Hexavalent chromium (Cr (VI)) is a hazardous heavy metal that induces hepatotoxicity and nephrotoxicity. Thus, this study was planned to explore the ameliorating capacity of Aloe vera leaf gel extract (AV) and their conjugated silver nanoparticles (AVNP) against Cr (VI) induced hepatotoxicity and renal toxicity. The organ indices, level of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, malondialdehyde, total protein, and creatinine in blood serum were measured. The histopathological and micrometric analysis of the hepatic and renal tissue sections were studied. The hepatosomatic index was raised significantly (0.098 ± 0.13 g) in Cr treated group. The blood serum level of AST (484 ± 10.7 U/L), ALT (163 ± 5.5 U/L), ALP (336.7 ± 9.5 U/L), MDA (642.3 ± 28.3 U/L), and creatinine (4.0 ± 0.1 mg/dL) were increased significantly, whereas total protein level was declined (2.8 ± 0.3 g/dL) significantly in Cr exposed group. In the histopathological study, necrosis, disturbed hepatic cords, impaired glomeruli, and Bowman's capsule were noted. Micrometric data from the liver and kidney revealed a significant surge in the size of hepatocytes and their nuclei (1188.2 ± 467.7 µ2 and 456.5 ± 205.6 µ2) and CSA of glomeruli and Bowman's capsule (9051.8 ± 249.8 µ2 and 11,835.5 ± 336.7 µ2) in Cr (VI) exposed group, whereas the brush border (10.2 ± 4.0 µ) size declined significantly. The administration of AV and AVNP reduced the oxidative stress induced by Cr (VI).

Arthritis is a common illness that affects joints and it may result in inflammation and pain. Even though arthritis usually affects older people, it can also affect children, adults, and both genders. Numerous arthritic mouse models have been developed but the CIA model of rheumatoid arthritis (RA) has received the most attention. With the use of steroids, DMARDs, and NSAIDs, therapy objectives such as reduced disease incidence and better pain management are achieved. Long-term usage of these therapeutic approaches may have negative side effects. Herbal medications are the source of several medicinal substances. Studies have explored the potential benefits of medicinal plants in treating RA. These benefits include up-regulating antioxidant potential, inhibiting cartilage degradation, down-regulating inflammatory cytokines such as NF-kB, IL-6, and TNF-α, and suppressing oxidative stress. In this review, we systematically discuss the role of traditional medicinal plants in rheumatoid arthritis (RA) disease treatment. The role of different medicinal plants such as Curcuma longa, Syzygium aromaticum, Zingiber officinale and Withania somnifera, against arthritis is discussed in this review.

