RESUMEN

INTRODUCTION: Using microelectrodes for recording purposes in deep brain stimulation (DBS) has proven to be very useful. Their efficiency can be improved by characterising the properties of extracellular action potentials (EAPs). PATIENTS AND METHODS: We analysed the records of nine patients who underwent surgery for epilepsy or aggressiveness under general anaesthesia. The properties of the EAPs of the centromedian, ventral intermediate, ventrocaudal and posteromedial hypothalamic nuclei of the thalamus have been determined. RESULTS: We have analysed 706 thalamic and 142 hypothalamic cells. The proportion of cell types was found to be specific to each cell nucleus. The most frequent cell type was P1P2N1 (59.5%), followed by N1P1N2 (23.1%). The first phase of the EAP is highly variable. The properties of the EAP phases of the same morphology differ greatly from one nucleus to another. CONCLUSIONS: We have shown that several deep brain nuclei have properties that are specific to the morphology of the EAPs. This will allow for improved localisation of these nuclei during DBS.

This study evaluated the effect of adding ultra-diluted and dynamized Arnica montana 6 cH, and its vehicle (0.3% ethanol) to the in vitro maturation (IVM) medium, in the absence (experiment 1) or presence (experiment 2) of heat stress (HS), on bovine oocyte maturation and in vitro embryo production (IVEP). In experiment 1 (n = 902 cumulus oocyte complexes, COCs), the treatments were 1) IVM medium (Control treatment), 2) IVM medium + 0.3% ethanol, and 3) IVM medium + Arnica montana 6 cH. In experiment 2 (n = 1064 COCs), the treatments were 1) IVM medium without HS, 2) IVM medium under HS, 3) IVM medium + ethanol under HS, and 4) IVM medium + Arnica montana under HS. In the absence of HS (experiment 1), the addition of Arnica montana to the IVM medium had a deleterious effect on the IVEP (cleavage and blastocyst rates) and the total cell number/blastocysts. On the other hand, ethanol (0.3%) increased IVEP in relation to the Control and Arnica montana treatments. However, in the presence of HS during IVM (experiment 2), the addition of ethanol or Arnica montana increased IVEP when compared to the HS treatment alone, and the Arnica montana treatment resulted in greater total cell number/blastocysts compared to the other treatments. In conclusion, this study showed for the first time that the negative or positive effect of Arnica montana 6 cH on IVEP depends on the culture condition (i.e., absence or presence of HS during IVM). On the other hand, ethanol showed beneficial and consistent results on IVEP regardless of exposure to HS.

RESUMEN

Although, antibiotics are effective in the treatment of bovine mastitis, they do not address the regeneration of mammary glandular tissue and have been associated to the increment in antimicrobial resistance worldwide. Considering the necessity of alternative therapies for this disease of high economic impact and the reported regenerative and antibacterial effects of mesenchymal stem cell (MSCs), we evaluated the safety and efficacy of an allogenic MSC-based intramammary therapy in dairy cows with experimentally induced Staphylococcus aureus clinical mastitis. In a safety trial, heifers were inoculated intramammarily with a 2.5 × 107-suspension of bovine fetal AT-MSCs on experimental days 1 and 10. Animals were evaluated clinically on a daily basis during a 20-day experimental period and blood samples were collected for hemogram determination and peripheral blood leukocytes (PBLs) isolation. In an efficacy trial, Holstein Friesian cows were inoculated with S. aureus and treated intramammarily with vehicle (NEG; days 4 and 10), antibiotics (ATB; days 4 and 5) or a suspension of 2.5 × 107 AT-MSCs (MSC; days 4 and 5). Cows were clinically evaluated daily and milk samples were collected for somatic cell count (SCC) and colony forming units (CFU). Blood samples were collected for serum haptoglobin and amyloid A determination. Intramammary administration of two doses of bovine fetal AT-MSCs in healthy cows did not induce changes in clinical or hematological variables, and gene expression profiles in PBLs associated to activation (CD4, CD8, CD25, CD62L and CD69) and proinflammatory cytokines (CCL2, CCL5, IL2, CXCL3, IFNγ, and TNFα). Quarters of MSC group of cows had similar SCC log/mL in milk compared to infected quarters of ATB or NEG cows. However, quarters of MSC cows had lower CFU log/mL in milk compared to quarters of NEG cows. Intramammarily inoculation of repeated doses of 2.5 × 107 allogenic AT-MSCs did not induce clinical or immunological response in healthy cows. Moreover, MSC-intramammary treatment reduced bacterial count in milk of cows with S. aureus clinical mastitis compared to untreated cows. This work provides initial evidence for the safety and efficacy of an allogenic MSC-based intramammary therapy for the treatment of bovine mastitis.

