ETHNOPHARMACOLOGICAL RELEVANCE: Syringa Pubescens Turcz. (SP), a member of the Oleaceae family, is a species of plant known as Syringa. Flowers, as the medicinal part, are commonly used in the treatment of hepatitis and tonsillitis. AIM OF THE STUDY: The research was the first to assess the antioxidant and anti-inflammatory potential of different parts of SP flowers (SPF) in vitro. The most promising fraction was ethyl acetate fraction of SP flower (SPFEA). The antioxidant, anti-inflammatory and analgesic activities of SPFEA were further studied, and the chemical components were identified. METHODS: HPLC was used to identify the major components in various fraction of SPF. DPPH and ABTS + radical scavenging assays as well as FRAP test and ß-carotene bleaching test were employed to assess the antioxidant potential of SPF fraction in vitro. The inhibitory effect on NO production in LPS-treated RAW264.7 cells and heat-induced protein denaturation test were used to evaluate the anti-inflammatory potential of SPF fraction. Further analysis of the biological activity of SPFEA was performed. Acute toxicity test was conducted to assess the toxicity of SPFEA. The anti-inflammatory effect was assessed by utilizing xylene induced ear edema model, carrageenan-induced foot edema model and peritonitis model in vivo. The analgesic effect of SPFEA was evaluated using hot plate test, tail immersion test, formaldehyde test as well as acetic acid-induced abdominal writhing pain experiment in vivo. In carrageenan induced foot edema model, ELISA kits were employed to measure levels of inflammation factors (NO, TNF-α, IL-6, COX-2, IL-1ß) in foot tissue as well as MDA, CAT, SOD, GSH-PX levels in liver tissue. RESULTS: HPLC results showed that there were significant differences in bioactive substances among different fractions of SPF, and SPFEA was rich in bioacitve components. Compared with other fractions of SPF, SPFEA exhibited better antioxidant and anti-inflammatory abilities. The 3000 mg/kg SPFEA group in mice had no obvious side effects. The xylene-induced ear edema model, carrageenan-induced foot edema and peritonitis models demonstrated that the SPFEA had significant anti-inflammatory effect. Moreover, inflammation factors including NO, TNF-α, IL-6, COX-2, IL-1ß were significantly reduced in SPFEA groups in foot tissue induced by carrageenan. Additionally, SPFEA effectively decreased liver tissue oxidative stress levels (MDA, SOD, GSH-PX and CAT). The bioactivities of SPFEA demonstrated a clear dose-dependent relationship. The results of the hot plate test, tail immersion test, formaldehyde test and acetic acid-induced abdominal writhing pain experiments indicated the SPFEA possessed an excellent analgesic effect, and this effect was in dose-dependent manner. CONCLUSION: The study provides a scientific foundation for understanding the pharmacological action of SPFEA. It has been indicated that SPFEA has excellent antioxidant, analgesic and anti-inflammatory effects.

The flower of Syringa pubescens Turcz. is used in Chinese folk medicine and also as a flower tea for healthcare. The effects of five drying methods on the active compound contents, the antioxidant abilities, anti-inflammatory properties and enzyme inhibitory activities were evaluated. The plant materials were treated using shade-drying, microwave-drying, sun-drying, infrared-drying and oven-drying. The seven active compounds were simultaneously determined using an HPLC method. Furthermore, the chemical profile was assessed using scanning electron microscopy, ultraviolet spectroscopy and infrared spectroscopy. The antioxidant capacities and protective effects on L02 cells induced with hydrogen peroxide were measured. The anti-inflammatory effects on lipopolysaccharide-induced RAW264.7 cells were investigated. The enzyme inhibitory activities were determined against α-amylase, α-glucosidase cholinesterases and tyrosinase. The results indicated that drying methods had significant influences on the active compound contents and biological properties. Compared with other samples, the OD samples possessed low IC50 values with 0.118 ± 0.004 mg/mL for DPPH radical, 1.538 ± 0.0972 for hydroxyl radical and 0.886 ± 0.199 mg/mL for superoxide radical, while the SHD samples had stronger reducing power compared with other samples. The SHD samples could be effective against H2O2-induced injury on L02 cells by the promoting of T-AOC, GSH-PX, SOD and CAT activities and the reducing of MDA content compared with other samples. Furthermore, SPF samples, especially the SHD sample, could evidently ameliorate inflammation through the inhibition of IL-6, IL-1ß and TNF-α expression. All the studied SPF samples exhibited evidently inhibitory effects on the four enzymes. The IC50 values of inhibitory activity on α-glucosidase and α-amylase from SHD sample were 2.516 ± 0.024 and 0.734 ± 0.034 mg/mL, respectively. SD samples had potential inhibitory effects on cholinesterases and tyrosinase with IC50 values of 3.443 ± 0.060 and 1.732 ± 0.058 mg/mL. In consideration of active compound contents and biological activities, it was recommended that SHD and SD be applied for drying SPF at an industrial scale.

