RESUMEN

The topographical organization of the efferent projections from the cat substantia nigra (SN) to the thalamus and the superior colliculus was examined using the anterograde transport of horseradish peroxidase-wheat germ agglutinin (HRP-WGA) and of labelled proteins. HRP-WGA or a mixture of [14C]amino acids was injected into various areas of the SN and the transported material visualized on coronal brain sections by histochemistry or autoradiography, respectively. The retrograde transport of [14C]gamma-aminobutyric acid ([14C]GABA) injected into thalamic nuclei was used also to determine the identity of the nigrothalamic projections. Identical results were found using either the anterograde transport of HRP-WGA or of labelled proteins. In the thalamus, dense nigral projections were observed in the nucleus ventralis medialis (VM) and in the rostromedioventral part of the nucleus ventralis lateralis (VL) whilst more limited projections were seen in the nuclei centralis lateralis (CL) and paracentralis (PC) as well as in the paralamellar zone of the nucleus medialis dorsalis (MD-Il). In addition, a patchy distribution of HRP-WGA or of radioactivity was found in the intermediate layer of the superior colliculus. More precisely, labelling of the VM was dense following injection of [14C]amino acids into the intermediate part of the SN pars reticulata (SNR) regardless of the depth of the injection site, whilst the intralaminar nuclei were labelled preferentially following injections made into the dorsal part of the intermediate SNR. Nigral projections to the intermediate layer of the superior colliculus were visualized over the whole mediocaudal and laterorostral extent when [14C]amino acids were injected into the rostral part of the SNR. Labelling of the superior colliculus was also seen following injection of [14C]amino acids into the intermediate part of the SNR but, in this case, ventral injections led to a more intense labelling than dorsal ones. Both the SNR and the SN pars compacta (SNC) were labelled when [14C]GABA was injected into the VM nucleus of the thalamus, confirming that the nigro VM projection is GABAergic and showing that recurrent collaterals of these GABAergic cells innervating the SNC also contained the transported radioactive material. In this condition ([14C]GABA injection into the VM), the thalamic reticularis nucleus also exhibited a dense labelling.(ABSTRACT TRUNCATED AT 400 WORDS)

Neurotransmitter-related messenger RNAs were detected by in situ hybridization in sections of rat and mouse brains by using 35S-radiolabelled RNA probes transcribed from cDNAs cloned in SP6 promoter-containing vectors. The distribution of messenger RNAs for glutamic acid decarboxylase, tachykinins (substance P and K), and tyrosine hydroxylase was examined in the striatum, pallidum, and substantia nigra. Dense clusters of silver grains were observed with the RNA probe complementary of the cellular messenger RNA for glutamic acid decarboxylase (antisense RNA) over most large neurons in the substantia nigra pars reticulata and medium-sized to large neurons in all pallidal subdivisions. A few very densely and numerous lightly labelled medium-sized neurons were present in the striatum. Among the areas examined, only the striatum contained neurons labelled with the antisense tachykinin RNA. Most of these neurons were of medium size, and a few were large. With the antisense tyrosine hydroxylase RNA, silver grains were found over neurons of the substantia nigra pars compacta and adjacent A10 and A8 dopaminergic cell groups. No signal was observed with RNAs identical to the cellular messenger RNA for glutamic acid decarboxylase or tachykinin (sense RNA). These results show a good correlation with immunohistochemical studies, suggesting that documented differences in the distribution and the level of glutamic acid decarboxylase, tyrosine hydroxylase, and substance P immunoreactivities in neurons of the basal ganglia are related to differences in the level of expression of the corresponding genes rather than to translation accessibility, stability, or transport of the gene products.

Following acute administration of sulpiride (100 mg/kg, i.p.), Met-enkephalin levels were increased in striatum but decreased in substantia nigra and nucleus accumbens. No effect was observed in hypothalamus or frontal cortex. In contrast, Leu-enkephalin content was decreased in the nucleus accumbens but not elsewhere. Administration of sulpiride (10 or 100 mg/kg, i.p.) for 21 days, followed by 2 days drug withdrawal, caused a dose-dependent increase in Met-enkephalin in nucleus accumbens but decreases in hypothalamus and frontal cortex. No change was observed in the striatum or substantia nigra. Repeated sulpiride administration caused a decrease in Leu-enkephalin content of the hypothalamus and cortex but not elsewhere.

RESUMEN

The distribution of immunoreactive alpha-neo-endorphin (alpha-Neo) in the rat brain was demonstrated by means of highly specific radioimmunoassay (RIA). The highest concentrations of immunoreactive (ir) alpha-Neo (100 pmol/g tissue) were found in the substantia nigra, the hypothalamic supraoptic nucleus (86 pmol/g) and the preoptic area (63 pmol/g); medium levels were found in the basal ganglia, mesencephalic reticular formation, nucleus accumbens and hippocampus. Other structures contained less alpha-Neo-ir. The possible anatomical interaction between alpha-Neo and monoaminergic nerve terminals or cell bodies in some structures is discussed.

