Selenium nanoparticle (Nano-Se) is a new type of selenium supplement, which can improve the deficiency of traditional selenium supplements and maintain its physiological activity. Due to industrial pollution and irrational use in agriculture, Cu overexposure often occurs in animals and humans. In this study, Nano-Se alleviated CuSO4-induced testicular Cu accumulation, serum testosterone level decrease, testicular structural damage, and decrease in sperm quality. Meanwhile, Nano-Se reduced the ROS content in mice testis and enhanced the activities of T-AOC, GSH, SOD, and CAT compared with CuSO4 group. Furthermore, Nano-Se alleviated CuSO4-induced apoptosis by increasing the protein expression of Cleaved-Caspase-3, Cleaved-Caspase-9, Cleaved-Caspase-12, and Bax/Bcl-2 compared with CuSO4 group. At the same time, Nano-Se reversed CuSO4-induced increase of γ-H2AX protein expression in mice testis. In conclusion, this study confirmed that Nano-Se could alleviate oxidative stress, apoptosis, and DNA damage in the testis of mice with Cu excess, thereby protecting the spermatogenesis disorder induced by Cu.

Inadequate Se intake can enhance vulnerability to certain health risks, with supplementation lessening these risks. This study investigated the bioavailability of Se and Se species in five Se compounds and in Se-rich Cardamine violifolia using in vitro digestion coupled with a Caco-2 cell monolayer model, which enabled the study of Se transport and uptake. Translocation results showed that SeCys2 and MeSeCys had high translocation rates in C. violifolia leaves (CVLs). The uptake rate of organic Se increased with time, and MeSeCys exhibited a higher uptake rate than that for SeCys2 and SeMet. The translocation mechanisms of SeMet, Se(IV), and Se(VI) were passive transport, whereas those of SeCys2 and MeSeCys were active transport. The bioavailability of organic Se was higher than that of inorganic Se, with a total Se bioavailability in CVLs of 49.11 %. This study would provide a theoretical basis for the application of C. violifolia in the functional food.

Pathological vascular remodeling is a hallmark of various vascular diseases. Previous research has established the significance of andrographolide in maintaining gastric vascular homeostasis and its pivotal role in modulating endothelial barrier dysfunction, which leads to pathological vascular remodeling. Potassium dehydroandrographolide succinate (PDA), a derivative of andrographolide, has been clinically utilized in the treatment of inflammatory diseases precipitated by viral infections. This study investigates the potential of PDA in regulating pathological vascular remodeling. The effect of PDA on vascular remodeling was assessed through the complete ligation of the carotid artery in C57BL/6 mice. Experimental approaches, including rat aortic primary smooth muscle cell culture, flow cytometry, bromodeoxyuridine (BrdU) incorporation assay, Boyden chamber cell migration assay, spheroid sprouting assay, and Matrigel-based tube formation assay, were employed to evaluate the influence of PDA on the proliferation and motility of smooth muscle cells (SMCs). Molecular docking simulations and co-immunoprecipitation assays were conducted to examine protein interactions. The results revealed that PDA exacerbates vascular injury-induced pathological remodeling, as evidenced by enhanced neointima formation. PDA treatment significantly increased the proliferation and migration of SMCs. Further mechanistic studies disclosed that PDA upregulated myeloid differentiation factor 88 (MyD88) expression in SMCs and interacted with T-cadherin (CDH13). This interaction augmented proliferation, migration, and extracellular matrix deposition, culminating in pathological vascular remodeling. Our findings underscore the critical role of PDA in the regulation of pathological vascular remodeling, mediated through the MyD88/CDH13 signaling pathway.

