RESUMEN

In this study, the fumigant antifungal activity of 10 Lamiaceae plant essential oils was evaluated against two phytopathogenic fungi, Raffaelea quercus-mongolicae, and Rhizoctonia solani. Among the tested essential oils, thyme white (Thymus vulgaris) and summer savory (Satureja hortensis) essential oils exhibited the strongest fumigant antifungal activity against the phytopathogenic fungi. We analyzed the chemical composition of two active essential oils and tested the fumigant antifungal activities of the identified compounds. Among the tested compounds, thymol and carvacrol had potent fumigant antifungal activity. We observed reactive oxygen species (ROS) generation in two fungi treated with thymol and carvacrol. Confocal laser scanning microscopy images of fungi stained with propidium iodide showed that thymol and carvacrol disrupted fungal cell membranes. Our results indicated that ROS generated by thymol and carvacrol damaged the cell membrane of R. querqus-mongolicae and R. solani, causing cell death.

Dried leaves of Satureja khuzestanica Jamzad are a popular herbal medicine and dental anesthetic among the nomadic inhabitants of southwestern Iran. The present study establishes criteria for identification of S. khuzestanica dried whole, fragmented and powdered leaves for specification as a herbal substance using macroscopic and microscopic characterization. Quantitative microscopy techniques were also considered. Macroscopically leaves exhibit a grayish-green color, are broadly ovate in shape with an acute apex, attenuate base, and ciliate margin and have a surface covered by an indumentum of glandular and non-glandular trichomes. Microscopically leaves have an isobilateral amphistomatic structure containing peltate glandular trichomes consisting of a multiseriate stalk (five cells) and an enlarged secretory head composed of 12 cells, capitate glandular trichomes of variable morphology together with two types of non-glandular trichomes. Oval-shaped hygromorphic diacytic stomata with an adaxial stomatal index of 13.54, collateral vascular bundles consisting of xylem, and three layers of sclerenchymatous tissue close to phloem together with cluster, prismatic and raphide calcium oxalate crystals were also identified as useful pharmacognostic parameters for identification of S. khuzestanica dried leaves.

In this study we looked into the cells and histological organization of leaves (Saturejae folium) as well as a phyto-chemical composition of overground parts (Saturejae herba) of endemic species Satureja subspicata Bartl. ex Vis. (Lamiaceae) collected during year 2003 on south slopes of mountain Velez in Herzegovina. Microscopic organization was analyzed in wet slides using light microscope. Estimation of stomata index was done according to Ph. Yug. IV. Chemical composition of overground material extracts was determined by thin layer chromatography (TLC) using thymol as a reference. In our research we found the following: Leaf structure of the analyzed species Satureja subspicata points at numerous specificities in anatomical and histological sense. In histological sense, leaf is of ventral type, with differentiated upper and lower epidermis and palisade and spongy tissue in between. Stoma index assigned according to Ph. Yug. IV leads to a conclusion that it is the case of diastitic stomata, which is common feature of most species from Lamiaceae family. Comparative qualitative analysis of essential oils in species Satureja subspicata showed similarities with other species from Lamiaceae family such as Thymus L. (thymol). In fact, we found more common substances that are part of the species Satureja montana L. extract, but in different concentrations.
