RESUMEN

Methionine (Met) is the first limiting amino acid in corn and soybean meal-based diets (containing L-Met) in broiler chickens, which are often supplemented with synthetic DL-Met or DL-Hydroxy Met (OH-Met). Our objective was to quantitatively assess the efficacy of synthetic Met sources and determine differences in growth rate of broilers fed at or below requirements in response to Met intake. A systematic literature search resulted in building a database containing 480 treatment means from 39 articles published between 1985 and 2019 globally. The database was divided into starter, grower, and finisher subsets based on the age of the broilers. For each subset, linear-plateau and quadratic-plateau models were fitted to determine Met or sulfur amino acid (SAA; Met + Cysteine) requirements using average daily gain as a response variable. For each phase, 4 new subsets were obtained by only retaining records with digestible Met or SAA intake at or below requirement by linear-plateau or quadratic-plateau models. Then, a linear model (without plateau) was fitted for all new subsets for each rearing phase using supplemental digestible synthetic Met or SAA intake (basal Met intake was subtracted from total Met intake) as independent variables. The basal diet was made of only raw materials without supplementation of any synthetic Met source. Finally, the models were extended to evaluate source of synthetic Met effects on the slope parameter. At all stages of model fitting, the inclusion of a random study effect was evaluated for each parameter. All models were fitted within a Bayesian framework, for which minimally informative priors were used. The best models, that is, the most accurate inclusion of random effects, were selected based on at least 10-point difference in leave-one-out cross-validation information criterion. Model selection criteria did not consistently favor either of the linear- and quadratic-plateau models to determine Met or SAA requirements across broiler growth phases. Extending models with covariates (e.g., dietary energy and amino acids) did not improve any model fit. Body weight gain response of broiler chickens to the 2 sources was not different when fed at or below Met requirements for any of the growth phases.

Providing adequate concentrations of AA in the prepartum diet is pivotal for the cow's health and performance. However, less is known about the potential in utero effects of particular AA on early-life performance of calves. This experiment was conducted to determine the effects on dairy calves when their dams were fed rumen-protected lysine (RPL; AjiPro-L Generation 3, Ajinomoto Heartland Inc.; 0.54% dry matter of total mixed ration as top dress) from 26 ± 4.6 d (mean ± standard deviation) before calving until calving. Seventy-eight male (M) and female (F) Holstein calves were assigned to 2 treatments based on their dams' prepartum treatment, RPL supplementation (PRE-L) or without RPL (CON). At the time of birth (0.5-2 h after calving), before colostrum was fed, blood samples were collected. An initial body weight was obtained at 1 to 3 h after birth. Calves were fed 470 g of colostrum replacer (Land O'Lakes Bovine IgG Colostrum Replacer, Land O'Lakes, Inc.) diluted in 3.8 L of water. Calves were provided water ad libitum and fed milk replacer (Advance Excelerate, Milk Specialties Global Animal Nutrition; 28.5% crude protein, 15% fat) at 0600 h and 1700 h until 42 d of age. Calves were measured weekly, at weaning (d 42), and at the end of the experimental period (d 56). Plasma concentrations of AA were measured on d 0, 7, and 14 d using ultra-performance liquid chromatography-mass spectrometry (Waters) with a derivatization method (AccQ-Tag Derivatization). Final body weight was greater for M (87 ± 11 kg) than F (79 ± 7 kg). Calves in PRE-L tended to have greater dry matter (814 ± 3 g/d) and crude protein (234 ± 6 g/d) intakes than those in CON (793 ± 9 g/d and 228 ± 11 g/d, respectively). Calves in PRE-L had greater average daily gain (0.96 ± 0.04 kg/d) than calves in CON (0.85 ± 0.03 kg/d) during wk 6 to 8. Calves in PRE-L tended to be medicated fewer days than CON (4.7 ± 1.2 d vs. 6.2 ± 3.4 d, respectively). Calves in PRE-L-M and CON-F (2,916 ± 112 µM and 2,848 ± 112 µM, respectively) had greater total AA concentration in plasma than calves in PRE-L-F and CON-M (2,684 ± 112 µM and 2,582 ± 112 µM, respectively). Calves in PRE-L-F and CON-M (4.09 ± 0.11% and 4.16 ± 0.11%, respectively) had greater concentration of Lys as a percentage of total AA compared with calves in CON-F and PRE-L-M (3.91 ± 0.11% and 3.90 ± 0.11%, respectively). Calves in PRE-L tended to have greater percentage of phagocytic neutrophils (39.6 ± 1.59%) than calves in CON (35.9 ± 1.59%). In conclusion, increasing the metabolizable lysine provided to prepartum dairy cows had modest effect over offspring performance, with the major result being a greater average daily gain for calves in PRE-L during the preweaning phase (wk 6-8).