Many lines of evidence demonstrate the associations of colorectal cancer (CRC) with intestinal microbial dysbiosis. Recent reports have suggested that maintaining the homeostasis of microbiota and host might be beneficial to CRC patients, but the underlying mechanisms remain unclear. In this study, we established a CRC mouse model of microbial dysbiosis and evaluated the effects of fecal microbiota transplantation (FMT) on CRC progression. Azomethane and dextran sodium sulfate were used to induce CRC and microbial dysbiosis in mice. Intestinal microbes from healthy mice were transferred to CRC mice by enema. The vastly disordered gut microbiota of CRC mice was largely reversed by FMT. Intestinal microbiota from normal mice effectively suppressed cancer progression as assessed by measuring the diameter and number of cancerous foci and significantly prolonged survival of the CRC mice. In the intestine of mice that had received FMT, there were massive infiltration of immune cells, including CD8+ T and CD49b+ NK, which is able to directly kill cancer cells. Moreover, the accumulation of immunosuppressive cells, Foxp3+ Treg cells, seen in the CRC mice was much reduced after FMT. Additionally, FMT regulated the expressions of inflammatory cytokines in CRC mice, including down-regulation of IL1a, IL6, IL12a, IL12b, IL17a, and elevation of IL10. These cytokines were positively correlated with Azospirillum_sp._47_25, Clostridium_sensu_stricto_1, the E. coli complex, Akkermansia, Turicibacter, and negatively correlated with Muribaculum, Anaeroplasma, Candidatus_Arthromitus, and Candidatus Saccharimonas. Furthermore, the repressed expressions of TGFb, STAT3 and elevated expressions of TNFa, IFNg, CXCR4 together promoted the anti-cancer efficacy. Their expressions were positively correlated with Odoribacter, Lachnospiraceae-UCG-006, Desulfovibrio, and negatively correlated with Alloprevotella, Ruminococcaceae UCG-014, Ruminiclostridium, Prevotellaceae UCG-001 and Oscillibacter. Our studies indicate that FMT inhibits the development of CRC by reversing gut microbial disorder, ameliorating excessive intestinal inflammation and cooperating with anti-cancer immune responses.

This study investigated the effects of phenolic acids with different functional groups (cinnamic acid: CIA, caffeic acid: CA, ferulic acid: FA) on corn starch (CS) digestibility by simulating dietary intake patterns (co-heating and non-co-heating) and their mechanism. Both treatments could reduce the digestibility of CS. Compared to the non-co-heating treatment, the resistant starch content of 10 % CA co-heating samples increased by 8.36 %. The co-heating case led to a decrease in the trough viscosity, peak viscosity, and final viscosity of CS. Phenolic acids reduced the short-range order of CS, which was due to the interaction through hydrogen bonding by co-heating. The contribution was most pronounced for CA which contained more hydroxyl groups on the benzene ring. Quartz Crystal microbalance tests further confirmed that different absorption of phenolic acids to CS was caused by their hydroxyl groups on the benzene ring. These results demonstrated that the functional groups of phenolic acids were a controllable factor in inhibiting starch digestion, and co-heating could be considered a promising method to control starch digestion and an advocating way to ingest phenolic supplements.

Understanding the effects of drying on the selenium (Se) content and Se bioaccessibility of Se-rich plants is critical to dietary supplementation of Se. The effects of five common drying methods (far-infrared drying (FIRD), vacuum drying (VD), microwave vacuum drying (MVD), hot air drying (HD), and freeze vacuum drying (FD)) on the content and bioaccessibility of Se and Se species in Cardamine violifolia leaves (CVLs) were studied. The content of SeCys2 in fresh CVLs was the highest (5060.50 µg/g of dry weight (DW)); after FIRD, it had the lowest selenium loss, with a loss rate of less than 19%. Among all of the drying processes, FD and VD samples had the lowest Se retention and bioaccessibility. FIRD, VD, and FD samples have similar effects on antioxidant activity.

