<b>Background and Objective:</b> Freshwater fish aquaculture in Indonesia has grown rapidly, especially the aquaculture of catfish (<i>Pangasianodon hypophthalmus</i>). This species is very good because it is fast-growing and very popular in the market and is important for national food security in many Asian countries. One of the problems faced by freshwater fish aquaculture is ectoparasite <i>Ichthyophthirius multifiliis</i> infection, which often results in significant economic losses to freshwater fish aquaculture. This study aimed to check the effect extract of betel leaf against the ectoparasite, <i>Ichthyophthirius multifiliis</i> in pangasius catfish in an eco-friendly manner. <b>Materials and Methods:</b> A total of 120 fishes with a mean weight of 4.17±0.96 g and a length of 8.5±0.67 cm were examined. Preliminary research was carried out to detect ectoparasites in fish. All fish was infected with ectoparasitic Ich (100%) and were identified as a salt-like granule white spot and a large C-shaped macronucleus. Infected fishes were transferred and equally distributed to the tank (20 L water) which had previously been treated with betel leaf extract for 24 hrs, 3 days, at doses 2.5, 5 and 7.5 g L<sup></sup><sup>1</sup> and control. <b>Results:</b> The results showed that the betel leaf extract solution effect decreased significantly to the number of ectoparasites <i>Ichthyophthirius multifiliis</i>, both in mucus and pangasius catfish and a dose of 7.5 g L<sup></sup><sup>1</sup> was the optimum dose. <b>Conclusion:</b> Betel leaf extract has the potential to control the decrease in the number of ectoparasites, though further phytochemical studies will need to be performed.

Some evidence has demonstrated that Ichthyophthirius multifiliis, etiologic agent of "white spot disease", causes severe bioenergetics dysfunction in the spleen of naturally infected silver catfish (Rhamdia quelen), which contributes directly to disease pathogenesis. Recently, several studies have demonstrated the efficacy of Melaleuca alternifolia essential oil, popularly known as tea tree oil (TTO), in the treatment of freshwater fish naturally or experimentally infected with I. multifiliis. In this sense, the aim of this study was to evaluate whether TTO is capable of preventing or reducing splenic bioenergetics dysfunction in silver catfish naturally infected with I. multifiliis. Splenic cytosolic and mitochondrial creatine kinase (CK) and pyruvate kinase (PK) activities decreased in infected animals compared to uninfected animals, while adenylate kinase (AK) activity increased. Treatment with TTO was able to prevent the inhibition on splenic CK and PK activities but was not able to prevent the stimulation of AK activity. Based on this evidence, treatment with TTO prevents the impairment on energetic metabolism via improvement of enzymes belonging to the phosphotransfer network, such as CK and PK. In summary, this treatment can be considered an interesting approach to prevent the bioenergetics imbalance in spleen of silver catfish naturally infected with I. multifiliis.

This study evaluated the activity of leaf essential oil (EO) from Hyptis mutabilis as well as its major constituent, (-)-globulol, in infections by the parasite Ichthyophthirius multifiliis (ich). Effects on hematological, biochemical, and immunological parameters of silver catfish, Rhamdia quelen, exposed to the same samples also were evaluated. In the first experiment, naturally infected fish were treated with EO (0, 10, and 20 mg · L-1) and ethanol, using several methods of exposure. Fish mortality and the number of trophonts per fish were assessed after 48 and 96 hr. Hour-long daily baths resulted in optimal survival, so this methodology was used for the second experiment, in which infected animals were exposed to (-)-globulol at 2.5 and 5 mg · L-1. The most effective concentrations in Experiments 1 and 2 were chosen for Experiment 3, in which healthy animals were subjected to hour-long daily baths with EO (20 mg · L-1) or (-)-globulol (2.5 mg · L-1). Additionally, an in vitro experiment was performed with EO and globulol at the same concentrations of the in vivo test. EO and (-)-globulol increased the survival of fish infected with ich and altered certain hematological and biochemical parameters. After 4 days, levels of hematocrit, erythrocytes, and leukocytes increased significantly in healthy animals exposed to EO. Exposure to (-)-globulol increased leukocyte number alone. No significant differences in nonspecific immunological parameters were detected when treated groups were compared to controls, but the leukocytosis observed in EO- and globulol-treated healthy animals indicates that EO and (-)-globulol increased innate immunity in these fish. An in vitro antiparasitic effect was observed in both samples.

