RESUMEN

Panus lecomtei is emerging as an edible mushroom found worldwide and particularly in the Northern Hemisphere. The mushroom contains a substantial amount of useful nutritional and medicinal compounds. In the present study, we have examined a specimen of P. lecomtei submitted to the ICAR-Directorate of Mushroom Research gene bank. The specimen was examined for taxonomical characters using classical and molecular tools. Attempts were made for cultivation of this mushroom under controlled conditions using sawdust-based substrate. The specimen was characterized by its purplish fruiting body having coarse, rigid, dense hairs on the cap, pubescent stipe, and abundant metuloids. Molecular identification through conserved ITS region was done and the sequence was deposited in NCBI GenBank under accession number MN332200. Nutritional profiling and biochemical analysis showed that the mushroom contained high carbohydrate but low fat contents. The mushroom showed the presence of phenolics, ß-carotene, and lycopene. The analysis also showed substantial antioxidant properties in the mushroom. The findings presented herein point out that P. lecomtei can be used as a potential edible mushroom for diversification of mushroom production in India.

RESUMEN

A case of fungal keratitis due to Coprinellus radians is reported. To our knowledge, fungal keratitis caused by this species was rare. Fungal hyphae were detected in corneal scrapings, and isolates were identified by morphology and by sequencing the internal transcribed spacer region of ribosomal DNA. The patient was treated with systemic and local antifungal therapy for 5 days, and lamellar keratoplasty was performed after no obvious improvement in symptoms. The in vitro antifungal susceptibilities of the case strain were tested for six antifungal agents. The results showed that 5-fluorouracil was resistant, fluconazole was moderately sensitive, and the other drugs assayed (amphotericin B, posaconazole, itraconazole and voriconazole) were highly effective against this fungus.

RESUMEN

Mushroom Phellinus linteus ("Sanghuang" in Chinese) is a popular medicinal polypore used to treat several disorders through its various biological functions. Inonotus sanghuang is claimed to produce general immune-potentiating and strengthening, anti-inflammatory, anti-tumor and anti-microbial properties, but its effect on acute lung inflammation and oxidative stress are not clearly understood. To determine the effect and mechanism of the polyphenols-rich ethyl acetate fraction from wild I. sanghuang extract (ISE) on acute lung injury (ALI) induced by bleomycin (BLM), female C57BL/6 mice were fed ISE (0%, 0.15% or 0.6% in diet) for 4 weeks prior to challenge with BLM. Bronchoalveolar lavage fluid (BALF) from lung, spleen and lung tissues were collected on day 3 after BLM challenge for histological, oxidative stress, molecular and biochemical analysis. ISE supplementation improved pathological features in lung injury scores and reduced lung wet-to-dry ratios. Moreover, ISE reduced inflammatory cell infiltration and the pro-inflammatory cytokines including IL-1ß, IL-6 and TNF-α in BALF, decreased the MPO activity and the MDA level and increased the SOD, CAT and GSH-Px activities in lung tissue homogenates. Further mechanism analysis demonstrated that dietary ISE inhibited NF-κB signal. Finally, peripheral immune function analysis showed that ISE had less effect on immune response including splenocyte producing inflammatory cytokines and T cell proliferation except for IL-1ß and IL-2. Our findings indicate the possibility that dietary ISE attenuates ALI induced by BLM through correcting the inflammation and oxidation balance at least in part via inhibiting NF-κB signal in vivo, suggesting that ISE might be a valuable medicinal food effective in improving lung injury.

This study is, to our knowledge, the first to investigate the pharmacological importance of wild Tunisian mushrooms. Ethanolic extracts of 5 Tunisian mushrooms-Phellinus torulosus, Fomes fomentarius, Trametes versicolor, Pisolithus albus, and Fomitopsis pinicola-were collected from the Kroumirie Region (North Tunisia). The dry basidomes of mushrooms were extracted using ethanol and evaluated for total polyphenol, flavonoid, flavonol, tannin, proanthocyanidin, and anthocyanin content. In addition, their antioxidant activities were determined using 3 assays (testing 2,2-diphenyl-1-picrylhydrazyl [DPPH] radical scavenging, the reducing power of iron, and the iron-chelating power). Their antimicrobial activities were assessed against 8 bacterial species. The results revealed the presence of significant differences between the secondary metabolites and biological activities of the different tested extracts. In addition, significant correlations were observed between antioxidant activities and phenolic contents. Crude ethanol extracts prepared from basidomes of F. fomentarius and Ph. torulosus have higher total phenolic content and antioxidant activity per the DPPH and metal-chelating activity assays. The reducing power assay showed that the ethanolic extract of F. pinicola had the highest activity. Ethanolic extracts of the 5 mushrooms have antibacterial activity against the evaluated strains.

