RESUMEN

This trial compared the rate and time of viral clearance in subjects receiving a combination of nitazoxanide, ribavirin, and ivermectin plus Zinc versus those receiving supportive treatment. This non-randomized controlled trial included 62 patients on the triple combination treatment versus 51 age- and sex-matched patients on routine supportive treatment. all of them confirmed cases by positive reverse-transcription polymerase chain reaction of a nasopharyngeal swab. Trial results showed that the clearance rates were 0% and 58.1% on the 7th day and 13.7% and 73.1% on the 15th day in the supportive treatment and combined antiviral groups, respectively. The cumulative clearance rates on the 15th day are 13.7% and 88.7% in the supportive treatment and combined antiviral groups, respectively. This trial concluded by stating that the combined use of nitazoxanide, ribavirin, and ivermectin plus zinc supplement effectively cleared the SARS-COV2 from the nasopharynx in a shorter time than symptomatic therapy.

The balance between tumor accumulation and renal clearance has severely limited the efficacy of mesoporous silica-based drug nanocarriers in cancer therapy. Herein, a pH-responsive dissociable mesoporous silica-based nanoplatform with efficient dual-drug co-delivery, tumor accumulation and rapid clearance for cancer therapy is achieved by adjusting the wetting of the mesoporous silica surface. At pH 7.4, the synthesized spiropyran- and fluorinated silane-modified ultrasmall mesoporous silica nanoparticles (SP-FS-USMSN) self-assemble to form larger nanoclusters (denoted as SP-FS-USMSN cluster) via hydrophobic interactions, which can effectively co-deliver anticancer drugs, doxorubicin hydrochloride (Dox) and curcumin (Cur), based on the mesopores within SP-FS-USMSN and the voids among the stacked SP-FS-USMSN. At pH 4.5-5.5, the conformational conversion of spiropyran from a "closed" state to an "open" state causes the wetting of the SP-FS-USMSN surface, leading to the dissociation of the SP-FS-USMSN cluster for drug release and renal clearance. The in vitro and in vivo studies demonstrate that the Cur and Dox co-loaded SP-FS-USMSN cluster (Cur-Dox/SP-FS-USMSN cluster) possesses great combined cytotoxicity, and can accumulate into tumor tissue by its large size-favored EPR effect and potently suppress tumor growth in HepG2-xenografted mice. This research demonstrates that the SP-FS-USMSN cluster may be a promising drug delivery system for cancer therapy and lays the foundation for practical mesoporous silica-based nanomedicine designs in the future.

Coccidiosis is a disease caused by Eimeria spp., resulting in approximately 3 billion US dollar loss in the poultry industry annually. The present study evaluated the effects of potential feed additives, 2-Nitro-1-propanol (NP) and nitroethanol (NE), on control of coccidiosis. An in vitro experiment indicated that both NP and NE inhibited the development of sporozoites in Madin-Darby bovine kidney cells (MDBK). The in vivo study was further conducted to evaluate the effects of NP and NE on growth performance, nitrogen-corrected apparent metabolizable energy (AMEn), and intestinal lesion scores of broilers challenged with Eimeria spps. Six treatments were tested in the study, including the nonchallenged control, challenged control, 100 ppm NP, 200 ppm NP, 100 ppm NE, and 200 ppm NE. Broilers were fed the treatment diets from day 12 until the end of the trial. All birds except the unchallenged control were challenged with Eimeria maxima, Eimeria tenella, and Eimeria acervulina on day 14. The growth performance was calculated, and the intestinal lesion was scored on day 20. The results showed that Eimeria challenge significantly reduced growth performance, increased intestinal lesion scores, and decreased AMEn compared with the nonchallenged control group. Birds fed with 200 ppm of NP had reduced growth performance compared with the nonchallenged control and challenged control. However, the supplementation of NP significantly improved AMEn and reduced cecal damage. Overall, NP and NE reduced sporozoites numbers in the MDBK cells. NP improved dietary digestibility of energy and reduces lesion scores in the ceca but could not maintain growth performance in broiler chickens infected with Eimeria spp.