Selenium (Se)-enriched peptides were isolated from Cardamine violifolia by enzymatic hydrolysis and ultrafiltration. S3 (molecular weight [MW] distribution of 3-5 kDa) exhibited the strongest inhibitory effect on HepG2 cells and was thus screened using the 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide assay; it was found to have a high organic Se content. Its amino acid sequence was determined using HPLC-MS/MS. We then examined its ability to inhibit tumor cell proliferation and found that it arrested tumor cells in the S phase; moreover, it could induce cancer cell apoptosis. Following S3 treatment, we observed a decrease in mitochondrial membrane potential and an increase in cell calcium content. Upon S3 treatment at 60 µg/ml, the relative activities of caspase-3 and caspase-9 increased by 1.48 times and 2.17 times, and the contents of PI3K and AKT decreased from 2.05 ng/L and 1.95 ng/L to 0.71 ng/L and 0.50 ng/L, respectively, when compared with the control group. Transcriptomic analysis revealed significant changes in the PI3K-AKT pathway following S3 treatment. This study thus established a foundation for additional development of Se-enriched peptides from C. violifolia as a functional food. PRACTICAL APPLICATION: Cardamine violifolia is a Se-tolerant cruciferous plant that can metabolically transform inorganic Se into organic Se that exists in the form of a selenoprotein. Se-enriched peptide obtained by extraction and enzymolysis of selenoprotein, as an organic combination of organic Se and peptide, possess valuable biological activities. In this paper, the effect of Se-enriched peptides of C. violifolia on tumor cells was studied via cell experiments, and its mechanism was preliminarily discussed, which should provide a theoretical basis for developing functional foods containing C. violifolia.

Selenium biofortification of edible and medicinal mushrooms is an effective way to produce selenium-enriched food supplements. Ganoderma lucidum is the typical one with excellent biological activity. This study investigated G. lucidum growth and bioactive metabolites alterations during liquid culture with different concentrations of selenite. Low selenium levels did not affect growth and mycelia morphology, whereas high selenium levels negatively influenced growth, dramatically decreased biomass, caused nucleic acid and protein leakage, damaged cell walls and membranes, and resulted in indicators such as degraded cells, a red color, and an unpleasant odor. Through headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) analysis, ten volatile Se compounds were identified in G. lucidum with 200 ppm selenite, which led to an odor change, whereas only three with 50 ppm selenite. SeMet was the major selenoamino acid in the 50 ppm selenite group by high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS), but more MeSeCys was produced with 200 ppm selenite. Polysaccharide yields were promoted and inhibited with 50 and 200 ppm selenite, respectively. These results provide comprehensive insights into the effects of selenite on G. lucidum in liquid culture and are beneficial for functional selenium-enriched mushroom production and improving nutritive values.

In this study, a tea polyphenol (TP) loaded beeswax gelator (TP gelator) was constructed and incorporated into soybean oil to improve the oxidative stability of oleogels. The effects of TP on the structure and storage stability of oleogels were investigated. The TP gelator prepared by embedding TP into beeswax improved the dispersity of TP in lipids. The thermal characteristics and X-ray diffraction analysis showed that the components of the TP gelator coexisted homogeneously and TP was well dispersed in beeswax. The Fourier-transform infrared spectra indicated that the phenolic hydroxyl group of TP had intermolecular force with the quaternary ammonium nitrogen in the choline portion of soybean lecithin. The formation of needle-like crystals by beeswax restricted the flow of liquid oil, and TP did not participate in the construction of the internal network structure of the oleogel. In the accelerated storage experiment at 60 °C, the peroxide value of the TP loaded oleogel decreased by 60.6% at the end of the storage period compared with soybean oil. TP was successfully embedded in the oleogel without changing its structure, which not only solved the problem of poor dispersion of TP in oil, but also showed a significant inhibitory effect on lipid oxidation.

Botanic bioactive substances have issues with their solubility, stability, and oral bioavailability in the application, which could be improved by nanotechnologies. In another hands, green synthesis of nanoparticles (NPs) with plant extract is also a promising technology for preparation of NPs due to its safety advantage, yet the bioactive botanic substances that could be more than the assistant of the green synthesis of NPs. Based on the above concerns, this review summarized the preparation of botanic NPs with various plant extract, their solubility, stability, and oral bioavailability; specific attention has been paid to the botanic Ag/Au NPs, their capacity of antioxidant, bioavailability, antimicrobial, anti-inflammatory, and anticancer.

