RESUMEN

Porcine circovirus 2 (PCV-2) is a primary agent of post-weaning multi-systemic wasting syndrome (PMWS), ubiquitous in pig herds. The course of viraemia and seroconversion in naturally infected pigs were investigated in piglets from the 2nd week of their life. Piglets were divided into seropositive (Ab(+)) and seronegative (Ab(-)) groups. Subsequently, after vaccination against PCV-2 (Ingelvac(®) CIRCOFLEX™, Böehringer Ingelheim), they were further divided into non-vaccinated seronegative (NVAC/Ab(-)) and seropositive (NVAC/Ab(+)), and vaccinated seronegative (VAC/Ab(-)) and seropositive (VAC/Ab(+)). PCV-2 colostral antibodies failed to prevent development of natural PCV-2 infection in conventional piglets; however, this occurred at a higher age in comparison with seronegative pigs. Neither colostral nor post-infection antibodies prevented development of viraemia, which persisted up to the end of the study (the 19th week), but without clinical signs of PMWS. Vaccination failed to prevent development of natural PCV-2 infection, but viraemia was limited to between the 8th and 10th week. The presence of colostral anti-PCV-2 antibodies did not show any untoward effect to vaccination; on the contrary, VAC/Ab(+) animals showed the lowest titre of viraemia.

Porcine circovirus type 2 (PCV2) can be vertically transmitted resulting in fetal infection with or without clinical signs and lesions. The primary objective of this study was to assess the prevalence of intrauterine PCV2 infection in clinically normal newborn piglets in conventional pork production facilities. Five commercial breeding herds located in the U.S. and Mexico were included in the study. A total of 125 sows and 3-5 neonatal piglets per sow were arbitrarily selected. Blood and colostrum samples were collected from sows. Blood was collected from piglets prior to suckling. All samples were analyzed for the presence of anti-PCV2 IgG antibodies and presence and amount of PCV2 DNA. In addition, PCV2 DNA positive samples were further subtyped into PCV2a and PCV2b. All (125/125) sow colostrum samples and 96.8% (121/125) of the sow serum samples and 21.4% (107/499) of the piglet pre-suckle serum samples were positive for anti-PCV2 IgG antibody. The overall PCV2 DNA prevalence was 47.2% (59/125) in sow serum, 40.8% (51/125) in sow colostrum, and 39.9% (199/499) in pre-suckle piglet serum. In the PCV2 DNA positive samples, PCV2b was detected at a higher frequency (69.5% for sow serum, 84.3% for sow colostrum, and 74.4% for piglet serum) compared to PCV2a (18.6% for sow serum, 9.8% for sow colostrum, and 15.6% for piglet serum). Concurrent PCV2a and PCV2b infection was detected in 11.9% of the sow serum, in 5.9% of the colostrum samples, and in 10.0% of the piglet serum samples. In conclusion, an unexpectedly high prevalence of PCV2 viremia was detected in healthy sows (serum and colostrum) and their pre-suckle piglets in the five breeding herds investigated and PCV2b was more prevalent than PCV2a. This information adds to the knowledge of PCV2 infection in breeding herds.

Groups (5 to 15 per group) of gnotobiotic swine were infected oronasally with porcine circovirus type 2 (PCV2) at 3 days of age and then given 1 of 6 different commercial Mycoplasma hyopneumoniae (M. hyopneumoniae) bacterins as either a single dose (7 d of age, 1 application products) or 2 doses (7 and 21 d of age, 2 application product). Control groups received PCV2 alone (n = 9) or were infected with PCV2 and immunized twice with keyhole limpet hemocyanin (KLH) emulsified in incomplete Freund's adjuvant (ICFA) (n = 7). Five of 7 (71%) PCV2-infected piglets immunized with KLH/ICFA developed mild or overt PMWS, whereas none of 9 piglets infected with PCV2 alone developed PMWS. Five of 12 (42%) piglets vaccinated with a commercial bacterin containing mineral oil adjuvant developed PMWS following vaccination. None of the PCV2-infected piglets in the other bacterin-vaccinated groups developed PMWS in this model of PCV2-associated disease. This difference in prevalence of PMWS in piglets given the mineral oil-adjuvanted M. hyopneumoniae bacterin and the other M. hyopneumoniae bacterin vaccination groups was statistically significant (P < 0.05).