The objective of this study was to evaluate the effect of supplementing a CP-reduced diet with rumen-protected methionine on growth performance of Fleckvieh bulls. A total of 69 bulls (367 ±â€¯25 kg BW) were assigned to three feeding groups (n = 23 per group). The control (CON) diet contained 13.7% CP and 2.11 g methionine/kg diet (both DM basis) and was set as positive control. The diet reduced in CP (nitrogen) (RED) diet as negative control and the experimental RED + rumen-protected methionine (MET) diet were characterised by deficient CP concentrations (both 9.04% CP). The RED + MET diet differed from the RED diet in methionine concentration (2.54 g/kg DM vs. 1.56 g/kg DM, respectively) due to supplementation of rumen-protected methionine. Rumen-protected lysine was added to both RED and RED + MET at 2.7 g/kg DM to ensure a sufficient lysine supply relative to total and metabolisable protein intake. Metabolisable energy (ME) and nutrient composition were similar for CON, RED, and RED + MET. Bulls were fed for 105 days (d) on average. Individual feed intake was recorded daily; individual BW was recorded at the beginning of the experiment, once per month, and directly before slaughter. At slaughter, blood samples were collected and carcass traits were assessed. Reduction in dietary CP concentration reduced feed intake, and in combination with lower dietary CP concentration, daily intake of CP for RED and RED + MET was lower compared with CON (P < 0.01). Daily ME intake was reduced in RED and RED + MET compared with CON (P < 0.01). Consequently growth performance and carcass weights were reduced (both P < 0.01) in both RED and RED + MET compared with CON. Supplemental rumen-protected methionine was reflected in increased serum methionine concentration in RED + MET (P < 0.05) as compared to RED but it did not affect growth performance, carcass traits and serum amino acid (AA) concentrations, except for lysine which was reduced (P < 0.01) compared to CON and RED. In conclusion, bulls fed RED or RED + MET diets were exposed to a ruminal CP deficit and subsequently a deficit of prececal digestible protein, but methionine did not appear to be the first-limiting essential AA for growth under the respective experimental conditions.

The preweaning period is vital in the development of calves on dairy farms and improving daily liveweight gain (DLWG) is important to both financial and carbon efficiency; minimising rearing costs and improving first lactation milk yields. In order to improve DLWG, veterinary advisors should provide advice that has both a large effect size as well as being consistently important on the majority of farms. Whilst a variety of factors have previously been identified as influencing the DLWG of preweaned calves, it can be challenging to determine their relative importance, which is essential for optimal on-farm management decisions. Regularised regression methods such as ridge or lasso regression provide a solution by penalising variable coefficients unless there is a proportional improvement in model performance. Elastic net regression incorporates both lasso and ridge penalties and was used in this research to provide a sparse model to accommodate strongly correlated predictors and provide robust coefficient estimates. Sixty randomly selected British dairy farms were enrolled to collect weigh tape data from preweaned calves at birth and weaning, resulting in data being available for 1014 calves from 30 farms after filtering to remove poor quality data, with a mean DLWG of 0.79 kg/d (range 0.49-1.06 kg/d, SD 0.13). Farm management practices (e.g. colostrum, feeding, hygiene protocols), building dimensions, temperature/humidity and colostrum quality/bacteriology data were collected, resulting in 293 potential variables affecting farm level DLWG. Bootstrapped elastic net regression models identified 17 variables as having both a large effect size and high stability. Increasing the maximum preweaned age within the first housing group (0.001 kg/d per 1d increase, 90 % bootstrap confidence interval (BCI): 0.000-0.002), increased mean environmental temperature within the first month of life (0.012 kg/d per 1 °C increase, 90 % BCI: 0.002-0.037) and increased mean volume of milk feeding (0.012 kg/d per 1 L increase, 90 % BCI: 0.001-0.024) were associated with increased DLWG. An increase in the number of days between the cleaning out of calving pen (-0.001 kg/d per 1d increase, 90 % BCI: -0.001-0.000) and group housing pens (-0.001 kg/d per 1d increase, 90 % BCI: -0.002-0.000) were both associated with decreased DLWG. Through bootstrapped elastic net regression, a small number of stable variables have been identified as most likely to have the largest effect size on DLWG in preweaned calves. Many of these variables represent practical aspects of management with a focus around stocking demographics, milk/colostrum feeding, environmental hygiene and environmental temperature; these variables should now be tested in a randomised controlled trial to elucidate causality.