Realgar, an arsenic-containing traditional Chinese medicine of As2S2, has significant therapeutic effects for hundreds of years. NiuHuangJieDu tablets (NHJDT) is one of the most commonly prescribed realgar-containing preparations for the treatment of sore throat, swelling, and aching of gums. However, realgar-containing TCMs raise great safety concerns due to the adverse effects reported by arsenic poisoning. In this study, the arsenic-related health risk assessment of NHJDT was conducted in healthy volunteers after single and multiple doses oral administration. Blood, plasma, and urine samples were collected after dosing at predetermined time points or periods. Simple, rapid, and sensitive methods were established for the quantification of total arsenic and arsenic speciation in biological samples. The total arsenic and arsenic speciation were determined by hydride generation-atomic fluorescence spectrometry (HG-AFS) and high-performance liquid chromatography-hydride generation-atomic fluorescence spectrometry (HPLC-HG-AFS), respectively. No significant fluctuation of total arsenic was observed in human blood, and no traces of arsenic speciation were found in human plasma. Dimethylarsenic acid was detected as the predominated arsenic species in human urine after dosing. Therapeutic dose administration of NHJDT was relatively safe in single dose for the limited blood arsenic exposure, but long-term medication may still pose health risks due to the accumulation of arsenics in blood and its extremely slow excretion rate. Therefore, arsenic exposure should be carefully monitored during realgar-containing TCM medication, especially for long-term regimen. The results obtained in this study will provide scientific references for the clinical application of realgar and its-containing TCMs.

Cardamine violifolia (Brassicaceae) is a novel selenium(Se) hyperaccumulation plant with rich nutrients, and serves as a good source of special vegetables in Enshi, China. The present study aimed to investigate the effects of the application of selenate, selenite, and Se yeast (50-800 mg/L) on the growth, Se accumulation, nutrient uptake, and antioxidant response of C. violifolia. The results showed that the Se accumulation efficiency was selenate > selenite > Se yeast, the maximum Se concentration could achieve over 7000 mg/kg, and about 90% was organic Se. The major Se speciation found was mainly SeCys2 and the proportion of various Se species were affected by the Se forms and concentrations. Besides, the plant growth, nutrition quality indexes, element uptakes, and antioxidant responses indicated that 200 mg/L selenate was optimum for C. violifolia to accumulate Se without much impacts, while to obtain more proportion of organic Se, 200 mg/L selenite might be a better choice.

Selenium-enriched peptides from Cardamine violifolia (CSP) have excellent antioxidant functions but little is known about their effects on obesity and associated metabolic disorders in mice fed with a high-fat diet (HFD). In this study, C57BL/6 mice were fed a HFD with or without CSP supplementation (CSPL: 26 µg Se per kg bw per d; CSPH: 104 µg per kg bw per d) for 10 weeks. The results showed that both CSPL and CSPH could ameliorate overweight gain, excess fat accumulation, serum lipid metabolism, and insulin resistance. The potential mechanism might be associated with the increase in thermogenesis, reduced oxidative stress, and inflammation, which regulated the gene expression in lipid and cholesterol metabolism. In addition, CSPL and CSPH also maintained the intestinal integrity and modulated the gut microbiota. Increased Blautia in CSP may be involved in the protective effect against obesity. Furthermore, a distinct increase in Lactobacillus was exclusively found in CSPH, suggesting that a more effective function of CSPH on metabolic disorders might be through the synergism of Blautia and Lactobacillus. Spearman's correlation analysis revealed that these specific genera were significantly correlated with the metabolic improvements. Taken together, CSP supplementation prevented HFD-induced obesity and metabolic disorders, probably by ameliorating oxidative stress and inflammation, regulating metabolic genes, and modulating the gut microbiota compositions.

Cardamine violifolia is a selenium (Se)-enriched plant found in China. In this study, the Se-enriched peptides of C. violifolia (CP) were isolated using a 1 kDa ultrafiltration membrane after enzymatic hydrolysis by alkaline and neutral proteases. The peptides were separated by DEAE-Sepharose FF anion-exchange chromatography and purified using preparative high-performance liquid chromatography (prep-HPLC). The component with the highest antioxidant activity, CPR13, was identified by comparing the 1,1-diphenyl-2-picrylhydrazyl (DPPHâ¢), hydroxyl (â¢OH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+ â¢), and superoxide ( O 2 - · ) radical scavenging ability of each fraction. At a concentration of 0.1 mg/mL, the DPPHâ¢, â¢OH, O 2 - · , and ABTS+ ⢠scavenging activities of CPR13 were 89.2%, 26.3%, 40.6%, and 42.9%, respectively. Amino acid sequences were obtained by liquid chromatography combined with mass spectrometry as follows: GRVGSSSC, GRAGGSYM, GHPNFKLNCSGG, GTKSCKA, ASSNARDMI, TAGGCYIPI, and KNCALQ. The seleno-amino acids were identified as selenomethionine, methylselenocysteine, and selenocysteine. Correlation analysis among organic Se content, peptide content, and antioxidant activity revealed that organic Se plays a greater role in free radical scavenging than peptides, and that the organic Se content of the Se-enriched peptides was positively correlated with their antioxidant ability (P < 0.05). It indicated that CP has a great potential as natural functional materials for dietary supplement.

