RESUMEN

Allergic rhinitis (AR) is a noninfectious inflammatory disease seriously affecting the quality of life. This study aimed to assess the efficacy of the Tuomin Zhiti decoction in allergic rhinitis and to provide a reference for clinical treatment. One hundred patients with AR treated in the Department of Otolaryngology of our hospital from January 2019 to December 2020 were recruited and assigned via a random number table method (1 : 1) to receive either oral loratadine and mometasone nasal spray (control group) or the Tuomin Zhiti decoction plus oral loratadine and mometasone nasal spray (study group). The total clinical efficacy was 86% (43/50) in the study group, which was significantly higher than that of 64% (32/50) in the control group. After treatment, the Total Nasal Symptoms Scores (TNSS) and Rhinoconjunctivitis Quality of Life Questionnaire (RQLQ) scores between the two groups were similar, but 12 weeks after treatment, the study group had significantly lower TNSS and RQLQ scores than the control group. After treatment, the study group obtained lower levels of interleukin (IL)-4 and higher levels of interferon-γ (IFN-γ) than the control group. Significantly lower post-treatment peripheral blood eosinophil count (EOS) and eosinophil cationic protein (ECP) levels were observed in the study group in contrast to those of the control group. The Tuomin Zhiti decoction for the treatment of AR patients alleviates their clinical symptoms, reduces the inflammatory responses, enhances the immune function of patients by regulating IL-4 and IFN-γ, and lowers the long-term recurrence rate.

Clinical studies have shown that large doses of prednisone could lower serum uric acid (SUA) in patients with decompensated heart failure (HF); however, the optimal dose of prednisone and underlying mechanisms are unknown. Thirty-eight patients with decompensated HF were randomized to receive standard HF care alone (n = 10) or with low-dose (15 mg/day, n = 8), medium-dose (30 mg/day, n = 10), or high-dose prednisone (60 mg/day, n = 10), for 10 days. At the end of the study, only high-dose prednisone significantly reduced SUA, whereas low- and medium-dose prednisone and standard HF care had no effect on SUA. The reduction in SUA in high-dose prednisone groups was associated with a significant increase in renal uric acid clearance. In conclusion, prednisone can reduce SUA levels by increasing renal uric acid clearance in patients with decompensated HF.

RESUMEN

Five new naturally occurring natural products, including two atisine-type diterpene alkaloids (1 and 2), two atisane-type diterpenes (3 and 4), and a new natural product spiramine C2 (5), along with nine known ones (6-14), were isolated from the ethanolic extracts of the whole plant of Spiraea japonica var. acuminata Franch. Their structures were elucidated by extensive spectroscopic analysis. The anti-tobacco mosaic virus (TMV) activities of all the compounds were evaluated by the conventional half-leaf method. Six compounds (2, 3, 6, 7, 11, and 12) exhibited moderate activities at 100 µg/mL with inhibition rates in the range of 69.4-92.9%, which were higher than that of the positive control, ningnanmycin. Their preliminary structure-activity relationships were also discussed.

Five new compounds (1-5), including three limonoids, one diterpenoid, and one phytosterol, along with four known limonoids (6-9), were isolated from the ethanolic extracts of whole plants of Munronia henryi. Their structures were elucidated by extensive spectroscopic analysis. In addition, the anti-tobacco mosaic virus (TMV) activities of all the isolated compounds were evaluated by the conventional half-leaf and leaf-disk methods along with Western blot analysis. Most of the tested compounds showed strong antiviral activities, with IC50 values in the range of 14.8-34.0µg/mL, compared with ningnanmycin as a positive control (IC50=44.6µg/mL), and their preliminary structure-activity relationships were also discussed.

Column chromatographies over silica gel, Sephadex LH-20, reverse phase C18, and MCI, and semi-preparative HPLC were used for separation and purification of constituents from Inula cappa. The 22 compounds were obtained and their strutures were determined by NMR and MS spectra data as nine flavonoids: luteolin (1), apigenin (2), chrysoeriol (3), artemetin (4), 2', 5-di- hydroxy-3, 6, 7, 4', 5'-pentamethoxyflavone (5), chrysosplenol C (6), apigenin-5-0-ß-D-glucopyranoside (7), luteolin-3-methyl, luteolin-3-methylether-4'-0-ß-D-glucopyranoside (8), luteolin-4'-0-ß-D-glucopyranoside (9); four triterpenes: darma-20, 24-dien- 3ß-0-acetate (10), darma-20, 24-dien-3ß-ol (11), epirfiedelanol (12), friedelin (13); three coumarins: scopoletin (14) , isosco- poletin (15) , scopolin(16) , and other types of compounds stigmasta-5, 22-dien-3ß-0-7-one (17), stigmasterol (18), palmitic acid (19), linoleic acid (20), linoleic acid methyl ester (21), (E) -9, 12, 13-trihydroxyoetadee-10-enoie acid (22). Compound 5 is a new natural product. Compounds 3-9, 15, 17, 21, and 22 were isolated from this genus for the first time.