Optimising heifer growth rate may offer an opportunity to improve lifetime milk yield per cow, enhancing the environmental and economic efficiency of dairy farming operations. The effect of dairy heifer pre-breeding average daily weight gain (ADGPB) on first lactation milk yield was investigated. This observational study employed a data set comprising 265 Holstein-Friesian, or Holstein-Friesian-cross-Jersey heifers from seven commercial, spring-calving, pasture-based dairy herds, where the major component of the diet was grazed grass. These were weighed at birth and prior to breeding and ADGPB was calculated. Milk recordings were performed throughout the heifers' first lactation and 305-day yield figures calculated from these records. Yields were corrected to 4% fat and 3.1% protein to create standardised 305-day milk yield (SMY), which was the outcome of interest. Median ADGPB was 0.72 kg/day. Median 305-day yield was 5 967 kg. Linear regression was used to investigate the effect of weight and genetic, age and first calving factors on SMY. Pre-breeding average daily weight gain, age at first calving and predicted transmitting abilities for milk protein production and calving interval were all significant in the final model, which also included the random effects of farm and month of calving within year. ADGPB was quadratically related to first lactation SMY, with an ADGPB of 0.82 kg/day corresponding to the maximum predicted SMY. The model predicted that a heifer growing at 0.82 kg/day would produce 1 120 kg more SMY than a heifer growing at 0.55 kg/day, 218 kg more than a heifer growing at 0.7 kg/day and 103 kg more than a heifer growing at 0.90 kg/day. Manipulation of heifer growth rate may offer a viable method of increasing first lactation milk yield.

BACKGROUND: Early weaning in yak calves is being attempted to improve yak reproduction rate. However, this has to be done with caution because of the high mortality rate of calves due to the lack of nutrients and the harsh environmental conditions. Twenty-four weaned male yak calves were used in a 60 day feeding trial in which astragalus root extract (ARE) was supplemented. They were assigned randomly to one of four dietary treatments (n = six per treatment) that differed in ARE level: 0 g kg-1 (control), ARE0 ; 20 g kg-1 , ARE20 ; 50 g kg-1 , ARE50; and 80 g kg-1 dry matter intake (DMI), ARE80 . RESULTS: Final bodyweight and average daily gain (ADG) were significantly higher and the DMI/ADG ratio was significantly lower in calves with ARE supplementation than control (ARE0 ) calves. Ruminal concentrations of acetate and propionate and serum concentration of superoxide dismutase in ARE80 calves were higher than in the other groups and serum concentration of insulin was higher in ARE80 calves than in ARE20 calves. Serum immunoglobulin G (IgG) and interleukin-2 (IL-2) concentrations in ARE-fed calves were higher than in controls. Serum tumor necrosis factor (TNF-α) concentration was higher in ARE50 and ARE80 groups than ARE0 calves and serum interleukin-6 (IL-6) concentration was higher in ARE80 than in ARE0 calves. Serum immunoglobulin A (IgA), IgG and immunoglobulin M (IgM) concentrations increased with age in ARE-fed calves. ARE supplementation increased the abundance of fiber degrading bacteria. CONCLUSION: ARE at a dosage of 5% to 8% DMI can be supplemented to early weaned yak calves to improve growth performance, antioxidant capacity and immunity. © 2020 Society of Chemical Industry.

The aim of this study was to evaluate the effects of maternal and dietary vitamin A (VA) level on growth performance, meat quality, antioxidant status, and immune function of offspring broilers. Chinese yellow-feathered breeder hens were fed a basal diet supplemented with 0, 5,400, 10,800, and 21,600 IU/kg VA for 8 wk, with 6 replicates of 22 hens per replicate. Then the offspring hatched from each of the 4 maternal groups were fed a basal diet supplemented with 0 or 5,000 IU/kg VA for 63 D. Overall, there were 8 treatment combinations, each with 6 replicate pens of 20 birds. Results showed that (1) providing VA in offspring diets increased final body weight (FW), average daily gain, and average daily feed intake but reduced feed-to-gain ratio and mortality of offspring broilers (P < 0.05), whereas maternal provision of VA did not significantly affect the growth performance and mortality of offspring broilers. Maternal or offspring VA did not affect proportion of breast or thigh muscle (P > 0.05). (2) Maternal feeding with 21,600 IU/kg VA increased (P < 0.05) pH 24 h postmortem of breast muscle, compared with those without maternal supplication of VA. Dietary provision of 5,000 IU/kg VA in the posthatching diet decreased (P < 0.05) drip loss, yellowness (b∗) value and lightness (L∗) value, and increased shear force and pH of breast muscle compared with those without dietary VA supplication. (3) Maternal or offspring VA did not affect the activities of total superoxide dismutase and glutathione peroxidase (GSH-Px) or the content of malondialdehyde; however, there was a significant interaction (P < 0.05) between maternal and offspring VA on the activity of GSH-Px in serum. (4) Dietary provision of 5,000 IU/kg VA increased (P < 0.05) the weight proportion of liver and bursa of fabricius, whereas maternal feeding with 21,600 IU/kg VA increased the hatchling BW. Maternal feeding with 5,400 and 21,600 IU/kg VA decreased (P < 0.05) splenic interferon-γ (IFN-γ) transcripts and increased (P < 0.05) those of interleukin-2 (IL-2) in the progeny. There were interactions (P < 0.05) between maternal and offspring VA on splenic IL-2, IL-1ß, and IFN-γ expression. In summary, maternal and offspring provision of VA both had influence on meat quality and immune function in progeny broilers. Dietary VA increased growth performance, whereas the maternal VA affected the initial body weight of progeny when hatched, but the difference in performance caused by maternal VA level was able to be eliminated by dietary VA supplementation. Therefore, offspring provision had greater importance than maternal VA in the production; however, both should be considered in broiler nutrition to achieve good meat quality and immune status of broilers.