First, the antibacterial, antibiofilm effect and chemical composition of burdock (Arctium lappa L.) leaf fractions were studied. Then, the efficiency of burdock leaf fractions in pork preservation was evaluated. The results showed that burdock leaf fraction significantly inhibited the growth and biofilm development of Escherichia coli and Salmonella Typhimurium. MICs of burdock leaf fractions on E. coli and Salmonella Typhimurium were both 2 mg/ml. At a concentration of 2.0 mg/ml, the inhibition rates of the fraction on growth and development of E. coli and Salmonella Typhimurium biofilms were 78.7 and 69.9%, respectively. During storage, the log CFU per gram of meat samples treated with burdock leaf fractions decreased 2.15, compared with the samples without treatment. The shelf life of pork treated with burdock leaf fractions was extended 6 days compared with the pork without treatment, and the sensory property was obviously improved. Compared with the control group, burdock leaf fraction treatment significantly decreased the total volatile basic nitrogen value and pH of the meat samples. Chemical composition analysis showed that the burdock leaf fraction consisted of chlorogenic acid, caffeic acid, p-coumaric acid, rutin, cynarin, crocin, luteolin, arctiin, and quercetin. As a vegetable with an abundant source, burdock leaf is safe, affordable, and efficient in meat preservation, indicating that burdock leaf fraction is a promising natural preservative for pork.

STATEMENT OF PROBLEM: Denture base resin may be exposed to different conditions for long periods, resulting in varying degrees of staining, discoloration, and distortion. PURPOSE: The purpose of this in vitro study was to evaluate whether an organic-inorganic hybrid coating for base resins can improve color stability and reduce water absorption and solubility. MATERIAL AND METHODS: An organic-inorganic hybrid coating was prepared, and resin sheets made from 4 types of base resin materials were divided into an experimental group (surface coating) and a control group (without coating). The water absorption and solubility in each group were tested. The resin sheets were immersed in tea, red wine, coffee, and cola. The color change (ΔE values) of resin sheets in different soaking solutions was evaluated at 1, 7, and 28 days. RESULTS: Statistical analysis showed that water absorption and solubility in the experimental group were significantly lower than in the control group (P<.05). The 4 staining solutions (tea, red wine, coffee, and cola) caused base resin discoloration in a time-dependent manner. This staining and discoloration was also influenced by the type of resin material. About 85% of ΔE values in the experimental group were significantly lower than those in the control group (P<.05). At 28 days, almost all ΔE values in the experimental group were significantly lower than those in the control group (P<.05). CONCLUSIONS: The organic-inorganic hybrid coating effectively reduced the water uptake and solubility of base resins and significantly improved the color stability of the denture base.

Disease caused by the parasitic helminths Dactylogyrus spp. results in significant economic damage to the aquaculture industry. Treatment using common chemicals (e.g. formalin) is usually dissatisfactory due to environmental problems, risk of residues, toxicity to fish, and the possibility of anthelmintic resistance. The search for an alternative drug is thus becoming more urgent. This study was designed to evaluate in vivo the anthelmintic efficacy of total saponin (TS), saikosaponin a (SSa), and saikosaponin d (SSd) from radix bupleuri (i.e. the dried root of Bupleurum sp.) based on our previous screening works, with the aim of determining which has commercial potential. Results showed that median effective concentration (EC50) values for TS, SSa, and SSd were 2.01, 1.46, and 0.74 mg l⻹, respectively. The acute toxicities against goldfish Carassius auratus for TS, SSa, and SSd were also determined, with median lethal concentration (LC50) of 8.99, 11.20, and 1.54 mg l-1, respectively. The resulting therapeutic indices (TIs) indicated that SSa (TI = 7.67) is a potential therapeutic agent for treating Dactylogyrus infection.

