This study investigated the sensitivity of two deepsea species using mortality of northern shrimp (Pandalus borealis) and polyp activity of stony coral (Lophelia pertusa) to dispersant, Corexit 9500 and aromatic hydrocarbons (toluene, 2-methylnaphthalene, phenanthrene) in 96-h tests. Resulting hydrocarbon toxicity data were fit to the Target Lipid Model to generate predictive models and determine species sensitivity. Toxicity of chemically enhanced water accommodated fractions of Alaskan North Slope crude oil (ANS-oil) was also investigated with shrimp using nominal loading, total petroleum hydrocarbons and biomimetic extraction (BE) as oil exposure metrics. Coral were more sensitive to dispersant than shrimp while similar sensitivity was observed for hydrocarbons. Study and literature findings indicate deepsea species exhibit acute sensitivities to dispersant, hydrocarbons and oil that are comparable to pelagic species. Results support use of passive sampling methods to quantify dissolved oil for interpreting oil toxicity tests and suggest models for predicting time-dependence of toxicity warrant re-evaluation.

A predominant concern following oil spills is toxicity to aquatic organisms. However, few data are available on effects in deep-sea cold water fishes. The present study had 3 major objectives. The first was to investigate the relative sensitivity of the deep-sea species Anoplopoma fimbria (sablefish) to acute effects of 3 aromatic compounds (toluene, 2-methylnaphthalene, and phenanthrene), dispersant alone, and chemically enhanced water accommodated fractions (CEWAFs) of Alaskan North Slope crude oil. The second was to determine the critical target lipid body burden (CTLBB) for sablefish by fitting aromatic hydrocarbon toxicity data to the target lipid model (TLM), which then allowed expression of CEWAF exposures in terms of dissolved oil toxic units. The final aim was to apply a passive sampling method that targets bioavailable, dissolved hydrocarbons as an alternative analytical technique for improved CEWAF exposure assessment. The results indicate that sablefish exhibit sensitivity to Corexit 9500 (96-h median lethal concentration [LC50] = 72.2 mg/L) within the range reported for other fish species. However, the acute CTLBB of 39.4 ± 2.1 µmol/goctanol lies at the lower end of the sensitivity range established for aquatic species. The utility of both toxic units and passive sampling measurements for describing observed toxicity of dispersed oil is discussed. The present study is novel in that a new test species is investigated to address the uncertainty regarding the sensitivity of deep-sea fishes, while also employing modeling and measurements to improve exposure characterization in oil toxicity tests. We have experimentally investigated the effects of repeated diesel spills on the bivalve Anomalocardia brasiliana, the gastropod Neritina virginea and the polychaete Laeonereis culveri, by monitoring the responses of oxidative stress biomarkers in a subtropical estuary. Three frequencies of exposure events were compared against two dosages of oil in a factorial experiment with asymmetrical controls. Hypotheses were tested to distinguish between (i) the overall effect of oil spills, (ii) the effect of diesel dosage via different exposure regimes, and (iii) the effect of time since last spill. Antioxidant defense responses and oxidative damage in the bivalve A. brasiliana and the polychaete L. culveri were overall significantly affected by frequent oil spills compared to undisturbed controls. The main effects of diesel spills on both species were the induction of SOD and GST activities, a significant increase in LPO levels and a decrease in GSH concentration. N. virginea was particularly tolerant to oil exposure, with the exception of a significant GSH depletion. Overall, enzymatic activities and oxidative damage in A. brasiliana and L. culveri were induced by frequent low-dosage spills compared to infrequent high-dosage spills, although the opposite pattern was observed for N. virginea antioxidant responses. Antioxidant responses in A. brasiliana and L. culveri were not affected by timing of exposure events. However, our results revealed that N. virginea might have a delayed response to acute high-dosage exposure. Experimental in situ simulations of oil exposure events with varying frequencies and intensities provide a useful tool for detecting and quantifying environmental impacts. In general, antioxidant biomarkers were induced by frequent low-dosage exposures compared to infrequent high-dosage ones. The bivalve A. brasiliana and the polychaete L. culveri are more suitable sentinels due to their greater responsiveness to oil and also to their wider geographical distribution.