Dietary electrolyte balance (dEB) is known to affect acid-base status and mineral metabolism, but is rarely considered in diet formulation for pigs. Yet, the use of a wide variety of local feedstuffs in Europe contributes to lowering the dEB and increasing the fibre content. Hence, mineral requirements may be modified and skeletal health affected. Therefore, the effects of a lower dEB and a higher dietary Ca level on acid-base balance and mineral status were assessed in young pigs fed a diversified diet. A total of twenty-four weaned pigs were fed a control moderate-dEB diet (C) or a diversified moderate-dEB (D), low-dEB (D-A) or low-dEB supplemented with Ca (D-CA) diet. Growth performance, venous blood gas and chemistry, urine pH, mineral balance and femur characteristics were determined. With an equivalent dEB compared with the C diet, the D diet caused an acidification of the urine and increased the excretion of P as a result of a higher dietary content of S. Low-grade metabolic acidosis occurred in piglets fed the D-A diet with changes at systemic and urine levels. A higher excretion of ammonia and P in urine was observed and some bone characteristics tended to be negatively affected. Ca supplementation partially counteracted the effects of low-grade acidosis. Urine excretion of P and ammonia was alleviated and bone characteristics improved. In conclusion, a higher Ca supply must be considered in more diversified diets to counteract the risk of evolving towards low-grade metabolic acidosis which can negatively affect bone.

It was shown previously that high dietary fiber (DF) and immune system stimulation (ISS) with systemic Escherichia coli lipopolysaccharide independently increased the threonine (Thr) requirement to maximize growth performance and protein deposition (PD). However, no additive effects on the Thr requirement were observed when both DF and ISS were present. The objective of the present study was to investigate whether supplementing Thr to meet previously estimated requirements for high DF and systemic immune challenge would maintain performance of pigs exposed to an enteric immune challenge when fed high DF. A total of 128 pigs (22.6 ± SD = 1.6 kg initial BW) were assigned to 1 of 4 dietary treatments in a 2 × 2 factorial arrangement in a randomized complete block design (n = 8 pens/treatment and 4 pigs/pen) for 28 d. Treatments were a low-fiber (LF; 13% total DF) or high-fiber (HF; 20% total DF) diet with either a standard (STD; 0.65% SID) or supplemental (SUP; 0.78% SID) Thr level. After a 7-d adaptation, pigs were orally inoculated with 2 mL (2.3 × 109 CFU/mL) of Salmonella typhimurium (ST). Blood samples and rectal swabs were obtained and rectal temperature recorded to determine clinical responses and ST shedding. On day 7 postinoculation, 1 pig/pen was euthanized and mesenteric lymph nodes, spleen, and digesta (ileum, cecum, and colon) were sampled to assess ST colonization and translocation. Body weight and feed intake were recorded on day 0, 7, and 21 postinoculation to calculate ADG, ADFI, and G:F. Rectal temperature increased (P < 0.05) 24 h postinoculation and remained elevated at day 6. Serum albumin concentration decreased (P < 0.05), whereas haptoglobin concentration increased (P < 0.05) postinoculation. There was no fiber or Thr effect (P > 0.05) on ST counts in the ileum and cecum, but a fiber × Thr interaction (P < 0.05) was observed in the colon. Supplemental Thr improved (P < 0.05) growth performance in LF- and HF-fed challenged pigs. However, performance of supplemented HF challenged pigs was less than (P < 0.05) supplemented LF challenged pigs. These results suggest that Thr supplemented to meet requirements for high DF and systemic immune challenge was not sufficient to maintain growth performance of pigs fed HF diets and challenged with an enteric pathogen.