RESUMEN

INTRODUCTION: The response of the thalamus during the study with somatosensory evoked potentials (SSEP) is not sufficiently understood. CASE REPORT: A 17-year-old man undergoing surgery for deep brain stimulation in the centromedian nucleus for drug-resistant epilepsy under complete sedation. During the intervention, the responses to SSEPs of the thalamic nuclei were recorded by means of four microelectrodes. These responses can be decomposed into three types: local field potentials (LFP), low amplitude fast oscillations (LFO), and high amplitude slow oscillations (HSO). LFO are widespread in much of the registered regions. However, HSOs are located at a single point in the registry and are closely associated with LFPs. CONCLUSIONS: Although the presence of LFO has been considered as an indicator of the presence of the sensory thalamus, its wide extension by different thalamic nuclei suggests that it is an unspecific response to SSEPs. However, the restricted spatial location of HSO and their association with LFP suggest that these newly described potentials are the markers for the presence of the sensory thalamus. Their identification may prove very useful in thalamic deep brain stimulation either in awake patients or especially in those requiring sedation.

TITLE: Nuevo potencial talamico asociado con potenciales evocados somatosensoriales.

UNLABELLED: The objective of this study was to evaluate enamel and dentin susceptibility to toothbrushing abrasion, after bleaching with 7.5% hydrogen peroxide (HP) gel supplemented or not with 0.5% calcium gluconate (Ca). Toothbrushing was performed immediately and 1h after bleaching, with two suspensions (high and low abrasivity). Bovine enamel and dentin specimens were divided into 12 groups (n=10) according to the bleaching gel (with and without Ca), slurry abrasivity (high or low) and elapsed time after bleaching (immediately and after 1h). As control, a group was not bleached, but abraded. The treatment cycle (7 d) consisted of bleaching (1h) and toothbrushing (135 strokes/day) immediatelly or after 1h of artificial saliva exposure. Surface roughness and surface loss (µm) were measured by profilometry and analysed by three-way ANOVA (5%). Surface roughness means were significantly influenced by slurry abrasivity (p<0.0001). For enamel loss, significant triple interaction was observed (p<0.0001). HP-bleached groups and immediately brushed with high-abrasive slurry exhibited increased loss (1.41±0.14) compared to other groups (µm). Control and HP+Ca-bleached groups brushed after 1h with low abrasive slurry presented the lowest loss (0.21±0.03/0.27±0.02). For dentin loss, significant interaction was observed for bleaching and interval factors (p<0.001). 7.5%HP-bleached groups and immediately brushed showed significantly higher loss (8.71±2.45) than the other groups. It was concluded that surface roughness increased when high abrasive was used, independently of bleaching. 7.5%HP increased enamel and dentin loss, mainly with high abrasive slurries. Calcium supplementation of bleaching gel reduced surface loss. Additionally, in order to minimize tooth wear susceptibility, it is recommended to delay brushing after bleaching. CLINICAL RELEVANCE: After bleaching gel application, postponing toothbrushing is recommended, as well as brushing with low abrasive dentifrices. Additionally, supplementation of hydrogen peroxide gel with calcium-based remineralizing agent potentially reduces tooth loss after abrasion.

