Copper (Cu) and selenium (Se) are antioxidants and essential trace elements that have mutual interaction and are reported to have beneficial effects at supranutritional levels. The experiment was executed to evaluate the individual impact of supranutritional levels of targeted elements with the effect of their interactions in buffalo calves. Twenty male Murrah buffalo calves of about 8-9 months (bodyweight 112.1 ± 7.69 kg) were distributed into four groups of five calves in each group and fed either a control (C) diet or supplemented with supranutritional levels of Cu (T1), Se (T2), or combination of both (T3) for 120 days. Higher (P = 0.015) values of packed cell volume were observed in group T2 at day 120; otherwise, all other hematological parameters remained comparable among groups. Over the period (day 120 vs. day 0), an enhancement in the percentage of lymphocytes (P = 0.006) with a reduction in neutrophils (P = 0.028) and hemoglobin (P = 0.024) values was observed in the control group. An enhancement in the percentage of monocytes (P = 0.031), with a reduced percentage of neutrophils (P = 0.022), was reported in groups T2 and T3, respectively. Interaction of Cu and Se at supranutritional level (T3) dramatically reduced plasma Cu (P = 0.008) level against the control values, with an improvement in Se markers (i.e., plasma Se, P = 0.041 and enzyme glutathione peroxidase, P = 0.057) over the values in calves fed supplemental Se alone (T2). Additionally, Cu (T1 and T3) was forced to decline (P < 0.05) Zn level in the plasma of buffalo calves. Cu (T1, P < 0.05) and Se (T2 and T3, P ≤ 0.01) supplementation was able to improve their respective plasma levels. The interaction of two trace elements at the supranutritional level further helped in reducing the lipid peroxidation (P = 0.01) values as well. Though antioxidant vitamins and cell-mediated immunity remained unaffected, humoral immunity against antigen P. multocida was high (P = 0.005) in the group T2. The conclusion may be drawn that supranutritional Cu and Se were capable to influence certain blood parameters with an additional interaction effect due to simultaneous supplementation in buffalo calves.

Sources of supplemental minerals in the diet of animals are of important significance. Bio-availability of organic sources is believed to be more in the body as compared to regularly used inorganic sources and hence environment-friendly due to reduced mineral excretion, which in turn reduces their requirements in the diet as well. Twenty-four male Murrah buffalo (Bubalus bubalis) calves (about 18-20 months of age and 318.54 ± 8.85 kg body weight) were divided randomly into four groups of six animals each. In the control group (C, InOrg100) zinc (Zn), copper (Cu), and manganese (Mn) were supplemented through an inorganic source, while in treatment groups, organic source at the rate of 50, 75, or 100% (in groups T1 (Org50), T2 (Org75), and T3 (Org100), respectively) was fed at level as supplemented in the control group. Feeding was continued for a period of 180 days with blood sampling at day 0 followed by a regular interval of 45 days. Plasma samples were analyzed for trace elements Cu, Mn, Zn, and iron (Fe), total antioxidant status, ceruloplasmin, and superoxide dismutase (SOD) with cell-mediated and humoral immune response. Plasma levels of different trace minerals like Fe, Mn, and Cu remained unaffected with two sources and different levels of organic minerals, except the level of Zn, which showed higher (P < 0.05) levels in the group Org100 compared to others, and remained indicative of higher bio-availability through the organic source. The concentration of plasma total antioxidants indicated no adverse effect on the reduction of supplemental levels up to half of these minerals. Also, the level of plasma SOD was high (P < 0.05) at each level of the organic source as compared to the 100% level of the inorganic source. Immune response in respect of cell-mediated as well as humoral immunity did not show any reduction in different groups. The study indicated beneficial impacts of the organic source in the form of superior plasma Zn level as well as SOD concentrations. In addition, no negative effect on most of the studied parameters was observed after reducing supplemental trace minerals to half indicating higher bio-availability of organic trace minerals.

The present study was performed to ascertain the synergistic effects of phytogenic feed additives (PFA-7) supplementation on immune status and haemato-biochemical profile of buffalo calves. The PFA-7 is a mixture of neem seed cake (Azadirachta indica), mahua seed cake (Madhuca longifolia), fennel seed (Foeniculum vulgare), harad (Terminalia chebula), fruit pulp of bahera (Terminalia bellirica), fruit pulp of amla (Phyllanthus emblica) and ajwain seed (Trachyspermum ammi) mixed in 2:2:2:1:1:1:1 proportion. Male buffalo calves (n = 21) having similar age and body weight were allotted to three groups in a completely randomised design. The dietary treatments were viz. T1: control (without PFA-7) and T2 and T3: provided with PFA-7 at 2 and 4% of dry matter intake (DMI), respectively, plus sodium sulphate at 0.06% of DMI. The feeding trial was carried out for 4 months, and serum isolation was done on days 0, 60 and 120 post-feeding. The concentrations of total protein, albumin, globulin, aspartate transaminase and alanine transaminase increased, whereas cortisol and glucose decreased in the supplemented groups as compared to the control. The levels of triglycerides, urea, albumin/globulin ratio, calcium, phosphorus and alkaline phosphatase were not affected by the supplementation of PFA-7. Both cell-mediated and humoral immune response increased in the supplemented groups. The results revealed that PFA-7 positively impacted haemato-biochemical profile and both cellular and humoral immunity of the growing calves. The PFA-7 can be used as an alternative for chemical feed additives in the diet of growing calves.