Dactylogyrus ctenopharyngodonid and Ichthyophthirius multifiliis are two important ectoparasites of freshwater fish. Co-infection by the two parasites leads to high fish mortality and results in heavy economic losses. This study aimed to evaluate the efficacy of medicated feed and a ginger extract bath against D. ctenopharyngodonid and I. multifiliis on grass carp and investigate the hematological response of grass carp co-infected by the two parasites. These results demonstrated that red blood cell (RBC) and thrombocyte percentage among leucocytes significantly decreased after grass carp were co-infected by D. ctenopharyngodonid and I. multifiliis. The monocyte and neutrophil percentages significantly increased with the increment of parasite mean intensities, while the lymphocyte percentage decreased. The activities of serum acid phosphatase (ACP), alkaline phosphatase (AKP), lysozyme (LZM), and superoxide dismutase (SOD) significantly increased after co-infection. When grass carp treated with medicated feed containing 4% of Astragalus membranaceus, Allium sativum, Morus alba, and Glycyrrhiza uralensis, the activities of ACP, AKP, LZM, and SOD were significantly enhanced, and the mean intensities of D. ctenopharyngodonid and I. multifiliis were significantly decreased. When grass carp was treated with medicated feed and a 4-mg/L ginger extract bath, all parasites were eliminated during 28 days. The bath of ginger extract at a concentration of 4 mg/L kept a low mean intensity of I. multifiliis and D. ctenopharyngodonid, then the two parasites were eliminated by oral administration of the medicated feed with an immunostimulant (Chinese medicine compound).

Since malachite green was banned for using in food fish due to its carcinogenic and teratogenic effects on human, the search of alternative drug to treat Ichthyophthirius multifiliis becomes urgent. This study aimed to (1) evaluate the ethanol extracts of medicinal plants Cynanchum atratum, Zingiber officinale, Cynanchum paniculatum, immunostimulant (A), and immunostimulant (B) for their efficacy against I. multifiliis, and (2) determine effects of medicated feeds with C. atratum, Z. officinale, C. paniculatum, and immunostimulant (A) to treat I. multifiliis in grass carp. The results in this study showed that the minimum concentrations of C. atratum, Z. officinale, and C. paniculatum extracts for killing all theronts were 16, 8, and 16 mg/L, respectively. In vivo experiments, fish fed with medicated feeds of C. atratum for 10 days, or Z. officinale for 3 days, or combination of three plants for 10 days resulted in a significant reduction in the I. multifiliis infective intensity on grass carp after theronts exposure. Grass carp fed with medicated feeds of immunostimulant (A) for 21 days showed no infection and 100 % of survival 15 days post theronts exposure. Therefore, immunostimulant (A) is a promising feed supplement to treated I. multifiliis with good antiparasitic efficacy.

In vitro effect of the Melaleuca alternifolia, Lavandula angustifolia and Mentha piperita essential oils (EOs) against Ichthyophthirius multifiliis and in vivo effect of M. alternifolia for treating ichthyophthiriasis in one of the most important South American fish, Piaractus mesopotamicus (Holmberg), were evaluated. The in vitro test consisted of three EOs, each at concentrations of 57 µL L(-1) , 114 µL L (-1) , 227 µL L(-1) and 455 µL L (-1) , which were assessed once an hour for 4 h in microtitre plates (96 wells). The in vitro results demonstrated that all tested EOs showed a cytotoxic effect against I. multifiliis compared to control groups (P < 0.05). The in vivo treatment for white spot disease was performed in a bath for 2 h day(-1) for 5 days using the M. alternifolia EO (50 µL L (-1) ). In this study, 53.33% of the fish severely infected by I. multifiliis survived after the treatment with M. alternifolia (50 µL L (-1) ) and the parasitological analysis has shown an efficacy of nearly 100% in the skin and gills, while all the fish in the control group died. Furthermore, the potential positive effect of M. alternifolia EO against two emergent opportunistic bacteria in South America Edwardsiella tarda and Citrobacter freundii was discussed.