RESUMEN

Purpose. We report the response rate in children older than 18 months with stage 4 Neuroblastoma, using a modified dose-intensive, response-adaptive, induction mN7 protocol. Methods. From 2005 to 2012, 24 patients were treated with the mN7 protocol. Phase 1 included five MSKCC N7 cycles and surgery and two high-dose cyclophosphamide-topotecan (HD-CT) cycles for those who did not achieve complete remission (CR) and negative bone marrow (BM) minimal residual disease (MRD) status (CR+MRD-). Phase 2 consisted of myeloablative doses of topotecan, thiotepa and carboplatin plus hyperfractionated RT. Phase 3 included isotretinoin and 3F8 immunotherapy plus GM-CSF. BM MRD was monitored using GD2 synthase, PHOX2B and cyclin D1 mRNAs. Results. After 3 cycles, all patients showed BM complete histological clearance and 6 (25 %) were MRD-. Twenty of 21 s-look surgeries achieved macroscopic complete resection. After 5 cycles and surgery, 123I-MIBG scan was negative in 15 (62.5 %) cases, BM disease by histology was negative in 23 (96 %) and 10 (42 %) patients were MRD-. Twelve (50 %) pts were in CR, 2 in very good partial response (VGPR), 9 partial response (PR) and one had progressive disease. With 2 HD-CT extra cycles, 17 (71 %) pts achieved CR+MRD- status moving to phase 2. Overall and event-free survival at 3 years for the 17 patients who achieved CR+MRD- is 65 and 53 %, respectively, median follow-up 47 months. Seven (29 %) patients never achieved CR+MRD-. Univariate Cox regression analysis shows CR+MRD- status after mN7 induction as the only statistically significant prognostic factor to predict overall survival. Conclusions. mN7 induction regimen produced a CR+MRD- rate of 71 %. CR+MRD- status following induction was the only predictive marker of long-term survival (AU)

RESUMEN

Magnetic materials, which have the potential for application in heating therapy by hyperthermia, were prepared. This alternative treatment is used to eliminate cancer cells. Magnetite, magnesium-calcium ferrites and manganese-calcium ferrites were synthesized by sol-gel method followed by heat treatment at different temperatures for 30 min in air. Materials with superparamagnetic behavior and nanometric sizes were obtained in all the cases. Thus, these nanopowders may be suitable for their use in human tissue. The average sizes were 14 nm for magnetite, 10 nm for both Mg(0.4)Ca(0.6)Fe(2)O(4) and Mg(0.6)Ca(0.4)Fe(2)O(4) and 11 nm for Mn(0.2)Ca(0.8)Fe(2)O(4). Taking into account that the Mg(0.4)Ca(0.6)Fe(2)O(4) and Mg(0.6)Ca(0.4)Fe(2)O(4) treated at 350 °C showed the lower coercivity values, these nanoparticles were selected for heating tests and cell viability. Heating curves of Mg(0.4)Ca(0.6)Fe(2)O(4) subjected to a magnetic field of 195 kHz and 10 kA/m exhibited a temperature increase up to 45 °C in 15 min. A high human osteosarcoma cell viability of 90-99.5% was displayed. The human osteosarcoma cell with magnesium-calcium ferrites exposed to a magnetic field revealed a death cell higher than 80% in all the cases.

RESUMEN

Pollen is routinely monitored, but it is unknown whether pollen counts represent allergen exposure. We therefore simultaneously determined olive pollen and Ole e 1 in ambient air in Córdoba, Spain, and Évora, Portugal, using Hirst-type traps for pollen and high-volume cascade impactors for allergen. Pollen from different days released 12-fold different amounts of Ole e 1 per pollen (both locations P < 0.001). Average allergen release from pollen (pollen potency) was much higher in Córdoba (3.9 pg Ole e 1/pollen) than in Évora (0.8 pg Ole e 1/pollen, P = 0.004). Indeed, yearly olive pollen counts in Córdoba were 2.4 times higher than in Évora, but Ole e 1 concentrations were 7.6 times higher. When modeling the origin of the pollen, >40% of Ole e 1 exposure in Évora was explained by high-potency pollen originating from the south of Spain. Thus, olive pollen can vary substantially in allergen release, even though they are morphologically identical.