This study aimed to simulate conditions in which dispersant (Dasic NS) might be used to combat an oil spill in coastal sub-Arctic water of limited depth and water exchange in order to produce input data for Net Environmental Benefit Analysis (NEBA) of Arctic and sub-Arctic coastal areas. Concentration dependent differences in acute responses and long-term effects of a 48h acute exposure to dispersed oil, with and without the application of a chemical dispersant, were assessed on the Arctic filter feeding bivalve Chlamys islandica. Icelandic scallops were exposed for 48h to a range of spiked concentrations of mechanically and chemically dispersed oil. Short-term effects were assessed in terms of lysosomal membrane stability, superoxide dismutase, catalase, gluthatione S-transferases, glutathione peroxidases, glutathione reductase, glutathione, total oxyradical scavenging capacity, lipid peroxidation and peroxisomal proliferation. Post-exposure survival, growth and reproductive investment were followed for 2 months to evaluate any long-term consequence. Generally, similar effects were observed in scallops exposed to mechanically and chemically dispersed oil. Limited short-term effects were observed after 48h, suggesting that a different timing would be required for measuring the possible onset of such effects. There was a concentration dependent increase in cumulative post-exposure mortality, but long-term effects on gonadosomatic index, somatic growth/condition factor did not differ among treatments.

In the context of an oil spill accident and the following oil spill response, much attention is given to the use of dispersants. Dispersants are used to disperse an oil slick from the sea surface into the water column generating a cloud of dispersed oil droplets. The main consequence is an increasing of the sea water-oil interface which induces an increase of the oil biodegradation. Hence, the use of dispersants can be effective in preventing oiling of sensitive coastal environments. Also, in case of an oil blowout from the seabed, subsea injection of dispersants may offer some benefits compared to containment and recovery of the oil or in situ burning operation at the sea surface. However, biological effects of dispersed oil are poorly understood for deep-sea species. Most effects studies on dispersed oil and also other oil-related compounds have been focusing on more shallow water species. This is the first approach to assess the sensitivity of a macro-benthic deep-sea organism to dispersed oil. This paper describes a toxicity test which was performed on the macro-benthic deep-sea amphipod (Eurythenes gryllus) to determine the concentration causing lethality to 50% of test individuals (LC50) after an exposure to dispersed Brut Arabian Light (BAL) oil. The LC50 (24 h) was 101 and 24 mg L(-1) after 72 h and 12 mg L(-1) at 96 h. Based on EPA scale of toxicity categories to aquatic organisms, an LC50 (96 h) of 12 mg L(-1) indicates that the dispersed oil was slightly to moderately toxic to E. gryllus. As an attempt to compare our results to others, a literature study was performed. Due to limited amount of data available for dispersed oil and amphipods, information on other crustacean species and other oil-related compounds was also collected. Only one study on dispersed oil and amphipods was found, the LC50 value in this study was similar to the LC50 value of E. gryllus in our study. Since toxicity data are important input to risk assessment and net environmental benefit analyses, and since such data are generally lacking on deep-sea species, the data set produced in this study is of interest to the industry, stakeholders, environmental management, and ecotoxicologists. However, studies including more deep-sea species covering different functional groups are needed to evaluate the sensitivity of the deep-sea compartments to dispersed oil relative to other environmental compartments.

Concentration dependent differences in acute and long-term effects of a 48 h exposure to mechanically or chemically dispersed crude oil were assessed on juvenile lumpsucker (Cyclopterus lumpus). Acute or post-exposure mortality was only observed at oil concentrations representing higher concentrations than reported after real oil spills. Acute mortality was more apparent in chemically than mechanically dispersed oil treatments whereas comparable EC50s were observed for narcosis. There was a positive correlation between EROD activity and muscle PAH concentration for the lower oil concentrations whereas higher concentrations inhibited the enzyme activity. The incidence of gill tissue lesions was low with no difference between dispersion methods or oil concentrations. A concentration dependent decrease in swimming- and feeding behavior and in SGR was observed at the start of the post-exposure period, but with no differences between corresponding oil treatments. Three weeks post-exposure, fish from all treatments showed as high SGR as the control fish.

In this study, impact of dispersed oil on cardiac mitochondrial function was assessed in a key species of Arctic marine ecosystem, the polar cod Boreogadus saida. Mature polar cod were exposed during 48 h to dispersed oil (mechanically and chemically) and dispersants alone. The increase observed in ethoxyresorufin-O-deethylase activity and polycyclic aromatic hydrocarbon metabolites in bile indicated no difference in contamination level between fish exposed to chemical or mechanical dispersion of oil. Oil induced alterations of O2 consumption of permeabilised cardiac fibres showing inhibitions of complexes I and IV of the respiratory chain. Oil did not induce any modification of mitochondrial proton leak. Dispersants did not induce alteration of mitochondrial activity and did not increase oil toxicity. These data suggest that oil exposure may limit the fitness of polar cod and consequently could lead to major disruption in the energy flow of polar ecosystem.