Minerals play important role in the diet of an animal. Bio-availability of minerals largely gets affected by absolute as well as the relative amount of each mineral present in the diet of an animal. Copper and selenium are two such an essential elements affect utilization of each other in the gastrointestinal tract. The present study elucidates the utilization of copper and selenium at supra-nutritional levels (higher than nutritional requirements). Male Murrah buffalo (Bubalus bubalis) calves (n = 10, 8-9 months, 111.7 ± 12.55 kg body weight) were divided equally into two groups and fed either a standard (Control) diet or the same diet supplemented with 0.3 ppm selenium (Se) and 10 ppm copper (Cu) (Treatment). Supplementation was made using liquid solutions of two inorganic mineral sources after mixing in the concentrate mixture and study lasts for a period of 80 days. Blood samples were collected just before starting supplementation (designated as 0 day of study) and at day 40 and 80 after starting supplementation. Blood samples were subjected to haematological parameters, plasma minerals and various oxidative stress-related parameters were determined with the cell-mediated and humoral immune response against antigen P. multocida (P52 strain). Supra-nutritional Se with Cu had higher blood monocytes (P < 0.05) and plasma selenium (P < 0.01) levels, while other hematological parameters and plasma minerals (except zinc, which was lower (P = 0.025) at day 80 in the treatment group) remained unaffected. Among markers for oxidative stress in blood, levels of lipid peroxidation were lesser (P < 0.01), at day 80 and overall mean values of the enzyme glutathione peroxidase and catalase were higher (P < 0.05) in the supra-nutritional group against control values. The overall mean activity of other oxidative stress markers including reduced glutathione, ceruloplasmin as well as the concentration of α tocopherol, retinol, and ß carotene remained unaffected due to supra-nutritional Se and Cu. Although cell-mediated immune response remained comparable (P > 0.05) between groups, higher (P < 0.05) overall mean antibody titer values, as well as the values at day 80, was reported in supra-nutritional Se + Cu group. The study concluded that supra-nutritional Se with Cu in the ration of growing Murrah buffalo calves was helpful to reduce the oxidative stress and to enhance the humoral immune response. Simultaneously, higher plasma Se level and number of monocytes in blood highlighted the additional role of selenium and copper in a ration of growing buffalo calves as compared to its normal recommended dose.

A study of 120 days was undertaken to ascertain the effect of mannan-oligosaccharides (MOS) and Lactobacillus acidophilus supplementation on growth performance, nutrient utilization and faecal characteristics in Murrah buffalo calves. Twenty Murrah buffalo calves of 5-7 days old and 31 ± 2.0 kg of body weight (BW) were randomly assigned into four groups. Group I served as the control (CON) in which only basal diet (concentrate mixture and green fodder) was provided, without any supplementation. Mannan-oligosaccharides at 4 g/calf/day were supplemented as prebiotic to Group II (PRE), whereas Group III (PRO) received Lactobacillus acidophilus in the form of fermented milk as probiotic at 200 ml/calf/day having 108  CFU/ml and Group IV (SYN) was supplemented with both MOS and Lactobacillus acidophilus as synbiotic at similar dose. Final BW (kg), dry matter intake, average daily gain, feed conversion efficiency and structural growth measurements were improved (p < .05) in the treatment groups compared to control. Digestibility of neutral detergent fibre was higher (p < .05) in SYN followed by PRE and PRO than control. The faecal lactobacilli and bifidobacterium population was higher (p < .05) in all the supplemented groups with a concomitant reduction in faecal coliform count as compared to control. Faecal ammonia, lactate and pH were also altered favourably (p < .05) in all the supplemented groups as compared to CON. The faecal volatile fatty acids were higher (p < .05) in PRE, PRO and SYN group than CON. The incorporation of MOS and Lactobacillus acidophilus in diet either individually or in combination as synbiotic has the potential to improve the performance and faecal characteristics in Murrah buffalo calves; however, the observed responses among the treatment groups were more evident in the synbiotic fed group compared to individual supplementation of MOS and Lactobacillus acidophilus.