Diabetes is a life-threatening disease that affects different parts of the body including the liver, kidney, and pancreas. The core root of diabetes is mainly linked to oxidative stress produced by reactive oxygen species (ROS). Berberis lyceum Royle (BLR) is the source of natural products. It comprises numerous bioactive compounds having antioxidant activities. In the current investigation, silver nanoparticles from BLR root extract were synthesized, characterized, and assessed for antidiabetic potential. UV spectrophotometry, Transmission electron microscopy (TEM), Fourier transform infra-red spectroscopy (FTIR), and x-ray diffraction (XRD) were applied for the characterization of NPs. It was evident from the morphological studies that the synthesized NPs were spherical and the average size was 11.02 nm. Results revealed that BLR-AgNPs showed higher radical scavenging activity as compared to BLR extract. Moreover, BLR-AgNPs displayed superior in vivo and in vitro antidiabetic activity in comparison to BLR extract. Glucose level (116.5 ± 5.1 mg/dL), liver function test (ALAT: 54.038 ± 6.2 IU/L; ASAT: 104.42 ± 13.9 IU/L; ALP: 192.6 ± 2.4 IU/L; bilirubin: 1.434 ± 0.14 mg/dL; total protein: 5.14 ± 0.24 mg/dL), renal function test (urea: 39.6 ± 0.63 mg/dL; uric acid: 21.4 ± 0.94 mg/dL; creatinine: 0.798 ± 0.03 mg/dL; albumin: 4.14 ± 0.2 mg/dL), lipid profile level (cholesterol: 101.62 ± 3 mg/dL; triglyceride: 110.42 ± 7 mg/dL; HDL-C: 29.7 ± 3 mg/dL; LDL-C: 47.056 ± 1 mg/dL; VLDL-C: 22.0 ± 1.3 mg/dL) and hematology (WBCs: 3.82 ± 0.24 103 /µL; RBCs: 4.78 ± 0.42 106 /µL; Hb: 12.6 ± 1.0 g/dL; Hematocrit: 39.4 ± 3.7%; MCV: 65.8 ± 3 fL; platelets: 312 ± 22.4; neutrophils: 34.8 ± 1.87; eosinophils: 3.08 ± 0.43; monocytes: 3.08 ± 0.28; lymphocytes: 75.6 ± 3.77) confirmed the significant antidiabetic potential of BLR-AgNPs. Histopathological examination authenticated that BLR-AgNPs caused a significant revival in the morphology of the liver, kidney, and pancreas. Hence, findings of the study suggested the BLR-AgNPs as a potent antidiabetic agent and could be an appropriate nanomedicine to prevent diabetes in future. Recent developments in the green synthesis of metallic nanoparticles (NPs) using phytoconstituents have attracted the attention of the global scientific community. The present study was designed to synthesize silver NPs (AgNPs) using Punica granatum and Plectranthus rugosus plant extracts. The fabricated AgNPs were characterized using UV-visible spectrophotometry (UV-Vis), Fourier-transform infrared (FTIR) spectroscopy, scanning electron microscopy (SEM), and energy-dispersive x-ray spectroscopy (EDS). The shift in the color of the silver nitrate (AgNO3 ) solution after the addition of P. granatum and P. rugosus extracts indicated the synthesis of AgNPs. The effect of AgNO3 concentrations and pH on the synthesis of AgNPs was also evaluated. The findings of this study suggest that AgNO3 concentration of 1 mM, reaction time of 1 h, and pH of 7 at room temperature were the best suited conditions for the synthesis of AgNPs. According to the FTIR analysis, amidic and carbonyl compounds were primarily responsible for the encapsulation of AgNPs. SEM investigations have shown irregularly shaped geometry with sizes of 35 nm (P. granatum) and 33 nm (P. rugosus) with low agglomeration. The prepared AgNPs exhibited good potential for 2,2-diphenyl-1-picrylhydrazyl radical scavenging, with values of 70% (P. granatum) and 68% (P. rugosus). Hence, we conclude that the leaves of P. granatum and P. rugosus are excellent material for designing of different plant-extracted-conjugated AgNPs for biomedical applications. Morphology (size, shape) and structural variations (bonding pattern, crystallography, and atomic arrangements) have significant impacts on the efficacy of the metallic nanoparticles. Fabrication of these metal nanoparticles through green synthesis using plant extracts has increased attention due to their low cost, less hazardous byproducts, and multiple applications. In present study, Eucalyptus globulus extract was used to synthesize silver nanoparticles (AgNPs). Change of color from light brown to reddish brown and UV-visible spectral peak at 423 nm confirmed the formation of AgNPs. The shifting of FTIR spectra peaks indicated the potential role of the functional groups in extract as capping agents. The DLS evaluated the average size and stability of the nanoparticles while the surface morphology, size and the elemental composition of the AgNPs was established by the FESEM and EDX analysis. The SEM images revealed spherical nanoparticles of size ranging from 40-60 nm. Biogenic AgNPs showed better DPPH radical scavenging activity with IC50 (13.44 ± 0.3) as compared to leaves extract with IC50 (10.57 ± 0.2). The synthesized AgNPs showed higher zones of inhibition (ZOI) by well diffusion method against Escherichia coli, Staphylococcus aureus and Klebsiella pneumoniae. Results of present study highlights the potential benefits of Eucalyptus globulus leaves extract-based AgNPs for various biomedical uses.