This study investigated the nutritive properties of the culinary-medicinal mushroom Auricularia auricula- judae. From the analysis, among the major minerals, potassium was found in the highest concentration, with a value of 172.03 ± 8.62 mg/kg, whereas manganese had the highest concentration among trace elements, with a value of 1.66 ± 0.11 mg/kg. Glutamic acid was the amino acid detected in the largest amounts in this study, with a value of 10.09 ± 1.86 mg/kg. Although glutamine and asparagine were not detected in this mushroom, cysteine and methionine had the lowest concentrations, with values of 0.34 ± 0.01 and 0.80 ± 0.03 mg/kg, respectively. The predominant oil was 9,12-octadecadienoic acid (Z,Z)methyl ester, with retention times of 21.246 and 21.715 minutes, and comprising percentages of the total of 4.309 and 47.385, respectively.

Invasive fungal infection is a serious complication following allogeneic hematopoietic stem cell transplantation. Pulmonary infection due to Hormographiella aspergillata is an uncommon condition associated with a high mortality rate. The susceptibility of H. aspergillata to available antifungal agents is not well established. We report for the first time a case of H. aspergillata lung infection that responded poorly to conventional treatment with liposomal amphotericin B (LAmB; 3 mg kg-1 of body weight per day) with renal damage at higher posology (5 mg kg-1 of body weight per day), but improved rapidly after addition of nebulized LAmB to intravenous LAmB (3 mg kg-1 of body weight per day). Successful treatment of our patient using nebulized LAmB would be worth evaluating in cases refractory to standard treatment or when the reference treatment may not be extended due to interaction or side effects.

The Thelephora ganbajun mushroom has been found to be a potential rich source of natural antioxidants. In this study, an ultrasound-assisted extraction (UAE) technique together with GRAS (generally recognized as safe) solvents (ethanol and water) was used to maximize the extraction of antioxidants from Thelephora ganbajun. Five extraction parameters (ethanol concentration, solvent to solid ratio, extraction time, temperature and ultrasound power) were investigated by single-factor experiments, and then a central composite rotatable design was employed to study interaction of three key extraction parameters. The optimum conditions were as follows: 57.38% ethanol, 70.15 mL/g solvent to solid ratio, 10.58 min extraction time, 40 °C extraction temperature and 500 W ultrasound power. Under the optimum conditions, the antioxidant activity obtained was 346.98 ± 12.19 µmol Trolox/g DW, in accordance with the predicted value of 344.67 µmol Trolox/g DW. Comparison of UAE with conventional maceration and Soxhlet extraction, the UAE method showed stronger extract efficiency in a shorter extraction time. These results showed that UAE was an effective technique to extract antioxidants from Thelephora ganbajun. Furthermore, the extracts obtained under the optimized conditions exhibited antiproliferative activities toward human lung (A549), breast (MCF-7), liver (HepG2) and colon (HT-29) cancer cells, especially for liver and lung cancer cells. In addition, rutin, 2-hydrocinnamic acid and epicatechin were identified in the extract, which might contribute to antioxidant and antiproliferative activities.

A recent study published in this journal demonstrates the pitfalls faced by researchers who utilize commercial products as their test samples without proper characterization. Labeling of commercial mushroom products is often incorrect, which can lead to erroneous interpretations and conclusions. Nine of the 10 samples of commercially branded products used in the study and identified as ground mushrooms were actually grain spawn: mycelium propagated on grain.

Disseminated infection by Hormographiella aspergillata is extremely rare and small intestine involvement has not been reported previously. A 51-year-old man with myelodysplastic syndrome developed pneumonia after cord blood cell transplantation. Fungal growth from the biopsied lung was identified as H. aspergillata by morphology and the gene analysis. Although antifungal agents including voriconazole and liposomal amphotericin B were administered, he died of disseminated H. aspergillata infection. We review the literature and discuss the treatment and prognosis.

