In this study, our objective was to conduct a comprehensive phytochemical analysis, determine toxicity levels, and assess the antioxidant and antimicrobial properties of extracts derived from the leaves of Dipteryx alata Vogel, a native species of the Brazilian cerrado flora. Three distinct extracts were prepared utilizing assisted ultrasound and the Soxhlet apparatus, namely, Ultrasound Crude Extract (UCE), Soxhlet Crude Extract (SCE), and the Soxhlet Ethanol Extract (SEE). The phytochemical analysis revealed the presence of flavonoids, tannins, phytosterols, and saponins in all extracts. Additionally, alkaloids were specifically identified in the SCE and SEE extracts. In the analysis using LC-DAD, the compounds gallic acid, rutin, quercetin, luteolin and kampefrol were determined in higher concentrations in the SCE, followed by the SEE and UCE, respectively. The GC-MS analysis revealed the presence of campesterol, stigmasterol and ß-sitosterol in all extracts, with UCE and SCE showing a higher concentration of ß-sitosterol. SCE showed the highest concentration of all identified compounds. In the analysis of antioxidant activity by DPPH• and ABTS•+, SEE showed greater efficiency (IC50 = 2.98 ± 2.92 and 6.57 ± 0.89 µg/mL, respectively). In the toxicity test with Allium cepa, all extracts stimulated root growth at 50 g/mL; UCE and SEE stimulated root growth at 250 g/mL; and SEE inhibited root growth at 750 g/mL. In the Artemia salina toxicity, all extracts were non-toxic. Antibacterial activity was identified in the microorganisms S. aureus and S. mutans; however, the extracts did not show antifungal action against the strain of C. albicans. The extracts of D. alata have therapeutic potential for applicability in dentistry.

Dipteryx odorata and Dipteryx punctata are species native to the Amazonian, traded by extractivists to obtain coumarin. We aimed to analyze the presence of coumarin in the ethanolic extracts of leaves, branches and fruits of D. odorata and D. punctata and to evaluate the antimicrobial activity of these extracts against phytopathogenic fungi and bacteria of clinical interest. Chemical analyses were performed by thin layer chromatography (TLC) and by gas chromatography coupled to mass spectrometry (GC-MS). For the antifungal assays, the fungi used were Cercospora longissima, Colletotrichum gloeosporioides, two isolates of Fusarium spp. and Sclerotium rolfsii, and the antibacterial assay was performed using the minimum inhibitory concentration (MIC) test with Burkholderia cepacia, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus bacteria. In D. odorata seed extracts and in D. punctata husks, endocarps, and seeds, we identified 1,2-benzopyrone. D. odorata endocarp extracts and D. punctata seeds provided the greatest decrease in mycelial growth of the evaluated phytopathogens, showing promise as an alternative control. The husk and endocarp extracts of both species had a weak effect on E. coli. This research is the first to compare the different parts of species of the genus Dipteryx and to evaluate the use of husks and endocarps of D. punctata fruits to obtain coumarin. Chemical analyses used to quantify the compounds existing in the extracts, and tests with phytopathogens in vitro and in vivo are currently being carried out.

Background: This study assessed the effects of Baru (Dipteryx alata Vog.) almond oil supplementation on vascular function, platelet aggregation, and thrombus formation in aorta arteries of Wistar rats. Methods: Male Wistar rats were allocated into three groups. The control group (n = 6), a Baru group receiving Baru almond oil at 7.2 mL/kg/day (BG 7.2 mL/kg, n = 6), and (iii) a Baru group receiving Baru almond oil at 14.4 mL/kg/day (BG 14.4 mL/kg, n = 6). Baru oil was administered for ten days. Platelet aggregation, thrombus formation, vascular function, and reactive oxygen species production were evaluated at the end of treatment. Results: Baru oil supplementation reduced platelet aggregation (p < 0.05) and the production of the superoxide anion radical in platelets (p < 0.05). Additionally, Baru oil supplementation exerted an antithrombotic effect (p < 0.05) and improved the vascular function of aorta arteries (p < 0.05). Conclusion: The findings showed that Baru oil reduced platelet aggregation, reactive oxygen species production, and improved vascular function, suggesting it to be a functional oil with great potential to act as a novel product for preventing and treating cardiovascular disease.