RESUMEN

Unilateral ablation of the frontal cortex results in a significant reduction (19.5%) of glutamic acid in the ipsilateral compared with the contralateral substantia nigra. The GABA level in the substantia nigra and both the glutamate and GABA levels in the hippocampus remain unchanged. The results suggest that glutamate is a transmitter in the cortico-nigral fibre tract.

The substantia nigra (SN) of the cat was shown, by the anterograde and retrograde horseradish peroxidase methods, to contain neurons which send their axons to the caudomedial portions of the suprageniculate nucleus and/or lateroposterior nucleus of the thalamus; these neurons were located in the restricted region in the lateral part of the SN, which corresponds to the pars lateralis of the SN.

RESUMEN

A case is presented in which the fourfold increase of the HI titer during the progression of the disease, and an increase in IgM content found at the beginning of the second week of the disease confirmed the diagnosis of tick-borne encephalitis. The light microscopic changes correspond to the findings accepted as characteristic to tick-borne encephalitis. Viruses, morphologically belonging to the Flavivirus genus were found by electron microscopy in the thalamus, substantia nigra, and cerebellum of the dissected brain. This paper presents the first demonstration of the virus in a case of a human tick-borne encephalitis.

RESUMEN

A radioenzymatic method for simultaneous determination of dopamine and DOPAC in small brain areas is described. By using this assay, 250 pg of dopamine and 150 pg of DOPAC can be estimated. The present method has been applied to compare the effect of different psychotropic drugs on the dopamine and DOPAC levels in the caudate nucleus, substantia nigra and medial basal hypothalamus.

The distribution of L-glutamate decarboxylase activity (GAD) and gamma-aminobutyric acid (GABA) was investigated in the hypothalamic nuclei and in parts of the substantia nigra in the rat. GAD varied markedly among these areas. The reticular part of the nigra showed the highest activity two-fold higher than any other nucleus. Among the hypothalamic nuclei, a 5-fold difference was found between the poorest and richest nuclei. High GAD was measured in the preoptic, anterior and dorsomedial nuclei. Low activity was found in arcuate and supraoptic nuclei. The lowest GAD activity was measured in the median eminence with only half the activity of the whole brain homogenate. This suggests that GABAergic neurones might not be involved in neuroendocrine regulation at the median eminence level. GABA was determined using the sensitive cycling microassay. The rats were killed by microwaves, procedure which was found to inactivate enzymatic processes within two sec without affecting the level or the distribution of GABA. Postmortem increments during the first 3 min following decapitation ranged up to 5 times the endogenous levels, and were proportional to the GAD activity of the corresponding nucleus. This confirms that GAD is the limiting factor in GABA synthesis and suggests that GABA turnover might be rapid. Endogenous GABA showed a uniform distribution within hypothalamic nuclei and nigra. No relationship appeared between endogenous GABA levels and GAD activities in the various nuclei. These results suggest that while GABA synthesis is likely to occur in non-evenly distributed nerve cells, most GABA may be stored in surrounding cells, presumably glia cells.

Monospecific antisera against the purified protein component of tissue thromboplastin (apoprotein-III) from human brain have been raised in goats and rabbits. The antisera neutralized tissue thromboplastin prepared from brain, thyroid gland and pulmonary tissue, indicating that apoproteins in the various preparations cross-reacted immunologically and therefore were similar or identical. Comparison of the activities of tissue thromboplastin preparations from 34 different areas of the brain demonstrated a characteristic distribution pattern and a wide range of activities. White and grey matter from the same areas had similar activities. Bulbus and tractus olfactorius, medulla oblongata, corpus pineale, hippocampus and hypothalamus contained 160-270% of the average activity, whereas cerebellum globus pallidus, nucleus ruber and substantia nigra contained 30-60%. The distinct distribution pattern was unrelated to tissue vascularization, and may suggest that apoprotein-III could serve other functions, apart from the coagulation of blood. The predominance in phylogenetically older brain regions would suggest that it represents a primitive or fundamental feature.

Using a sensitive radioimmunoassay we have studied the regional distribution of substance P. The level of substance P is higher in the mesencephalon, hypothalamus and preoptic area than in other regions of the brain. Substance P is found in especially high concentrations in the reticular part of the substantia nigra and the interpeduncular nucleus. It is present in large amounts in several septal, preoptic and hypothalamic nuclei as well.

RESUMEN

The distribution of calcium, copper, magnesium and zinc in five anatomical regions of both hemispheres of the human brain were investigated. No asymmetry was observed with the exception of the distribution of copper in the white matter. The concentrations of these elements in the substantia nigra from both sides of the midbrain were also measured.