Due to a northward shift in oil and gas activities, there is an increasing need to understand the potential anthropogenic impacts of oil-related compounds on sub-Arctic and Arctic organisms, particularly those in coastal habitats. Capelin (Mallotus villosus), a key fish species in the Barents Sea ecosystem, undertakes aggregated spawning at both intertidal and subtidal coastal localities in northern Norway. To investigate the sensitivity of capelin embryos to oil compounds, newly fertilized capelin eggs were collected from a spawning beach and exposed until hatch (32 days) to either the water soluble fraction of crude oil or the single PAH compound, pyrene. Threshold levels for egg mortality, development and hatching success were determined. Concentrations of 40 µg/L crude oil (∑26 PAHs) and 55 µg/L pyrene significantly increased embryonic mortality rates and decreased hatching success, compared with controls, indicating that a potential oil spill in the vicinity of capelin spawning grounds may cause significant impacts. No significant incidence of adverse effects such as yolk sac oedema, pericardia oedema, haemorrhages, craniofacial abnormalities, premature hatch or inhibited growth was observed. Histological studies of hatched larvae did not reveal specific sublethal effects in tissues and organs. Developmental delays and subsequent embryo death were noticed at the period of eye pigmentation in affected groups. Early life-history stages of capelin are sensitive indicators of PAH impacts, but the mechanisms responsible for the toxic effects require further investigation.

In an international collaborative effort, an impact analysis tool is being developed to predict the effect of accidental oil spills on recruitment and production of Atlantic cod (Gadus morhua) in the Barents Sea. The tool consisted of three coupled ecological models that describe (1) plankton biomass dynamics, (2) cod larvae growth, and (3) fish stock dynamics. The discussions from a series of workshops are presented in which variables and parameters of the first two ecological models were listed that may be affected by oil-related compounds. In addition, ecotoxicological algorithms are suggested that may be used to quantify such effects and what the challenges and opportunities are for algorithm parameterization. Based on model exercises described in the literature, survival and individual growth of cod larvae, survival and reproduction of zooplankton, and phytoplankton population growth are denoted as variables and parameters from the ecological models that might be affected in case of an oil spill. Because toxicity databases mostly (67%) contain data for freshwater species in temperate environments, parameterization of the ecotoxicological algorithms describing effects on these endpoints in the subarctic marine environment is not straightforward. Therefore, it is proposed that metadata analyses be used to estimate the sensitivity of subarctic marine species from available databases. To perform such analyses and reduce associated uncertainty and variability, mechanistic models of varying complexity, possibly aided by new experimental data, are proposed. Lastly, examples are given of how seasonality in ecosystems may influence chemical effects, in particular in the subarctic environment. Food availability and length of day were identified as important characteristics as these determine nutritional status and phototoxicity, respectively.

Polar cod Boreogadus saida an indicator species for biomonitoring in the Arctic was exposed to crude oil in waterborne and dietary experiments. Ethoxyresorufin O-deethylase (EROD) activity was measured in liver and gills of polar cod at weeks 0, 2 and 4 of exposure and following 2 weeks of depuration. EROD increased significantly and dose-dependently in both tissues through both exposure routes. Levels were very low in gills compared to liver reflecting the tissue-specific metabolism capacities and tissue-specific response kinetics were also observed. Furthermore, a significant increase of gill EROD was shown in dietary exposed fish, demonstrating a substantial transport of PAHs via the systemic circulation. To conclude, this study gave some preliminary information on the EROD response in terms of levels, dose dependency and timing, in gills of PAH exposed polar cod.

In order to mimic the biological effects of an oil spill in Arctic waters, we examined several types of biomarkers (genes, enzymes, metabolites, and DNA damage) in polar cod Boreogadus saida experimentally exposed to the water soluble fractions of crude oil. During 4 weeks of exposure, induction of the studied biomarkers exceeded baseline levels. The mRNA expression of the cytochrome P4501A1 (cyp1a1) gene was the most promising biomarker, with glutathione S-transferase (gst) as a suitable complement. The delayed ethoxyresorufin O-deethylase (EROD) and GST activities and their persistence following 2 weeks of depuration may allow detection of previous exposures in field samples. The composition of PAH metabolites in the bile indicated the bioavailability of different PAH size-classes. Although mRNA expressions of antioxidant defense genes were induced at start of the exposure, with the strongest responses from catalase and cytosolic superoxide dismutase, they were poor for oil monitoring purposes due to their very short response times. Significant DNA damage demonstrated genotoxicity even at low PAH concentrations (<15microgL(-1)) and was correlated with benzo(a)pyrene and pyrene metabolites in the bile.

Polar cod Boreogadus saida were exposed weekly to two doses of dietary crude oil for 4 weeks followed by 2 weeks of depuration. Administered doses corresponded on average to 4 and 9microgSigmaPAHsg(-1)fishweek(-1). Cytochrome P4501A1 (cyp1a1) and glutathione S-transferase (gst) mRNA expression, ethoxyresorufin O-deethylase (EROD) activity and metabolites in the bile showed strong and dose-dependent inductions at 2 and 4 weeks of exposure. Following 2 weeks depuration, mRNA expression of cyp1a1 and gst and PAH metabolites returned to basal levels while EROD activity and GST activity were still induced in the high oil treatment. The mRNA expressions of antioxidant defense genes (catalase, glutathione peroxidase and cytosolic and mitochondrial superoxide dismutase) did not change significantly during the experiment. Catalase activity was significantly depressed at week 2 in the high oil treatment. We conclude that the cyp1a1 mRNA expression, EROD activities and bile metabolites were the most reliable biomarkers of exposure while gst mRNA expression and GST activity were less sensitive and are considered only as complementary. Antioxidant defenses were poor biomarkers to assess effects of crude oil exposure in polar cod.