Ichthyophthirius multifiliis (Ich) is a ciliate parasite that infects many species of freshwater fishes worldwide and causes heavy economic losses in aquaculture. Currently, parasiticides for controlling this parasite are limited, and few pond-practical chemical therapies exist. Hence, the search for new parasiticides is urgently needed. One challenge confronting the screening of potential parasiticides is the difficulty in raising enough parasite for efficacy testing as Ich is an obligate parasite. This study used species of Tetrahymena, Ich-related and cultivable ciliate protozoa, to evaluate two in vitro methods to test parasiticides. Plate counting and MTS assays (CellTiter 96® AQueous Non-Radioactive Cell Proliferation Assay) were used to compare lethal concentrations or median lethal concentrations (LC50) of copper sulphate, formalin and malachite green between T. thermophila and Ich theronts or between T. thermophila and Ich tomonts. The parasiticides that killed T. thermophila have been demonstrated to kill theronts or tomonts. These in vitro methods using T. thermophila can be used to screen novel parasiticides against Ich.

Ichthyophthirius multifiliis (Ich) is an important ciliate that parasitizes gills and skin of freshwater fish and causes massive fish mortality. In this study, two flavonoids (kuwanons G and O) with anti-Ich activity were isolated by bioassay-guided fractionation from the root bark of Morus alba, an important plant for sericulture. The chemical structures of kuwanons G and O were elucidated by spectroscopic analyses. Kuwanons G and O caused 100% mortality of I. multifiliis theronts at the concentration of 2 mg/L and possessed a median effective concentration (EC50) of 0.8 ± 0.04 mg/L against the theronts. In addition, kuwanons G and O significantly reduced the infectivity of I. multifiliis theronts at concentrations of 0.125, 0.25, 0.5, and 1 mg/L. The median lethal concentrations (LC50) of kuwanons G and O to grass carp were 38.0 ± 0.82 and 26.9 ± 0.51 mg/L, which were approximately 50 and 35 times the EC50 for killing theronts. The results indicate that kuwanons G and O have the potential to become safe and effective drugs to control ichthyophthiriasis.

The present study aims to evaluate the antiparasitic activity of active components from Costus speciosus against Ichthyophthirius multifiliis. Bioassay-guided fractionation was employed to identify active compounds from C. speciosus yielding 2 bioactive compounds: Gracillin and Zingibernsis newsaponin. In-vitro assays revealed that Gracillin and Zingibernsis newsaponin could be 100% effective against I. multifiliis at concentrations of 0.8 and 4.5 mg L(-1), with median effective concentration (EC50) values of 0.53 and 3.2 mg L(-1), respectively. All protomonts and encysted tomonts were killed when the concentrations of Gracillin and Zingibernsis newsaponin were 1.0 and 5.0 mg L(-1). In-vivo experiments demonstrated that fish treated with Gracillin and Zingibernsis newsaponin at concentrations of 1.0 and 5.0 mg L(-1) carried significantly fewer parasites than the control (P<0.05). Mortality of fish did not occur in the treatment group (Zingibernsis newsaponin at 5.0 mg L(-1)) during the trial, although 100% of untreated fish died. Acute toxicities (LD50) of Gracillin and Zingibernsis newsaponin for grass carp were 1.64 and 20.7 mg L(-1), respectively. These results provided evidence that the 2 compounds can be selected as lead compounds for the development of new drugs against I. multifiliis.