Hericium erinaceus possesses multiple medicinal values. To date, however, there have been few studies of the systemic screening of H. erinaceus strains, and the neuroprotective effects of H. erinaceus prepared from homogenized, fresh fruiting bodies are not fully understood. In this study, 4 random primers were selected and used in random amplified polymorphic DNA (RAPD) polymerase chain reaction (PCR) to screen and evaluate the genetic diversity of 19 commercial strains of H. erinaceus from different localities in China. A total of 66 bands were obtained, and the percentage of polymorphic loci reached 80.30%. Five dendrograms were constructed based on RAPD by Jaccard cluster and within-group linkage analysis. Primer S20 as well as all 4 primers had great potential as specific primers for RAPD-PCR molecular identification and differentiation of H. erinaceus strains. Based on the results of submerged culture and fruiting body cultivation, strains HT-N, HT-J1, HT-C, and HT-M were identified as superior among the 19 H. erinaceus strains. Further study showed that the oral preparation of homogenized, fresh fruiting bodies of H. erinaceus could attenuate the Aß25-35-triggered damage in PC12 cells by significantly increasing cell viability and by decreasing the release of lactate dehydrogenase. In conclusion, RAPD-PCR combined with liquid and solid cultures can be used well in the screening and identification of H. erinaceus strains, and products prepared from homogenized, fresh fruiting bodies of H. erinaceus had neuroprotective effects on PC12 cells.

Hormographiella aspergillata is a rare causative agent of invasive filamentous breakthrough infection, mostly arising after echinocandin exposure. We report a neutropenic patient who developed a severe sino-orbito-cerebral H. aspergillata infection while receiving empirical caspofungin, successfully controlled by an aggressive strategy associating surgical debridement and combined high-dose regimen of antifungal drugs.

RESUMEN

The author, as a reviewer of many international journals, describes his long-standing experiences with incorrect identification of mushroom and fungal species and the resultant incorrect naming of those species that served as experimental models. From his own praxis, he selected several characteristic examples that sometimes ended in a curious situation. Some recommendations to authors of publications and persons responsible for the proper naming of mushrooms under study are summarized.

Antecedentes. Agaricus subrufescens Peck es un hongo cuyo cultivo ha despertado gran interés en todo el mundo en los últimos años, adquiriendo gran popularidad. Sus propiedades medicinales y culinarias hacen prever una rápida expansión del cultivo en todo el mundo. Objetivos. El trabajo plantea como objetivo la evaluación del efecto sobre los principales parámetros de producción de 3 cepas de Agaricus subrufescens que se han hecho fructificar sobre 5 capas de cobertura diferentes. Métodos. Se ha llevado a cabo un ciclo de cultivo de Agaricus subrufescens en condiciones controladas en el que se han evaluado los principales parámetros de producción. Resultados. Los mejores resultados han sido proporcionados por la cepa ABL 99/30. Las coberturas basadas en turba presentan mejor comportamiento que las basadas en suelo mineral. El mayor rendimiento (6,75 kg/m−2, eficiencia biológica 27,57 kg/dt) ha sido proporcionado por la combinación ABL 99/30-Euroveen. Conclusiones. Nuestros resultados evidencian que la combinación de la cepa ABL 99/30 utilizando una capa de cobertura basada en turba (Euroveen) ofrece un alto potencial para ser utilizada a escala comercial por el sector productor de hongos. La disponibilidad de alternativas a las especies de hongos comestibles cultivadas habitualmente puede suponer un mejor aprovechamiento de recursos y una mayor rentabilidad económica de la actividad (AU)


Background. Agaricus subrufescens Peck is a mushroom whose cultivation has aroused great interest worldwide in recent years, and is becoming increasingly popular. A rapid expansion of culture throughout the world is foreseen because of its medicinal and culinary properties. Aims. This work assesses the effect of 5 different casing layers on the production of 3 strains of Agaricus subrufescens. Methods. A growth cycle of Agaricus subrufescens under controlled conditions has been carried out. The main production parameters were evaluated. Results. The best results were provided by the ABL 99/30 strain. Peat-based casings have a better yield than those based on mineral soil. The highest yield (6.75 kg/m2, biological efficiency 27.57 kg/dt) was provided by the combination ABL 99/30-Euroveen. Conclusions. Our results suggest that the combination of the strain ABL 99/30 using a peat-based casing layer (Euroveen) offers a high potential for use on a commercial scale by the edible mushroom production sector. The availability of alternatives to the usually cultivated species can make better use of resources, and increase the profitability of this activity (AU)

