Objective: To observe the effects of estradiol on expression of plasma membrane Ca2+-ATPase isoform 2 in inner ear of rats. Methods: Twenty-five Three-months-old female Sprague-Dawley rats were randomly and equally divided into five groups by random number table mathod,with five rats in each group. Animals in Sham group were sham-operated while others were bilateral ovariactmized. One month after modeling, the OVX groups were supplemented with estradiol (E2 group), progesterone (P group), estradiol and progesterone (E2+P group)and vehicle sesame oil (Veh group), while the Sham operation group (Sham group) was supplemented with vehicle sesame oil.All rats were sacrificed and otocysts were obtained immediately. Enzyme-linked immunosorbent assay was used to detect the changes in serum estradiol and progesterone levels of each group of rats before operation, before treatment and before sacrifice. Western blot and quantitative real-time polymerase chain reaction were used to detect the expression of total PMCA2 protein and mRNA in the inner ear of each group. Results: There was no significant difference in serum estradiol and progesterone levels among the five groups before operated(P>0.05). Before treatment, the serum estradiol and progesterone levels of rats in each group were significantly lower than those in Sham group (P<0.05). The serum estradiol level in E2 group and E2+P group was not significantly different from that in Sham group (P>0.05), while the serum estradiol level in P group and Veh group was significantly different from that in Sham group (P<0.05). The level of progesterone in P group and E2+P group was higher than that in Sham group (P<0.05), while the level of progesterone in Veh group and E2 group was lower than that in Sham group (P<0.05). Protein and mRNA expression of PMCA2 in P and Veh groups were significantly decreased compared with that of Sham group (P<0.05) while the expression levels underwent no significantly change in E2 and E2+P groups (P<0.05). Conclusion: The decrease of serum estradiol level can reduce the expression of otolith regulatory protein PMCA2 in rats, and then affect otolith metabolism, which may be an important link of estrogen affecting otolith metabolism.

Objective: To analyze the data of the Bethesda system for reporting thyroid cytopathology applied in a comprehensive cancer center and to evaluate the diagnostic ability of fine needle aspiration (FNA). Methods: We retrospectively reviewed the medical records of 5 729 cases applying this reporting system at Cancer Hospital, Chinese Academy of Medical Sciences. The series were from 5 011 patients including 1 174 men and 3 837 women, and their median age was 45 years (range, 7-88 years). FNA results were correlated with final histological diagnosis after surgery and the accuracy of FNA diagnosis and the malignancy rates for each of categories were also analyzed. Results: Among 5 729 thyroid aspirates, aside from 456 (8.0%) cases with nondiagnostic or unsatisfactory (ND/UNS) outcomes, 1 055 (18.4%) cases were benign, 409 (7.1%) cases showed atypical of undetermined significance or follicular lesions with undetermined significance (AUS/FLUS), 80 (1.4%) cases were follicular neoplasm or suspicious for follicular neoplasm (FN/SFN), 982 (17.1%) cases were suspicious for malignancy (SUS), and 2 747 cases were malignant (47.9%). Of 5 729 cases, 3 239 had received thyroidectomies after FNA, 95.99% of them were proven histologically to be malignant, with following malignancy rates in individual FNA categories: ND/UNS 75.00%; benign 40.91%; AUS/FLUS 77.67%; FN/SFN 41.67%; SUS 96.86%; and malignant 99.96%. FNA predicted malignancy with sensitivity, specificity, accuracy, positive predictive value and negative predictive values of 98.8%, 60.5%, 97.7%, 98.9% and 59.1%, respectively. Conclusions: The data of the Bethesda reporting system indicates high proportion of malignant diagnosis and high risk of malignancy at all FNA diagnostic categories. FNA offers high diagnostic accuracy and positive predictive value for the diagnosis of thyroid diseases.

