RESUMEN

Herba Epimedii, known for its rich array of bioactive ingredients and widespread use in ethnopharmacological practices, still lacks a comprehensive understanding of its gastrointestinal biotransformation. In this study, we qualitatively explored the dynamic changes in Epimedium sagittatum components during in vitro simulated digestions, with a quantitative focus on its five major flavonoids. Notably, significant metabolism of E. sagittatum constituents occurred in the simulated small intestinal fluid and colonic fermentation stages, yielding various low molecular weight metabolites. Flavonoids like kaempferol glycosides were fully metabolized in the simulated intestinal fluid, while hyperoside digestion occurred during simulated colon digestion. Colonic fermentation led to the production of two known bioactive isoflavones, genistein, and daidzein. The content and bioaccessibility of the five major epimedium flavonoids-icariin, epimedin A, epimedin B, epimedin C, and baohuoside I-significantly increased after intestinal digestion. During colon fermentation, these components gradually decreased but remained incompletely metabolized after 72 h. Faecal samples after E. sagittatum fermentation exhibited shift towards dominance by Lactobacillus (Firmicutes), Bifidobacterium (Actinobacteria), Streptococcus (Firmicutes), and Dialister (Firmicutes). These findings enhance our comprehension of diverse stages of Herba Epimedii constituents in the gut, suggesting that the primary constituents become bioaccessible in the colon, where new bioactive compounds may emerge.

Objectives were to determine the effects of supplementing rumen-protected choline (RPC) from an established source with low (L, 28.8%) or a prototype with less lipid coating protection and high (H, 60.0%) concentrations of choline chloride on digestibility of fat and supra-mammary lymph metabolome in feed-restricted cows. Pregnant, nonlactating Holstein cows (n = 33; 11/treatment) at mean (±standard deviation) 231 ± 4.7 days of gestation were blocked by body condition (4.23 ± 0.47) and assigned to receive 0 (CON) or 25.8 g/d of choline ion from L (L25.8) or H (H25.8). Cows were adapted to the diet and then fed-restricted to 42% of the net energy of lactation required for maintenance and pregnancy for 9 days. Intake of metabolizable methionine was maintained at 19 g/d. On Day 9, cows were fed 450 g of saturated fatty acids (SFA), and feces and blood were sampled continuously for 24 h. Supra-mammary lymph was sampled 6 h after feeding SFA and metabolome was characterized. Feeding RPC increased digestibility of fat (CON = 80.4 vs. RPC = 86.0 ± 1.9%) and reduced the concentration of haptoglobin in serum (CON = 174 vs. RPC = 77 ± 14 µg/ml) independent of source of RPC fed. Feeding RPC increased the concentrations of triacylglycerol in serum (CON = 15.1 vs. RPC = 17.8 ± 1.9 mg/dl) in feed-restricted cows after feeding SFA, and the increment tended to be greater for cows fed H25.8 than L25.8. Supplementing RPC tended to increase the concentrations of triacylglycerol (CON = 11.4 vs. RPC = 15.8 ± 3.4 mg/dl) in supra-mammary lymph. Feeding RPC increased the concentration of choline and affected the concentrations of analytes involved in metabolic pathways associated with amino acid metabolism and biosynthesis of phospholipids in lymph compared with CON. Feeding RPC, independent of source used, increased fat digestibility with some changes in lymph metabolome in cows under negative nutrient balance.

The objective of this experiment was to evaluate the influence of nanoselenium (NANO-Se) addition on milk production, milk fatty acid synthesis, the development and metabolism regulation of mammary gland in dairy cows. Forty-eight Holstein dairy cows averaging 720 ± 16.8 kg of body weight, 66.9 ± 3.84 d in milk (dry matter intake [DIM]) and 35.2 ± 1.66 kg/d of milk production were divided into four treatments blocked by DIM and milk yields. Treatments were control group, low-Se (LSe), medium-Se (MSe) and high-Se (HSe) with 0, 0.1, 0.2 and 0.3 mg Se, respectively, from NANO-Se per kg dietary dry matter (DM). Production of energy- and fat-corrected milk (FCM) and milk fat quadratically increased (p < 0.05), while milk lactose yields linearly increased (p < 0.05) with increasing NANO-Se addition. The proportion of saturated fatty acids (SFAs) linearly decreased (p < 0.05), while proportions of monounsaturated fatty acids (MUFAs) linearly increased and polyunsaturated fatty acids (PUFAs) quadratically increased. The digestibility of dietary DM, organic matter (OM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF) quadratically increased (p < 0.05). Ruminal pH quadratically decreased (p < 0.01), while total VFA linearly increased (p < 0.05) with increasing NANO-Se addition. The acetic to propionic ratio decreased (p < 0.05) linearly due to the unaltered acetic molar percentage and a quadratical increase in propionic molar percentage. The activity of CMCase, xylanase, cellobiase and pectinase increased linearly (p < 0.05) following NANO-Se addition. The activity of α-amylase increased linearly (p < 0.01) with an increase in NANO-Se dosage. Blood glucose, total protein, estradiol, prolactin, IGF-1 and Se linearly increased (p < 0.05), while urea nitrogen concentration quadratically decreased (p = 0.04). Moreover, the addition of Se at 0.3 mg/kg from NANO-Se promoted (p < 0.05) mRNA and protein expression of PPARγ, SREBP1, ACACA, FASN, SCD, CCNA2, CCND1, PCNA, Bcl-2 and the ratios of p-ACACA/ACACA and BCL2/BAX4, but decreased (p < 0.05) mRNA and protein expressions of Bax, Caspase-3 and Caspase-9. The results suggest that milk production and milk fat synthesis increased by NANO-Se addition by stimulating rumen fermentation, nutrients digestion, gene and protein expressions concerned with milk fat synthesis and mammary gland development.

