RESUMEN

Iron-salen, i.e., µ-oxo-N,N'-bis(salicylidene)ethylenediamine iron (Fe(Salen)) was a recently identified as a new anti-cancer compound with intrinsic magnetic properties. Chelation therapy has been widely used in management of metallic poisoning, because an administration of agents that bind metals can prevent potential lethal effects of particular metal. In this study, we confirmed the therapeutic effect of deferoxamine mesylate (DFO) chelation against Fe(Salen) as part of the chelator antidote efficacy. DFO administration resulted in reduced cytotoxicity and ROS generation by Fe(Salen) in cancer cells. DFO (25 mg/kg) reduced the onset of Fe(Salen) (25 mg/kg)-induced acute liver and renal dysfunction. DFO (300 mg/kg) improves survival rate after systematic injection of a fatal dose of Fe(Salen) (200 mg/kg) in mice. DFO enables the use of higher Fe(Salen) doses to treat progressive states of cancer, and it also appears to decrease the acute side effects of Fe(Salen). This makes DFO a potential antidote candidate for Fe(Salen)-based cancer treatments, and this novel strategy could be widely used in minimally-invasive clinical settings.

Oil-based mud, a complex mixture containing amines in emulsifiers, is used in offshore drilling operations. It is a skin irritant that occasionally gives rise to allergic contact sensitivity. In patch testing patients with allergy to drilling mud, we have identified polyamine (diethylenetriamine and triethylenetetramine) sensitivity in 5 patients. All 5 patients were also allergic to emulsifiers. These emulsifiers are cross-linked fatty acid amido-amines, in which unreacted amine groups are thought to cross-sensitize with these constituent polyamines. Cross-reactivity between ethylenediamine, diethylenetetramine and triethylenetetramine was found in 9 subjects.

During an examination of 47 workers engaged in ethylenediamine production subjective complaints of the skin were registered in 39% of them. Professional contact allergic dermatitis (26%) prevail over the contact irritative (6.5%) and lead very often to a change of employment for the operators. The epicutanic test with ethylenediamine and diethylene-triamine gives respectively 32 and 20% positiveness with high relative part of latent allergy. At compensated pH of the skin the sample for alkalic neutralization is prolonged in 42.9% of the workers. The ethylenediamine dermal absorption at concentrations in the air of the working area about MAC, is shown with methemoglobinemia registered in 15 operators. The raised methemoglobin in blood could be used with success as indirect test for exposure in hygiene-toxicological studies of workers exposed to ethylenediamine and aliphatic amines.

To study the preventive effect of supplementary iodine on interdigital phlegmon, 50 Swedish red and white calves weighing approximately 100 kg each were given 1 mg ethylenediamine dihydroiodide (EDDI) per kg bodyweight daily. Fifty calves in the same herd were untreated controls. As a result of severe side effects the experiment was interrupted after five weeks. No case of interdigital phlegmon was observed, but 31 calves in the EDDI group and one in the control group were treated for respiratory disease. Serum protein bound iodine increased on average from 68.0 to 213.0 micrograms/litre in the EDDI group compared to an increase from 59.9 to 70.7 micrograms/litre in the control group.
