RESUMO

Salvia splendens is a popular ornamental plant in China with extensive potentials, including value in traditional Chinese medicine and in environmental restoration function (Li et al. 2008). In September 2019, leaf blight disease was observed on road side plants of S. splendens in Bayi park, Nanchang city, Jiangxi province, China. The typical symptoms appeared as irregular necrotic spots or leaf blight, accompanied by extensive scorch necrosis or ultimately defoliation. Small segments cut from diseased leaves were surface sterilized in a 2% sodium hypochlorite solution for 2 min and rinsed three times with sterile distilled water. Then, the samples were placed on potato dextrose agar (PDA) plates incubated at 25°C in darkness. Pure cultures were obtained by the hyphal tip method. Morphologically, all 11 colonies were identical to each other on PDA. Two strains, YZU 191468 and YZU 191481, were selected for further study and deposited in the Fungal Herbarium of Yangtze University (YZU), Jingzhou, Hubei, China. The 7-day-old colonies were circular, 53 to 56 mm in diameter, and consisted of white mycelium with a buff margin, and were cinnamon colored in the center of the reverse side. To examine conidial morphology, the mycelium was transferred onto potato carrot agar (PCA) and incubated at 23°C with a period of 8 h light/16 h dark for 7 days. Conidia were normally solitary or two in a chain, ellipsoid or long ellipsoid, beakless, 10 to 23×30 to 60 µm in size (n=50). Based on morphology, the isolates were consistent with Stemphylium lycopersici (Yamamoto 1960). To confirm the identification, genomic DNA was extracted from both isolates and used to amplify the internal transcribed spacer rDNA region (ITS), glyceraldehydes-3-phosphate dehydrogenase (GAPDH) and calmodulin (CAL) genes with primer pairs ITS5/ITS4, gpd1/gpd2, and CALDF1/CALDR2, respectively (Woudenberg et al. 2017). Sequences were deposited in GenBank with accession numbers OP564983 and OP564984 (ITS), OP892529 and OP892530 (GAPDH), OP584970 and OP584971 (CAL). A neighbor-joining tree was constructed with Mega 7.0 based on the combined dataset with 1,000 bootstrap replicates. The resulting phylogenetic tree showed that the strains from S. splendens clustered with S. lycopersici (CBS 122639 and CBS 124980) supported with 100% bootstrap values. The molecular analyses confirmed that the species causing leaf blight symptoms was S. lycopersici. To test pathogenicity, healthy leaves of S. splendens were surface sterilized and inoculated by mycelium blocks (6 mm in diameter) and spore suspension (1×106 spore/mL) of representative strains YZU 191468 and YZU 191481, respectively. Controls were inoculated with blocks of PDA and sterile water. Each strain was inoculated on three leaves of a plant. One clean plant was used as control. The test was replicated three times. After inoculation, the plants were covered with plastic bags and incubated in a greenhouse (25℃, 80 % relative humidity, 8 h light/16 h dark). After 5 days, the inoculated leaves exhibited dark brown spots with white mycelium, followed by withering of necrotic tissues. There were no symptoms observed on the controls. The fungal isolates inoculated leaves had the same morphological characteristics as the strains used for inoculation. S. lycopersici has been found on eggplant and Zinnia elegans in China (He et al. 2019; Yang et al. 2017). To the best of our knowledge, this is the first report of S. lycopersici causing leaf blight on S. splendens in China. This finding offers a new reference for the management and control of S. splendens leaf diseases in China.

OBJECTIVE: The study aims to investigate and visualize the hotspots of acupuncture for Allergic rhinitis (AR) over the past two decades and pinpoint future trends in this field. METHOD: We conducted a systematic search of English-language articles or reviews on acupuncture for AR in the Web of Science Core Collection from 2002 to 2022. Using Citespace, VOSviewer, and Bibliometrix, we analyzed and visualized the publications, countries, institutions, authors, journals, and keywords from various angles. RESULT: The study identified 197 documents, 80 journals, 458 keywords, and 928 authors associated with acupuncture for AR. Although article publication fluctuated over the past 20 years, an overall upward trend emerged, with rapid growth during the second decade. China contributed the most to acupuncture research on AR and had the closest collaborations with the United States and Germany. China Medical University was the most prolific institution, and Benno Brinkhaus was the most productive and influential author. The most published journal was Medicine, while the Journal of Allergy and Clinical Immunology was the most frequently cited journal. The highest frequency keywords included acupuncture, allergic rhinitis, and asthma. Randomized controlled trials and alternative & complementary medicine remained significant research hotspots, while rhinoconjunctivitis is expected to be the emerging focus of future investigations. CONCLUSION: acupuncture has experienced robust development for the treatment of allergic rhinitis over the last two decades, with rhinoconjunctivitis and clinical research being the anticipated trends and frontiers of future research.