The objective of the study was to investigate the effect of stocking density and extract from Cassia abbreviata stem bark on growth performance, oxidative stress and liver function of indigenous chickens. A total of 420 1-day-old female Ovambo chicks with initial body weight of 0.32 ± 0.036 kg (mean ± SD) were used in the study. Birds, which were cooped in stocking densities consisting 5, 10 and 20 birds/m2, were orally administered with 0, 50, 200 and 500 mg/kg of extract from C. abbreviata stem bark. Each stocking density per dosage level of extract was repeated three times. Average feed intake was lower (P < 0.05) in birds housed at 20 birds/m2. There was a low (P < 0.05) average daily gain in birds housed at 20 birds/m2. Malondialdehyde was higher (P < 0.05) in 20 birds/m2. Average daily gain was high (P < 0.05) in birds administered with 0 and 50 mg/kg of C. abbreviata stem bark extract. Birds administered with 0 and 50 mg/kg of C. abbreviata stem bark extract had a higher (P < 0.05) gain to feed ratio. Birds dosed with 500 and 200 mg/kg had high (P < 0.05) superoxide dismutase activity. Birds administered with 500 mg/kg of C. abbreviata stem bark extract had the lowest (P < 0.05) malondialdehyde. A 500 mg/kg of stem bark extract from C. abbreviata resulted to higher (P < 0.05) activities of aspartate transaminase and alanine transaminase. There was a significant (P < 0.05) interaction between the stocking density and C. abbreviata extract on catalase activity. High stocking density of 20 birds/m2 reduced growth performance and induced stress in indigenous chickens. High dosage of level 500 mg/kg of stem bark extract of C. abbreviata reduced oxidative stress while depressing growth performance and causing hepatotoxicity in birds. There is a need to precisely determine the maximum dosage level of C. abbreviata extract to improve growth performance and reduce oxidative stress and hepatotoxicity in indigenous chickens in high stocking density.

This study was conducted to evaluate the effects of supplementation with different levels of mineral-vitamin supplement (MVS) on performance and health of fattening lambs. Thirty-nine fattening male Ghezel× Afshar lambs (averaging body weight 31.02 ± 3.46 kg) were used in a completely randomized design for a period of 84 days. Treatments consisted of 0 (CON), 15 (MVS15), or 30 (MVS30) g/day of MVS mixed into the concentrate. The results showed that inclusion of MVS in the ration linearly enhanced body weight (BW) on day 29 (P < 0.002), 45 (P = 0.05), and 70 (P < 0.002). Dry matter intake (DMI) was increased linearly (P < 0.02) and quadratically (P < 0.001) with increasing supplement between days 58 and 70. Overall DMI tended (P = 0.1) to be greater for the MVS 30 treatment. Total average daily gain (ADG), ADG between days 16 and 29, days 58 and 70, total gain: feed ratio, gain: feed ratio between days 16-29 and days 58-70 linearly increased with increasing amount of MVS in the ration (P < 0.01). The days with loose fecal scores were not affected by treatments (P > 0.05). There was a linear and quadratic (P < 0.01) effect on serum Ca concentration. Phosphorus concentration tended to quadratically increase (P = 0.06) with MVS. Blood serum Zn increased linearly (P = 0.01) with increasing level of MVS. Collectively, the results showed that, under the conditions of the current experiment, MVS addition to ration improved performance of fattening lambs.

Plant extracts can affect the rumen microbiome and ADG in ruminants, and studies of the association between the rumen microbiome and ADG provide information applicable to improving ruminant growth performance. The objectives were to investigate the effects of Allium mongolicum Regel extracts on the rumen microbiome and ADG and their association in sheep. Forty healthy, male, small-tailed Han sheep (6 mo, 34 ± 3.5 kg body weight) were randomly assigned to 1 of the following 4 dietary treatments: basal diet as control group (CK, n = 10), basal diet supplemented with 3.4 g·sheep-1·d-1A. mongolicum Regel powder extract as PAM group (PAM, n = 10), basal diet supplemented with 10 g·sheep-1·d-1A. mongolicum Regel powder as AM group (AM, n = 10), and basal diet supplemented with 10 g·sheep-1·d-1A. mongolicum Regel powder extract residue as RAM group (RAM, n = 10). The ADG for individual sheep was calculated using the sum of the ADGs observed during the experimental period divided by the number of days in the experimental period. At the end of the experiment, sheep were randomly selected from each treatment for slaughter (n = 6), and the rumen fluids were collected and stored immediately at -80 °C. Illumina HiSeq was subsequently used to investigate the changes in the rumen microbiome profile, and the associations with ADG were analyzed by Spearman correlation coefficient analysis. The results demonstrated that, compared with that in CK group, the ADG in AM and RAM significantly increased (P = 0.0171). The abundances of Tenericutes and Mollicutes ([ρ] = 0.5021, P = 0.0124) were positively correlated with ADG. Within Mollicutes, the abundances of Anaeroplasmatales ([ρ] = 0.5458, P = 0.0058) and Anaeroplasmataceae ([ρ] = 0.5458, P = 0.0058) were positively correlated with ADG. The main negatively correlated bacteria were Saccharibacteria ([ρ] = -0.4762, P = 0.0187) and Betaproteobacteria ([ρ] = -0.5669, P = 0.039). Although Anaeroplasmatales and Anaeroplasmataceae were positively correlated with ADG, Saccharibacteria and Betaproteobacteria were negatively correlated with ADG. In conclusion, supplementation with A. mongolicum Regel powder and extracts will influence the rumen microbiome and increase the ADG.