With expanding oil and gas activities into the Arctic region, there is a need to evaluate the induction capacity of polycyclic aromatic hydrocarbon (PAH) biomarkers on Arctic marine organisms and to test analytical methods that have been optimized for their temperate counterparts. Polar cod Boreogadus saida were injected intraperitoneally with cod liver oil (solvent control), 6.6+/-3.7, 85+/-48 or 378+/-190 microg kg(-1) wet weight of benzo(a)pyrene (B(a)P), or not injected (control), and liver and bile were sampled at 0 and 16 h and 1, 2, 4 and 7d. The mRNA expression of cytochrome P4501A1 (cyp1a1) and glutathione S-transferase (gst) genes showed a dose-dependent induction in the first 16 h following the injection and a return to basal levels after 4d. The aryl hydrocarbon receptor 2, however, showed no change in mRNA expression. The protein quantification of cytochrome P4501A (CYP1A), through Western blot analysis and the enzyme-linked immunosorbent assay (ELISA), presented similar but weaker and time-delayed responses (4-7d) compared to the gene (16 h to 2d). Ethoxyresorufin O-deethylase (EROD) and glutathione S-transferase (GST) activities increased significantly at day 7 following the gene induction and increase in protein levels. Overall, these biomarkers showed dose-dependent but weak responses to B(a)P and low levels of bile metabolites. The mRNA expressions of oxidative stress genes, superoxide dismutases (sod(Cu/Zn) and sod(Mn)), catalase (cat) and glutathione peroxidase (gpx), were all up-regulated between 16 h and 2d of B(a)P exposure with cat (72-fold) and sod(Cu/Zn) (20-fold) giving the strongest responses in the highest dose. Finally, CAT protein level and enzyme activities showed less clear responses than the genes. The mRNA expression showed the earliest responses, followed by the protein levels. The enzymatic activities were the least sensitive and responded to the exposure after 7d. The study shows the induction capability of biomarkers in polar cod at very low bioavailable doses of B(a)P and provides new information on the selected biomarkers for use in oil monitoring in the Arctic.

Increasing offshore oil and gas activities in the European Arctic has raised concerns of the potential anthropogenic impact of oil-related compounds on the polar marine ecosystem. We measured cellular energy allocation (CEA) in the sea ice amphipod Gammarus wilkitzkii after exposure for one month to the water soluble fraction (WSF) of oil. The CEA biomarker measures the energy budget of organisms by biochemically assessing changes in carbohydrates, protein and lipid content as well as the electron transport system activity. A significantly higher protein content was observed in the medium dose compared to controls, while the total energy budget was not affected in G. wilkitzkii. This indicates that parts of the energy budget of G. wilkitzkii, which is a key species in the Arctic ecosystem, is affected by a WSF of oil.

Offshore oil and gas activities have gained momentum in the European Arctic, raising concerns of the potential impact of oil-related chemicals on the polar marine ecosystem, notably on sea ice communities. Herein, malformations on embryos of the Arctic sea ice amphipod Gammaruswilkitzkii exposed to the water soluble fraction of oil were studied. The females ranged from development stage three to nine. No differences in reproductive stage were observed among the different treatments after 30 days of exposure. Frequency of embryo aberrations was significantly higher in the high-dose compared to controls, indicating that the embryos of G. wilkitzkiii were affected by oil.

We studied cellular energy allocation (CEA) in three Arctic benthic species (Gammarus setosus (Amphipoda), Onisimus litoralis (Amphipoda), and Liocyma fluctuosa (Bivalvia)) exposed to oil-related compounds. The CEA biomarker measures the energy budget of organisms by biochemically assessing changes in energy available (carbohydrates, protein and lipid content) and the integrated energy consumption (electron transport system activity (ETS) as the cellular aspect of respiration). Energy budget was measured in organisms subjected to water-accommodated fraction (WAF) of crude oil and drill cuttings (DC) to evaluate whether these compounds affect the energy metabolism of the test species. We observed significantly lower CEA values and higher ETS activity in G. setosus subjected to WAF treatment compared to controls (por=0.19). Different responses to oil-related compounds between the three test species are likely the result of differences in feeding and burrowing behavior and species-specific sensitivity to petroleum-related compounds.
