGP soil ratios on PAH removals was also investigated. Results showed that the FAME mixture synthesized by our lab was more efficient than the cyclodextrin and the two surfactants used for PAH removal from the spiked soils with individual PAH concentrations of 200 and 400 mg kg(-1). However, the difference among three PAH removals by the FAME, soybean oil and methanol was not quite pronounced. The FAME synthesized and market biodiesel exhibited better performance for PAH removals (46% and 35% of total PAH) from the weathered contaminated MGP soil when compared with the other agents (0-31%). Individual PAH removals from the weathered MGP soil were much lower than those from the spiked soils. The percentages of total PAH removals from the MGP soil were 59%, 46%, and 51% for the FAME:MGP soil ratios of 1:2, 1:1, and 2:1, respectively. These results showed that the FAME could be a more attractive alternative to conventional surfactants in ex situ washing of PAH-contaminated soils.