In this study, we evaluate the influence of post surface pre-treatments on the bond strength of four different cements to glass fiber posts. Eighty extracted human maxillary central incisors and canines were endodontically treated and standardized post spaces were prepared. Four post pre-treatments were tested: (i) no pre-treatment (NS, control), (ii) sandblasting (SA), (iii) silanization (SI) and (iv) sandblasting followed by silanization (SS). Per pre-treatment, four dual-cure resin cements were used for luting posts: DMG LUXACORE Smartmix Dual, Multilink Automix, RelyX Unicem and Panavia F2.0. All the specimens were subjected to micro push-out test. Two-way analysis of variance and Tukey post hoc tests were performed (α=0.05) to analyze the data. Bond strength was significantly affected by the type of resin cement, and bond strengths of RelyX Unicem and Panavia F2.0 to the fiber posts were significantly higher than the other cement groups. Sandblasting significantly increased the bond strength of DMG group to the fiber posts.

In order to selectively remove caffeine from green tea, a microwave-enhanced vacuum ice water extraction (MVIE) method was proposed. The effects of MVIE variables including extraction time, microwave power, and solvent to solid radio on the removal yield of caffeine and the loss of total phenolics (TP) from green tea were investigated. The optimized conditions were as follows: solvent (mL) to solid (g) ratio was 10:1, microwave extraction time was 6 min, microwave power was 350 W and 2.5 h of vacuum ice water extraction. The removal yield of caffeine by MVIE was 87.6%, which was significantly higher than that by hot water extraction, indicating a significant improvement of removal efficiency. Moreover, the loss of TP of green tea in the proposed method was much lower than that in the hot water extraction. After decaffeination by MVIE, the removal yield of TP tea was 36.2%, and the content of TP in green tea was still higher than 170 mg g(-1). Therefore, the proposed microwave-enhanced vacuum ice water extraction was selective, more efficient for the removal of caffeine. The main phenolic compounds of green tea were also determined, and the results indicated that the contents of several catechins were almost not changed in MVIE. This study suggests that MVIE is a new and good alternative for the removal of caffeine from green tea, with a great potential for industrial application.

Vertical and temporal distributions of N and P in soil solution in aquatic-terrestrial ecotone (ATE) of Taihu Lake were investigated, and the relations among N, P, ORP (oxidation reduction potential), TOC, root system biomass and microorganism were studied. As a whole, significant declines in TN, NO3- -N, DON (dissolved organic nitrogen) and TP concentration in soil solution have occurred with increase of the depth, and reached their minima at 60 cm depth, except for NH4+ -N, which increased with depth. The concentration of TP increased gradually from spring to winter in the topsoil, the maximum 0.08 mg/L presented in the winter while the minimum 0.03 mg/L in spring. In the deeper layer, the concentration value of TP fluctuated little. As for the NO3- -N, its seasonal variation was significant at 20 cm depth, its concentration increased gradually from spring to autumn, and decreased markedly in winter. Vertical and temporal distribution of DON is contrary to that of NO3- -N. The results also show that the variation of N and P in the percolate between adjacent layers is obviously different. The vertical variation of TN, TP, NO3- -N, NH4+ -N and DON is significant, of which the variation coefficient of NO3- -N along the depth reaches 100.23%, the highest; while the variation coefficient of DON is 41.14%, the smallest. The results of correlation analysis show that the concentration of nitrogen and phosphorus correlate significantly with TOC, ORP, root biomass and counts of nitrifying bacteria. Most nutrients altered much from 20 to 40 cm along the depth. However, DON changed more between 60 and 80 cm. Results show that soil of 0-60 cm depth is active rhizoplane, with strong capability to remove the nitrogen and phosphorus in ATE. It may suggest that there exists the optimum ecological efficiency in the depth of above 60 cm in reed wetland. This will be very significant for ecological restoration and reestablishment.