A high-performance liquid chromatograph with diode array detector was established for the simultaneous determination of five phenylethanoid glycosides in Syringa pubescens Turcz. The optimal chromatographic conditions were achieved on a Zorbax C18 column using gradient elution with 0.5% aqueous acetic acid and acetonitrile as the mobile phase at the flow rate of 1.0 mL/min. The detection wavelength was developed as follows: 0-10 min, 276 nm; 10-45 min, 332 nm. The validation of the method including linearity, precision, stability, accuracy, repeatability and recovery was tested. The chemometric analysis including hierarchical cluster analysis and principal component analysis was employed to investigate the similarity and difference of samples from different geographical origin. The results revealed that S. pubescens samples were divided into four clusters based on the phenylethanoid glycosides contents. Antioxidant activity of extract was measured using three different methods including α,α-diphenyl-ß-picrylhydrazyl and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) radical scavenging assays, and ferric reducing antioxidant power assay. Furthermore, different phenylethanoid glycosides exhibited different contribution to antioxidant capacities. This study provides a foundation for the quality evaluation and offers scientific data for the utilization of S. pubescens resources.

Syringa pubescens Turcz is commonly used folk medicinal herb in west of Henan Province of China. In this work, water and various concentration of methanol, ethanol and acetone in water were used as solvent to extract echinacoside and oleuropein from S. pubescens. The antioxidant properties of different extracts were evaluated using various in vitro assays. The highest yields of echinacoside and oleuropein were obtained by using the 60% aqueous methanol and 80% aqueous ethanol, respectively. The extracts of water, aqueous ethanol or methanol showed strong antioxidant abilities. Furthermore, the high correlation between echinacoside content and antioxidant properties was found. The contribution of oleuropein content was not significant to antioxidant abilities. These findings indicate that S. pubescens can be used as a new natural antioxidant resource.

In the present study, the ultrasound-assisted extraction (UAE) process of polysaccharide from Fructus corni (FCP) was opitimized using the Box-Behnken design (BBD). The main parameters including ultrasound time (min), temperature (oC) and solvent to raw material ratio (mL/g) were chosen as the process variables for the optimization of UAE process. The results of analysis of variance indicated that the equation obtained from the experiments could represent the data and the predicted responses satisfactorily. The optimum conditons obtained by BBD were ultrasonic time (51min), temperature (69oC) and solvent to solid ratio (20mL/g) with actual yield (12.68±0.16%), which was good agreement with value predicted by the model. The antioxidant properties of FCP were assessed in vitro based on scavenging effect of the DPPH radical, hydroxyl radical and super oxide radical tests. The FCP possessed strong antioxidant abilities on DPPH and super oxide radical at the high concentration. The results on hydroxyl radical demonstrated that FCP exhibited high scavenging effect when the concentration was over 3000µg/mL. The findings suggested that the FCP had antioxidant capacities and could be developed as a source of natural antioxidants and functional food material.