Baru (Dipteryx alata) almond is an emerging nut from the Brazilian savannah, that presents unique flavor and an interesting specialty oil. In this study, we aimed at investigating the effects of pressure, temperature, type (alcohol and/or water), and concentration of polar cosolvent on the extraction yield and tocopherol contents of baru oil obtained by supercritical-CO2 extraction (SC-CO2); and to investigate the effect of temperature and pressure on phytosterol, phenolic, and volatile compounds' profile in the oil when H2O was the cosolvent. Baru oil extracted with SC-CO2 using alcohol as a cosolvent showed a higher extraction yield (20.5-31.1%) than when using H2O (4.16-22.7%). However, when 0.3% H2O was used as cosolvent, baru oils presented the highest γ-tocopherol (107 and 43.7 mg/100 g) and total tocopherol (212 and 48.7 mg/100 g) contents, depending on the temperature and pressure used (50°C and 10 MPa or 70°C and 30 MPa, respectively). Consequently, the lowest pressure (10 MPa) and temperature (50°C) values resulted in baru oils with better γ/α-ratio, and the highest contents of ß-sitosterol (107 mg/100 g) and phenolic compounds (166 mg/100 g). However, the highest pressure (30 MPa) and temperature (70°C) values improved the volatile profile of oils. Therefore, although alcohol as a cosolvent improved oil yield, small amounts of H2O provided a value-added baru oil with either high content of bioactive compounds or with a distinctive volatile profile by tuning temperature and pressure used during SC-CO2 extraction.

Baru nuts (Dipteryx alata Vog.) are a native species from Brazil, rich in phenols and other antioxidants, with high socioeconomic value and possible pharmaceutical applications. Here we investigated baru nut ethanolic extract (BNEE) antioxidant and wound healing activities in human NCI-H441 and A549 lung epithelial cell lines for a possible use in conditions related to oxidative stress and wound healing impairments, such as chronic obstructive pulmonary disease (COPD). BNEE was characterised with high DPPH free radical scavenging activity and high total phenolics content, amongst them gallic acid, that was identified and quantified by HPLC. BNEE was not cytotoxic at concentrations studied, reduced the levels of reactive oxygen species before and during oxidative stress and increased wound healing in cell monolayers. These are the first steps to investigate the beneficial properties of baru in diseases related to oxidative stress and wound healing impairments such as COPD.

The aim of this study was to evaluate the topical application of alcoholic extracts of Dipteryx alata Vogel almonds and bark in skin wound healing in mice. Fifty-four C57BL/6 mice were equally distributed into three groups: Control, Almond, and Bark. A 9 mm skin fragment was resected from the dorsal region of the animals' thorax. The wounds were submitted to topical application of base cream (vehicle), 10% hydroalcoholic almond extract, or bark extract twice a day. Macroscopic, histological, and immunohistochemical evaluations were conducted on the 7th, 14th, and 21st postoperative days. No significant difference was observed regarding skin wound area among groups, with the parameter presenting only a temporal effect on healing (p>0.05). The almond and control groups exhibited more intense collagenization than the bark group (p<0.05). Dipteryx alata Vogel showed to be inert in the wound healing process in mice.(AU)


O objetivo deste estudo foi avaliar a aplicação tópica do extrato alcoólico da semente e da casca da Dipteryx alata Vogel na cicatrização de feridas cutâneas, em camundongos. Um total de 54 camundongos C57BL/6 foram utilizados neste estudo, distribuídos em três grupos de 18 animais (controle, semente e casca). Em todos os animais, um fragmento de pele foi ressecado da região dorsal do tórax utilizando-se instrumento de punção de 9mm de diâmetro, após o qual foi realizada aplicação tópica de creme base (veículo), extrato hidroalcoólico 10% de semente ou casca, duas vezes ao dia. As avaliações macroscópica, histológica e imuno-histoquímica foram realizadas no sétimo, 14º e 21º dias de pós-operatório. Não foi observada diferença significativa quanto à área da ferida cutânea entre os grupos, apenas um efeito temporal na cicatrização (P>0,05), indicando estágio possivelmente mais avançado desse processo. Porém, na avaliação histológica, os grupos semente e controle apresentaram colagenização mais intensa que o grupo casca (P<0,05). Dipteryx alata Vogel mostrou-se inerte no processo de cicatrização de feridas em camundongos.(AU)