Neurodegenerative disorders, caused by progressive neuron loss, are a global health issue. Among the various factors implicated in their pathogenesis, dysregulation of acetylcholinesterase activity has been recognized as a key contributor. Acetylcholinesterase breaks down the neurotransmitter acetylcholine, important for neural transmission. Evaluating phyto-compounds from Bacopa monnieri Linn. through in vitro and in silico analysis may expand their role as alternative therapeutic agents by modulating the function of acetylcholinesterase and complementing existing treatments. To accomplish this objective, chemical structures of phyto-compounds were retrieved from PubChem database and subjected to in silico and in vitro approaches. Virtual screening was performed through molecular docking and molecular dynamic simulation resulting in four top hit compounds including quercetin, apigenin, wogonin, and bacopaside X (novel lead compound for acetylcholinesterase inhibitor) with least binding score. Further, dose dependent acetylcholinesterase inhibition biochemical assay depicted that bacopaside X, apigenin, quercetin, and wogonin exhibited strong potential against acetylcholinesterase with IC50 values of 12.78 µM, 13.83 µM, 12.73 µM and 15.48 µM respectively, in comparison with the donepezil (IC50: 0.0204 µM). The in silico and in vitro research suggests that B. monnieri phyto-compounds have the potential to modulate molecular targets associated with neurodegenerative diseases and have a role in neuroprotection.

The present study was aimed to prepare the potent silk sericin-based hydrogels in combination with plant extracts (curcumin and banana peel powder) and silver nanoparticles (AgNPs) to accelerate the acute wound healing process. Experimental excision wounds were created in mice by biopsy puncture, and the wound healing potential of silk sericin (2%)-based hydrogel and its combinations with curcumin (2%), banana peel powder (2%), and AgNPs (2%) was estimated by calculating the percent wound contraction, healing time, histology of skin tissues, and different biochemical tests. The results showed that the mice treated with sericin-based hydrogels showed significantly (P < 0.001) high percent wound contraction as compared to negative control, and wounds were healed in 11 days. The histological evaluation also showed that wounds covered with hydrogels were healed more than the uncovered wounds. Furthermore, the results of biochemical tests revealed that the treatment groups showed a significant (P < 0.001) decrease in the serum level of pro-inflammatory cytokines (IL-6). A significant (P < 0.001) increase in anti-inflammatory cytokines (IL-10) and anti-oxidant enzymes was observed in treatment groups. The highest wound healing potential was observed by sericin-based hydrogel containing banana peel powder, leaving behind the commercially available ointment polyfax (positive control). It can be concluded that the silk sericin-based hydrogels in combination with plant extract and AgNPs can be used as natural biomaterials in wound dressing for the rapid healing of acute wounds.

The complications associated with diabetic wounds make their healing process prolonged. Hydrogels could be ideal wound dressings therefore present research was conducted to prepare silk sericin (an adhesive protein polymer) based hydrogels in combination with plant extracts and to evaluate its effectiveness against wound healing process in mice with alloxan induced diabetes. Excision wounds were formed via a biopsy puncture (6 mm). Experimental hydrogels were prepared and applied topically on the diabetic wounds. All the hydrogel treatment groups showed significantly higher (P < 0.001) percent wound contraction from day 3 to day 11 as compared to the negative diabetic control group. The serum level of anti-inflammatory cytokine (Interleukin-10) and tissue inhibitor metalloproteinase (TIMP) was significantly higher (P < 0.001), while the level of pro-inflammatory cytokines (tumor necrosis factor-α, Interleukin-6) and matrix metalloproteinases (MMP-2, MMP-9) was significantly lower (P < 0.001) in hydrogels treatment groups as compared to diabetic control group. Although all the hydrogels showed effective results, however the best results were shown by 4 % sericin+4 % banyan+4 % onion based hydrogel. It can be concluded that Sericin based hydrogel enriched with banyan and onion extracts can be used as an effective remedy for the treatment of diabetic wounds due to their high healing and regenerative properties.