INTRODUCTION: The Paeonia ostii T. Hong & J. X. Zhang seed shell, characterised by a high content of oligostilbenes, is one of the two most important by-products in the preparation of seed oil. Oligostilbenes are considered characteristic constituents of the genus Paeonia, and can be used in fingerprinting to determine the geographical origin and the quality of raw materials. OBJECTIVE: To develop and optimise a simple and reproducible high-performance liquid chromatography diode array detection (HPLC-DAD) method for the simultaneous determination of seven oligostilbenes in P. ostii seed shell from different geographical areas, and to associate the cultivation area. METHODOLOGY: A validated HPLC method coupled with a DAD detector was performed for the detection and determination of target compounds in the samples. Optimal chromatographic conditions were achieved using an Agilent Zorbax Eclipse SB-AQ-C18 column and a gradient elution with acetonitrile and potassium dihydrogen phosphate solution. RESULTS: The proposed quantitative method showed appropriate accuracy and precision, and was successfully applied to the routine analysis of seven oligostilbenes and the quality evaluation of 50 P. ostii seed shell samples. There were significant differences between the contents of the seven oligostilbenes in different samples (P < 0.01). CONCLUSION: The results demonstrated that the oligostilbenes were main secondary metabolites in the P. ostii seed shells, and the content of seven components in P. ostii seed shells sourced from different cultivation areas in China was different.

OBJECTIVE: To investigate the expression of the sperm-specific cation channel (CatSper1) in the epididymal sperm of varicocele (VC) rats and the effect of L-carnitine (LC) on the CatSper1 level. METHODS: Seventy male rats were equally randomized into groups A (normal control), B (VC model control), C (VC treated with normal saline), D (VC treated with low-dose LC), E (VC treated with medium-dose LC), F (VC treated with high-dose LC), and G (VC treated by prolonged medication of high-dose LC). The VC model was established by partial ligation of the left renal vein. At 12 weeks after modeling, the model rats in group C were treated intragastrically with normal saline at 1 ml/kg/d, those in groups D, E and F with LC at 0.05, 0.1 and 0.2 g/kg/d respectively, all for 5 consecutive weeks, and those in group G with LC at 0.2 g/kg/d for 7 successive weeks. Then, all the animals were sacrificed and their epididymides harvested for obtainment of the semen parameters by computer-assisted semen analysis (CASA) and determination of the mRNA and protein expressions of CatSper1 in the sperm by RT-PCR and Western blot. RESULTS: Compared with the rats in group A, those in group B showed significantly decreased percentage of grade a+b sperm (P < 0.01), sperm viability (P < 0.01), sperm concentration (P < 0.01) and expressions of CatSper1 mRNA (1.44 ± 0.67 vs 0.71 ± 0.38, P < 0.01) and protein (1.87 ± 0.67 vs 0.84 ± 0.42, P < 0.01). In comparison with the animals in group C, those in the four LC intervention groups exhibited a markedly increased percentage of grade a+b sperm, sperm viability and mRNA and protein expressions of CatSper1, even more remarkably in groups F and G (P < 0.01). No statistically significant difference, however, was observed in sperm concentration between group C and the LC intervention groups (P > 0.05), nor in the mRNA and protein expressions of CatSper1 between groups F and G. CONCLUSIONS: The expression of CatSper1 is decreased in the epididymal sperm of varicocele rats, and L-carnitine can increase the sperm viability, percentage of grade a+b sperm and CatSper1 expression of the rats.

The aim of the present work was to investigate the trace elements and the correlation with flavonoids from Sparganii rhizoma. The ICP-AES and ultraviolet-visible spectroscopy were employed to analyze trace elements and flavonoids. The concentrations of trace elements and flavonoids were calculated using standard curve. The content of flavonoids was expressed as rutin equivalents. The cluster analysis was applied to evaluate geographical features of S. rhizoma from different geographical regions. The correlation analysis was used to obtain the relationship between the trace elements and flavonoids. The results indicated that the 15 trace elements were measured and the K, Ca, Mg, Na, Mn, Al, Cu, and Zn are rich in Sparganii rhizome. The different producing regions samples were classified into four groups. There was a weak relationship between trace elements and flavonoids.