Baru (Dipteryx alata Vogel) is a native tree plant, widely distributed in Brazil, and has a growth and development in acidic soils like Cerrado, indicating a probable tolerance to adverse soil conditions, such as the high concentration of metals and the acidic pH. Due to the lack of information about the tolerance of this species to metals and the possibility of being used in the recovery of degraded areas and/or in phytoremediation, this work was developed with the objective of evaluating the in vitro germination and growth capacity of baru in medium supplemented with different concentrations of aluminum, iron, and manganese, as well as through chemical analysis, to determine the concentration of metals accumulated in cultivated plants in these conditions. The treatments consisted in different concentrations of metals: aluminum, Al3+ (0, 3.5, 7.0, 10.5, 21.0, or 42.0 mg L-1); iron, Fe3+ (0, 2.5, 4.9, 7.4, 14.7, or 29.4 mg L-1); and manganese, Mn2+ (0, 0.4, 0.8, 1.2, 2.4, or 4.8 mg L-1) added to the medium WPM. The tested values were based on using the lower concentration as the limit value, calculated based on risk to human health in accordance with CONAMA resolution 420/2009 for groundwater. At 60 days of cultivation, the percentage of germination, the average number of leaves, the length of the main root and the aerial part, the fresh and dry mass of the aerial part and the root system and the cations concentration Al3+, Fe3+ and Mn2+ in the plant biomass, were evaluated. The results showed that under the conditions in which the experiment was conducted, germination and in vitro growth of baru were not affected by the presence in high concentrations of any of the evaluated metals, with no differences in the percentage of germination and plant growth, as well as typical toxicity characteristics were not observed, such as changes in root morphology, chlorosis, or tissue oxidation. The absence of toxicity symptoms in baru plants, in the presence of Al3+, Fe3+, and Mn2+, indicate that the species is tolerant to these metals. The accumulation of Al3+ and Fe3+ in the plant biomass at the beginning of growth, simultaneously with the increase in the concentrations of these elements in the culture medium, indicates that this species can be used for phytoremediation, because it is a probable accumulator of these elements throughout its development, given the presence in significant concentrations of these elements also in the seeds.

Fruits are sources of bioactive compounds that are responsible for several biological activities. Therefore, this study aimed to identify the chemical composition of the pulp of the Brazilian Savanna fruit Dipteryx alata; evaluate its toxic effects, influence on the life expectancy of the nematode Caenorhabditis elegans, and its antioxidant activities in vitro and in vivo; and describe the mechanisms involved. The chemical compounds identified include phenols, terpenes, fatty acid derivatives, vitamins, and a carboxylic acid. The in vitro antioxidant activity was demonstrated by radical scavenging methods. In vivo, the D. alata fruit pulp was not toxic and promoted resistance to oxidative stress in nematodes exposed to a chemical oxidizing agent. Furthermore, it promoted an increased life expectancy in wild-type nematodes and increased the expression of superoxide dismutase and the nuclear translocation of DAF-16. These results suggest that the beneficial effects identified are related to these two genes, which are involved in the regulation of metabolic activities, the control of oxidative stress, and the lifespan of C. elegans. These beneficial effects, which may be related to its chemical constituents, demonstrate its potential use as a functional and/or nutraceutical food.