RESUMEN

OBJECTIVES: People increasingly desire tooth whitening. Considering the wide range of whitening products on the market, this study evaluated the efficacy of whitening toothpastes and mouth rinses compared with the 10% carbamide peroxide (CP) whitening gel. METHODS: We obtained 120 cylindrical specimens from bovine teeth, which were darkened for 24 hours in a coffee solution. The color measurement was performed by a spectrophotometer using the CIE L*a*b* system, and specimens were divided into six groups according to the use of the following agents: group 1, conventional fluoridated toothpaste; group 2, Close Up White Now; group 3, Listerine Whitening; group 4, Colgate Plax Whitening; group 5, experimental mouth rinse with Plasdone; and group 6, 10% CP Whiteness Perfect. After the simulation of 12 weeks of treatment for groups 1 to 5 and 14 days of treatment for group 6, the specimens were subjected to a new color reading. RESULTS: Data were subjected to one-way analysis of variance (α=0.05), which showed significant differences among groups after 12 weeks for ΔE (p=0.001). Results of the Tukey test revealed that groups 3, 4, and 6 presented significantly higher color alteration than groups 1, 2, and 5. CONCLUSIONS: The whitening toothpaste Close Up White Now and the experimental mouth rinse with Plasdone showed similar color alteration as conventional toothpaste after a 12-week treatment simulation. These groups presented significantly lower color alteration compared with whitening mouth rinses Listerine and Colgate Plax Whitening, which showed similar results to those observed after 14 days of bleaching with 10% CP treatment.

RESUMEN

OBJECTIVE: To investigate the clinical, laboratory and histological manifestations of patients who received illegal injections of foreign substances for cosmetic purposes. PATIENTS AND METHODS: We studied patients who met the following inclusion criteria: 1) history of application of foreign substances for cosmetic purposes, 2) clinical data of autoimmune disease or non-specific autoimmune manifestation (i.e. arthralgias, myalgia, malaise, fever, and weight loss), 3) detection of autoantibodies in patients' sera, 4) histological evidence of chronic inflammation and/or granulomatous reaction to foreign body. RESULTS: Fifty female patients aged 44.4 ± 10 years were studied. The mean time between application of foreign substances and onset of symptoms was 4.5 ± 4.3 years. Patients were followed for 12 ± 7.5 years. Forty-one patients were injected with mineral oil, nine patients received other substances: three iodine gadital, one guayacol, one guayacol plus silicone fluid, two collagen, two silicone fluid. The sites of application were: buttocks (36), legs and/or thighs (11), breasts (eight) hands and face (one), face (two) (seven patients received an injection to more than one site). Thirty patients presented with non-specific autoimmune manifestations, whereas 20 patients fulfilled the criteria for a defined autoimmune disease such as systemic lupus erythematosus, rheumatoid arthritis, systemic sclerosis, overlap syndrome, autoimmune hemolytic anemia, autoimmune thyroiditis, autoimmune hepatitis, and ulcerative colitis. CONCLUSIONS: Cases of human adjuvant disease following illegal injections of oil substances for cosmetic purposes are reported. Patients presented with defined autoimmune diseases as well as with non-specific autoimmune manifestations. Illegal injection of these substances could lead to serious local and systemic complications, even to death. These cases represent another model of Autoimmune/inflammatory Syndrome Induced by Adjuvants (ASIA). The use of these substances should be prohibited.

Introduction: Inulin and FOS are prebiotics with potential benefit in cardiovascular risk factors. Alphalinolenic acid (ALA) is the metabolic precursor of the long chain n-3 fatty acid eicosapentaenoic acid (20: 5n-3),this fatty acid has anti-inflammatory properties. The aim of our study was to evaluate the response of the cardiovascular risk profile in obese patients after inclusion in the diet of an ALA, FOS and inulin enriched-cookie. Material and methods: 36 patients were randomized in both branches: group I (inulin, FOS and ALA enriched cookie) Gullon SL® and group II (control cookie). Previous and after 1 month of the treatment, a nutritional and biochemical study was realized. Results: 15 patients finished the procotol in each group. In group I, a significantly increase in soluble fiber (2.3 ±0.8 g/day vs 7.7 ± 0.8 g/day: p < 0.05) and ALA (0.6 ± 0.5g/day vs 3.8 ± 0.5 g/day; p < 0.05) intakes was detected. In this group a significant decrease of total cholesterol (238.1± 45.3 mg/dl vs 210.5 ± 38.1 mg/dl: p < 0.05), LDL cholesterol(153.6 ± 23.2 mg/dl vs 127.1 ± 27.9 mg/dl: p < 0.05)and C reactive protein (6.6 ± 1.4 mg/dl vs 4.4+7-1.8 mg/dl:p < 0.05) was reached in males. Anthropometric parameters did not change in both groups. The increase in soluble fiber and ALA dietary intakes did not produce any gastrointestinal adverse effect. Conclusion: The increase of 2 grams per day of inulin,3.1 grams per day of FOS and 3.2 grams per day of alphalinolenic (ALA) dietary intakes from an enriched-cookie, improved total cholesterol, LDL cholesterol and C reactive protein levels in obese males. As far as we know, this is the first study that has evaluated the effect on risk factors of an ALA enriched cookies (AU)