Fourteen new limonoids, munronins A-N (1-14), and eight known limonoids (15-22) were isolated from the whole plants of Munronia henryi. The structures of the new compounds were elucidated by 2D NMR spectroscopy and mass spectrometry, and the structure of 8 was confirmed by single-crystal X-ray diffraction analysis. Compound 1 represents the first limonoid found with a novel 7-oxabicyclo[2.2.1]heptane moiety produced by incorporating C-11 and C-14 via an oxygen atom. All compounds were evaluated for their anti-tobacco mosaic virus (TMV) activity and in vitro cytotoxicity against the human cancer HL-60, SMMC-7721, A-549, MCF-7, and SW-480 cell lines. Among them, compounds 2, 8, 9, 10, 11, 12, 18, and 20 showed significant anti-TMV activity, with IC50 values in the range 19.6-44.4 µg/mL. Compounds 1 and 18 exhibited cytotoxic effects for all five cancer cell lines, with IC50 values between 0.4 and 4.8 µM.

Schisanhenol (Sol) was isolated from Schisandra rubriflora, and a series of derivatives (1-16, 15a-16a, and 15b-16b) were designed and prepared by chemical modification. The curative and protective effects of these dibenzocyclooctadiene lignan analogues against tobacco mosaic virus (TMV) were evaluated. Most analogues exhibited stronger protective effects than the positive control ningnanmycin. Dibromoschisanhenol (6) at 0.25mM exhibited the strongest protective activity (83.5±1.8% at 0.25mM), and 14-(3, 5-dibenzyloxy)-benzoyloxyschisanhenol (16) showed a significant curative effect (78.0±3.8% at 0.15mM) that was much stronger than that of the commercial virucide ningnanmycin. This study is the first to demonstrate that natural dibenzocyclooctadiene lignans and analogues are active against plant viruses.

To investigate their cytotoxicity, seventeen C21-steroidal pregnane sapogenins 1-17 were isolated from the hydrolytic extracts of the roots of Cynanchum wilfordii. Among them, sapogenins 1-7 are new compounds, whose structures were determined by extensive analysis of spectroscopic data and X-ray crystallographic analysis. Especially, sapogenins with salicyl or vanilloyl group, and a new aberrant pregnane skeleton with ether linkage between C-12 and C-20 were found for the first time. Compound 1 revealed significant cytotoxicities on HL-60 (IC50 6.72µM) and MCF-7 (IC50 2.89µM), and compounds 14 and 15 also revealed strong inhibitory activities against K-562 (IC50 6.72µM) or MCF-7 (IC50 2.49µM), respectively.

Hydrogen sulfide (H2S) is a signaling gasotransmitter, involved in various physiological and pathological processes. H2S-donating drugs have been tested to conjugate the beneficial effects of H2S with other pharmaceutical properties. It has been shown that the endogenous cystathionine-γ-lyase (CSE)/H2S pathway participates in myocardial ischemia injury in isolated hearts in rats. The present study aimed to investigate the cytoprotective action of H2S against acute myocardial ischemia injury in rats. Isolated rat hearts were perfused and subjected to ischemic conditions for 4 h. The hearts were assigned to five groups: Sham, model, infarct plus low-dose (5 µmol/l) NaHS, infarct plus middle-dose (10 µmol/l) NaHS and infarct plus high-dose (20 µmol/l) NaHS. The administration of NaHS enhanced the activity of CSE, increased the content of H2S and reduced infarct volumes following myocardial ischemia injury. Furthermore, the administration of NaHS attenuated the injury to organelles (including the mitochondria, nucleus and myofilaments) by reducing lactate dehydrogenase activity, decreasing the level of mitochondrial malondialdehyde and increasing the activities of superoxide dismutase and glutathione peroxidase in the ischemic myocardial mitochondria. These protective effects of H2S against myocardial ischemia injury appeared to be mediated by its antioxidant activities and the preservation of mitochondrial function.