Green synthesis of nanoparticles is an emerging field due to it's biosafety and promising results. Biological systems due to their biodiversity are employed in different production processes. In this study Spirulina platensis mediated silver nanoparticles (S-AgNPs) production was done. The Uv spectra, FTIR and SEM analysis was performed for the characterization of biosynthesized S-AgNPs. The biocompatibility evaluation of S-AgNPs was done through hemolysis analysis. S-AgNPs were also evaluated for anticoagulant and thrombolytic potential. In addition to the medical applications of S-AgNPs, silver nanoparticles have been known to show potential industrial applications among which one application is the utilization of silver nanoparticles in the degradation of toxic industrial dyes. Therefore, degradation assay of Eosin Y and Methylene Blue dyes was estimated. The SEM analysis of S-AgNPs showed the particle size of 50-65 nm, whereas the biocompatibility analysis showed that these S-AgNPs are biocompatible at ≤400 µM concentration. The S-AgNPs showed good anticoagulant potential and thrombolytic potential and were able to degrade 44% of the thrombus. The S-AgNPs significantly degraded 76% of Eosin Y within 30 min, whereas Methylene Blue was 80% degraded within 20 min (P-value ≤ .001). To the best of our knowledge, the dye degradation of Eosin Y, thrombolytic activity and anticoagulant activity of S-AgNPs produced from the biomass of Spirulina platensis has been reported for the first time. In the current study, we conclude that our biosynthesized S-AgNPs showed promising medical and industrial applications and these nanoparticles can be further evaluated and upscaled for large scale applications.

Fabrication, characterization and evaluation of the larvicidal potential of novel silk protein (sericin)-based silver nanoparticles (Se-AgNPs) were the prime motives of the designed study. Furthermore, investigation of the sericin as natural reducing or stabilizing agent was another objective behind this study. Se-AgNPs were synthesized using sonication and heat. Fabricated Se-AgNPs were characterized using particle size analyzer, UV spectrophotometry, FTIR and SEM which confirmed the fabrication of the Se-AgNPs. Size of sonication-mediated Se-AgNPs was smaller (7.49 nm) than heat-assisted Se-AgNPs (53.6 nm). Being smallest in size, sonication-assisted Se-AgNPs revealed the significantly highest (F4,10 = 39.20, p = .00) larvicidal activity against fourth instar lab and field larvae (F4,10 = 1864, p = .00) of dengue vector (Aedes aegypti) followed by heat-assisted Se-AgNPs and positive control (temephos). Non-significant larvicidal activity was showed by silver (without sericin) which made the temperature stability of silver, debatable. Furthermore, findings of biochemical assays (glutathione-S transferase, esterase, and acetylcholinesterase) showed the levels of resistance in field strain larvae. Aforementioned findings of the study suggests the sonication as the best method for synthesis of Se-AgNPs while the larvicidal activity is inversely proportional to the size of Se-AgNPs, i.e., smallest the size, highest the larvicidal activity. Conclusively, status of the sericin as a natural reducing/stabilizing agent has been endorsed by the findings of this study. RESEARCH HIGHLIGHTS: Incorporation of biocompatible and inexpensive sericin as a capping/reducing agent for synthesis of Se-AgNPs. A novel sonication method was used for the fabrication of Se-AgNPs which were thoroughly characterized by particle size analyzer, UV-visible spectrophotometry, SEM and FTIR. Analysis of enzymatic (GSTs, ESTs) levels in field and lab strains of Aedes aegypti larvae for evaluation of insecticides resistance.

