Spermatogenesis process is sensitive to heat stress because the testicular temperature is 2 to 4 °C lower than the core body temperature. The current study aimed to investigate the effects of iron oxide nanoparticles containing curcumin on spermatogenesis in mice induced by long-term scrotal hyperthermia. In this experimental study, 18 mice were equally divided into the following three groups: control, scrotal hyperthermia, and scrotal hyperthermia + curcumin-loaded iron particles (NPs) (240 µL) (mice were treated for 20 days). Hyperthermia was induced by exposure to the temperature of 43 °C for 20 min every other day for 5 weeks. Afterward, the animals were euthanized; sperm samples were collected for sperm parameters analysis, and testis samples were taken for histopathology experiments, evaluation of serum testosterone level, and RNA extraction in order to examine the expression of c-kit, STRA8 and PCNA genes. Our study showed that curcumin-loaded iron particles could notably increase the volume of testis, length of seminiferous tubules, sperm parameters, and stereological parameters (i.e., spermatogonia, primary spermatocyte, round spermatid, and Leydig cells) thereby increasing serum testosterone level; in addition, TUNEL-positive cells showed a significant decrease in curcumin-loaded iron particle group. Thus, based on the obtained results, the expression of c-kit, STRA8, and PCNA genes was significantly increased in treatment groups by curcumin-loaded iron particles compared with scrotal hyperthermia-induced mice. In conclusion, curcumin-loaded iron particles can be considered an alternative treatment for improving the spermatogenesis process in scrotal hyperthermia-induced mice.

For the purpose to develop of method for treatment of male hypofertility the efficiency of gadolinium orthovanadate nanoparticles (NP GdVO4) in rats with neonatal induced reproductive disease, has been investigated. The progeny of intact rats parents were exposed to emotional stress (Maternal separation stressn) and received an excessive amount of the blend of phytoestrogens (Ph) with mother's milk (1st group) from 3 to 22 day life. At the age of 10 months males received NP GdVO4 (0.33 mg/kg; 2nd group) or speman (158 mg/kg; 3rd group) with feed over the 70 days. Sexual behavior and fertility of males were investigated; the integrated indicator of reproductive potential (FI) was calculate and was estimated with the data of animals from group Control who were given vehicle (water). In the 1st group sexual behavior of males was characterized by accelerating sexual responses on female (double shortening of mounting and intromission latency) and a reduced ability to achieve ejaculation and to start the second round of copulations. The less females have been inseminated by these males (67% vs. 91% in Control group), but almost all became pregnant. As a result of the neonatal induction deterioration in the quality of male gametes intrauterine pregnancy loss increased more than twice, and the total number of foetus decreased (on 21%). These changes have led to a reduction of the male integrated indicator of reproductive potential (Fi) in 2.6 times compared with control animals. Using a NP GdVO4 rehabilitated male sexual behaviour indicators to Control group level, resulting in an increase in the number of fertilized females (up to 95%). In pregnant females decreased fetal losses (up to 15.2% vs. 18.1% in control, p<0,05) and increase in the number of fetuses (on 25%, p<0,05) that shows an improvement in the quality of sperm. The integrated indicator of reproductive potential (F1) increased to (8.3 ± 0.5) vs. (3.0 ± 0.3) u. in the 1st group (without treatment), that statistically higher than in groups Control and 3rd. Application of NP GdVO4 in rats with neonatal inductive pathology of the reproductive function and metabolic disorders normalizes sexual behaviour, the quality of sperm that restores male fertility and improves reproductive potential much more efficiently than the reference drug speman.

In this study, purification, preliminary characterization and biological activities of water-soluble polysaccharides from Ostrea rivularis Gould (ORP) were investigated. Firstly, crude ORP was extracted by enzyme-assisted extraction and then sequentially purified by chromatography of DEAE-52 and Sephadex G-100, producing one main purified fractions of ORPp. Furthermore, the preliminary characterization of ORPp was studied, and its antioxidant and spermatogenesis activities were evaluated. Experimental results showed that ORPp was mainly composed of glucose (76.3%) and galactose (23.7%). The average molecular weight of ORPp was 118 kDa. Besides, ORPp showed strong antioxidant activities in vitro. For the experiments of antioxidant activities in vivo, ORPp can significantly inhibited the formation of MDA in rats' serums, and raised the activities of antioxidant enzymes and the level of total antioxidant capacity (TAOC). Furthermore, ORPp could significantly increase the weights of male rats' sexual organs, promote sperm motility and raise epididymal sperm counts. These results suggest that ORPp could be a new source of natural antioxidants and spermatogenic agent with its potential usage in developing novel supplements and medicines.

Antitumour, antifertility and histopathological investigations were carried out on male rats by the use of organotin complexes. The organotin complexes were synthesized by the alkylation of [Sn(TAML(n))Cl(2)] (n=1-4 and TAML(n) represents the tetraazamacrocyclic ligands) in the presence of CH(3)I or C(2)H(5)Br. The structures of all the complexes have been established on the basis of elemental analyses, conductivity measurements, IR, (1)H NMR, (13)C NMR, (119)Sn NMR and X-ray spectral data. The antitumour effect of the compounds was examined on swiss mice. The results obtained clearly indicated that the compounds, [C(2)H(5)Sn(TAML(3))C(5)H(5)N] and [C(2)H(5)Sn(TAML(4))C(5)H(5)N] display effective antitumour activity. The emphasis has been given on in vivo study on male albino rats (Rattus norvegicus) by performing serum analyses, blood analyses and fertility test.