The digestion behavior of lipids plays a crucial role in their nutritional bioaccessibility, which subsequently impacts human health. This study aims to investigate potential variations in lipid digestion profiles among individuals of different ages, considering the distinct physiological functions of the gastrointestinal tract in infants, aging populations, and healthy young adults. The digestion fates of high oleic peanut oil (HOPO), sunflower oil (SO), and linseed oil (LINO) were investigated using in vitro digestion models representing infants, adults, and elders. Comparatively, lipid digestion proved to be more comprehensive in adults, leading to free fatty acid (FFA) levels of 64.53%, 62.32%, and 57.90% for HOPO, SO, and LINO, respectively. Besides, infants demonstrated propensity to selectively release FFAs with shorter chain lengths and higher saturation levels during the digestion. In addition, in the gastric phase, particle sizes among the elderly were consistently larger than those observed in infants and adults, despite adults generating approximately 15% FFAs within the stomach. In summary, this study enhances our fundamental comprehension of how lipids with varying degrees of unsaturation undergo digestion in diverse age groups.

The current study was conducted to explore the productive performance and health status of lactating buffaloes fed diets supplemented with probiotic and/or fibrolytic enzymes. Forty multiparous lactating Egyptian buffaloes (body weight 451 ± 8.5 kg) were equally assigned to four experimental groups: (1) the first group fed control diet, (2) second experimental group fed control diet plus 4 g of probiotic/kg dry matter (DM) (probiotic), (3) third experimental group fed control diet plus 4 g of fibrolytic enzymes/kg DM (enzymes) and (4) fourth experimental group fed control diet plus 2 g of probiotic + 2 g fibrolytic enzymes/kg DM (Mix), The experiment was extended for 63 days. Nutrients digestibility was estimated, daily milk yield was recorded and milk samples were analyzed for total solids, fat protein, lactose and ash. Blood serum samples were analyzed for glucose, total protein, albumin, urea-N, aspartate transaminase, alanine transaminase and cholesterol concentrations. Results showed that adding probiotic and/or fibrolytic enzymes improved nutrients digestibility (p < 0.05). The probiotic, enzymes and mix groups did not affect (p > 0.05) concentrations of serum total protein, albumin (A), globulin (G), albumin/globulin (A/G) ratio and urea-N concentrations. An improvement in daily milk yield (p < 0.0001) and energy-corrected milk (p = 0.0146) were observed with the probiotic and mix groups compared with the control. In conclusion, this study suggests that supplementing lactating buffaloes' diets with probiotic alone or in combination with fibrolytic enzymes would improve their productive performance without adversely impacting their health.

Carbohydrase supplementation in grow-finish pig diets improves energy, nutrient digestibility, and gastrointestinal function, but their efficacy in gestation diets is understudied. The experimental objective was to evaluate the efficacy of a multicarbohydrase to improve digestion, energetics, and various physiological functions in gestating sows fed soluble and insoluble fiber diets. On day 28 of gestation, 36 sows (186 ±â€…4.6 kg body weight), blocked by parity, were randomly assigned to a 2 × 2 factorial arrangement of dietary treatments (n = 9). Factors included fiber type of insoluble (IF; 20% dried distiller grains with solubles) or soluble fiber (SF; 20% sugar beet pulp) and with (+) or without (-) enzyme (0.05%, Rovabio Advance P10; Adisseo, Antony, France). Diets were fed from days 28 to 109 of gestation at a feeding level of 2.1 kg (SID-Lys 11 g/d and 4.5 net energy-Mcal/d). Two separate 9-d metabolism periods were conducted on days 50 to 59 (mid) and 99 to 108 (late) of gestation. During each period, days 1 to 3 served as an adaptation period, days 4 to 7 total urine and feces were collected (96-h) and followed by a 48-h lactulose-mannitol study. Serum and plasma were collected on days 50 and 99. Data were analyzed as repeated records using a linear mixed model with block as a random effect and fiber type, enzyme, and period and their interactions as fixed effects. Sows fed SF+ had increased serum IL-1ra (Fiber × Enzyme P = 0.035), and IL-2 (Fiber × Enzyme P = 0.042). In the presence of IF, multicarbohydrases increased serum lipopolysaccharide-binding protein, but not when supplemented with SF (Fiber × Enzyme P = 0.028). Circulating IL-8 and TNF-α were decreased in sows fed multicarbohydrases (P < 0.05). Multicarbohydrase supplementation increased the apparent total tract digestibility (ATTD) of gross energy (GE), dry matter, and neutral detergent fiber by 2.8%, 3.4%, and 8.3%, respectively (P < 0.05). Compared to IF-, the ATTD of hemicellulose was 5.3% greater in sows fed IF+ but did not differ from SF- and SF+ (Fiber × Enzyme P = 0.037). Sows fed IF+ had the greatest ATTD of insoluble dietary fiber (Fiber × Enzyme P = 0.011). Sows fed multicarbohydrases excreted less energy in their urine (519 vs. 469 GE kcal/d; Enzyme P = 0.033) and in their feces (985 vs. 900 GE kcal/d; Enzyme P = 0.003). This resulted in an improvement in both digestible energy (Enzyme P < 0.01) and metabolizable energy (Enzyme P = 0.041), irrespective of fiber type. In conclusion, multicarbohydrase supplementation increased the digestibility and energetic contribution of fiber, irrespective of adaptation time or fiber type, but modulation of inflammatory responses was unique to dietary fiber type.