The parasitic ciliate Ichthyophthirius multifiliis infests all species of freshwater fish and can cause severe economic losses in fish breeding. The present study aims to evaluate the antiparasitic activity of the active components from Toddalia asiatica against I. multifiliis. Bioassay-guided fractionation and isolation of compounds with antiparasitic activity were performed on the methanol extract of T. asiatica yielding two bioactive compounds: chelerythrine and chloroxylonine identified by comparing spectral data (NMR and ESI-MS) with literature values. Results from in vitro antiparasitic assays revealed that chelerythrine and chloroxylonine could be 100% effective against I. multifiliis at the concentration of 1.2 mg L(-1) and 3.5 mg L(-1), with the median effective concentration (EC50) values of 0.55 mg L(-1) and 1.90 mg L(-1) respectively. In vivo experiments demonstrated that fish treated with chelerythrine and chloroxylonine at the concentrations of 1.8 and 8.0 mg L(-1) carried significantly fewer parasites than the control (P<0.05). The acute toxicity (LC50) of chelerythrine for goldfish was 3.3 mg L(-1).

Ichthyophthirius multifiliis (also called "ich") is an external protozoan parasite that may infest almost all freshwater fish species and caused significant economic damage to the aquaculture industry. Since the use of malachite green was banned, there have been relatively few effective alternative strategies for controlling I. multifiliis infections. The present study was designed to screen potential antiparasitic medicinal plants based on our previous studies, and comprehensively evaluate in vitro and in vivo anti-ich activity of selected plant extracts. The screening results showed that the methanol extract of Psoralea corylifolia had the highest activity against I. multifiliis theronts. In vivo theront trials demonstrated that 1.25 mg/L or more concentrations of P. corylifolia methanol extract caused 100 % mortality during the 4-h exposure period, and the subsequent in vitro trials indicated that the minimum concentration of P. corylifolia methanol extract that prevented the initial infestation was 2.50 mg/L. Protomonts and encysted tomonts surviving trials suggested that encysted tomonts were less susceptible to P. corylifolia methanol extract than protomonts, and the methanol extract of P. corylifolia at a concentration of 5.00 mg/L could kill 100 % of protomonts and 88.89 % of encysted tomonts. It was also observed that after 12-h exposure of protomonts or encysted tomonts to 2.50 mg/L of P. corylifolia methanol extract, the theronts emerged from encysted tomonts led to more infection level than the ones in the other groups. The results suggested that whether the protomonts finish encystment is crucial to the survival, reproduction, and theronts infectivity. In addition, our results showed that long duration (24 h) and high concentration (5.00 mg/L) significantly reduced the survival and reproduction of I. multifiliis tomont exited from the fish after in-bath treatment, and it is indicated that P. corylifolia methanol extract had a potential detrimental effect on I. multifiliis trophont in situ.

Ichthyophthiriasis is a widespread disease in aquaculture and causes mass mortalities of fish. The development of new antiprotozoal agents for the treatment of Ichthyophthirius multifiliis infections is of increasing interest. The aim of the present study was to investigate the efficacy of 30 medicinal plants against I. multifiliis. The results showed that the methanol extracts of Magnolia officinalis and Sophora alopecuroides displayed the highest antiprotozoal activity against theronts, with 4-h LC(50) values estimated to be 2.45 and 3.43 mg L(-1), respectively. Concentrations of 2.5, 5.0, 10.0, and 20.0 mg L(-1) of M. officinalis extracts resulted in tomont mortality of 9.7, 43.7, 91.3, and 100% at 20 h, respectively. From 40 to 320 mg L(-1) of S. alopecuroides extracts, tomont mortality increased from 29.7 to 100%. Antiprotozoal efficacy against settled tomonts (2 and 10 h) was also applied; the results indicated that encysted I. multifiliis tomonts were less susceptible to these plant extract treatments. In vivo experiments demonstrated that high concentrations of M. officinalis and S. alopecuroides extracts could kill tomonts, and M. officinalis significantly reduced its reproduction (P < 0.05). These results suggested that the methanol extracts of M. officinalis and S. alopecuroides have the potential to be used as an eco-friendly approach for the control of I. multifiliis.