Global warming has aggravated the problem of lake eutrophication. As a typical large, eutrophic, shallow lake in China, the issue of cyanobacterial harmful algal blooms (cyanoHABs) was particularly prominent in Lake Taihu. We took Lake Taihu as the study area, using the meteorological (temperature, wind speed, rainfall, and sunshine hours), water quality (total nitrogen, total phosphorus, conductivity, pH, and chemical oxygen demand), and biological (chlorophyll-a in phytoplankton) monitoring data from 1992 to 2020. We built a simulation model of chlorophyll-a based on the Bayesian network model with continuous variables to study the chlorophyll-a level of Lake Taihu under different meteorological and water quality conditions. The 75th percentile of chlorophyll-a concentration was used as the threshold to judge the risk of cyanobacterial bloom. When the probability of chlorophyll-a concentration below this threshold was greater than 75%, it was regarded as "low risk" of cyanobacterial bloom outbreak. The results showed that the average level of "temperature wind ratio" (ratio of air temperature to wind speed) in spring was 6.67℃·s·m-1, and the probability of high chlorophyll-a was less than 75% when the total phosphorus concentration was less than 0.130 mg·L-1. The average "temperature wind ratio" level in summer was 10.52℃·s·m-1, and the probability of high chlorophyll-a was less than 75% when the total phosphorus concentration was less than 0.257 mg·L-1. The average level of total phosphorus concentration in autumn was 0.154 mg·L-1, and the probability of high chlorophyll-a was less than 75% when the "temperature wind ratio" was less than 6.30℃·s·m-1. Based on the above research, the chlorophyll-a model constructed by the Bayesian network model with continuous variables was further used to simulate the nutrient control objectives under different climate change backgrounds. In order to control chlorophyll-a in Lake Taihu at the:"low risk" level of cyanoHABs, the target concentration thresholds of total phosphorus needed to be controlled under the climate level background from 1992 to 2000, 2001 to 2010, and 2011 to 2020 were given. From 1992 to 2000, the threshold value of total phosphorus concentration was 0.135 mg·L-1 in spring, 0.174 mg·L-1 in summer, and 0.171 mg·L-1 in autumn. From 2001 to 2010, the threshold value of total phosphorus concentration was 0.115 mg·L-1 in spring, 0.164 mg·L-1 in summer, and 0.162 mg·L-1 in autumn. From 2011 to 2020, the threshold value of total phosphorus concentration was 0.059 mg·L-1 in spring, 0.145 mg·L-1 in summer, and 0.145 mg·L-1 in autumn. The results showed that the control of cyanoHABs in eutrophic lakes required more stringent nutrient control strategies with global warming. It provided a reference for preventing and controlling cyanoHABs and eutrophication in Lake Taihu. Previous studies have used multiple regression models, hydrodynamic numerical models, and other methods to predict chlorophyll-a concentrations or cyanobacterial blooms in lakes. However, there has been no study on the prediction of cyanoHABs in lakes based on the Bayesian network model with continuous variables and the "dynamic" evaluation of nutrient thresholds. Therefore, based on the seasonal meteorological, water quality, and biological monitoring data of Lake Taihu from 1992 to 2020, the chlorophyll-a model of Lake Taihu was constructed for the first time based on the Bayesian network model with continuous variables to simulate the chlorophyll-a concentration of Lake Taihu under different climate indicators and total phosphorus concentrations. The weight of its influencing factors was also analyzed, and the nutrient control objectives under different climate scenarios were "dynamically" evaluated.

ETHNOPHARMACOLOGICAL RELEVANCE: Acacetin is widely distributed in traditional Chinese medicine and traditional herbs, with strong biological activity. Perhaps there are many potential effects that have not been explored. In the field of drug discovery, Mainstream methods focus on chemical structure. Traditional medicine cannot adapt to the mainstream prediction methods due to its complex composition. AIM OF THE STUDY: Our aim is that provide a prediction method more suitable for traditional medicine by graph representation learning and transcriptome data. And use this method to predict acacetin. MATERIALS AND METHODS: Our method mainly consists of two parts. The first part is to use the method of graph representation learning to vectorize drugs as a database. The original data of this part comes from transcriptome data on Gene Expression Omnibus. The method of graph representation learning is an unsupervised learning. If there is no prior knowledge as the label data, the training effect cannot be analyzed. Therefore, we define a standard score to evaluate our results through the idea of Jaccard index. The second part is to put the target drug into our database. The potential similarity between drugs was evaluated by the Euclidean distance between vectors, and the potential efficacy of the target drug is predicted by combining the chemical-disease relationship data in the Comparative Toxicogenomics Database. The target drug in this paper uses acacetin. We compared the predicted results with existing reports, and we also experimentally verified the efficacy of improving insulin resistance in the predicted results. RESULTS: The prediction results are relatively consistent with the existing reports, which demonstrated that our method has a certain degree of predictive performance. And for the efficacy of improving insulin resistance in the predicted result, we verified it through experiments. CONCLUSIONS: We propose a method to predict the potential efficacy of drugs based on transcriptome data, using Graph representation learning, which is very suitable for traditional medicine. Through this method, we predicted the efficacy of acacetin, and the results are relatively consistent with the current reports. This provides a new idea for unsupervised learning to apply medical information.