Present study was aimed to assess the bactericidal potential of sericin-capped silver nanoparticles (Se-AgNPs) synthesized by heat, light, and sonication. Se-AgNPs were characterized by size analyzer, UV spectrophotometry, energy-dispersive X-ray spectroscopy, and Fourier transform infrared spectroscopy. Average size of Se-AgNPs synthesized by heat, light and sonication was 53.60, 78.12, and 7.49 nm, respectively. All (10) bacterial strains were exposed to Se-AgNPs prepared from different methods to compare their antibacterial potentials. Largest zone of inhibition (13 ± 1.15 mm) was observed for sonication-based nanoparticles (NPs) against Klebseilla pneumoniae while the smallest zone of light assisted NPs against Serratia rubidaea (5 ± 1 mm). Bacterial strains were also exposed to different concentrations (0.2%, 0.3%, and 0.6%) of Se-AgNPs which showed largest zone (12 ± 1 mm) of inhibition for 0.4% of Se-AgNPs against Protius mirabilis and smallest zone (5 ± 1.154 mm) for 0.3% of Se-AgNPs against Escherichia coli. Furthermore, effect of different temperatures (5°C, 37°C, and 60°C) and pH (3, 7, and 12) on the efficacy and stability of Se-AgNPs was also evaluated against different bacterial strains. Sonication mediated NPs showed highest bactericidal results against K. pneumoniae (F3,8 = 6.154; p = 0.018) with smallest size NPs (7.49 nm) while lowest bactericidal results against S. rubidaea (5 ± 1 mm) were shown with largest size (78.12 nm) NPs prepared by natural light. These variations of bactericidal activities of NPs with difference size endorse that the Se-AgNPs with smallest size have highest antibacterial activity than larger size NPs. Moreover, Se-AgNPs maintain their bactericidal potency at wide range of temperature and pH, hence seemed stable.

The antibacterial activity of synthetic antimicrobial agents is well known, but most of them have several side effects and are effective against selective microbes. Recently, major concern for the microbiologists is to investigate for some stable, non-toxic, cheap, and eco-friendly antimicrobial agents with a wide range of bactericidal potential. A cost-effective and environmentally friendly alternate has been proposed in the form of green synthesized nanoparticles. The Present study was designed to fabricate sericin-coated silver nanoparticles (S-AgNPs) using sericin as stabilizer and reductant of silver ions and their antibacterial potential was evaluated at various concentrations and temperatures (8, 40, and 50°C). Antimicrobial activities were assessed by the agar well diffusion method. Antibacterial activity of S-AgNPs was measured at different concentrations (1-6 mg/ml) whereas; antifungal activity was tested at 5-20 mg/ml of S-AgNPs. Nanoparticles were characterized by UV-visible spectrophotometer, Fourier transform infrared spectroscopy, and scanning electron microscopy. These nanoparticles significantly subdued the growth of Clostridium difficile (18.7 ± 0.9 mm), Proteus mirabilis (12.3 ± 0.3 mm) and Bacillus licheniformis (10.7 ± 0.9 mm) and Aspergillus flavus (18.7 ± 2.0 mm), Mucor mycetes (13 .0 ± 1.5 mm), Candida albicans (15.3 ± 0.3 mm) and Aspergillus niger (10.0 ± 0.6 mm). S-AgNPs were stable at all temperatures and the maximum growth inhibition was found at 8°C for all pathogenic strains. We concluded that the S-AgNPs could be a potential candidate to inhibit the growth of bacterial and fungal pathogens at a wide range of environmental conditions like temperature. In various biomedical applications including antimicrobial and wound dressings, S-AgNPs can be used in the future to treat various bacterial and fungal infections.