This work aimed to contribute to the nutritional and functional characterization of roasted baru nuts, a seed widely consumed and produced in Brazil. Baru nut was characterized in terms of its nutritional value and volatile composition (SPME-GC-MS analysis). The ultrasound assisted extraction was used to extract free and bound phenolic compounds that were identified by LC-DAD-ESI-MS/MS method. Bioactivity assays were carried out to evaluate the antioxidant activity (ORAC and HOSC assay) and anticancer effect (inhibition of HT29 cell growth and targeting of cancer stemness) of baru nut extracts and phenolic compounds. Results showed that baru is a good source of protein and monounsaturated fatty acids, specifically oleic acid (47.20 g/100 g). The predominant volatile compounds are hexanal (71.18%) and 2,5-dimethyl-pyrazine (9.43%). The main phenolic compounds identified were gallic acid and its derivatives, such as gallic acid esters and gallotannins. Among all, gallic acid and methyl gallate seemed to be the main compounds responsible for the high antioxidant activity. The antiproliferative effect evaluated of baru extracts in HT29 cell line showed ability to impair cell growth in both monolayer and spheroid cultures and to reduce ALDH+ population. These results supply new information about the functional compounds presents in baru nut, which are important sources of natural antioxidants and antiproliferative compounds.

The presence of glandular appendages in the anthers is a rare condition in angiosperms. In Leguminosae it occurs in species of the Mimosoid clade and in early-branching clades of papilionoids such as Dipterygeae. In Dipterygeae such appendages surprisingly exhibit a secretory cavity instead of secretory emergences as is the case for the Mimosoid clade. Thus, the objective of this study was to elucidate the function of anther glands in Dipteryx alata and Pterodon pubescens, species in the Dipterygeae clade that exhibit a pollen release mechanism that is intermediate between the explosive and valvular types. Flower buds and flowers were processed for surface, anatomical, histochemical and ultrastructural analyses. Anther glands consist of a cavity secreting sticky substances (oleoresins and polysaccharides) that play a key role during the flower's lifespan by aggregating pollen grains and attaching them to the floral visitor's body. Other floral features that are important for understanding the pollen release mechanism that is intermediate between the valvular and the explosive types are: (i) keel petals intertwined with tector trichomes; (ii) glandular appendages in the abaxial and lateral sepals and in petals composed of secretory ducts; and (iii) a continuous secretion process of the anther glands followed by an asynchronous dehiscence of anthers. The well-adapted papilionoid flag blossom with anther glands and keel petals intertwined with trichomes provided the foundation for a successful canalisation toward a pollen release mechanism intermediate between the explosive and valvular types inside early-branching papilionoids.

Worldwide prevalence of noncommunicable chronic degenerative diseases is among the main causes of death worldwide. The consumption of some foods such as nuts and seeds may be beneficial in preventing these diseases. Dipteryx alata Vogel (DA), known popularly as Baru, belongs to the family Fabaceae and is a native fruit tree from the Brazilian savanna. The purpose of this study was to evaluate the use of seeds of DA on the metabolic and oxidative profile of Wistar rats. Animals were divided randomly into four groups (n = 10): G1 (control group), and G2 (treated with DA 20%), G3 (treated with DA 30%), and G4 (treated with DA 40%). After 40 days, animals were euthanized and metabolic and oxidative profiles were analyzed (glycemia, cholesterol, triglycerides [TGs], high-density lipoprotein-cholesterol [HDL-c], very low-density lipoprotein-cholesterol [VLDL-c], low-density lipoprotein-cholesterol [LDL-c], C reactive protein, aspartate aminotransferase, alanine aminotransferase, Lee index, weight, visceral fat, ferric reducing ability of plasma, and ferric-xylenol orange method. The use of the seeds was effective in reducing TGs, VLDL-c, LDL-c, and increasing HDL-c but did not interfere in the percentage of weight gain, visceral fat, levels of total cholesterol, and oxidative stress. Based on our results, it is possible to say that the use of DA may improve the lipid profile of Wistar rats and we may suggest that the consumption of DA almonds or products prepared with them may be an effective option for the intake of healthy products.