AIM: To study the bufadienolides in the Chinese traditional drug "Ch'an Su" and their cytotoxic activity. METHOD: Various chromatographic techniques were used to isolate the constituents, and their structures were elucidated through physical and spectroscopic data. RESULTS: Twenty compounds were isolated, and eighteen were evaluated in vitro for their cytotoxic activity against A-549 and K-562 cells. CONCLUSION: Compound 1 (bufalin 3ß-acrylic ester) was a new bufadienolide and exhibited the most potent activity against the two tumor cell lines with IC50 values of 7.16 and 6.83 nmol · L(-1). The relationships between structure and activity are discussed.

Four new triterpenoids, named Toosendansins A-D (1-4), along with nine known ones (5-13) were isolated from the fruits of Melia toosendan Sieb. et Zucc. Their structures were established on the basis of spectroscopic data. The isolation of compounds 1-12 were reported for the first time from this plant. All compounds were analyzed for the anti-Tobacco Mosaic Virus (TMV) activity and protective effect on H2O2-induced damage of SH-SY5Y cells. Compound 7 showed evident anti-TMV activity. Compounds 2 and 9 exhibited modest protection against H2O2-induced damage of SH-SY5Y cells.

RESUMEN

A new hasubanan alkaloid, hernsubanine E (1), as well as two known compounds p-hydroxybenzaldehyde (2) and (-)-syringaresinol (3) have been isolated from the whole plants of Stephania hernandifolia by various column chromatographic methods. Their structures were identified by physicochemical properties and spectral analyses. Compounds 2 and 3 were isolated from the genus of Stephania for the first time.

Fifteen new seco-pregnane steroidal glycosides cynanosides A-O (1-15) together with twenty-seven known ones were isolated from the roots of Cynanchum atratum. The structures of 1-15 were determined by extensive analysis of spectroscopic data. The anti-tobacco mosaic virus (TMV) activity of these steroidal glycosides was screened by the conventional half-leaf method, enzyme-linked immunosorbent assay, and Western blot methods, most of them showed potent anti-TMV activity. Among them, compounds 1, 7, 13, 28 and 31 showed significantly anti-TMV activity with an IC50 value of 20.5, 18.6, 22.0, 19.2 and 22.2 µg/mL, respectively, and were much more effective than the positive control, ningnanmycin (IC50=49.6 µg/mL).

Five new limonoids, named munronoids K-O (1-5), together with three known limonoids were isolated from Munronia unifoliola Oliv. These limonoids were involved in the skeletons of evodulone, gedunin, and peieurianin types of limonoids, and their structures were established on the basis of spectroscopic data. Compound 5 featuring a γ-lactone ring instead of the ß-substituted furan ring was found in the peieurianin type for the first time. The antitobacco mosaic virus (anti-TMV) activities of compounds 1-8 were also evaluated with half-leaf, enzyme-linked immunosorbent assay, and Western blot methods, and limonoids 1, 5, and 8 showed stronger anti-TMV treatment activities than the positive control ningnanmycin. Six compounds (1-5 and 8) exhibited infection inhibition activities against TMV.

OBJECTIVE: To investigate the effect of hydrogen sulfide (H2S) on mitochondrial function in acute myocardial ischemia in rats. METHODS: Acute myocardial ischemia models were established by ligating the left anterior descending coronary artery (LADC) of rats. Fourty-eight male SD rats were randomly divided into 6 groups (n = 8): sham operation group, ischemia group, ischemia + sodium hydrosulfide (NaHS) low, middle and high dose groups and ischemia + DL-proparglycine(PPG) group. The ultrastructures of myocardial mitochondria were observed with electron microscope. The content of H2S in plasma and the activity of cystathionine-gamma-lyase (CSE) in myocardial tissue of rats were respectively detected. The swelling and activity of myocardial mitochondria were determined. The activities of ATPase, GSH-Px, SOD and the content of malondial-dehyde (MDA) in myocardial mitochondria of rats were also measured. RESULTS: Compared with those of the sham operation group, the content of H2S in plasma, the activity of CSE in myocardial tissue and the activity of myocardium mitochondria were significantly decreased. The activities of ATPase, SOD, GSH-Px in myocardial mitochondria were significantly decreased, The content of malondial dehyde(MDA) in myocardial mitochondria and the swelling of mitochondria were distinctly increased in the ischemia group (P < 0.01). Compared with those of the ischemia group, the content of H2S in plasma and the activity of CSE in myocardial tissue were increased, and the activities of mitochondria, ATPase, SOD, and GSH-Px in myocardial mitochondria were significantly increased in ischemia + NaHS low, middle and high-dose groups; the swelling of mitochondria and the content of MDA in myocardial mitochondria were significantly decreased in ischemia + NaHS middle and high-dose groups (P < 0.05 or P < 0.01). The administration of PPG could partially reduce the myocardial protection of hydrogen sulfide (P < 0.05 or P < 0.01). CONCLUSION: It could be concluded that the administration of hydrogen sulfide could enhance the activities of mitochondrial ATPase, SOD, GSH-Px, decrease the level of mitochondrial lipid peroxidation, and play a protective effect against acute myocardial ischemia.