Organic additives are recently being used in animal diets owing to their ability to control metabolic issues and result in better animal performance. Specifically, the organic additive Fator P® presents an additional advantage that is to cause a lesser greenhouse gas emission. This study evaluated whether Fator P® intake changes ruminal parameters or animal performance of beef cattle. Evaluations were carried out in a feedlot experiment divided into growing (46 days; two diets [control mix-CM and standard mix-SM] and finishing (lasted 83 days; four diets: CM, SM, Fator P® + virginiamycin, and Fator P® alone [FP]) trials. Animal performance study involved 48 animals allocated to 12 collective pens in completely randomized experimental design. Ruminal parameters were evaluated in separate metabolism study developed carried out using individual pen with four steers. During growing trial, FP diet resulted in higher (p < 0.05) dry matter intake (DMI) and ruminating time. In the finishing trial, diets containing Fator P® resulted in higher DMI than obtained with CM. Most of the ruminal parameters did not differ (p > 0.05) among dietary treatments. Therefore, Fator P® represents a viable and safe strategy for supplementation to beef cattle finished using high-concentrate diet in feedlot systems.

The present study was carried out to evaluate the effect of dietary supplemental vitamin D3 (VD3) on P absorption and utilisation as well as its related mechanisms in the small intestine of broilers. A total of 384 1-d-old Arbor Acres male broilers were assigned randomly into four treatments following a completely randomised design with a 2 (dietary non-phytate P (NPP) contents: 0·43 and 0·22 %)×2 (dietary VD3 supplemental levels: 0 and 87·5 µg/kg) factorial arrangement. The experiment lasted for 22 d. The results showed that P contents in serum from the hepatic portal vein and tibia ash of broilers were higher (P<0·05) for 0·43 % NPP than for 0·22 % NPP. The type IIb Na-dependent phosphate cotransporter (NaP-IIb) protein expressions in the duodenum and ileum were higher (P<0·05) also for 0·43 % NPP than 0·22 % NPP. Supplementation of VD3 enhanced (P<0·05) tibia P retention rate and type III Na-dependent phosphate cotransporter (PiT)-1 protein expression in the duodenum of all broilers. Moreover, VD3 supplementation decreased (P<0·002) mortality and increased (P<0·02) serum P content from the hepatic portal vein after 4 h of feeding, tibia ash content, tibia ash P content and protein expressions of NaP-IIb and PiT-1 in the jejunum of broilers fed diet with 0·22 % NPP. Thus, dietary supplemental VD3 promoted intestinal P absorption and bone P utilisation, and this effect might be associated with enhanced PiT-1 levels in the duodenum and PiT-1 and NaP-IIb levels in the jejunum respectively when dietary NPP is limiting.