Dipteryx alata Vog. (barueiro) is a native fruit of the Cerrado used in food and medicine. The species has never been domesticated and is currently at risk of extinction. Plant tissue culture is an important tool for the conservation of germplasm and for quick, large-scale propagation of quality seedlings. While, this technique has never been used with barueiro, it could be a valuable tool in the domestication of this species. The objective of this work is to evaluate concentrations of MS medium salts (25, 50, 75 and 100%) supplemented with coconut water (100 mL L-1), dwarf banana pulp (60 g L-1) or the absence of any supplement in the in vitro establishment of barueiro seeds (Dipteryx alata Vog). The experiment used a completely randomized design (CRD) with a 3x4 factorial scheme (three cultivation media types and four concentrations of MS salts) and three repetitions. The following characteristics were evaluated: germination rate, shoot length , largest root length , stem diameter , number of leaves , chlorophyll A , chlorophyll B , total chlorophyll, and the fresh and dry mass of the seedlings. The coconut water and banana pulp supplements did not improve the in vitro development of the barueiro seedlings and the full concentration of MS salts did not improve the initial development of the seedlings. However, the pure MS medium at a concentration of 25% MS salts was the best option for the in vitro establishment of Dipteyx alata.


O barueiro (Dipteryx alata Vog.) é uma fruteira nativa do bioma Cerrado e possui várias aplicabilidades, desde alimentares até medicinais. No entanto ainda não é uma espécie domesticada e encontra-se sob risco projetado de extinção. A cultura de tecidos vegetais constitui uma importante ferramenta para a conservação de germoplasma, além de proporcionar a rápida propagação de mudas de qualidade em larga escala. Porém, esta técnica não vem sendo utilizada em barueiro, podendo ser aplicada de forma valiosa contribuindo para seu processo de domesticação. Objetivou-se neste trabalho avaliar concentrações de sais do meio MS (25, 50, 75 e 100%) suplementados com água de coco (100 mL L-1), polpa de banana nanica (60 g L-1) ou ausência de suplemento no estabelecimento in vitro de sementes de barueiro (Dipteryx alata Vog). O delineamento experimental utilizado foi o inteiramente casualizado (DIC) em esquema fatorial 3x4 (três meios de cultivo e quatro doses de sais MS) com três repetições. Foram avaliadas as características: taxa de germinação, comprimento da parte aérea e da maior raiz, diâmetro do caule, número de folhas, teores de clorofila A, B e total , massa fresca e massa seca das plântulas. A água de coco e a polpa de banana como suplementos não forneceram vantagens no desenvolvimento in vitro de plântulas de barueiro e a concentração plena dos sais MS não forneceram vantagens ao desenvolvimento inicial das plântulas. Portanto, o meio MS puro a uma concentração de 25% dos sais MS é a formulação que melhor se aplica para o estabelecimento in vitro de Dipteyx alata.

Selective logging in Brazil allows for the removal of up to 90% of trees above 50 cm diameter of a given timber species, independent of a species' life history characteristics or how quickly it will recover. The genetic and demographic effects of selective logging on two Amazonian timber species (Dipteryx odorata Leguminosae, Jacaranda copaia Bignoniaceae) with contrasting ecological and reproductive characteristics were assessed in the same forest. Genetic diversity and gene flow were characterized by genotyping adults and seed sampled before and after logging, using hypervariable microsatellite markers. Overall, there were no short-term genetic impacts on the J. copaia population, with commercial application of current Brazilian forest management regulations. In contrast, for D. Odorata, selective logging showed a range of genetic impacts, with a 10% loss of alleles, and reductions in siring by pollen from trees within the 546-ha study area (23-11%) and in the number of pollen donors per progeny array (2.8-1.6), illustrating the importance of the surrounding landscape. Asynchrony in flowering between D. odorata trees led to trees with no breeding partners, which could limit the species reproduction and regeneration under current regulations. The results are summarized with other published studies from the same site and the implications for forest management discussed. The different types and levels of impacts associated with each species support the idea that ecological and genetic information by species, ecological guild or reproductive group is essential in helping to derive sustainable logging guidelines for tropical forests.