The objective of this study was to examine the effect of feeding diets containing fat supplements enriched in either saturated fatty acids (n = 10), Ca salts of trans-octadecenoic fatty acids (tFA, n = 10) or Ca salts of safflower oil fatty acids (SFL, high in linoleic acid, n = 9) on performance, metabolic, and endocrine responses of periparturient Holstein cows. Dietary treatments were initiated at approximately 28 d before calculated calving dates and continued through 49 d postpartum. Blood samples for metabolite and hormone analyses were collected weekly beginning 1 wk before estimated calving date through 7 wk postpartum. Incorporation of tFA or SFL into the peripartum diet had no detectable effects on body weight or body condition score. Cows fed the SFL-enriched diet produced less milk fat and established a positive energy balance sooner after calving than those fed the tFA supplement. Analysis for individual fatty acids resulted in increased concentrations of trans 18:1 fatty acid and conjugated linoleic acid isomers in milk fat from cows supplemented with SFL. Across weeks, the average nonesterified fatty acids concentration in plasma was lower in cows fed the SFL-enriched diet than in those consuming the tFA-supplemented diet. Mean concentrations of plasma glucose, insulin-like growth factor-I, and progesterone were greater in cows fed the SFL-enriched diet compared with those fed the saturated fatty acid-supplemented diet. Feeding fat supplements that can suppress milk fat production during the early postpartum period may help minimize negative energy balance, reduce adipose tissue mobilization, and improve circulating concentrations of insulin-like growth factor-I and progesterone. Whether the SFL supplement would have similar effects without a decrease in milk fat production remains to be determined and warrants further investigation.

The addition of oil sources and dietary vitamin E supplementation was evaluated on the characteristics of fresh boar sperm. Twenty-four mature Dalboar 85 boars of proven fertility and in routine semen production for artificial insemination were randomly distributed in a factorial arrangement 2 X 3, with two oil sources, soy and salmon, and three antioxidant levels: 150, 300, and 450mg/kg of vitamin E. Volume, total motile sperm, hyposmotic swelling test, percentage of live cell, and morphology did not differ (P<0.05) between the animals from the treatments. Salmon oil improved (P<0.05) sperm vigor. The vitamin E added to the diet improved (P<0.05) the sperm concentration, and there was no difference between the oil sources (P>0.05). Salmon oil treatment showed the lowest (P<0.05) concentration of total antioxidants in the semen. Vitamin E and concentration of total antioxidant showed a linear effect (P<0.05). The semen vigor and the concentration of total antioxidants were improved by oil sources, but the sperm concentration was dependent on the levels of vitamin E. Therefore, salmon oil can be used in the diet of male pigs.

RESUMEN

The objective of this work was to evaluate the feasibility of ethanol production by fermentation of coffee husks by Saccharomyces cerevisiae. Batch fermentation studies were performed employing whole and ground coffee husks, and aqueous extract from ground coffee husks. It was observed that fermentation yield decreased with an increase in yeast concentration. The best results were obtained for the following conditions: whole coffee husks, 3 g yeast/l substrate, temperature of 30 degrees C. Under these conditions ethanol production was 8.49 +/- 0.29 g/100 g dry basis (13.6 +/- 0.5 g ethanol/l), a satisfactory value in comparison to literature data for other residues such as corn stalks, barley straw and hydrolyzed wheat stillage (5-11 g ethanol/l). Such results indicate that coffee husks present excellent potential for residue-based ethanol production.