This study evaluated the effects and mechanisms of supplementing multicarbohydrases to gestating sow diets containing either corn-dried distiller grains or sugar beet pulp fibrous co-products. Findings revealed significant energy, dry matter, and fiber digestibility enhancements with carbohydrase supplementation, irrespective of adaptation time. However, the interaction between enzyme supplementation and fiber type led to distinct immune marker responses. Specifically, granulocyte-macrophage colony-stimulating factor, interleukin-1 receptor antagonist, and interleukin-2 responses differed based on the presence of insoluble or soluble fiber. Sows in late gestation have greater nutrient and energy digestibility and altered nitrogen balance relative to day 50 of gestation. These findings highlight the potential benefits of incorporating carbohydrases into gestating sow diets to improve nutrient utilization and metabolic efficiency. Moreover, the study underscores the intricate interplay between enzyme supplementation, adaptation time, dietary fiber characteristics, and immune responses, offering valuable insights into the underlying mechanisms governing sow physiology during gestation and the influence of enzyme supplementation.

Growth performance, gizzard weight, ceca digesta short-chain fatty acids (SCFA), and apparent retention (AR) of components were investigated in broilers and turkeys in response to cereal grain type, fiber level, and multienzyme supplement (MES) fed from hatch to 28 d of life. 480-day-old male broiler chicks and equal number of turkeys were placed separately in metabolism cages (10 birds/cage) and allocated to 8 diets. The species-specific diets were a corn or wheat-based basal diet without (LF) or with 10% corn DDGS or wheat middlings (HF) and fed without or with MES. This effectively created a 2 (grain types) × 2 (fiber levels) × 2 (MES) factorial arrangement of treatments. The diets had TiO2 as an indigestible marker. Body weight, feed intake, and mortalities were recorded to calculate body weight gain (BWG) and feed conversion ratio (FCR). Excreta samples were collected on d 25 to 27 for AR, and all birds were necropsied for gizzard weight and ceca digesta on d 28. The interaction between grain and MES in broilers was such that wheat diets with MES had the lowest (P = 0.005) FCR. In broilers, LF diets had better (P = 0.010) FCR than HF diets. The wheat diets had the highest (P = 0.006) concentration of butyric acid in broilers. Broilers fed HF and corn diets had heavier gizzard than broilers-fed LF and wheat diets. The MES improved (P < 0.05) AMEn in HF, corn, and wheat diets in broilers. The turkeys fed wheat diets had the lowest (P = 0.019) FCR. Turkeys fed HF wheat diets had the heaviest (P < 0.001) gizzard. In turkeys, the MES improved AMEn in HF and LF corn diets, and only in LF wheat diets compared to respective controls. Treatments had no effect on turkeys cecal SCFA. In conclusion, grain type, fiber, and MES did not affect growth in both species. However, species exhibited differing FCR, gizzard, and energy utilization to fiber and MES.