Ichthyophthirius multifiliis is a holotrichous protozoan that invades the gills and skin surfaces of fish and can cause morbidity and high mortality in most species of freshwater fish worldwide. The present study was undertaken to investigate the antiparasitic activity of crude extracts and pure compounds from the leaves of Macleaya microcarpa. The chloroform extract showed a promising antiparasitic activity against I. multifiliis. Based on these finding, the chloroform extract was fractionated on silica gel column chromatography in a bioactivity-guided isolation affording two compounds showing potent activity. The structures of the two compounds were elucidated as dihydrosanguinarine and dihydrochelerythrine by hydrogen and carbon-13 nuclear magnetic resonance spectrum and electron ionization mass spectrometry. The in vivo tests revealed that dihydrosanguinarine and dihydrochelerythrine were effective against I. multifiliis with median effective concentration (EC(50)) values of 5.18 and 9.43 mg/l, respectively. The acute toxicities (LC(50)) of dihydrosanguinarine and dihydrochelerythrine for richadsin were 13.3 and 18.2mg/l, respectively. The overall results provided important information for the potential application of dihydrosanguinarine and dihydrochelerythrine in the therapy of serious infection caused by I. multifiliis.

The ciliate Ichthyophthirius multifiliis is one of the most pathogenic parasites of fish maintained in captivity. In this study, effects of crude extracts, fractions, and compounds from the leaves of Macleaya cordata against I. multifiliis were investigated under in vitro conditions by bioactivity-guided isolation method. The dried ethanol extract of M. cordata was extracted successively in a separating funnel with petroleum ether, ethyl acetate, chloroform and n-butanol. Among them, only the chloroform extract showed promising activity and therefore, was subjected to further separation and purification using various chromatographic techniques. Four compounds were isolated from chloroform extract, but only one compound showed potent activity. The structure of the active compound was elucidated as sanguinarine by hydrogen, carbon-13 nuclear magnetic resonance spectrum and electron ionization mass spectrometry. In vitro antiparasitic efficacy tests exhibited that sanguinarine was 100% effective against I. multifiliis at a concentration of 0.7 mg l(-1), with LC(50) and LC(90) values of 0.437 and 0.853 mg l(-1) after 4 h of exposure. In vivo antiparasitic efficacy tests showed that the number of I. multifiliis on the gills in the treatment group (in 0.9 mg l(-1) sanguinarine) was reduced by 96.8%, in comparison to untreated group at 25°C for 48 h. Mortality of fish did not occur in the treatment group during the trail, although 40% of untreated fish died. Our results indicate that the studied plant extracts, as well as sanguinarine might be potential sources of new antiparasitic drug for the control of I. multifiliis.

The ciliate Ichthyophthirius multifiliis is among the most pathogenic parasites of fish maintained in captivity. In the present study, the effects of the crude methanolic extract of leaves of Mucuna pruriens and the petroleum-ether extract of seeds of Carica papaya against I. multifiliis were investigated under in vivo and in vitro conditions. Goldfish (Carassius auratus auratus) infected with the parasites were immersed for 72 h in baths with M. pruriens extract, and for 96 h in baths with C. papaya extract. There was a 90% reduction in numbers of I. multifiliis on fish after treatment in baths of each plant extract at 200 mg l(-1 )compared to untreated controls. Consequently, parasite-induced fish mortality was reduced significantly. A complete interruption of trophont recruitment was achieved by immersion in the M. pruriens extract. In vitro tests led to a 100% mortality of I. multifiliis in 150 mg/l M. pruriens extract, and in 200 mg/l of C. papaya extract after 6 h. Although the active constituents of the medicinal plant extracts are still unknown, we have demonstrated that they have potential for effective control of I. multifiliis.