Foodborne disease caused by Salmonella Enteritidis (SE) is one of the important public health and economic concerns. A study was conducted to determine the effect of supplementation with 2-nitroethanol (NE) and 2-nitropropanol (NP) on Salmonella recovery of internal organs as well as on the immune gene expression in the ileum of laying hens. Thirty-six White Leghorns were orally gavaged with nalidixic acid resistant Salmonella Enteritidis (SENR). Hens were housed individually in wire-laying cages and randomly assigned to six dietary treatments: T1 = SENR unchallenged (negative control), T2 = SENR challenged (positive control), T3 = SENR challenged + 100 ppm NE, T4 = SENR challenged + 200 ppm NE, T5 = SENR challenged + 100 ppm NP, and T6 = SENR challenged + 200 ppm NP. Hens were sampled at 7 days post inoculation (dpi). Ceca, liver with gall bladder (L/GB), and ovary samples were collected for bacteriology, and ileum samples were collected for analysis of immune gene expression. T3 and T6 significantly reduced (P < 0.05) cecal SENR count, whereas T4 and T5 were not different from T2, the SENR challenged control. There was no significant difference in SENR reduction in the L/GB or ovary after supplementation of either nitrocompounds. Pro- and anti-inflammatory cytokines such as interferon (IFN)-γ, interleukin (IL)-1B, IL-6, toll-like receptors (TLR)-4, and IL-10 all were significantly upregulated (P < 0.05) after SENR challenge. Supplementation at both levels of NE and NP showed a significant immune gene expression response in the ileum with reduction of IFN-γ, IL-6, TLR-4, and IL-10 mRNA expression. Overall, nitrocompounds such as NE and NP can be used in the intervention strategy to reduce Salmonella infection in hens.

A variety of tissue biomolecules and intracellular structures are known to be autofluorescent. However, autofluorescent signals in brain tissues often confound analysis of the fluorescent markers used for immunohistochemistry. While investigating tissue and cellular pathologies induced by 3-nitropropionic acid, a mitochondrial toxin selective for striatal neurons, we encountered many autofluorescent signals confined to the lesion core. These structures were excited by blue (wavelength = 488 nm) and yellow-orange (555 nm), but not by red (639 nm) or violet (405 nm) lasers, indicating that this autofluorescence overlaps with the emission spectra of commonly used fluorophores. Almost all of the autofluorescence was localized in activated microglia/macrophages, while reactive astrocytes emitted no detectable autofluorescence. Amoeboid brain macrophages filled with autofluorescent granules revealed very weak expression of the microglial marker, ionized calcium-binding adaptor molecule 1 (Iba1), while activated microglia with evident processes and intense Iba1 immunoreactivity contained scant autofluorescent granules. In addition, immunolabeling with two lysosomal markers, ED1/CD68 and lysosomal-associated membrane protein 1, showed a pattern complementary with autofluorescent signals in activated microglia/macrophages, implying that the autofluorescent structures reside within cytoplasm free of intact lysosomes. A correlative light- and electron-microscopic approach finally revealed the ultrastructural identity of the fluorescent granules, most of which matched to clusters of lipofuscin-like inclusions with varying morphology. Thus, autofluorescence in the damaged brain may reflect the presence of lipofuscin-laden brain macrophages, which should be taken into account when verifying any fluorescent signals that are likely to be correlated with activated microglia/macrophages after brain insults.

The small molecule 4EGI-1 was identified as an inhibitor of cap-dependent translation initiation owing to its disruption of the eIF4E/eIF4G association through binding to eIF4E. 4EGI-1 exhibits growth-inhibitory and apoptosis-inducing activity in cancer cells; thus, we were interested in its therapeutic efficacy in human lung cancer cells. 4EGI-1, as a single agent, inhibited the growth and induced apoptosis of human lung cancer cells.When combined with the death ligand tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), enhanced apoptosis-induced activity was observed. As expected, 4EGI-1 inhibited eIF4E/eIF4G interaction and reduced the levels of cyclin D1 and hypoxia-inducing factor-1alpha (HIF-1alpha), both of which are regulated by a cap-dependent translation mechanism. Moreover, 4EGI-1 induced CCAAT/enhancer-binding protein homologous protein-dependent DR5 expression and ubiquitin/proteasome- mediated degradation of cellular FLICE-inhibitory protein (c-FLIP). Small interfering RNA-mediated blockade of DR5 induction or enforced expression of c-FLIP abrogated 4EGI-1's ability to enhance TRAIL-induced apoptosis, indicating that both DR5 induction and c-FLIP down-regulation contribute to enhancement of TRAIL-induced apoptosis by 4EGI-1. However, inhibition of eIF4E/eIF4G interaction by knockdown of eIF4E effectively reduced the levels of cyclin D1 and HIF-1alpha but failed to induce DR5 expression, downregulate c-FLIP levels, or augment TRAIL-induced apoptosis. These results collectively suggest that 4EGI-1 augments TRAIL-induced apoptosis through induction of DR5 and down-regulation of c-FLIP, independent of inhibition of cap-dependent protein translation.