ETHNOPHARMACOLOGICAL RELEVANCE: Hericium is a genus of mushrooms (fungus) in the Hericiaceae family. Hericium erinaceus (HE) has been used for the treatment of digestive diseases for over 2000 years in China. HE possesses many beneficial functions such as anticancer, antiulcer, antiinflammation and antimicrobial effects, immunomodulation and other activities. The aim of the studies was to evaluate the anticancer efficacy of two extracts (HTJ5 and HTJ5A) from the culture broth of HE against three gastrointestinal cancers such as liver, colorectal and gastric cancers in both of in vitro of cancer cell lines and in vivo of tumor xenografts and discover the active compounds. MATERIALS AND METHODS: Two HE extracts (HTJ5 and HTJ5A) were used for the studies. For the study of chemical constituents, the HTJ5 and HTJ5A were separated using a combination of macroporous resin with silica gel, HW-40 and LH-20 chromatography then purified by semipreparative high-performance liquid chromatography (HPLC) and determined by nuclear magnetic resonance (NMR) spectra. For the in vitro cytotoxicity studies, HepG2 and Huh-7 liver, HT-29 colon, and NCI-87 gastric cancer cell lines were used and MTT assay was performed to determine the in vitro cytotoxicity. For in vivo antitumor efficacy and toxicity studies, tumor xenograft models of SCID mice bearing liver cancer HepG2 and Huh-7, colon cancer HT-29 and gastric cancer NCI-87 subcutaneously were used and the mice were treated with the vehicle control, HTJ5 and HTJ5A orally (500 and 1000 mg/kg/day) and compared to 5-fluorouraci (5-FU) at the maximum tolerated dose (MTD, 25-30 mg/kg/day) intraperitoneally daily for 5 days when the tumors reached about 180-200 mg (mm(3)). Tumor volumes and body weight were measured daily during the first 10 days and 2-3 times a week thereafter to assess the tumor growth inhibition, tumor doubling time, partial and complete tumor response and toxicity. RESULTS: Twenty-two compounds were obtained from the fractions of HTJ5/HTJ5A including seven cycli dipeptides, five indole, pyrimidines, amino acids and derivative, three flavones, one anthraquinone, and six small aromatic compounds. HTJ5 and HTJ5A exhibited concentration-dependent cytotoxicity in vitro against liver cancer HepG2 and Huh-7, colon cancer HT-29, and gastric cancer NCI-87 cells with the IC50 in 2.50±0.25 and 2.00±0.25, 0.80±0.08 and 1.50±0.28, 1.25±0.06 and 1.25±0.05, and 5.00±0.22 and 4.50±0.14 mg/ml; respectively. For in vivo tumor xenograft studies, HTJ5 and HTJ5A showed significantly antitumor efficacy against all four xenograft models of HepG2, Huh-7, HT-29 and NCI-87 without toxicity to the host. Furthermore, HTJ5 and HTJ5A are more effective than that of 5-FU against the four tumors with less toxicity. CONCLUSION: HE extracts (HTJ5 and HTJ5A) are active against liver cancer HepG2 and Huh-7, colon cancer HT-29 and gastric cancer NCI-87 cells in vitro and tumor xenografts bearing in SCID mice in vivo. They are more effective and less toxic compared to 5-FU in all four in vivo tumor models. The compounds have the potential for development into anticancer agents for the treatment of gastrointestinal cancer used alone and/or in combination with clinical used chemotherapeutic drugs. However, further studies are required to find out the active chemical constituents and understand the mechanism of action associated with the super in vivo anticancer efficacy. In addition, future studies are needed to confirm our preliminary results of in vivo synergistic antitumor efficacy in animal models of tumor xenografts with the combination of HE extracts and clinical used anticancer drugs such as 5-FU, cisplatin and doxurubicin for the treatment of gastrointestinal cancers.

Los macromicetos han sido parte de la cultura humana desde hace miles de años y aparecen descritos como alimento humano en las más importantes civilizaciones de la historia. Se han descrito muchísimas propiedades nutricéuticas de los macromicetos, como sus propiedades anticancerígenas y antitumorales, hipocolesterolémicas, antivirales, antibacterianas, o inmunomoduladoras, entre otras. Dado que la producción de hongos por cultivo tradicional y la extracción de los metabolitos bioactivos en algunos casos son muy dispendiosas, la biotecnología es fundamental para el desarrollo de técnicas rentables y productivas para la obtención de estos metabolitos. Es el desarrollo de esta tecnología y la facilidad que proporciona en cuanto al manejo de sus variables, lo que ha permitido realizar el cultivo en medio líquido del micelio de macrohongos con significativa reducción de tiempo y aumento en la producción de sus metabolitos, lo que ha impulsado aún más su obtención y el estudio de compuestos con potencial como medicamentos, nutricéuticos y cuasifarmacéuticos tanto del medio agotado como del micelio. El objetivo de esta revisión es el de ofrecer una visión general de la utilización de la fermentación en estado líquido como herramienta tecnológica para la obtención de hongos comestibles, su estudio y el de sus bioactivos, mediante la descripción de las diferentes condiciones de cultivo que en los últimos años se han empleado, así como los resultados obtenidos. Se discutirá lo correspondiente a los géneros Agaricus, Flammulina, Grifola, Pleurotus y Lentinula, con énfasis en este último, dado que el Shiitake ha sido considerado desde siempre como el hongo medicinal por excelencia(AU)