Iron is routinely supplemented in broiler feeds intending to prevent dietary deficiencies. The present research was conducted with the objective of assessing Fe requirements of broilers when fed supplemental phytase. A total of 1,280 1-d-old male Cobb × Cobb 500 were distributed in a 2 × 5 factorial arrangement (phytase-supplemented feeds × 5 graded increases of supplemental Fe) in 80 battery cages, eight replications of eight chicks each. The trial was replicated once. Chicks were fed a Fe-deficient diet without phytase (Fe analyzed at 31.30 ± 3.79 mg/kg) from placement to 7 d and then randomly distributed into battery cages with corresponding dieting treatments with or without phytase and graded increases of supplemental Fe. Feeds were formulated with corn and soybean meal (SBM), laboratory-grade calcium carbonate, and phosphoric acid; therefore, the vast majority of dietary Fe originated from corn and SBM (analyzed diet had 53.3 ± 1.41 mg/kg Fe). Phytase was added in excess to the producer recommendation of 1,000 FYT (4,452 ± 487 FYT/kg analyzed) such that phytate degradation was expected to be maximized. Supplemental Fe was from laboratory-grade ferrous sulfate heptahydrate (FeSO47H2O) which was increasingly added to the feeds (analyzed Fe in the supplemented feeds were: 53.3 ± 1.41, 65.5 ± 0.59, 77.2 ± 1.97, 87.6 ± 1.72, 97.7 ± 1.33 mg/kg). There were no interactions between phytase and dietary Fe for any response throughout the study (P > 0.05). Supplementing phytase had no effects on Fe intake or Fe excretion, as well as on hematocrit (Ht), hemoglobin (Hb), ferritin, Fe contents in the liver or thigh muscle color (P > 0.05). However, phytase-supplemented feeds produced better live performance as well as higher ileal digestible energy and Fe digestibility (P < 0.05). No effects were found for dietary Fe in live performance at day 28 (P > 0.05). On the other hand, increasing dietary Fe led to linear increases in Fe retention and excretion, Fe contents in livers, as well as Ht and Hb at 14 d (P < 0.05). Quadratic responses (P < 0.05) were observed for Hb at 21 d, serum ferritin on days 14, 21, and 28 (maximum responses were 83.3, 104.0, 91.9, and 88.3 mg/kg Fe, respectively). In conclusion, supplementing Fe adding to a total of 97.7 mg/kg dietary Fe did not affect live performance traits. However, the average of Fe-related blood parameters was maximized at 91.9 mg/kg dietary Fe. Supplementing phytase provided a significant increase in Fe digestibility.

From the eighth day, a total of 1,280 male broilers were distributed in a combination of diets supplemented with phytase or not and 5 graded increases in dietary Fe. Diets were formulated with corn and soybean meal, laboratory-grade calcium carbonate, and phosphoric acid. Phytase was added in excess (4,452 ± 487 FYT/kg analyzed) to facilitate complete degradation of dietary phytate. Laboratory-grade ferrous sulfate heptahydrate was increasingly added to the feeds to provide Fe. Iron in the experimental diets was present at 53.3 ± 1.41, 65.5 ± 0.59, 77.2 ± 1.97, 87.6 ± 1.72, 97.7 ± 1.33 mg/kg. Supplementing diets with phytase resulted in enhanced live performance, along with increased digestibility of ileal energy and Fe. Linear increases in Fe retention and excretion, hepatic Fe contents, and serum ferritin were observed with the progressive increase in dietary Fe. The supplementation of a total of 97.7 mg/kg of Fe in diets was found to have no significant impact on live performance traits. However, the Fe-related blood parameters reached their maximum levels at a dietary Fe level of 91.9 mg/kg. Phytase supplementation provided a significant increase in the digestibility of Fe and other nutrients evaluated.

Lipids are a concentrated source of energy with at least twice as much energy as the same amount of carbohydrates and protein. Dietary lipids provide a practical alternative toward increasing the dietary energy density of feeds for high-performing modern broilers. However, the digestion and absorption of dietary lipids are much more complex than that of the other macronutrients. In addition, young birds are physiologically limited in their capacity to utilise dietary fats and oils effectively. The use of dietary emulsifiers as one of the strategies aimed at improving fat utilisation has been reported to elicit several physiological responses including improved fat digestibility and growth performance. In practical terms, this allows for the incorporation of lipids into lower-energy diets without compromising broiler performance. Such an approach may potentially lower feed costs and raise revenue gains. The current review revisits lipids and the different roles that they perform in diets and whole-body metabolism. Additional information on the process of dietary lipid digestion and absorption in poultry; and the physiological limitation brought about by age on lipid utilisation in the avian gastrointestinal tract have been discussed. Subsequently, the physiological responses resulting from the dietary supplementation of exogenous emulsifiers as a strategy for improved lipid utilisation in broiler nutrition are appraised. Suggestions of nascent areas for a better understanding of exogenous emulsifiers have been highlighted.

Three experiments evaluated effects of adaptation diet and exogenous ß-glucanase and xylanase on TMEn of barley and rye. Single Comb White Leghorn roosters were fed adaptation diets based on corn/soybean meal (SBM), barley/SBM with and without ß-glucanase, or rye/corn/SBM with and without xylanase for 4 wk. In Experiments 1 and 2, after the adaptation period, TMEn was determined using a 48 h precision-fed rooster assay for 100% barley or 100% rye diets with or without ß-glucanase or xylanase, respectively. Experiment 3 consisted only of feeding adaptation diets for 4 wk. Cecal samples were collected at the end of experiments for microbial ecology, short-chain fatty acid (SCFA) profiles, and enzyme activity analyses. In Experiments 1 and 2, ß-glucanase increased (P < 0.05) TMEn of barley, and there was no significant effect of adaptation diet on TMEn values. Total cecal Eubacteria and Ruminococcaceae were decreased (P < 0.05) and Escherichia coli were increased (P < 0.05) at the end of the TMEn assay compared with the end of the adaptation period (with no TMEn assay). There was a large decrease (P < 0.05) for most cecal SCFA at the end of the TMEn assay compared with the end of the adaptation period. Both cecal ß-glucanase and xylanase activity were increased for birds fed adaptation diets containing the respective enzyme. In Experiment 3, there were no consistent effects of adaptation diet on cecal microbial profiles or SCFA but cecal ß-glucanase activity was increased (P < 0.05) by exogenous ß-glucanase for barley and cecal xylanase activity was increased (P < 0.05) by exogenous xylanase for rye. Overall, the results indicated that TMEn of barley was increased by exogenous ß-glucanase, adaptation diet did not significantly influence the TMEn response to the dietary enzymes, and cecal fermentation (based on cecal SCFA) was greatly reduced by the TMEn assay. Cecal ß-glucanase and xylanase activity, however, were often increased by feeding high barley and high rye diets containing exogenous enzymes.