Diabetes is involved in delayed wound healing that can be cured by natural products such as garlic, turmeric, and fibroin extracts. Alloxan monohydrate is used for inducing diabetes in mice. The percent wound contraction of garlic (150 mg/ml), turmeric (100 mg/ml), and fibroin (50 mg/ml), individually and in combinations garlic (150 mg/ml) + fibroin (50 mg/ml), turmeric (100 mg/ml) + fibroin (50 mg/ml), garlic (150 mg/ml) + turmeric (100 mg/ml), and garlic (150 mg/ml) + turmeric (100 mg/ml) + fibroin (50 mg/ml) was checked by evaluating the healing time, % wound contraction and histological analysis. The serum level of MMPs (MMP 2, MMP7, MMP 9), pro-inflammatory cytokines (TNF-α, IL-6, IL-8), and TIMPs were evaluated. With the combination of three extracts (Ga+Tu+Fi) garlic (150 mg/ml), turmeric (100 mg/ml) and fibroin (50 mg/ml), wounds healed in 12 days and had 97.3 ± 2.2% wound contraction. While the positive control (polyfax) and diabetic control (saline) wounds healed in 17- and 19-days with wound contraction of 96.7 ± 1.4% and 96.3 ± 1.1%, respectively. Histological analysis showed that the combination of Ga+Tu+Fi exhibited an increase in the growth of collagen fibers, fibroblasts number, and keratinocytes, and lessened inflammation of blood vessels. The combination of Ga+Tu+Fi significantly alleviated the serum concentration of TNF-α (14.2 ± 0.7 pg/ml), IL-6 (10.0 ± 1.0 pg/ml), IL-8 (16.0 ± 1.5 pg/ml), MMP2 (228.0 ± 18.1 pg/ml), MMP7 (271.0 ± 9.9 pg/ml), and MMP9 (141.0 ± 5.3 pg/ml) to diabetic control. The level of TIMPs (193.0 ± 9.1 pg/ml) was increased significantly with respect to diabetic control. We conclude that the combination of these biomaterials possessed high regenerative and healing capabilities and can be an effective remedy in the healing of chronic wounds in diabetic patients.

Burns wounds are one of the most health-threatening problems worldwide. Sericin is a natural protein synthesized by the silkworm Bombyx mori L. It is a natural biomaterial that helps in the management of wounds by promoting the proliferation of fibroblasts and keratinocytes. Jasminum grandiflorum L leaves contain high mucilage content and have been used to cure various dermal wounds in ancient times. The present study was aimed to investigate the individual and synergetic effect of sericin and J. grandiflorum leaves extract on burn injury in mice. Ten-week-old mice weighing 22 g ± 5 g were randomly divided into eight groups, each group containing 10 mice. Burn injury in mice was induced by using a hot metal rod. Wounds were evaluated morphologically and histologically. In comparison with controls, 4% sericin + jasmine (S + J) expressively improved wound contraction area. Restoration of the epidermis was also faster in S + J 4% group (P ≤ .01) than in other groups. Histopathological analysis also verified the efficacy of the 4% S + J cream. It is concluded that a mixture of sericin and jasmine leaves effectively improved the healing process of skin burns.

A novel therapeutic strategy for cancer treatment is to target altered tumor metabolism. Glucose- 6-phosphate dehydrogenase (G6PD) has been recently discovered to be implicated in apoptosis and angiogenesis, making it an excellent target in cancer treatment. The current study aimed to screen the plant extracts library to find potent hits against G6PD through enzymatic assay. Protein expression was induced by IPTG and purified using Ni-NTA columns after transformation of the pET-24a-HmG6PD plasmid into E. coli BL21-DE3 strain. An enzymatic assay was established by using purified rG6PD protein, for the screening of G6PD inhibitors. Out of 46 plant extracts screened, the sixteen plant extracts have shown inhibitory activity against the G6PD enzyme. At doses from 1 to 4 µg/ml, this extract demonstrated concentration-dependent inhibition of G6PD with an IC50 value of I.397 µg/ml. Moreover, the anticancer activity evaluation against HepG2 cells determined Smilax china as a potent inhibitor of cancer cells (IC50 value of 16.017 µg/ml). The acute and subacute toxicities were not observed in mice with various concentrations (50, 100, 200 and 2000 mg/kg). Furthermore, to identify the compounds from Smilax china as G6PD inhibitors, a literature-based phytochemical investigation of Smilax china was conducted, and sixty compounds were docked against the NADP+ and G6P binding sites of G6PD. The results of this study showed that three compounds were Scirpusin A, Smilachinin and Daucosterol with MolDock Score of -156.832, -148.215, and -145.733 respectively, against NADP+ binding site of G6PD. Conclusively, Smilax china root extract could be a safer drug candidate for the treatment of hepatocellular carcinoma.