Snakebite is a neglected disease and serious health problem in Brazil, with most bites being caused by snakes of the genus Bothrops. Although serum therapy is the primary treatment for systemic envenomation, it is generally ineffective in neutralizing the local effects of these venoms. In this work, we examined the ability of 7,8,3'-trihydroxy-4'-methoxyisoflavone (TM), an isoflavone from Dipteryx alata, to neutralize the neurotoxicity (in mouse phrenic nerve-diaphragm preparations) and myotoxicity (assessed by light microscopy) of Bothrops jararacussu snake venom in vitro. The toxicity of TM was assessed using the Salmonella microsome assay (Ames test). Incubation with TM alone (200 µg/mL) did not alter the muscle twitch tension whereas incubation with venom (40 µg/mL) caused irreversible paralysis. Preincubation of TM (200 µg/mL) with venom attenuated the venom-induced neuromuscular blockade by 84% ± 5% (mean ± SEM; n = 4). The neuromuscular blockade caused by bothropstoxin-I (BthTX-I), the major myotoxic PLA2 of this venom, was also attenuated by TM. Histological analysis of diaphragm muscle incubated with TM showed that most fibers were preserved (only 9.2% ± 1.7% were damaged; n = 4) compared to venom alone (50.3% ± 5.4% of fibers damaged; n = 3), and preincubation of TM with venom significantly attenuated the venom-induced damage (only 17% ± 3.4% of fibers damaged; n = 3; p < 0.05 compared to venom alone). TM showed no mutagenicity in the Ames test using Salmonella strains TA98 and TA97a with (+S9) and without (-S9) metabolic activation. These findings indicate that TM is a potentially useful compound for antagonizing the neuromuscular effects (neurotoxicity and myotoxicity) of B. jararacussu venom.

Choibá (Dipteryx oleifera Benth) es un árbol de la familia Fabaceae (Papilionoideae), con una distribución geográfica reportada desde Nicaragua hasta Colombia a una altura de hasta 1000 msnm. Crece en bosque húmedo, muy húmedo o premontano húmedo. Esta especie es considerada vulnerable debido a la sobreexplotación de su madera, ya que es un árbol altamente apetecido por esta y por sus frutos. Su almendra almacena una buena cantidad de aceites con potencial para la industria alimentaria, lo que podría resultar en una nueva fuente alimenticia, por lo cual el cultivo in vitro de vegetales con el propósito de producir compuestos de interés, marca un punto de partida para reducir el uso del suelo y lograr componentes bioactivos bajo condiciones controladas. En este trabajo, como una primera etapa experimental, se evaluó el crecimiento celular en suspensiones, a partir de callo inducido en explantes de cotiledón; se ensayaron 6 tratamientos diferentes, la mitad de estos con MS como medio basal y la otra mitad con B5, cada uno de los dos grupos con un control y la combinación hormonal de 2.5 mg/L de 2,4-D y 1 mg/L de BAP o kinetina, suplementado con adenina, biotina, glutamina y ácido pantoténico y 30 g/L de sacarosa, bajo completa oscuridad. Se encontró que dos tratamientos con MS en combinación con 2.5 mg/L de 2,4-D y 1 mg/L de kinetina o BAP fueron los mejores.


Choibá (Dipteryx olifera) is a tree of the Fabaceae family, with a geographical distribution reported from Nicaragua to Colombia, nearly 1.000 msnm in a tropical rain forest. This species is a highly desired tree for its timber and fruits, the kernel store a lot of important oils for the food industry, resulting in a new possible food source, so we are making in vitro cultivation of vegetables with the purpose of producing compounds of interest and that mark a starting point on the reduction of land use and it achieves bioactive components under controlled conditions. In this work, as a first experimental step was evaluated the tissue cell growth in suspension, using fragments of cotyledon and testing 6 different treatments, the half of those with MS as basal medium and the other half with B5, each of the two groups with one control and hormone combination of 2.5 mg/L 2,4-D and 1 mg/L BAP or kinetin, supplemented with adenine, biotin, pantothenic acid and glutamine and 30 g l sucrose under complete darkness. It was found that two treatments with MS in combination with 2.5 mg/L 2,4-D and 1 mg/L kinetin or BAP were the best.