This study determined the apparent ileal digestibility (AID) and standard ileal digestibility (SID) of crude protein (CP) and amino acids (AA) of six cottonseed meal (CSM) samples in pregnant and non-pregnant sows. Two CSM samples were processed by expelling with a CP level of 40.67% (ECSM41) and 44.64% (ECSM45), and four samples were processed by solvent-extracted which contained graded CP levels of 45% (SECSM45), 51.16% (SECSM51), 56.44% (SECSM56), and 59.63% (SECSM60). Landrace ×Yorkshire third parity sows, 7 at gestation and 14 non-pregnant, were fitted with T-cannula in the distal ileum. Pregnant sows were allotted to a 7 × 6 Latin square design with a cornstarch-based nitrogen-free (NF) diet and the six CSM diets, and non-pregnant sows were allotted to a replicated 7 × 3 Latin square design with seven diets and three periods, respectively, resulting in a total of six replicates per treatment. All experimental sows were fed 3.0 kg/d of the experimental diets. The AID of CP in ECSM41 (75.58%) was lower than in SECSM51 (80.42%), SECSM56 (80.50%), and SECSM60 (82.44%) diets for pregnant sows (P < 0.05). The AID of CP in ECSM41 (77.88%) was significantly lower than in SECSM60 (81.87%) diets for non-pregnant sows (P < 0.05). The physiological phase did not affect the AID of CP (P > 0.05). The SID of CP was affected by diets for both pregnant (P < 0.01) and non-pregnant sows (P = 0.06). The physiological phase also affected the SID of CP (P < 0.01). The AID of histidine, leucine, methionine, threonine, and tryptophan significantly differed between different CSM samples in both pregnant (P < 0.05) and non-pregnant sows (P < 0.05). The AID of dispensable AA aspartic acid, cysteine, glutamic acid, serine, and tyrosine differed between different CSM samples of both pregnant (P < 0.05) and non-pregnant sows (P < 0.05). For pregnant sows, the indispensable AA cysteine, glycine, proline, and tyrosine had significantly different SID between different groups (P < 0.05). For non-pregnant sows, the SID of arginine, lysine, methionine, threonine, aspartic acid, cysteine, and serine had different values among different diets (P < 0.05). In conclusion, the current study presented that the ileal AA digestibility of CSM fed to pregnant and non-pregnant sows increased with the decreased of fiber content, and the current findings can contribute to a precise formulation of diets for sows using CSM.


As a protein-rich cottonseed byproduct, cottonseed meal (CSM) is considered a vegetable protein source that can substitute soybean meal in the feed of livestock animals. However, the presence of free gossypol and high fiber levels in CSM have been limiting factors for its use in growing and finishing pigs, yet its nutritive value is still uncertain for sows. There is a lack of standard ileal digestibility (SID) of amino acids (AA) for plant proteins because fitting a T-cannula in the distal ileum is difficult. Therefore, this study evaluated the apparent ileal digestibility and SID of 18 AA of CSM in sows at two physiological stages (gestation and non-pregnancy). We found that CSM with different chemical compositions impacted the SID of AA when fed to pregnant and non-pregnant sows. Additionally, the physiological stage of the sow has a substantial impact on the SID of some AA. The current findings of this study provided a basis for the precise formulation of sow diets with CSM.

This study was conducted to evaluate the effect of polyacrylamide (PAM) supplementation on the intake, digestion, weight gain, metabolism and growth of lambs. A total of ten 30 days old male small-tailed Han lambs with a body weight of 7.7±0.5 kg were divided into two equal groups (n = 5 each) and fed a basal diet or diet supplemented with 2.0 g of PAM per kg diet. The duration of the experiment was 210 days and experimental diets were fed ad libitum throughout the experimental period. Voluntary feed intake (VFI) was measured on daily basis, while body weight was measured on every ten days of the experiment.Two digestive and metabolic trials were conducted at the lamb's age of 95 to 103 days (Trial 1) and at the age of 210 to 218 days (Trial 2). At the end of experiment, all lambs were slaughtered to determine carcass characteristics. Results of the current study showed that supplementation of PAM in the diet of lambs increased the VFI and daily body gain by 14.4% (P < 0.05) and 15.2% (P < 0.01), respectively. In Trial 1, PAM supplementation in the diet increased the digestibility of dry matter (DM), organic matter (OM), crude protein (CP), cellulose, energy, and nitrogen retention by 7.9%, 5.4%, 6.4%, 9.6%, 4.3% and 30.3% (P < 0.01), respectively, and in Trial 2, PAM supplementation in the diet increased the digestibility of DM, OM, CP, cellulose, energy, and nitrogen retention by 9.3%, 7.9%, 7.7%, 11.6%, 6.9% and 38.5% (P < 0.01), respectively. Results of carcass parameter explored that supplementation of PAM in the diet increased the carcass, net meat and lean meat weights by 24.5%, 25.5%, and 30.6% (P < 0.01), respectively, however, PAM supplementation in the diet did not influence the contents of DM, OM, or CP in fresh liver, leg muscle, and rumen tissue; in addition, the CP contents in the Longissimus dorsi muscle was decreased by the supplementation of PAM in the diet. In summary, supplementation of 2.0 g of PAM per kg diet increased the VFI, nutrient digestibility, nitrogen retention, and carcass yield of lambs.