Introduction and development. Depression is the most prevalent of all psychiatric disorders. The majority of patients respond to drug therapy and psychotherapy and electroconvulsive therapy are effective in a significant group of medicallyresistant patients. Despite these approaches, there are still patients who remain severely disabled. For these patients other therapeutic options must be considered. The advent of deep brain stimulation has revolutionized the treatment of movementdisorders. Because of its efficacy, reversibility and safety, the use of deep brain stimulation is becoming more widespread. As a result of our increasing understanding of the mood and affective circuits and their dysfunction in pathologic states, we havebeen able to identify potential targets for deep brain stimulation for treatment resistant depression. Conclusion. Despite the limited studies on the role of deep brain stimulation for treatment resistant depression, their results have been positive andthey have provided the base for future trials to investigate deep brain stimulation as a treatment for resistant depression

RESUMEN

After parturition, immune functions such as lymphocyte response to mitogens and production of antibodies are depressed in dairy cows. Dietary regimens that improve the immune function of dairy cows after calving may improve uterine health and lead to earlier breeding after parturition. The objective of this study was to examine the effect of feeding a calcium salt of trans isomers of fatty acids (tFA) to periparturient Holstein cows on plasma biomarkers of inflammation. Dietary treatments were initiated approximately 28 d before expected calving date and continued through d 21 postpartum. Prepartum and postpartum diets were formulated to be isolipidic, containing 1.5% saturated fats (n = 15) or 1.8% tFA (n = 15). Multiparous cows were heavier at calving (+32%) and produced more milk (+17%) than primiparous cows. Periparturient tFA supplementation increased plasma PGF(2alpha) metabolite concentration in multiparous cows, but not in primiparous cows. Concentrations of prostaglandin E(2), tumor necrosis factor-alpha, and interleukin-4 in plasma did not differ between diets and parities. Results raise the possibility that peripartum tFA supplementation may affect uterine health and reproductive efficiency of early lactation dairy cows through alteration of peripheral PGF(2alpha) concentration.

RESUMEN

This paper presents the physicochemical and nutrimental characterization of fresh nopal (Opuntia ficus indica, Redonda variety) and nopal powder produced at different stages of development. Nopal powder was obtained by dry vacuum technique using 10(2) Torr and low temperature (40 degrees C). The results showed that the nutrimental and mineral composition of nopal changes as a function of the maturation as follow: The ash content increases from 18.41 for nopalitos (60 g of weight) to 23.24% (nopal pads 200 g); calcium content increases from 1.52 to 3.72%, while phosphorous exhibits an opposite trend: 0.43 to 0.27%, respectively. Calcium oxalate was determined by X-ray diffraction and SEM microscopy and quantified by using atomic absorption spectroscopy. Calcium oxalate decreases from 7.95 to 3.47 mg/g and the Ca/P ratio varies from 3.6 to 11. The soluble fibre decreases from 25.22 to 14.91%, while insoluble fibre increases from 29.87 to 41.65%. These results suggest that nopal could be an important source of minerals within the diets of people in Mexico and the rest of Latin America.

RESUMEN

A three-stage process was developed to produce polyhydroxyalkanoates (PHAs) from sugar cane molasses. The process includes (1) molasses acidogenic fermentation, (2) selection of PHA-accumulating cultures, (3) PHA batch accumulation using the enriched sludge and fermented molasses. In the fermentation step, the effect of pH (5-7) on the organic acids profile and productivity was evaluated. At higher pH, acetic and propionic acids were the main products, while lower pH favoured the production of butyric and valeric acids. PHA accumulation using fermented molasses was evaluated with two cultures selected either with acetate or fermented molasses. The effect of organic acids distribution on polymer composition and yield was evaluated with the acetate selected culture. Storage yields varied from 0.37 to 0.50Cmmol HA/Cmmol VFA. A direct relationship between the type of organic acids used and the polymers composition was observed. Low ammonia concentration (0.1Nmmol/l) in the fermented molasses stimulated PHA storage (0.62Cmmol HA/Cmmol VFA). In addition, strategies of reactor operation to select a PHA-accumulating culture on fermented molasses were developed. The combination of low organic loading with high ammonia concentration selected a culture with a stable storage capacity and with a storage yield (0.59Cmmol HA/Cmmol VFA) similar to that of the acetate-selected culture.