Bauxite mining on karst generates numerous ecological and environmental problems, including metal pollution, water and soil erosion and destruction of vegetation. Among these, the most important environmental problem is soil metal pollution. Higher plants have a great ability to adsorb metals and can be used as biological indicators. However, the study of bioindicators for soil contamination in karst bauxite is not clear. Plants and their soil were collected from an abandoned karst bauxite area at Shangdong, Guizhou Province, southwestern China. Plants were collected and identified as Pteris vittata, Pinus massoniana, Miscanthus floridulus, Coriaria nepalensis, Artemisia argyi and Senecio scandens. The content of metals in plant roots were in the order: P. vittata > M. floridulus > C. nepalensis, other plants roots had no consistent pattern. Concentrations of metal in plants (P. vittata and M. floridulus) and soil were: soil > root > leaf > stem. Levels of metals in soil samples easily exceeded background values, indicating that soil had been contaminated. Al and Fe were highest in soil samples of P. vittata, with a good correlation. Results show that the metal content determined in plants is relatively high, particularly in P. vittata. Data also suggest that P. vittata colonies were able to tolerate and accumulate high levels of metal elements, which evidences their suitability for use as bioindicatord of soil metal contamination caused by mining activities.

Objective: To investigate the clinicopathological features, diagnosis and differential diagnosis of dedifferentiated liposarcoma (DDLPS) with inflammatory myofibroblastic tumor (IMT)-like features. Methods: Five cases of DDLPS with IMT-like features were collected from the First Affiliated Hospital of Nanjing Medical University, the Affiliated Hospital of Nanjing University of Traditional Chinese Medicine and the First People's Hospital of Qinzhou between 2013 and 2018. EnVision method and fluorescence in situ hybridization (FISH) were used to detect the immunophenotype of the tumor cells and the profile of MDM2 gene amplification respectively. Results: All five cases were male and the median age was 61 (range 53 to 65) years. The clinical symptoms were mainly related to the space-occupying lesions. The tumors were located in duodenal mesentery (two cases), intestinal wall (one case), retroperitoneum (one case), and spermatic cord (one case). Grossly, the tumors were not well encapsulated, ranging from 3 to 13 cm (median 6.7 cm) in diameter, with tan to gray and firm cut surface. Histologically, the dedifferentiated component closely resembled inflammatory myofibroblastic tumor (IMT), with spindle/polygonal/stellate-shaped cells arranged in storiform, sheet-like, or random pattern, with varying degrees of chronic inflammation and fibrosis. All three major patterns seen in IMT (myxoid, cellular and hypocellular fibrous) were observed, the hypocellular fibrous pattern was the most common. Well-differentiated liposarcomatous component was found in the peripheral areas of all the tumors. One case had high grade dedifferentiated component. Four cases were strongly positive for MDM2 and p16. Two cases were positive for SMA, and one case was focally positive for desmin and one for CD34. None of the cases stained for ALK-1. FISH demonstrated MDM2 gene amplification in all five cases. Clinical follow-ups were available in all five cases and the interval ranged from 3 to 66 months (median 23 months). Two patients developed recurrences and one patient had metastasis. The remaining two patients were alive with no evidence of tumor recurrence at 3 and 14 months after surgery respectively. Conclusions: DDLPS with IMT-like features is a more aggressive neoplasm than its histological mimic (IMT), and should not be misdiagnosed as other intermediate or low-grade malignant tumors, such as IMT, sclerosing liposarcoma, inflammatory liposarcoma, aggressive fibromatosis, solitary fibrous tumors, low-grade myofibroblastic sarcoma, and low-grade fibrosarcoma.

OBJECTIVE: We aimed at studying the effect of adjuvant therapy with low-dose vitamin A on the function of T lymphocytes in neonatal pneumonia. PATIENTS AND METHODS: We recruited 60 cases of neonatal pneumonia which were randomly divided in two equal groups. The control group was treated with conventional anti-inflammatory therapy and aerosol inhalation. The experimental group received oral vitamin A soft capsules for 7 days. RESULTS: Pre-treatment levels vitamin A level and vitamin A deficiency disorders (VADD) percentage revealed no differences between the two groups. The treatment course for the experimental group was shorter than the control group. Serum IgM, IgG, and glutathione peroxidase (GSH-Px) levels were increased, whereas the levels of malondialdehyde were decreased in the experimental group after treatment. The control group showed no changes in these factors. After treatment, both groups showed increased percentages of CD4+ and CD8+ T cells, but the experimental group showed a larger increase. CONCLUSIONS: Neonatal pneumonia is often accompanied by a low level of vitamin A, and adjuvant therapy can shorten its disease course, improve IgM and IgG levels, and improve anti-oxidative and cellar immune function.