Energy and P utilization in faba beans and peas were evaluated in 3 broiler chicken experiments. In Exp. 1, 240 birds were allotted to 5 diets in a randomized complete block design with BW as a blocking factor on d 18 post hatching to determine the regression-derived energy utilization of faba beans (FB) and field peas (FP). In each of the respective Exp. 2 and 3, regression-derived P utilization in FB and FP were determined with 162 birds assigned to each of 3 diets in a randomized complete block design with BW as a blocking factor on d 19 post hatching. There were 8 replicate cages with 6 birds per cage in Exp. 1, and 6 replicate cages with 9 birds per cage in Exp. 2 and 3. The test ingredients were added to a corn-soybean meal-based diet at 15% or 30% in Exp. 1, whereas FB was included at 21%, 42%, or 63% and FP at 16%, 32%, or 48% in Exp. 2 and 3, respectively. In Exp. 1, the apparent ileal digestibility (AID) of gross energy (GE) and the ileal digestible energy (IDE) in the diets decreased linearly (P < 0.01). There was a quadratic response or a linear decrease (P < 0.05) with increasing concentrations of FB or FP, respectively, on the apparent total tract utilization (ATTU) of GE, metabolizable energy (ME), and nitrogen-corrected ME (MEn). The respective IDE, ME, and MEn determined were 2,541, 2,628, and 2,394 kcal/kg DM in FB and 2,254, 2,540, and 2,331 kcal/kg DM in FP. In Exp. 2 and 3, the ileal digestible and retainable P intake were linearly increased (P < 0.01). The estimated true ileal digestibility and true total tract utilization of P in FB were 66.5% and 66.7%, respectively. The respective corresponding values for FP were 73.4% and 73.8%. In conclusion, the information on utilization of energy and P in FB and FP provided could enhance proper diet formulation when using these ingredients.

The objective of this study was to compare the effects of post-ruminal provision of Ca-butyrate (CaB) when delivered via abomasal dosing, and Ca-gluconate (CaG) when provided ruminally using a rumen protected form or using an unprotected form via abomasal dosing on short-chain fatty acid (SCFA) concentration throughout the GIT, nutrient digestibility, GIT barrier function, ruminal SCFA absorption, ruminal morphometrics, intestinal brush border enzyme activity, and blood parameters for beef heifers. Thirty-two beef heifers fitted with ruminal cannulas were used in a randomized complete block design and assigned to one of four treatments: 1) negative control (ruminal infusion of double-distilled water; CON); 2) abomasal infusion of CaB (AB; 0.0029% of BW); 3) abomasal infusion of CaG (AG; 0.0077% of BW); and 4) ruminal infusion of a hydrogenated fat-embedded CaG (RG; 0.0192% of BW) to provide ruminal protection. Excluding CON, treatments were designed to deliver the same amount of butyrate in the small intestine. Heifers were housed in individual pens and DMI was limited to 95% of voluntary intake to minimize a potential confounding effect of DMI on treatment responses. Total GIT barrier function was assessed on day 17 and SCFA disappearance was evaluated on day 21 using the temporarily isolated and washed reticulo-rumen technique. On day 28, heifers were slaughtered, and ruminal and colonic digesta were collected to assess SCFA concentration. Additionally, ruminal, jejunal, and colonic tissues were collected to assess SCFA fluxes and regional barrier function ex vivo using the Ussing chamber technique. For colonic digesta, both AB and CaG treatments reduced the proportion of acetate (P < 0.05) and increased the proportion on propionate (P < 0.05) compared to CON. Relative to CON, AB but not CaG treatments increased in vivo ruminal disappearance of total SCFA (P = 0.01), acetate (P = 0.03), propionate (P = 0.01), and butyrate (P > 0.01). Treatments did not affect (P ≥ 0.10) acetate and butyrate fluxes in the ruminal and colonic tissues when measured ex vivo; however, when compared with CON, AB tended to decrease (P = 0.09) mannitol flux across ruminal tissue. In addition, mannitol flux was affected (P < 0.01) by region, with greater mannitol flux across the jejunum than rumen and colon. We conclude that while both abomasal infusion of CaB and CaG affect the molar proportion of acetate and propionate in the colon, only abomasal CaB stimulated ruminal SCFA absorption for growing beef heifers.