Nos últimos anos, uma quantidade substancial de evidências tem indicado o papel chave dos radicais livres e outros oxidantes como grandes responsáveis pelo envelhecimento e pelas doenças degenerativas associadas ao mesmo. Por outro lado, substâncias fenólicas são reconhecidamente detentoras de pronunciada atividade antioxidante, muitas vezes envolvidas em tratamentos de pigmentação que resultam em hiperpigmentação ou hipopigmentação cutânea. Para o tratamento desses problemas de pigmentação vários produtos cosméticos e farmacêuticos são utilizados, porém, não são totalmente eficazes ou seguros, o que justifica a intensa pesquisa na busca de novos agentes ativos, principalmente àqueles envolvidos na melanogênese, como a tirosinase. Considerando que algumas substâncias obtidas de plantas apresentam essa atividade, a flora brasileira constitui-se uma importante fonte de pesquisa de novas substâncias. Assim, este trabalho foi realizado para avaliar os fenóis (método de Folin-Ciocalteau), a atividade antioxidante (CE50) (método de seqüestro do radical livre DPPH), a capacidade de quelação dos íons cobre, e a capacidade de inibição da tirosinase do extrato das folhas da espécie Dipteryx alata Vogel. Os resultados de fenóis totais mostraram uma concentração de 112,3 mg EAG.g-1 no extrato etanólico e 45 mg EAG.g-1 no extrato hexânico. A capacidade antioxidante dos extratos indica que o extrato etanólico, em comparação ao hexânico e ao BHT, possui maior teor de compostos antioxidantes, apresentando os respectivos valores sobre a quantidade de extrato necessária para decrescer a concentração inicial de DPPH em 50%: 52,9 ± 1,3 ppm, 169,1 ± 2,3 ppm, e 181± 6 ppm. Já a capacidade de quelação dos íons cobre mostrou que o extrato etanólico possui capacidade de quelação insignificante. No ensaio de inibição da tirosinase o extrato etanólico demonstrou um percentual de inibição da enzima de 42% após uma hora.


In recent years, a substantial amount of evidence has shown the key role of free radicals and other oxidants as largely responsible for aging and associated degenerative diseases. On the other hand, phenolic substances are known to hold pronounced antioxidant activity, often involved in pigmentation treatments, which result in skin hyperpigmentation or hypopigmentation. For the treatment of these pigmentation problems several cosmetic and pharmaceutical products have been used; however, they are not fully effective or safe, which justifies intense research to find new active agents, especially those involved in melanogenesis such as tyrosinase. Considering that some substances obtained from plants have this activity, the Brazilian flora constitutes an important source of research for new substances. Thus, this study was conducted to evaluate the phenols (Folin-Ciocalteau assay), the antioxidant activity (EC50) (DPPH free radical scavenging assay), the chelation capability of copper ions, and the inhibition capability of tyrosinase from leaf extract of the species Dipteryx alata Vogel. Results for total phenols showed concentration of 112.3 mg GAE.g-1 in ethanol extract and 45 mg GAE.g-1 in hexane extract. The antioxidant capacity of extracts indicates that ethanol extract, compared to hexane extract and BHT, has higher content of antioxidant compounds, showing the respective values of the necessary amount of extract to decrease the initial DPPH concentration by 50%: 52.9 ± 1.3 ppm, 169.1 ± 2.3 ppm and 181 ± 6 ppm. On the other hand, the chelation capacity of copper ions showed that the ethanol extract has insignificant chelation capacity. In the tyrosinase inhibition test, the ethanol extract had 42% enzyme inhibition after one hour.

BACKGROUND: Dipteryx alata Vogel popularly known as "baru" is an important commercial leguminous tree species from the Brazilian Cerrado, which possess medicinal properties, besides its fruits consumption by animals and humans. The use of the "naturally occurring plants" as herbal remedies and foods mainly from leaves, seeds, flowers and roots of plants or extracts require precautions before ensuring these are safe and efficacious. The objective of this study was to evaluate the safety of D. alata barks extract. METHODS: Vegetal drugs of D. alata barks were submitted to quality control assays and further to the safety assays under 1) in vitro parameter by Salmonella (Ames) mutagenicity, and 2) in vivo parameter on the pregnancy of rats. RESULTS: The extract was non-mutagenic to any of the assessed strains TA97a, TA98, TA100 and TA102 even after metabolic activation (+S9). All in vivo parameters (reproductive ability evaluation, physical development of rat offsprings, and neurobehavioral development assays) showed no changes related to control group. CONCLUSION: D. alata barks extract is neither mutagenic by the Ames test nor toxic in the pregnancy of rats, with no physical-neurobehavioral consequences on the rat offsprings development.