Soil erosion in karst areas not only destroys the natural environment but also accelerates the decline in land productivity together with the associated increase in poverty for local communities. There are no simple or straightforward answers to controlling soil erosion on karst. Such erosion has become a serious problem in China. This study addresses both the diversity of vegetation (mosses, herbs, shrubs and trees) on karst and the underlying soil chemical characteristics, in order to provide a scientific basis by which suitable plant species can be selected for recovery and restoration of karst degraded by soil erosion. Vegetation diversity and soil chemistry were assessed in areas with five different grades of soil erosion in Guiyang Karst Park, Guizhou, China. Mosses are more tolerant than vascular plants of soil erosion and associated environmental degradation of karst and the order of species diversity was: mosses > herbs > shrubs > trees. Mosses were found to play a major role in ecological restoration of microhabitats on karst. Soil microbial biomass carbon was found to be significantly higher in soil under mosses than in bare soil associated with other plant categories. Mosses were more effective in converting unavailable potassium to available potassium. Vascular plants were found to have a positive effect on total nitrogen fixation and the availability of phosphorus in the soil. Increasing soil degradation was associated with lower levels of total Nitrogen in soil underlying mosses than in soil underlying vascular plants. Thus, based on their different but complementary contributions to soil chemistry, mosses and vascular plants in combination can provide the most practical outcome for the repair and restoration of areas of karst affected by soil erosion. The combination of the moss species, Homomallium plagiangium, Cyrto-hypnum pygmaeuman and Brachythecium perminusculum, the herbs Veronica arvensis and Youngia japonica, and the tree Prunus salicina, are recommended as suitable pioneer plant species to cultivate for use in restoration of regions of karstic soil erosion.

Two experiments were conducted to evaluate effects of coated compound proteases (CC protease) on apparent total tract digestibility (ATTD) of nitrogen (N) and energy, and apparent ileal digestibility (AID) of amino acids (AA) and nutrients in diets for pigs. In Exp. 1, 12 crossbred barrows (initial body weight: 20.14±1.71 kg) were housed in individual metabolism crates and allotted into 2 treatments with 6 piglets per treatment according to weight in a randomized complete block design. The 2 diets were corn-soybean meal basal diets with (0.2 g/kg) or without CC protease supplementation. The CC protease supplementation increased (p<0.05) the digestible and metabolizable N and energy values and the digestibility and retention rate of N in the diet. The ATTD of energy and nutrients had been improved (p<0.05) in the diet supplemented with CC protease. In Exp. 2, 12 crossbred barrows (initial body weight: 20.79±1.94 kg), fitted with T-cannulas at the distal ileum, were blocked by body weight into 2 groups with 6 pigs each. The diets were the same as those in Exp. 1. The CC protease increased (p<0.05) the AID of crude protein and some essential AA including arginine, isoleucine and leucine. The AID and ATTD of energy and nutrients had been improved (p<0.05) by supplemental CC protease, but the hindgut digestibility of nutrients was unaffected. Overall, the CC protease improved the ATTD of N and energy and AID of some indispensible AA and nutrients in the corn-soybean meal diet for pigs. Therefore, the CC protease supplement could improve the utilization of protein in the corn-soybean meal diet and thus contribute to lower N excretion to the environment.

Systemic administration of tumour necrosis factor (TNF)-alpha induces the release of norepinephrine in the paraventricular nucleus (PVN) of hypothalamus and an increase in expression of corticotrophin-releasing factor (CRF) and CRF type 1 receptors. We explored the hypothesis that CRF and norepinephrine in PVN mediate the cardiovascular and sympathetic responses to acute systemic administration of TNF-alpha. In anaesthetised rats, the increases in arterial pressure and heart rate induced by intracarotid artery injection of TNF-alpha were attenuated by intracerebroventricular (ICV) injection of either the alpha 1-adrenergic antagonist prazosin or the CRF antagonist alpha-helical CRF. Prazosin blocked the TNF-alpha-induced increase in renal sympathetic nerve activity (RSNA), whereas alpha-helical CRF substantially reduced the RSNA response. Conversely, CRF and the alpha 1-adrenergic agonist phenylephrine, administered ICV, both elicited increases in PVN neuronal activity, RSNA, arterial pressure and heart rate. Microinjection of CRF and phenylephrine directly into PVN evoked smaller responses. These results are consistent with the hypothesis that norepinephrine and CRF in the PVN mediate the cardiovascular and sympathetic responses to acute systemic administration of TNF-alpha.