Butyrate, a short-chain fatty acid (SCFA), has received attention due to its ability to promote gastrointestinal (GIT) health and development. However, butyrate in its free form presents a strong odor, limiting its use in diet formulation. Supplementation of butyrate precursors, such as gluconate, have been studied to enhance butyrate production in the GIT. This study evaluated the effects of post-ruminal infusion of Ca-butyrate (AB; 0.0029% of BW) and Ca-gluconate (AG; 0.0077% of BW) and ruminal infusion of a hydrogenated fat-embedded Ca-gluconate (RG; 0.0192% of BW) relative to control (CON; ruminal infusion of double-distilled water). Thirty-two beef heifers fitted with ruminal cannulas were fed for 28 d and GIT barrier function and ruminal SCFA absorption were assessed. At slaughter, the rumen, jejunum, and colon tissues were collected and barrier function and SCFA fluxes were assessed ex vivo. Relative to CON, AB but not AG and RG increased in vivo ruminal SCFA absorption and tended to increase ex vivo barrier function. Thus, the data presented in this study shows that butyrate and gluconate do not function through the same mode of action in the GIT of beef heifers.

We aimed to evaluate the interaction between dietary starch concentration, varied by replacing wheat bran with dry ground corn, and replacement of corn silage (CS) with shredded beet pulp (BP) on production, milk fat output, milk fatty acid profile, and body reserves in dairy cows. Sixty-four Holstein dairy cows (140 ± 26 d in milk) were randomly assigned to 8 pens (8 animals per pen). Treatments were arranged in a 2 × 2 factorial arrangement with 2 concentrations of starch and 2 sources of fiber and were allotted to 8 pens (2 pens per treatment). Treatments were (1) 15% dry ground corn and 24% CS, (2) 15% dry ground corn and 24% BP replacing CS, (3) 30% dry ground corn and 24% CS, and (4) 30% dry ground corn and 24% BP replacing CS. The trial lasted for 47 d and final 7 d of experimental period was used for data and sample collection. Cows fed the BP-based diets had greater dry matter intake than those offered the CS-based diets, whereas no effects were observed with starch concentration. Milk yield increased by 1.8 kg/d with BP-based diets compared with CS-based diets and by 2.5 kg/d when cows received the high-starch compared with low-starch diets. Interactions between dietary starch concentration and forage substitution were detected for milk fat concentration and yield as BP inclusion lowered milk fat output with high-starch diet. Milk trans-18:1 concentration was lower with 15% dry ground corn and 24% CS compared with other diets. In conclusion, the effects of dietary starch concentration (22 and 32% dry matter) and forage substitution on production responses were independent except for milk fat output and milk trans 18:1 isomers. Substituting CS with BP is effective at increasing milk yield regardless of starch concentration; however, milk fat yield is lower when BP is used with high-starch concentration.

This study was conducted to evaluate the effects of supplementation of exogenous enzymes in broiler diets that includes fermented canola meal on performance, nutrient digestibility, biochemical indication, antioxidative capacity, digestive enzyme activity, immune responses, and gut health. Five hundred 1-day-old Ross 308 broiler chicks were randomly allocated into five experimental groups (5 replicate/group), the first group: a control (CON) contained a basal diet, and the second to the fifth groups were fed diets as follows: containing 20% canola meal (CM), contains 20% fermented canola meal (FCM), contains 20% canola meal and exogenous enzymes at 0.02%/kg feed (ECM), and contains 20% fermented canola meal and exogenous enzymes at 0.02%/kg feed (EFC), respectively. At the finisher phase, the best body weight gain, feed conversion ratio, and nutrient utilization were associated with chickens fed EFC compared to other groups (P < 0.05). Total protein, albumin, alanine aminotransferase, and superoxide dismutase levels increased (P < 0.05), while cholesterol and malondialdehyde levels decreased in chickens fed on EFC. Likewise, there was a significant increase in the relative weight of the bursa of Fabricius and antibody titer against Newcastle disease, whereas the weight of abdominal fat decreased in the EFC group compared to other groups. Furthermore, there was a significant improvement in the activity of lipase and amylase enzymes (P < 0.05) in the EFC group. Fermented canola meal addition improved gut health (decreased Escherichia coli, increased Lactobacillus, and the highest values of villus height). Overall, these results confirmed that supplementing a fermented canola meal diet with exogenous enzymes improved growth performance through enhancing nutrient digestibility, immunity, antioxidant capacity, and gut health. Thus, adding enzymes to a diet containing fermented canola meal can be recommended as an alternative protein source that could be safely used to replace up to 20% soybean meal in broiler diets.