In the current study, we compared the therapeutic effects of a non-steroidal and a steroidal anti-inflammatory drug on the production of pro-inflammatory cytokines, interleukin-1ß (IL-1ß), interleukin-6 (IL-6), interleukin-12p40 (IL-12p40), interferon gamma (IFNγ), and tumor necrosis factor alpha (TNF-α) in the blood of water buffalo (Bubalus bubalis) calves naturally infected by bronchopneumonia. Twenty-seven buffalo calves (7 ± 2-month-old, 163 ± 12 kg) reared in smallholder farms in El-Dakahlia province in Egypt were identified to have bronchopneumonia and randomly allocated into three equal groups. Ten clinically healthy buffalo calves with negative bronchoalveolar lavage results were served as negative control. Diseased calves were treated with tulathromycin alone, a combination of tulathromycin with dexamethasone (steroidal anti-inflammatory drug) or tulathromycin with flunixin meglumine (non-steroidal anti-inflammatory drug). The results revealed significant elevations (P < 0.05) in the production of selected cytokines in all diseased calves in comparison with healthy animals. Six days post-treatment, a significant inhibition (P < 0.05) in the production of all assessed cytokines was observed in the blood of all treated calves. Interestingly, the serum concentrations of IL-1ß and IL-12p40 were returned to the normal levels in pneumonic calves treated with the combination therapy of tulathromycin and flunixin meglumine. A strong significant positive correlation (P < 0.05) was detected between clinical sum scoring and IL-12p40 and TNF-α concentrations. The obtained results indicate the selectively potent anti-inflammatory effect of flunixin meglumine on the production of pro-inflammatory cytokines in pneumonic buffalo calves and highlight the efficacy of flunixin meglumine in the treatment of bronchopneumonia in buffalo calves when used in combination with tulathromycin.

Failure to grow (ill-thrift) in calves has a negative effect on animal production and health. The present study was carried out from November, 2009 to May, 2013 to investigate the risk factors of ill-thrift in buffalo calves. A total of 344 calves at 78 smallholder farms were selected randomly. A questionnaire was designed to include managemental, nutritional and disease risk factors. Serum selenium, copper, zinc, iron, calcium, phosphorus and magnesium were measured. Data were subjected to logistic regression analysis and results were expressed as p value, odds ratio (OR) and confidence interval (CI). Fifty-five calves (15.9%) showed ill-thrift. On animal level, the final multivariate logistic regression model showed a significant association between ill-thrift and early weaning (p < 0.01; OR: 45.755; CI: 4.35-480.25), diarrhea (p < 0.05; OR: 41.315; CI: 1.710-998.0), indoor management (p < 0.05; OR: 63.56; CI: 2.701-14.96) and low serum phosphorus (p < 0.01; 292.0; CI: 5.256-16.23). On farm level, inadequate mineral supplementation (p < 0.001; OR: 18.62; CI: 3.89-88.9) and irregular use of anthelmintics (p < 0.05; OR: 7.95; CI: 1.53-41.23) were the potential factors. Clinically, ill-thrift calves were more likely to have alopecia (p < 0.01), recumbency (p < 0.01), emaciation (p < 0.001), hypothermia (p < 0.01), inappetance (p < 0.001), lacrimation (p < 0.001), hypomotile rumen (p < 0.001), and pale mucous membrane (p < 0.001). The results of the present study indicate that ill-thrift in buffalo calves could occur as a result of interaction between management errors and disease factors. Identification of the risk factors associated with ill-thrift may provide useful information, which assist to construct the suitable preventive measures.

This study validated the use of mutant prevention concentration (MPC) and pharmacokinetic and pharmacodynamic (PK-PD) modeling approach for optimization of dose regimen of enrofloxacin to contain the emergence of Pasteurella multocida resistance. The PK and PD characteristics of enrofloxacin were investigated in buffalo calves after intramuscular administration at a dose rate of 12 mg/kg. The concentration of enrofloxacin and ciprofloxacin in serum were determined by high-performance liquid chromatography. The serum peak concentration (Cmax), terminal half-life (t1/2K10), volume of distribution (Vd(area)/F) and mean residence time (MRT) of enrofloxacin were 1.89 ± 0.35 µg/ml, 5.14 ± 0.66 h, 5.59 ± 0.99 l/kg/h and 8.52 ± 1.29 h, respectively. The percent metabolite conversion ratio of ciprofloxacin to enrofloxacin was 79. The binding of enrofloxacin to plasma proteins was 11%. The MIC, MBC and MPC for enrofloxacin against P. multocida were 0.05, 0.06 µg/ml and 1.50 µg/ml.In vitro and ex-vivo bactericidal activity of enrofloxacin was concentration dependent. Modeling of ex-vivo growth inhibition data to the sigmoid Emax equation provided AUC24h/MIC values to produce bacteriostatic (19 h), bactericidal (43 h) and bacterial eradication (64 h). PK-PD data in conjunction with MPC and MIC90 data predicted dosage schedules for enrofloxacin that may achieve optimum efficacy in respect of bacteriological and clinical cure and minimize the risk of emergence of resistance.