The green method is not only harmonious to our environment but saves time and is comparatively cheaper than other methods. The current study is aimed to synthesize the silver nanoparticles (AgNPs) using the green method by using the leaves of Loranthus pulverulentus Wall (LPW) which functioned as a reducing as well as capping agent. The synthesis of biogenic AgNPs was confirmed by UV-Vis spectroscopy (UV-Vis.), Scanning electron microscopy (SEM), X-ray diffraction (XRD), energy dispersive X-ray (EDX), and Fourier transform infrared (FTIR). The absorbance maxima of the synthesized AgNPs were observed in the range of 405-435 nm. The SEM was used to find out the size and shape of the synthesized AgNPs, which revealed that the NPs were rectangular having sizes from 05 to 16 nm. XRD revealed that the synthesized NPs were crystalline and face-centered cubic (FCC). EDX analysis reflected the elemental composition of the synthesized product and showed that the percentage of silver = 92. FTIR was used to confirm the functional groups responsible for the reduction and stabilization of silver ions. It revealed that biomolecules present in plant extract were responsible for the reduction and stabilization of silver ions. The effect of temperature, pH, reaction time, silver nitrate concentration, and plant extract concentration on the synthesis of AgNPs were also investigated. The synthesized silver nanoparticles were tested against four bacterial strains which showed strong antibacterial activity. We conclude that biogenic silver nanoparticles could be used as antibacterial agents to treat various diseases. Effects of pH 3, 4, 6, 9, and 11, reaction time 5, 10, 20, 40 min, 1, 2, and 3 h, temperature 30, 40, 50, 60°C, silver nitrate concentration 0.5, 1.0, 2.0, and 3.0 mM and plant extract concentration 0.5, 1.0, 2.0, and 3.0 g on the AgNPs synthesis were also studied Scanning electron microscopy analysis revealed a small size of synthesized silver nanoparticles (8 nm). The synthesized silver nanoparticles were found effective against various bacterial pathogens.

Phthalates are multifunctional compounds with extensive applications and emerging environmental pollutants. Due to their ubiquity in the environment and unavoidable exposure to humans, concerns have been voiced about public health dangers. This study was aimed to explore the diethyl phthalate (DEP) toxicity and the potential protective effect of black coffee in female Swiss albino mice. Four-week-old mice, weighing 12 ± 1 g were segregated into five groups (n = 10), designated as G-I (without any treatment), G-II (treated with corn oil), G-III (exposed to 1.5 mg/g body wt. (B.W.) DEP), G-IV (received 2 µg/g B.W coffee), and G-V (co-administrated with 1.5 mg/g DEP and 2 µg/g B.W coffee). Before dose administration, the coffee extract was assessed for its antioxidant potential through FRAP, TPC, and GC-MS analyses. Respective phthalates/coffee doses were administrated orally, once a day for 8 weeks consecutively starting from the prepubescent stage. After 56 days, mice were acclimated for 4 days then dissected. Morphological assessments showed an irregular shape of the ovaries in DEP-treated mice as compared to the control. The average bodyweight of DEP-intoxicated mice (p ≤ 0.05) increased notably against control, while DEP plus coffee group showed a regular gain in the average weight of mice. The gonado-somatic index showed non-significant variations among all groups. Micrometric studies showed that the diameter of secondary follicles (115 µm) in the ovaries of DEP-exposed mice (p ≤ 0.001) decreased significantly as compared to control (204 µm); conversely, follicular diameter in the coffee control group (248) increased significantly. Serum FSH and LH levels were significantly increased in DEP-exposed mice with a noteworthy decrease in estrogen level while hormonal levels of all other groups were comparable to control. Histological sections of DEP-exposed mice ovaries showed anatomical disruptions contrary to other groups, which were comparable with control. Antioxidant potential was checked in ovaries homogenates; FRAP values showed a notable decrease in DEP group in comparison with the control group, in contrast to G-V, when DEP was co-administrated with coffee. This study concluded that black coffee has protective effect, against DEP-instigated reproductive toxicity in Swiss albino female mice.