Leishmaniasis from Leishmania infantum is a parasitary zoonotic disease and a serious problem to public health. Guidelines from Italian Health Authority (Istituto Superiore di Sanità) suggest to control the zoonotic visceral leishmaniasis canine reservoir in endemic areas using an association of preventive and therapeutic tools. Moreover, in literature there are no studies about the long term effects on the disease seroprevalence and incidence in relation to this "holistic" approach. Past research has considered the effects of the alternative employment of preventive or therapeutic treatment, usually for limited periods. In this retrospective study the patterns of seroprevalence and incidence of leishmaniasis in a dog shelter sited in an endemic area of Central Italy are described throughout a 4-year period. Both preventive spot-on tools (imidacloprid/permetrin) and therapeutic protocols based on antimonials and allopurinol were administered. The results showed a progressive reduction of prevalence and incidence of serological reactivity to L. infantum, corroborating the effectiveness of the treatment administered to the animals. Significant improvements from the beginning to the end of the 4-year period were reported, considering both prevalence and incidence. A very low rate of relapses (8% in a pool of 67 subjects positive since 2004; 10.2% among all subjects enrolled in the study) was achieved.

Behavior was altered and survivorship was reduced when parasitoids, Anagyrus pseudococci (Girault) (Hymenoptera: Encyrtidae), were fed flowers from buckwheat, Fagopyrum esculentum L. (Polygonaceae), treated with soil applications of imidacloprid (Marathon 1% G). Parasitoids at 1 d had significantly reduced survivorship of 38 +/- 6.7% on label rate and 17 +/- 4.2% on twice label rate compared with 98 +/- 1.2% on untreated flowers. Parasitoids trembled 88% on label rate and 94% on twice label rate compared with 0% on untreated flowers. Residue analysis on a composite sample of 425 flowers showed that imidacloprid concentration was 6.6 +/- 1.0 ppm (16 ppb/flower) in label rate, 12.3 +/- 2.7 ppm (29 ppb/flower) in twice label rate, and 0 ppb in untreated flowers. The hydroxy metabolite concentration was 1.1 ppm (2.4 ppb/flower) in label rate, 1.9 ppm (4.4 ppb/flower) in twice label rate, and 0 ppm in untreated flowers. The olefin metabolite concentration was 0.2 ppm (0.5 ppb/flower) in label rate, 0.5 ppm (1.1 ppb/flower) in twice label rate, and 0 ppm in untreated flowers. Soil-applied imidacloprid used at flowering may be translocated to nectar in higher concentration compared with the imidacloprid seed treatment Gaucho. Considerable research has studied effects of Gaucho-treated canola, sunflower, and maize on behavior and mortality of Apis mellifera L. In our laboratory, we showed that translocation of imidacloprid to flowers reduced survivorship and altered behavior of pink lady beetle, Coleomegilla maculata DeGeer (Smith and Krischik 1999) and green lacewing, Chrysoperla carnea Stephens (Rogers et al. 2007).

Nobiletin and 3,5,6,7,8,3',4'-heptamethoxyflavone (HPT), isolated from the peel of Citrus plants, were examined for the anti-tumor-initiating activity on two-stage carcinogenesis of mouse skin tumors induced by a nitric oxide donor, (+/-)-(E)-methyl-2-[(E)-hydroxyimino]-5-nitro-6-methoxy-3-hexenamide, as an initiator and 12-O-tetradecanoylphorbol-13-acetate as a promoter. HPT exhibited the remarkable anti-tumor-initiating effect on mouse skin and it suggested the possibility of HPT being a chemopreventive agent against nitric oxide (NO) carcinogenesis.

The effects on thyroid function of an inhibitor of tyrosine dehalogenase, 3-nitro-L-tyrosine (MNT) have been investigated in rats. In preliminary studies, marked inhibition of iodotyrosine deiodination was demonstrated in rats drinking 8 mM MNT. A series of experiments was then performed in which rats received Remington low iodine diet and 8 mM MNT as drinking fluid. This regimen had the following effects, compared to the effects of a low iodine diet alone: (a) a decrease in serum protein-bound iodine, elevation of serum thyrotropin level, goiter, and growth inhibition all prevented or reversed by iodine supplements: (b) on initiation of MNT, a 2- to 3-fold increase in the rate of release of radioiodine from the thyroid and concomitant urinary excretion of large amounts of organic iodine: and (c) after 2 wk of MNT, a greatly increased rate of thyroidal uptake and release of (131)I, an increase in the ratio of monoiodotyrosine-(131)I to diiodotyrosine-(131)I in thyroid proteolysates and the appearance of labeled iodotyrosines in serum. Acute administration of MNT intraperitoneally to rats on either an iodine-deficient or iodine-sufficient diet did not inhibit thyroidal uptake of (131)I or alter the distribution of (131)I among thyroidal iodoamino acids. It is concluded that MNT is an effective inhibitor of iodotyrosine deiodination in vivo, without other important actions on thyroid function. Thus, MNT treatment affords a model for the human dehalogenase defect. By provoking iodotyrosine secretion and consequent urinary loss of iodine, MNT can exaggerate the effects of a low iodine intake, producing goitrous hypothyroidism despite a rapid rate of iodine turnover in the thyroid.