Amburana cearensis is a medicinal plant known as "cumaru". It is used in Northeast Brazil in the treatment of respiratory diseases. This was a randomized, double-blind, placebo-controlled study, with the aim of evaluating the efficacy and safety of cumaru syrup as complementary therapy in mild persistent asthma. The study consisted of 3 phases, pre-treatment, treatment and post-treatment. The primary efficacy outcome was comparison of the changes reported by patients of the cumaru and placebo groups after treatment, using the "Asthma Quality of Life Questionnaire" (AQLQ). The secondary outcome was the effect of cumaru syrup on lung function based on spirometry. The results showed that in the cumaru group, the proportion of patients who had global improvement in asthma symptoms was significantly greater (61.90%, P=0.0009) than in the placebo group (9.52%). Only the spirometric parameters Forced Vital Capacity (FVC) and Forced Expiratory Volume in 1 second (FEV1) showed significant intergroup differences in post-treatment (P<0.05). The hematological and serum chemistry tests performed in the pre-treatment and post-treatment showed no statistically significant differences (P>0.05). Adverse events were reported by 3 patients (14.29%) in the cumaru group and 3 patients (14.29%) in the placebo group. All adverse events were considered non-serious and mild.


Amburana cearensis é uma planta medicinal conhecida como "cumaru". No Nordeste do Brasil é usada no tratamento de doenças respiratórias. Este é um estudo randomizado, duplo-cego e controlado por placebo, com o objetivo de avaliar a eficácia e segurança do xarope de cumaru como terapia complementar da asma persistente leve. O estudo consistiu de três fases, pré-tratamento, tratamento e pós-tratamento. A variável primária para determinação da eficácia foi a comparação das mudanças referidas pelos pacientes dos grupos cumaru e placebo após o tratamento, usando o "Questionário sobre Qualidade de Vida na Asma" (QQVA). A variável secundária foi o efeito do xarope de cumaru na função pulmonar baseado na espirometria. Os resultados mostraram que no grupo cumaru, a proporção de pacientes com melhora global dos sintomas da asma foi significativamente maior (61,90%, P=0.0009) que no grupo placebo (9,52%). Somente os parâmetros espirométricos, capacidade vital forçada (CVF) e volume expiratório forçado no primeiro segundo (VEF1), mostraram diferença intergrupo significtivas no pós-tratamento (P<0.05). Os testes hematológicos e do soro realizados no pré-tratamento e pós-tratamento não mostraram diferenças estatisticamente significativas (P>0.05). Eventos adversos foram reportados por 3 pacientes (14,29%) no grupo cumaru e 3 (14,29%) no grupo placebo. Todos os eventos adversos foram não sérios e leves.

In Brazil, medicinal plants are widely used by the indigenous people, which leads to a constant requirement for toxicity tests to be performed on the plant extracts. Although the current Brazilian Directive 90/2004 on the preclinical toxicity testing of phytotherapeutics recommends only in vivo tests, some Brazilian researchers would like to change this situation by implementing the Three Rs in the toxicological testing of medicinal plants. The present study evaluated the cytotoxic and genotoxic potentials of bark extracts from Dipteryx alata Vogel, a medicinal plant of the Brazilian cerrado, by using CHO-K1 (Chinese hamster ovary) cells. An IC50 value was obtained, which corresponded to 0.16mg/ml of plant extract, and from this the equivalent LD50 was determined as 705mg/kg. In order to determine the genotoxic potential of the sample, the frequency of micronucleus formation was assessed. CHO-K1 cells were exposed, during targeted mitosis, to different concentrations of plant extract and cytochalasin B, in the presence and absence of an appropriate metabolic activation system (an S9 mix). The results obtained indicated that it might be possible to implement the Three Rs in assessing the potential human hazard of medicinal plants. The publication of such data can increase awareness of the Three Rs by showing how to optimise the management of animal use, if in vivo toxicological experiments are required.