Huaiju is one of the most famous and widely used Flos Chrysanthemi (FC) for medicinal purposes in China. Although various investigations aimed at phenolics extraction from other FC have been reported, a thorough optimization of the phenolics extraction conditions from Huaiju has not been achieved. This work applied the widely used response surface methodology (RSM) to investigate the effects of 3 independent variables including ethanol concentration (%), extraction time (min), and solvent-to-material ratio (mL/g) on the ultrasound-assisted extraction (UAE) of phenolics from FC. The data suggested the optimal UAE condition was an ethanol concentration of 75.3% and extraction time of 43.5 min, whereas the ratio of solvent to material has no significant effect. When the free radical scavenging ability was used as an indicator for a successful extraction, a similar optimal extraction was achieved with an ethanol concentration of 72.8%, extraction time of 44.3 min, and the ratio of solvent to material was 29.5 mL/g. Furthermore, a moderate correlation between the antioxidant activity of TP extract and the content of extracted phenolic compounds was observed. Moreover, a well consistent of the experimental values under optimal conditions with those predicted values suggests RSM successfully optimized the UAE conditions for phenolics extraction from FC. PRACTICAL APPLICATION: The work of the research investigated the plant phenolics in Flos Chrysanthemi and antioxidant capacities. These results of this study can support the development of antioxidant additive and relative food.

A simple, sensitive, and validated high-performance liquid chromatography-photodiode array method has been established for the simultaneous determination of anthraquinones and resveratrol in Polygonum Cillinerve (Nakai) Ohwi (Zhushaqi in Chinese). The evaluation was performed using a Sunfire C18 reversed-phase column with 30°C column temperature. The mobile phase was composed of a gradient elution of 0.5% acetic acid (solvent A) and methanol (solvent B) with flow rate of 1.0 mL/min. The detection wavelength was at 254 nm. The method developed and validated is simple, shows good linearity, sensitivity, precision, and recovery, and is applied to analyze anthraquinones and resveratrol in 13 batches of Zhushaqi. The results show that the target compounds of Zhushaqi are significantly different among these samples. Based on results of the statistical analysis, the samples collected from Funiu Mountain were clustered together, and the samples obtained from Bozhou Market were close together. The developed method can be a useful tool in quality control and used to evaluate difference and to identify the geographical area of Zhushaqi, and also to provide technical support for the pharmacological and clinical research of related drugs.

Paeonia ostii is a traditional medicinal plant popularly used in China. This study intended to evaluate the antioxidant properties and the chemical components of the flavonoid-rich extracts from the flowers of P. ostii. The results showed that the flavonoid-rich extracts from the flowers of P. ostii had strong scavenging capacities on 2,2'-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS), hydroxyls, superoxide anions, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals in a dose-dependent manner. Five flavonoids, dihydrokaempferol (1), apigenin-7-O-ß-d-glucoside (2), apigenin-7-O-ß-d-neohesperidoside (3), kaempferol-7-O-ß-d-glucopyranoside (4), and kaempferol-3-O-ß-d-glucopyranosyl-7-O-ß-d-glucopyranoside (5), were isolated from the flavonoid-rich extracts of the flowers of P. ostii. High-performance liquid chromatography (HPLC) analysis revealed that compounds 3 and 4 were abundant in the P. ostii flower and in flavonoid-rich extracts. The main components of the flower of P. ostii are flavonoids. The high antioxidant activity of the flavonoid-rich extracts may be attributed to the high content of flavonoids. The five isolated flavonoids were the primary antioxidant ingredients, and may play important roles in the strong antioxidant activities of this flower. Based on the obtained results, the flower of P. ostii could be a potential source of natural antioxidants in food and pharmaceutical applications.

In this study, the crude polysaccharides from Sparganium stoloniferum Buch.-Ham were prepared using hot-water extraction and further deproteinzated by Sevage method. The purified fraction of crude polysaccharide was obtained using a DEAE-52 cellulose chromatography and named with WSSP. Then, the antioxidant capacities of WSSP were assessed in vitro and in vivo. The results in vitro indicated that the WSSP possessed notably free radical scavenging capacity. And the antioxidant abilities were dose-dependent and increased with increasing dose of sample. The findings in vivo showed the gavage administration of WSSP can increase SOD and TAOC activities, and decrease MDA levels in tissue and serum of mice. Therefore, the WSSP may serve as a potential antioxidant.

