RESUMEN

Self-cleaning surface-enhanced Raman scattering (SERS) substrates dependent on versatile two-dimensional semiconductors offer an efficient channel for the sensitive monitoring and timely degradation of hazardous molecules. Herein, a kind of sophisticated SERS-active nanocomposites was developed by incorporating Au-Ag nanoparticles onto black phosphorus (BP) nanosheets via photo-induced self-reduction. Combining the substantial electromagnetic "hot spots" triggered by bimetallic plasma coupling effect and the efficient charge transfer from BP to probe molecules, the proposed nanocomposites featured attractive SERS enhancement, facilitating a limit of detection down to 4.5 × 10-10 M. Attributed to the remarkable restriction of electron-hole recombination stemming from "Schottky contact", the photocatalytic activity of BP was prominently boosted, demonstrating a complete degradation time as short as 65 min. Furthermore, the disgusting instability of BP was considerably hindered by inserting the nanocomposites into various bilayer matrices with diverse hardness and viscosity inspired by cling film principle. Moreover, a significantly elevated collection rate high to 93.1% for in-situ detection was also achieved by the as-manufactured flexible SERS chips based on tape. This study illustrates a clear perspective for the development of versatile BP-based SERS chips which might facilitate sensitive analysis and treatment of perilous contaminants in complicated real-life scenarios.

It is tremendously desirable for the timely and effective detection of cancer to facilitate the ultra-highly sensitive monitoring of tumor marker in clinical serum sample. In this study, an electromagnetic and chemical synergistically enabled recyclable immunoassay based on surface-enhanced Raman scattering (SERS) was proposed by realizing the anisotropic growth of sea-urchin-like gold nanoflowers (Au NFs) on two-dimensional red phosphorus (RP) nanoplates. Besides the achieved enhancement factor as high as 2.24 × 106, it was found that the photocatalytic and SERS activities were kept at a high level for the hybrid substrate of RP/Au NFs throughout 7 cycles of immunoassay. In combination with a non-metallic immunoprobe, the limit of detection was drove to 7.41 × 10-5 IU·mL-1 for cancer antigen 19-9. The comparative experiments of nonspecific monitoring verified the promising selectivity of this strategy. Considering the intriguing features of high sensitivity, recyclability, and specificity, the proposed multifunctional RP/Au NFs exhibited its superior role in the early detection of cancer and can be adapted for point-of-care diagnosis.

Sensory-guided analysis of the volatile fraction isolated from a freshly prepared green tea beverage (Camellia sinensis; type Jingshan cha) revealed 58 odor-active compounds after application of an aroma extract dilution analysis. Among them, 3-methylnonane-2,4-dione, (Z)-1,5-octadien-3-one, 3-(methylthio)propanal, trans-4,5-epoxy-(E)-2-decenal, methanethiol, dimethyl sulfide, and indole appeared with the highest flavor dilution factors. A quantitation of 42 aroma compounds by means of stable isotope dilution assays followed by the calculation of odor activity values (OAV; ratio of concentration to odor detection threshold) showed 27 key aroma compounds with OAVs ≥ 1. By far, the highest OAV of 458 was calculated for the asparagus-like/putrid smelling dimethyl sulfide followed by (E,E)-2,4-heptadienal (46). Finally, an aqueous recombinate containing all 27 aroma compounds in the concentrations measured in the beverage successfully mimicked the overall aroma profile of the tea infusion. Quantitative measurements were then performed on authentic tea material to elucidate changes in key aroma compounds during each processing step (fresh leaves, withering, pan-firing, rolling, and drying). The results indicated that dimethyl sulfide, one of the important aroma compounds, was significantly increased by withering of the fresh leaves, however, a major part was lost during drying. Linalool, geraniol, and hexanal showed the highest concentrations in the fresh tea leaves, while significantly lower concentrations were measured in the final tea. The same was observed for all lipid degradation products, such as (E,E)-2,4-heptadienal.

Two natural homogalacturonan (HG) pectins (MW ca. 20 kDa) were isolated from green tea based on their immunomodulatory activity. The crude tea polysaccharides (TPS1 and TPS2) were obtained from green tea leaves by hot water extraction and followed by 40% and 70% ethanol precipitation, respectively. Two homogenous water soluble polysaccharides (TPS1-2a and TPS1-2b) were obtained from TPS1 after purification with gel permeation, which gave a higher phagocytic effect than TPS2. A combination of composition, methylation and configuration analyses, as well as NMR (nuclear magnetic resonance) spectroscopy revealed that TPS1-2a and TPS1-2b were homogalacturonan (HG) pectins consisting of a backbone of 1,4-linked α-D-galacturonic acid (GalA) residues with 28.4% and 26.1% of carboxyl groups as methyl ester, respectively. The immunological assay results demonstrated that TPS1-2, which consisted mainly of HG pectins, showed phagocytosis-enhancing activity in HL-60 cells.

Two homogeneous water-soluble polysaccharides (TPSR4-2B and TPSR4-2C) were obtained from preinfused green tea. Their average molecular weights were estimated to be 41 kDa and 28 kDa, respectively. A combination of composition, methylation, and configuration analysis, as well as NMR spectroscopy, indicated that both TPSR4-2B and TPSR4-2C were poly-(1-4)-α-d-galactopyranosyluronic acid in which 30.5 ± 0.3% and 28.3 ± 0.5%, respectively, of uronic acid existed as methyl ester. Two sulfated derivatives (Sul-R4-2B and Sul-R4-2C) from TPSR4-2B and TPSR4-2C were prepared after sulfation with a 2:1 chlorosulfonic acid-pyridine ratio. The anticomplementary assay showed that Sul-R4-2B and Sul-R4-2C demonstrated a stronger inhibitory effect on the complement activation through the classic pathway, compared to that of heparin. Preliminary mechanism studies by using complement component depleted-sera indicated that both Sul-R4-2B and Sul-R4-2C selectively interact with C1q, C1r, C1s, C2, C5, and C9 but not with C3 and C4. The relationship between DS and the anticomplementary activity of sulfated derivatives of homogalacturonans showed that low sulfated derivatives of homogalacturonans also exhibited potent anticomplementary effect, which might greatly reduce the side effects related to heparin and oversulfated chondroitin sulfate, such as anticoagulant activity and allergic-type reaction. These results suggested that sulfated derivatives of homogalacturonans might be promising drug candidates for therapeutic complement inhibition.

OBJECTIVE: In the Chinese Pharmacopoeia (2010 edition), the minimum limit of verbascoside (acteoside) in Rehmanniae Radix Praeparata (RRP) was set at 0.20%, the rationality of the standard was evaluated in this paper. METHODS: 10 samples of Rehmanniae Radix (RR) and RRP were collected or prepared. According to the traditional method, different samples of RRP were prepared from the same batch of RR, and the content of verbascoside was determined by HPLC. RESULTS: The average content of verbascoside in RR and RRP was 0.0226% and 0.0097%, respectively, and their difference was up to the level of P < 0.01. The content of verbascoside was found decreased with processing time increasing. Long-time storage also results in substantial loss of verbascoside. CONCLUSION: Processing results in verbascoside decreasing significantly. It is unreasonable that RR and RRP have the same standard in the content of verbascoside. It is suggested that the maximum limit of verboscoside should be set in the new version of Chinese pharmacopoeia.