Three new diterpenoids, ingenol-5ß,20-O,O-isopropylidene-3ß-palmitate, ingenol-5ß,20-O,O-isopropylidene-3ß-myristinate and 3ß,19-dihydroxy-1(10),15-rosadien-2-one, were isolated from the roots of Euphorbia ebracteolata Hayata. Their structures were deduced by spectroscopic means and analytic techniques.

Adverse reactions induced by Chinese herbal injections have been frequently reported. However, the precise causes of these adverse reactions are not yet fully understood. The aim of the present study was to determine the role of chlorogenic acid (a ubiquitous component of Chinese herbs) in the toxicity of Chinese herbal injections. Beagle dogs were given chlorogenic acid, Yuxingcao injection, or Qingkailing injection (the latter two both containing chlorogenic acid) by intravenous (i.v.) injection, once a day for 7 or 9 days. The systemic toxicity was evaluated. An additional ultrastructural observation on liver and kidney was performed. Anaphylactoid reactions were obvious in dogs treated with Yuxingcao injection. Varying degrees of ultrastructural changes in liver and kidney were observed in the treated dogs, especially in dogs treated with Chinese herbal injections. Our study has led to the view that chlorogenic acid is not an allergen when administrated by i.v. injection, but liver and kidney injury induced by Chinese herbal injections can be partly attributed to chlorogenic acid.

RESUMEN

OBJECTIVE: To study the chemical constituents from the aerial part of Euphorbia chrysocoma. METHODS: All compounds were isolated and purified by many methods, including siliga gel and reversed phase RP-18 column chromatographies, preparative thin layer chromatography, Sephadex LH-20, and recrystallization. Their structures were mainly elucidated by ESI-MS and NMR spectra and their physical characters. RESULTS: Six compounds were isolated from the petroleum ether section from 75% ethanol extraction of the material. Their structures were identified as taraxerol (1), epitaraxerol (2), beta-sistosterol (3), beta-sitostenone (4), jolkinolide E (5), and sesamin (6). CONCLUSION: Compounds 1, 2, 4, 5, and 6 are isolated from this plant for the first time.

The distributing and changing characteristics and content of soil heavy metals was studied using methods of field survey and sampling, indoor analysis, and pollution index were used to investigate the soil environmental quality in the tea plantations of Changsha Baili Tea Zone. The results showed that the content of soil total Pb, Hg, Cd, Cr, As, Ni basically was in the soil background value, their averages were 42.7, 0.068, 0.074, 92.2, 12.4 and 19.5 mg/kg respectively. The content of heavy metals was lower than the standard of Environmental Qualification of Nuisance Free Tea Producing Area (NY 5020-2001). Simultaneity, soil environmental quality in tea plantations of Baili Tea Zone, it answered for the second grade of State Environmental Quality Standard for Soils (GB 15618-1995), achieved safe class, and the content of soil Hg, Cd, Ni accorded to the first grade of GB 15618-1995, these results showed the Changsha Baili Tea Zone were propitious to develop nuisance free tea production. Besides the single pollution index of Cr was 0.837 and in warning class at the tea plantation of Xiangfeng, the others were all less than 0.7, and in the safe class. The integrated index of 6 tea bases was all less than 0.7, in the safe class, the soil environmental quality was cleanness on the whole at Changsha Baili Tea Zone, and the soils were suitable for non-polluted agricultural production.