This study was conducted to investigate the protective effects of l-threonine (l-Thr) supplementation on growth performance, inflammatory responses and intestinal barrier function of young broilers challenged with lipopolysaccharide (LPS). A total of 144 1-d-old male chicks were allocated to one of three treatments: non-challenged broilers fed a basal diet (control group), LPS-challenged broilers fed a basal diet without l-Thr supplementation and LPS-challenged broilers fed a basal diet supplemented with 3·0 g/kg l-Thr. LPS challenge was performed intraperitoneally at 17, 19 and 21 d of age, whereas the control group received physiological saline injection. Compared with the control group, LPS challenge impaired growth performance of broilers, and l-Thr administration reversed LPS-induced increase in feed/gain ratio. LPS challenge elevated blood cell counts related to inflammation, and pro-inflammatory cytokine concentrations in serum (IL-1ß and TNF-α), spleen (IL-1ß and TNF-α) and intestinal mucosa (jejunal interferon-γ (IFN-γ) and ileal IL-1ß). The concentrations of intestinal cytokines in LPS-challenged broilers were reduced by l-Thr supplementation. LPS administration increased circulating d-lactic acid concentration, whereas it reduced villus height, the ratio between villus height and crypt depth and goblet density in both jejunum and ileum. LPS-induced decreases in jejunal villus height, intestinal villus height:crypt depth ratio and ileal goblet cell density were reversed with l-Thr supplementation. Similarly, LPS-induced alterations in the intestinal mRNA abundances of genes related to intestinal inflammation and barrier function (jejunal toll-like receptor 4, IFN- γ and claudin-3, and ileal IL-1 ß and zonula occludens-1) were normalised with l-Thr administration. It can be concluded that l-Thr supplementation could attenuate LPS-induced inflammatory responses and intestinal barrier damage of young broilers.

This study aimed to investigate the effects of dietary live yeast (LY) supplementation on growth, intestinal permeability and immunological parameters of piglets challenged with enterotoxigenic Escherichia coli K88 (ETEC). Piglets weaned at 21 d were allocated into three treatments with six pens and six piglets per pen, receiving the control diet (CON), diets supplemented with antibiotics plus zinc oxide (ANT-ZnO) and LY (Saccharomyces cerevisiae strain CNCM I-4407), respectively, for a period of 2 weeks. On day 8, thirty-six piglets were selected as control without ETEC (CON), CON-ETEC, ANT-ZnO-ETEC and LY-ETEC groups challenged with ETEC until day 10 for sample collections. Piglets fed ANT-ZnO diet had the highest average daily gain and average daily feed intake (P<0·05) during the 1st week, but ADG of piglets fed the ANT-ZnO diet was similar as piglets fed LY diet during the second week. Piglets with LY-ETEC or ANT-ZnO-ETEC had markedly lower diarrhoea score (P<0·05) than piglets with CON-ETEC during the 24 h after ETEC challenge. Relative to piglets with CON, the counts of E. coli, urinary ratio of lactulose to mannitol, plasma IL-6 concentration, mRNA abundances of innate immunity-related genes in ileum and mesenteric lymph node tissues were increased (P<0·05), whereas the villous height of jejunum and relative protein expression of ileum claudin-1 were decreased (P<0·05) in piglets with CON-ETEC; however, these parameters did not markedly change in piglets with LY-ETEC or ANT-ZnO-ETEC. In summary, dietary LY supplementation could alleviate the severity of diarrhoea in piglets with ETEC, which may be associated with the improved permeability, innate immunity and bacterial profile.

One hundred and fifty 7-day-old Arbor Acres broilers were randomly assigned into five groups: group 1 served as a control that was fed a basal diet without selenium (Se) supplementation; groups 2, 3 and 4 were fed the basal diet supplemented with 0.15, 0.5 and 1.5 mg Se as Se-enriched Saccharomyces cerevisiae (SSC) per kg of diet; and group 5 was fed the basal diet supplemented with 0.15 mg per kg of Se as sodium selenite (SS). Growth performance, glutathione peroxidase (GPX ) and superoxide dismutase (SOD) activities, total antioxidant capacity (T-AOC), and malondialdehyde (MDA) content in plasma and liver, and cellular glutathione peroxidase (GPX -1) and phospholipid hydroperoxide glutathione peroxidase (GPX -4) mRNA levels in liver were determined. Compared with group 1, groups 2-4 exhibited higher body weights (p < 0.05), lower feed/gain ratios, and higher GPX activities in plasma (p < 0.05) and GPX and SOD activities and GPX -1 and GPX -4 mRNA levels in liver (p < 0.05). Compared with group 5, group 2 exhibited higher GPX activity in plasma on day 21 (p < 0.05). Compared with group 2 and 5, group 3 exhibited lower MDA content in plasma on day 7 (p < 0.05), higher GPX activity in plasma, SOD activity and GPX -1 mRNA levels in liver on day 14 and 21 (p < 0.05), and higher GPX -4 mRNA levels on day 14 (p < 0.05). Compared with group 4, group 3 exhibited lower MDA contents in plasma on day 14 (p < 0.05) and in liver on day 21 (p < 0.05), higher T-AOC in plasma and higher GPX -1 mRNA levels on day 14 and 21 (p < 0.05), and higher SOD activity in plasma and higher SOD and GPX activities in liver on day 21 (p < 0.05). Thus, SSC improves growth and antioxidant status of broilers; the short-term bioavailability of SS was faster than that of SSC, but the long-term bioavailability of SSC was greater than SS.