The present study was aimed to investigate the comparative antidiabetic potential of Nigella sativa seed extract and oil in an in vivo trial using rat animal model. The levels of antioxidants analysed in this study included catalase, vitamin C and bilirubin. NS methanolic extract and its oil were tested for their hypoglycemic effect against alloxanized diabetic rabbits (120mg/kg). The crude methanolic extract and the oil (2.5ml/kg/day) were given orally for 24 days that resulted in a significant reduction in glycaemia, particularly during the first 12 days of treatment (reductions of 58.09% and 73.27%, respectively), whereas the oil treated group normalised the levels of catalase (-69.23%), vitamin C (27.30%) and bilirubin (-51.48%) and the extract treated group normalised the levels of catalase (-65.38), vitamin C (24.15%) and bilirubin (-26.19%) at the end of the trial. The results have shown that the seed oil more significantly normalized the levels of serum catalase, serum ascorbic acid and total serum bilirubin as compared to the methanolic extract of Nigella sativa, so Nigella sativa seed oil (NSO) may be used as part of antidiabetic remedies against diabetes and utilized as a nutraceutical.

BACKGROUND: Ajuga bracteosa is a traditional herb used against various diseases. OBJECTIVES: Current research aimed to investigate the anti-diabetic and hepato-protective effect of green synthesized silver nanoparticles (ABAgNPs) using Ajuga bracteosa aqueous extract (ABaqu). METHODS: In vitro anti-diabetic and cytotoxic effects were carried out via α- glucosidase inhibition, brine shrimp lethality, and protein kinase inhibition assays. For in vivo screening of 200 mg/kg and 400 mg/kg of both ABAgNPs and ABaqu in alloxan-induced and CCl4-induced Swiss albino mice were used. Liver and kidney functional markers, hematology, and histopathological studies were carried out after 14 days of administration. RESULTS: In vivo antidiabetic and anti-cancerous effects showed valuable anti-hyperglycemic and hepatoprotective potential when mice were treated with ABaqu and ABAgNPs. A significant reduction in the blood glucose level was recorded when ABaqu and ABAgNPs were administrated orally compared to Glibenclamide treated group. Significant reduction in ALT, AST, ALP, urea, uric acid, and creatinine was recorded in ABaqu and ABAgNPs treated diabetic mice. The hepato-protective findings indicated that ALT, ALP, AST were elevated in CCl4-induced mice while declined in both ABAgNPs and ABaqu treated CCl4-induced mice. Histopathological examination revealed that ABAgNPs have hepato-protective activity. CONCLUSION: It was concluded that ABAgNPs and ABaqu possessed strong anti-diabetic and hepatoprotective phytoconstituents, which could be used in the prevention of diseases.

BACKGROUND: Thalassemia is an inherited hematological disorder categorized by a decrease or absence of one or more of the globin chains synthesis. Beta-thalassemia is caused by one or more mutations in the beta-globin gene. The absence or reduced amount of beta-globin chains causes ineffective erythropoiesis which leads to anemia. METHODS: Beta-thalassemia has been further divided into three main forms: thalassemia major, intermedia, and minor/silent carrier. A more severe form among these is thalassemia major in which individuals depend upon blood transfusion for survival. The high level of iron deposition occurs due to regular blood transfusion therapy. RESULTS: Overloaded iron raises the synthesis of reactive oxygen species (ROS) that are noxious and prompting the injury to the hepatic, endocrine, and vascular system. Thalassemia can be analyzed and diagnosed via prenatal testing (genetic testing of amniotic fluid), blood smear, complete blood count, and DNA analysis (genetic testing). Treatment of thalassemia intermediate is symptomatic; however; it can also be accomplished by folic supplementation and splenectomy. CONCLUSION: Thalassemia major can be cured through regular transfusion of blood, transplantation of bone marrow, iron chelation management, hematopoietic stem cell transplantation, stimulation of fetal hemoglobin production, and gene therapy.