We have reported recently that tissue culture induced a high level of genetic variation at the primary nucleotide sequence in regenerants of medicinal plant Codonopsis lanceolata. It is not known, however, whether epigenetic variation in the form of alteration in DNA methylation also occurred in these plants. Here, we investigated possible alterations in level and pattern of cytosine methylation at the CCGG sites in the same set of regenerants relative to the donor plant, by the MSAP method employing a pair of isoschizomers, HpaII and MspI, which recognize the same restriction site but are differentially sensitive to cytosine methylation at the CCGG sites. A total of 1,674 MSAP profiles were resolved using 39 primer combinations. Of these, 177 (10.5%) profiles were polymorphic among the regenerants and/or between the regenerant(s) and the donor plant, in EcoRI + HpaII or EcoRI + MspI digest but not in both, indicating alteration in cytosine methylation patterns of specific loci, though their estimated total level of methylation remained more or less the same as the donor plant. Gel blot analysis validated most of the variant MSAP profiles as bona fide alteration in methylation patterns. Correlation analysis between the MSAP data and the previously reported ISSR and RAPD data revealed significant correlations, suggesting their possible intrinsic interrelatedness. Thirty-seven typical variant MSAP profiles were isolated and sequenced, of which 5 showed significant homology to known-function genes, 2 to chloroplast sequences, whilst the rest 30 did not find a match in the database.

The contribution of the conflict of 1999 to the environmental levels of uranium in the Balkan area was evaluated by means of lichens used as biomonitors. The average U concentration found in lichens in the present study was in line with the values reported for lichens from other countries and well below the levels found in lichens collected in areas with natural or anthropogenic sources of U. Measurement of isotopic ratios 235U/238U allowed to exclude the presence of depleted uranium. According to these results, we could not detect widespread environmental contamination by depleted uranium in the Balkan area.

Connectivity between the rat posterior insula and the ventrobasal thalamus has been demonstrated anatomically. Neurons convergent for baroreceptor and nociceptive input have also been identified in the homologous anterior insula of the primate. Whether similar convergent cells exist in the ventrobasal thalamus was investigated in 30 urethane anesthetized male Sprague--Dawley rats. Six classes of cells were identified in the right ventrobasal thalamus: (a) 83/159 (52%) baroreceptive and nociceptive convergent units; (b) 2/159 (1%) convergent cells responding to baroreceptor activation and light touch; (c) 44/159 (28%) purely nociceptive units; (d)10/159 (6%) purely baroreceptive units; (e) 1/159 (0.6%) cells responding to brush alone and (f) 19/159 (12%) unresponsive units. Of the viscerosomatic convergent cells, 66/85 (78%) were situated in the ventroposterolateral nucleus (VPL), 6/85 (7%) in the ventroposterolateral parvicellular nucleus (VPLpc), and 13/85 (15%) in the ventroposteromedial nucleus (VPM). Fifteen right ventrobasal thalamic units were antidromically activated and 34 units orthodromically activated by right posterior insular microstimulation. Cobalt injection into the right ventrobasal thalamus blocked the right insular response to baroreceptor activation by >70%. These data indicate: (a) baroreceptive and somatosensory nociceptive convergent units exist in the ventrobasal thalamus; (b) thalamic convergent neurons project directly to the ipsilateral posterior insula and receive reciprocal insulothalamic projections; and (c) a significant proportion of baroreceptor input relays to the posterior insula through the ipsilateral ventrobasal thalamus.

OBJECTIVE: To investigate the resources distribution and morphological histology of Centella asiatica distributed in China, and offer evidences for exploiting the plant resources and drafting the quality standards. METHOD: Sample collection, document investigation and microscopic section observation. RESULT: A table of resources distribution was worked out, the medicinal properties and the microscopic characteristics of the stem, leaf and stipe were described. CONCLUSION: This study supports the viewpoint of Urban and can be used as a reference for the quality standard of Centella asiation.