OBJECTIVE: To study the influence of different drying methods on the quality of Sparganii Rhizoma and obtain the optimal one. METHODS: Based on multi-index evaluation technology, Sparganii Rhizoma was processed using five drying methods including drying in the shade, drying in the sun, drying in the oven, infrared drying and microwave drying. The contents of the functional ingredients such as seven phenolic compounds including p-hydroxybenzaldehde, vanillic acid,p-coumaric acid, ferulic acid, rutin, kaempferol and formononetin, polysaccharide, total saponins and total polyphenols were determined by the validated procedures of HPLC and ultraviolet spectrophotometry. RESULTS: The contents of seven phenolic compounds, polysaccharide and total polyphenols were the highest using microwave drying, while the content of the total saponins was the highest by drying in the oven. CONCLUSION: Different drying methods have significant influence on the quality of Sparganii Rhizoma.

RESUMEN

Cell wall polysaccharides in the petioles and pedicels of Qian (Euryale ferox Salisb.) (EFPP) were extracted using ultrasound-assisted technique. Response surface methodology (RSM) based on Box-Behnken design (BBD) was employed to optimize extraction parameters for the maximum purity of polysaccharides. The results showed that the optimum extraction conditions were extraction temperature of 80 °C, extraction time of 32 min, ultrasonic power of 270W and liquid-to-solid ratio of 40 mL/g. Under the optimal conditions, the experimental purity of polysaccharides was 62.57% ± 1.68%, which was very close to the predicted. The crude EFPP were isolated using DEAE-52 column and four major fractions (EFPP-1, EFPP-2, EFPP-3 and EFPP-4) were obtained. Typical functional groups of polysaccharides were characteristic for EFPP-1, EFPP-3 and EFPP-4 from FT-IR spectrum. Furthermore, the crude EFPP and three fractions (EFPP-1, EFPP-3 and EFPP-4) possessed appreciable in vitro antioxidant effects on α,α-diphenyl-ß-picrylhydrazyl (DPPH), 2,2'-azinobis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS), hydroxyl radical scavenging and reducing powers. Then, the crude EFPP and EFPP-4 could effective against H2O2-induced injury on HUVEC and VSMC through enhancement of T-AOC, SOD and CAT activities and decrease of MDA content.

OBJECTIVE: To study the effect of Shengjingsan on spermatogenic function following testicular torsion/detorsion in rats and its action mechanism. METHODS: Forty SD male rats were equally randomized to groups A (sham operation), B (control), C (low-dose Shengjingsan), D (medium-dose Shengjingsan) and E (high-dose Shengjingsan). The model of testicular torsion was established by 720 degrees clockwise torsion of the left testis for 4 hours. An hour before operation, the rats of group B received daily gavage of normal saline at 1 ml per kg per d, while those in groups C, D and E that of Shengjingsan at 0.01, 0.02 and 0.03 g per kg per d, all for 35 days. Then all the rats were sacrificed for measuring the semen parameters by CASA and detecting the expression of the CatSper1 gene in the sperm by RT-PCR. RESULTS: Compared with group A, Sperm concentration, the percentage of grade a + b sperm, sperm vitality and CatSper1 expression were significantly lower in group B ([15.30 +/- 6.30] %, [44.42 +/- 6.36] %, [21.00 +/- 6.14] x 10(6)/ml and 1.12 +/- 0.50) than in A ([51.30 +/- 6.60]%, [69.01 +/- 7.20]%, [40.53 +/- 7.01] x 10(6)/ml and 2.04 +/- 0.77) (P < 0.01). Compared with group B, the four parameters were increased remarkably in groups D ([51.63 +/- 3.20] %, [72.09 +/- 2.20]%, [55.30 +/- 5.90] x10(6)/ml and 2.11 +/- 0.20) andE ([55.93 +/- 3.17]%, [73.01 +/- 2.11]%, [58.33 + 4.90] x 10(6)/ml and 2.31 +/- 0.17) (P < 0.01), but not significantly in C ([18.02 +/- 0.23]%, [48.04 +/- 7.01]%, [22.87 +/- 2.10] x 10(6)/ml and 1.19 +/- 0.51) (P > 0.05). CONCLUSION: Shengjingsan can improve sperm parameters following testicular torsion/ detorsion in male rats by regulating their spermatogenic function and improving the expression of CatSper1 in the sperm.