Two in vitro and in vivo experiments were accomplished to investigate the impacts of integrated feed additives (IFA, combination of protected fat, lysine, and methionine plus yucca extract, eucalyptus essential oil, and direct-fed microbial) on gas production (GP), ruminal fermentation and degradability parameters and lamb performance, digestibility, and nitrogen balance. In the in vitro experiment, responses of graded doses (0, 25, 50, 75, 100, and 125 g IFA/kg DM) were evaluated. In the in vivo experiment, 15 Barki male lambs (30.43 ± 0.74 kg BW ± SE) were individually allotted in complete randomized design into three treatments (five animal/treatment) as control (basal diet without additive), low dose (50 g/kg DM) and high dose (100 g/kg DM) for 120 days. In vitro results showed that both doses (100 and 125 g/kg DM) quadratically decreased (p < 0.001) GP and ammonia nitrogen (NH-3N) concentration. In vivo experiment revealed that dry matter intake was increased (p = 0.016) with low dose compared to high dose. Inclusion of high dose decreased blood serum glucose concentrations (p > 0.05) and ruminal protozoal populations (p = 0.094) compared with low dose and control diet. Both doses of IFA increased (p < 0.05) serum urea, creatinine, and triglyceride concentrations, while decreased (p < 0.001) ruminal NH3-N concentrations. These results suggested that, although IFA was effective to modify in vitro ruminal fermentation process and feed degradability, such aspects did not improve in vivo performance of growing lambs.

Direct fed microbial may enhance the utilization of halophyte forages leading to improved animal growth and productivity. This study was conducted to evaluate Atriplex hay-based diet supplemented with yeast (Saccharomyces cerevisiae; SC) or bacteria (Bacillus subtilis and Lactobacillus casei; BAC) on lamb growth performance, digestibility, rumen fermentation, and carcass characteristics. Fifteen Barki lambs (90 ± 7 days of age and 18.6 ± 0.41 kg SE body weight; BW) were randomly assigned to three treatments for 120 days as follows: Control (basal diet without supplementation), SC and BAC diets, the basal diet supplemented with SC or BAC at 2 g/head/day, respectively. All lambs had similar dry matter (DM) intake, while lambs fed SC or BAC dies had higher (P < 0.05) total gain and average daily gain than those fed the control diet. Supplementation of SC or BC increased (P < 0.05) the digestibility of DM, organic matter, and acid detergent fiber, tended to decrease (P < 0.05) the urine N excretion and enhanced the N balance compared to the control. Ruminal pH, acetate, total volatile fatty acids concentrations, and bacterial protein were increased (P < 0.05), while creatinine and urea concentrations were decreased (P < 0.05) by both additives. Compared to other diets, the BAC diet reduced (P < 0.05) triglycerides, total lipids, kidney fat, and eye muscle fat. In conclusion, both additives resulted in similar positive growth performance and feed utilization, while only the BAC additive had a beneficial advantage in reducing the fat content of the carcass.

Despite being touted as a rich source of nutrients and functional bioactive compounds, the amount of brown seaweed (Ecklonia maxima) that can be included in diets of Boschveld indigenous chickens is unknown. This study, therefore, investigated the effect of feeding graded levels of brown seaweed meal (BSM) on apparent nutrient digestibility, growth performance, and physiological and meat quality parameters in Boschveld cockerels. A total of 225, five-wk-old Boschveld cockerels (316.4 ± 23.01 g live weight) were raised on 5 isoenergetic and isonitrogenous experimental diets formulated by incorporating BSM in a standard grower diet at a concentration of 0 (BSM0), 20 (BSM2), 40 (BSM4), 60 (BSM6), and 80 g/kg (BSM8). Feeding graded levels of dietary BSM induced neither quadratic nor linear effects (P > 0.05) on apparent nutrient digestibility, growth performance, hematological parameters, and meat quality characteristics in Boschveld cockerels. However, it resulted in linear increases for overall feed intake (R2 = 0.397; P = 0.021), ceca weight (R2 = 0.417; P = 0.013), duodenum length (R2 = 0.537; P = 0.04), and small intestine length (R2 = 0.305; P = 0.041). Negative quadratic responses were recorded for alanine aminotransferase (R2 = 0.530; P = 0.0009) and ileum length (R2 = 0.457; P = 0.045) as BSM levels increased. In conclusion, dietary inclusion of BSM improved feed intake and some internal organ sizes, altered alanine transaminase levels, but had no significant effect on apparent nutrient digestibility, growth performance, and carcass and meat quality attributes of Boschveld indigenous cockerels.

Wheat is the staple crop for 35% of the world's population, providing a major source of calories, mainly in the form of starch. The digestibility of wheat starch varies between different flours and products. Wheat products that are rapidly digested elicit large post-prandial glucose peaks associated with metabolic disorders. We investigated the impact of protein on starch digestion in three commercial flours with different grain hardness. A soluble extract of wheat proteins reduced starch digestion, even following gastric proteolysis. This extract was enriched in proteinaceous α-amylase inhibitors which were partially degraded during gastric proteolysis. Starch digestion kinetic analysis was carried out for flour samples pre-treated with different pepsin activities. The rate of starch digestion was altered following pepsin pre-digestion, and the extent of starch digestion increased in response to pepsin pre-digestion. We conclude that soluble proteinaceous alpha-amylase inhibitors present in wheat can escape gastric digestion and significantly contribute to reducing starch digestion in the small intestine.