The Strong fluorescence signals were obtained in pollen tube of Lilium davidii Duch with labeled anti-VnR integrin serum, and anti-beta 3, alpha v integrin subunit cytoplasmic domain serum separately. The highest density of immunolabel was in the tip of pollen tube. There was little or no immunolabel in control experiment using non-immune serum, second antibody alone and anti-FnR, LnR integrin serum separately. In pollen of Prunus persica f. rubro-piena Schneid, fluoresence signals were also obtained in tube using labeled anti-beta 1, beta 3 integrin subunit cytoplasmic domain serum separately and in apertures using anti-beta 1 serum. Preliminary results show that during the germination of pollen and the growth of pollen tube, there may be integrin-like proteins in pollen tube, consisting of alpha v and beta s integria subunits in the pollen tube of Lilium davidii Duch, which is the receptor of vitronectin-like protein.

RESUMEN

The inhibitory effect of selenium (Se) and change of glutathione peroxidase activity during the development of brain tumors was investigated in Wistar rats. Four rat groups classified to match by age and weight were fed a diet containing 0, 0.5, 2.0, and 4.0 micrograms Se/g. After 6 wk, the rats were injected with 3 x 10(6) C6 cells into the right frontal lobe parenchyma. Survival was significantly longer in the 0.5 and 2.0 micrograms Se/g groups than in the Se-free and 4.0 micrograms Se/g groups. The activity of glutathione peroxidase after development of tumors was significantly higher in the high Se group at 18 and 30 d.

RESUMEN

According to the pharmacological results five compounds were isolated from the herb of Commelina communis. Based on physico-chemical constants and spectral data, four of them were identified as n-triacontanol, p-hydroxycinnamic acid, daucosteril and D-mannitol. p-hydroxycinnamic acid shows antibacterial activity and D-mannitol shows antitussive effect.

In the evaluation of Chinese herbs (A), ear-acupuncture (B) and epidural morphine (C) to relieve postoperative pain and abdominal distension, sixteen male patients with primary liver cancer were observed. This study was conducted by means of orthogonal experiment and double blind, randomized design. The patients received various treatments according to the display of the orthogonal table L16(2)15 which corresponds to 2(3) factorial experiment design. C+ (morphine 2 mg) was given before the peritoneum was sutured. A+ (orally administered) and B+ were given 24 hours after operation. 50-100 mg of pethidine was given when the pain intensity VAS (0-100) exceeded 50-70. The observation parameters included plasma leucine enkephalin (LEK), postoperative total dosage of narcotics administered for 5 days, VAS for pain and pain reliever, abdominal distension, urinary retention, constipation, etc. The results were as follows: a. Patients who had received A (A+B+C+, A+B+C-, A+B-C-, A+B-C+); C (C+A+B+, C+A+B-, C+A-B+, C+A-B-), or B (B+A+C+, B+A+C-, B+A-C+, B+A-C-) produced better analgesic effects than those who had received placebo. The A, B, and C reduced narcotics 650, 450 and 550 mg respectively when compared with placebo. The effects of A and C were of statistical significance (P < 0.05), while AB, BC, and AC interactions were not found; b. A and B minimized abdominal distension and urinary retention, while C prolonged them. As compared with the placebo, A and B accelerated restoration of bowel peristalsis (P < 0.05, ANOVA). Both A and B decreased it for 165 hours, while epidural morphine prolonged it for 49 hours; and c.(ABSTRACT TRUNCATED AT 250 WORDS)

In order to assess the clinical availability of early pregnancy detection with Sphygmogram of smooth pulse, pulse pictures and endometrial tissues were simultaneously examined respectively with sphygmograph and pathological method in 205 cases of clinically diagnosed early pregnancy. Among them, 196 cases were pathologically confirmed as early pregnancy, 192 displayed smooth pulse on the Shygmogram. This shows the precision of sphygmogram of smooth pulse in detecting early pregnancy was 97.6%, its sensitivity 97.9%, Jordan index 0.868, specificity 88.9%, and misdiagnosis 11.1%. Though its specificity was not quite satisfactory, sphygmogram of smooth pulse has proved to be a sensitive index with high precision and easy management. Therefore, it is well worth to be popularized in China.