RESUMEN

A reversed-phase high-performance liquid chromatographic method was developed for the simultaneous quantification of seven phenolic compounds from Rhizoma Sparganii. The samples were separated on a Waters SunFire C18 column with a temperature of 30°C. Gradient elution was applied, using 0.8% acetic acid (solvent A) and methanol (solvent B) with a flow rate of 1.0 mL/min, and detection wavelength was 280 nm. The validation of the method included linearity, precision, repeatability, stability and recovery. The calibration curves showed good linear regression (R(2) > 0.9996) within the test range. The developed method indicated good precision and accuracy, with the overall intra-day and inter-day variation at less than 3%. The range of recoveries for the seven analytes was 95.34-100.06% with relative standard deviation < 3.08 %. The established method was successfully applied for the determination of seven phenolic compounds in 12 batches of Rhizoma Sparganii. The study may be useful in the quality evaluation of Rhizoma Sparganii, and can provide technical support for pharmacological and clinical research of related drugs.

Plant-derived bioactive polysaccharides have a long history of application in traditional Chinese medicine (TCM). The polysaccharides of Fructus Corni, secondary metabolites from Cornus officinalis Sieb. Et Zucc, possess various pharmacological activities, including immune regulation, anti-oxidation, anti-tumor, and anti-aging effects. The present review is trying to summarize the extraction process of polysaccharides of Fructus Corni, structural features and related patents.

The present study reports on the extraction of phenolic compounds from sparganii rhizome. Box-Behnken Design (BBD), a widely used form of response surface methodology (RSM), was used to investigate the effect of process variables on the ultrasound-assisted extraction (UAE). Three independent variables including ethanol concentration (%), extraction time (min) and solvent-to-material ratio (mL/g) were studied. The results showed that the optimal UAE condition was obtained with an ethanol concentration of 75.3%, an extraction time of 40min and a solvent-to-material ratio of 19.21mL/g for total phenols, and an ethanol concentration of 80%, an extraction time of 33.54min and solvent-to-material ratio of 22.72mL/g for combination of ρ-hydroxybenzaldehyde, ρ-coumaric acid, vanillic acid, ferulic acid, rutin and kaempferol. The experimental values under optimal conditions were in good consistent with the predicted values, which suggested UAE is more efficient process as compared to solvent extraction.

OBJECTIVE: To establish a method for determining anticoagulation potency of Sparganii Rhizoma, and evaluate the effect of Sparganii Rhizoma herbs from different producing areas on promoting blood circulation and removing blood stasis; and study the material basis of Sparganii Rhizoma through the correlation analysis on its anticoagulation potency, ferulic acid and total flavonoid content. METHOD: The anticoagulation time of Sparganii Rhizoma from different producing areas with activeated partial thromboplastin time for their active extracts. Their biopotency was calculated by using the method of "parallel lines of dose effect" (3, 3). The degree of correlation between their anticoagulation potency and chemical constituents were calculated by using Pearson correlational analysis method. RESULT: Sparganii Rhizoma and is control drugs had a good linear relationship between dose and effect (Y = 172.76X - 193.39, R2 = 0.9955). The method had better accuracy (RSD 4.7%), repeatability (RSD 2.3%) and intermediate precision (RSD 5.4%), finding that the biopotency of Sparganii Rhizoma from different producing areas ranged between 52.33-238.58 U x g(-1), and all of them passed the test on reliability. The results of correlation analysis showed no remarkable relationship between the anticoagulation potency of Sparganii Rhizoma and the contents of the two chemical constituents. CONCLUSION: This biopotency determination method established in the experiment can be used as one of approaches for qulaity evaluation on Sparganii Rhizoma.