This study was conducted on 78 13-month-old crossbred beef heifers that weighed 215 kg in Southern Rio Grande do Sul (RS) State, Brazil. We evaluated the performance of beef heifers that were reared in a pasture system that received a mineral supplement energy-type protein with added sodium lasalocid (LAS). The heifers were randomly and uniformly divided into 2 groups, with 39 animals in each group. One group of animals received a mineral supplement energy-type protein without sodium lasalocid (CON), and the other group received a mineral supplement energy-type protein with added LAS. The mean feed intake, the body weight (BW), the average daily gain (ADG), the body condition score (BCS), and ovarian cyclicity were recorded, and economic parameters were calculated. No differences in supplement intake were observed between the groups, which ensures adequate intake of the other components of the mineral mixture, which are part of the nutritional requirements for the production process. Similarly, no difference in the ADG was observed between treatments. We observed that the heifers in the LAS group had a higher BW gain (51 kg) that the CON heifers (40 kg; P < 0.05). In addition, LAS-supplemented heifers had a higher BCS (3.53) than CON heifers (3.38) at the end of the experiment (P < 0.05). The heifers supplemented with LAS had a higher profitability than the CON heifers, even with the higher cost of the supplement containing LAS; this effect was due to the higher live BW at the end of the study. We concluded that the administration of a mineral supplement energy-type protein with added LAS has beneficial effects on beef heifers in terms of production and economic feasibility.

The aim of this study was to investigate whether supplementing branched-chain amino acids (AA) (BCAA) along with a reduced-protein diet increases piglet growth, and whether elevated feed intake and muscle growth-promoting effect contribute to this improvement. In Expt 1, twenty-eight weanling piglets were randomly fed one of the following four diets: a positive control (PC) diet, a reduced-protein negative control (NC) diet, an NC diet supplemented with BCAA to the same levels as in the PC diet (test 1 (T1)) and an NC diet supplemented with a 2-fold dose of BCAA in T1 diet (test 2 (T2)) for 28 d. In Expt 2, twenty-one weanling piglets were randomly assigned to NC, T1 and pair-fed T1 (P) groups. NC and T1 diets were the same as in Expt 1, whereas piglets in the P group were individually pair-fed with the NC group. In Expt 1, the NC group had reduced piglet growth and feed intake compared with the PC group, which were restored in T1 and T2 groups, but no differences were detected between T1 and T2 groups. In Expt 2, T1 and P groups showed increases in growth and mass of some muscles compared with the NC group. Increased feed intake after BCAA supplementation was associated with increased mRNA expressions of agouti-related peptide and co-express neuropeptide Y (NPY) and phosphorylation of mammalian target of rapamycin (mTOR) and ribosomal protein S6 kinase 1 (S6K1), as well as decreased mRNA expressions of melanocortin-4 receptor and cocaine- and amphetamine-regulated transcript and phosphorylation of eukaryotic initiation factor 2α in the hypothalamus. No differences were observed among PC, T1 and T2 groups except for higher NPY mRNA expression in the T2 group than in the PC group (Expt 1). Phosphorylation of mTOR and S6K1 in muscle was enhanced after BCAA supplementation, which was independent of change in feed intake (Expt 2). In conclusion, supplementing BCAA to reduced-protein diets increases feed intake and muscle mass, and contributes to better growth performance in piglets.

The aim of this study was to investigate the effect of providing supplementary Cr-enriched Bacillus subtilis (CEBS) to mice with regard to their growth performance, caecal microbiology, tissue Cr concentration, insulin receptor (IR) expression and plasma biochemical profile. A total of ninety-six Kunming strain mice were allocated to four different groups: control, CEBS, inorganic Cr and B. subtilis. After 15 d of treatment, mice that received CEBS or normal B. subtilis had higher body weights than control mice, and after 30 d mice given either CEBS or B. subtilis had greater body weights than control mice or those given inorganic Cr. The concentration of Cr in tissues (heart, liver, spleen, kidney and skeletal muscle) increased after CEBS supplementation. B. subtilis and CEBS supplementation caused a significant increase in the numbers of Lactobacillus and Bifidobacterium in the caecum, whereas the numbers of Escherichia coli and Staphylococcus decreased significantly compared with the control. The levels of IR RNA and protein in skeletal muscles increased significantly. Plasma glucose, total cholesterol, TAG and LDL-cholesterol levels declined significantly in the CEBS group compared with the control group, whereas plasma insulin and HDL-cholesterol levels increased significantly. In conclusion, CEBS supplementation enhanced the regulation of body growth, increased tissue organic Cr concentrations, altered caecal microbiota and enhanced IR expression to produce significant changes in plasma biochemistry.