Macromycetes have been part of the human culture for thousand years, and have been reported as food in the most important civilizations in history. Many nutraceutical properties of macromycetes have been described, such as anti-cancer, anti-tumour, cholesterol lowering, antiviral, antibacterial, or immunomodulatory, among others. Given that production of mushrooms by traditional cultivation and extraction of bioactive metabolites is very difficult in some cases, biotechnology is essential for the development of profitable and productive techniques for obtaining these metabolites. It is the development of this technology, and the ease in which it enables the use of its variables that has allowed mycelium to be cultivated in liquid medium of macrofungi, with a significant reduction in time and an increased production of metabolites. This increased production has led to the study of compounds that have medicinal, nutriceutical and quasi-farmaceutical potential, in the exhausted media and the mycelium. The aim of this review is to provide an overview of the use of liquid-state fermentation as a technological tool for obtaining edible fungi, and the study of these and their metabolites, by describing the different cultivation conditions used in recent years, as well as the results obtained. The relevance of Agaricus, Flammulina, Grifola, Pleurotus and Lentinula genera, will also be discussed, with emphasis on the last one, since Shiitake has been always considered as the ultimate medicinal mushroom(AU)

This study reports that acidic polysaccharide (PL) isolated from Phellinus linteus alleviated the septic shock induced by high dose lipopolysaccharide (LPS) injection in mice. To examine the origin of this effect, we investigated cytokine production in serum and the expression of MHC II in B cells and macrophages in areas of inflammation. Pretreatment with PL 24 h before LPS administration resulted in a significant inhibition of up to 68% of circulating tumor necrosis factor (TNF)-alpha, a moderate reduction of 45% of interleukine (IL)-12 and 23% of IL-1beta, but no significant reduction in IL-6. In addition, the expression of MHC II in B cells and macrophages was examined. Our results show that LPS-stimulated cytokine release and the level of MHC II can be modulated by in vivo administration of soluble PL in mice. The decrease of IL-1beta, IL-12 and TNF-alpha in sera and the down-modulation of MHC II during septic shock may contribute to the long survival of mice by PL. Administration of PL in vivo decreases IL-2, IFN-gamma and TNF-alpha production in splencotyes and enhances spontaneous cell apoptosis in macrophages and lymphocytes stimulated with LPS in vitro. Thus, part of the anti-inflammatory effects of PL treatment in vivo may result from the enhanced apoptosis of a portion of the activated macrophages and lymphocytes. The ability of PL to significantly reduce the TNF-alpha production indicates the potential of the polysaccharides in possible therapeutic strategies that are based on down-regulation of TNF-alpha.

Natural killer (NK) cells are directly cytotoxic for tumor cells and play a primary role in regulating immune responses. We monitored levels of NK cell cytotoxic activity in cancer patients receiving D-Fraction extracted from maitake mushrooms (Grifola frondosa). Elevated levels of cytotoxic activity were maintained for one year. To elucidate the mechanisms underlying long-term activation of NK cells during treatment with D-Fraction, we examined tumor volume and levels of IFN-gamma and TNF-alpha in MM46-bearing C3H/HeN mice to which D-Fraction was administered for 19 d. D-Fraction markedly suppressed tumor growth, corresponding with increases in TNF-alpha and IFN-gamma released from spleen cells and a significant increase in TNF-alpha expressed in NK cells. This suggests that the D-Fraction activates NK cells even on the 20th day after treatment. Furthermore, D-Fraction increased macrophage-derived interleukin (IL)-12, which serves to activate NK cells. These results suggest that NK cells are not only responsible for the early effects of D-Fraction on tumor growth, but also for the long-term tumor-suppressive effects of D-Fraction through increased IL-12 released from macrophages.

The current study was carried out to investigate the in vitro effects of an 85% methanol extract of dried Morus alba leaves on melanin biosynthesis, which is closely related to hyperpigmentation. These extracts inhibited the tyrosinase activity that converts dopa to dopachrome in the biosynthetic process of melanin. Mulberroside F (moracin M-6, 3'-di-O-beta-D-glucopyranoside), which was obtained after the bioactivity-guided fractionation of the extracts, showed inhibitory effects on tyrosinase activity and on the melanin formation of melan-a cells. This compound also exhibited superoxide scavenging activity that is involved in the protection against auto-oxidation. But its activity was low and was weaker than of kojic acid. These results suggest that mulberroside F isolated from mulberry leaves might be used as a skin whitening agent.