Soil Phosphorous (P) availability is a limiting factor for plant growth and regulates biological metabolism in plantation ecosystems. The effect of variations in soil microbial P cycling potential on the availability of soil P during succession in plantation ecosystems is unclear. In this study, a metagenomics approach was used to explore variations in the composition and diversity of microbial P genes along a 45-year recovery sequence of Robinia pseudoacacia on the Loess Plateau, as well soil properties were measured. Our results showed that the diversity of P cycling genes (inorganic P solubilization and organic P mineralization genes) increased significantly after afforestation, and the community composition showed clear differences. The gcd and ppx genes were dominant in inorganic P transformation, whereas phnM gene dominated the transformation of organic P. The abundance of genes involved in inorganic P solubilization and organic P mineralization was significantly positively correlated with P availability, particularly for phnM, gcd, ppx, and phnI genes, corresponding to the phyla Gemmatimonadetes, Acidobacteria, Bacteroidetes, and Planctomycetes. The critical drivers of the microbial main genes of soil P cycling were available P (AP) and total N (TN) in soil. Overall, these findings highlight afforestation-induced increases in microbial P cycling genes enhanced soil P availability. and help to better understand how microbial growth metabolism caused by vegetation restoration in ecologically fragile areas affects the soil P cycling.

Objective: To perform a bibliometric analysis of published research on acupuncture for neurodevelopmental disorders and to provide new insights for future studies. Methods: Web of Science Core Collection was used to search for articles on acupuncture for neurodevelopmental disorders in children, from inception to Dec 4, 2022. VOSviewer and CiteSpace software were used for bibliometric analyses. VOSviewer was used to analyze and visualize the knowledge maps of the articles' countries, authors, and institutions of origin, the journals and keywords. CiteSpace was used to visualize the dual-map overlay of the journals in which the articles were published and those publishing the articles they cited. Results: A total of 119 papers were retrieved. The highest number of publications came from China, followed by the United States and South Korea. The most frequently cited article was from the United States, followed by China. The most publications were from KyungHee University, followed by Sichuan University. Author Cho, Seung-hun from KyungHee University published the most articles. The Journal of Alternative and Complementary Medicine and Medicine published the most articles. The top three most frequently used keywords were "acupuncture", "children", and "complementary". Conclusion: Research intensity and recognition, as well as collaboration within the field of acupuncture for treating neurodevelopmental disorders in children has increased. Research is generally diverse and comprehensive, and the neuro-endocrine-immune mechanism should be a new direction for further development. More basic research is also needed, to elucidate the therapeutic mechanisms, to standardize and validate the use of acupuncture for neurodevelopmental disorders.

BACKGROUND: Cardiovascular diseases are the major cause of mortality in patients with advanced chronic kidney diseases. The predominant abnormality observed among this population is cardiac dysfunction secondary to myocardial remodelings, such as hypertrophy and fibrosis, emphasizing the need to develop potent therapies that maintain cardiac function in patients with end-stage renal disease. AIMS: To identify potential compounds and their targets as treatments for cardiorenal syndrome type 4 (CRS) using molecular phenotyping and in vivo/in vitro experiments. METHODS: Gene expression was assessed using bioinformatics and verified in animal experiments using 5/6 nephrectomized mice (NPM). Based on this information, a molecular phenotyping strategy was pursued to screen potential compounds. Picrosirius red staining, wheat germ agglutinin staining, Echocardiography, immunofluorescence staining, and real-time quantitative PCR (qPCR) were utilized to evaluate the effects of compounds on CRS in vivo. Furthermore, qPCR, immunofluorescence staining and flow cytometry were applied to assess the effects of these compounds on macrophages/cardiac fibroblasts/cardiomyocytes. RNA-Seq analysis was performed to locate the targets of the selected compounds. Western blotting was performed to validate the targets and mechanisms. The reversibility of these effects was tested by overexpressing Osteopontin (OPN). RESULTS: OPN expression increased more remarkably in individuals with uremia-induced cardiac dysfunction than in other cardiomyopathies. Lobetyolin (LBT) was identified in the compound screen, and it improved cardiac dysfunction and suppressed remodeling in NPM mice. Additionally, OPN modulated the effect of LBT on cardiac dysfunction in vivo and in vitro. Further experiments revealed that LBT suppressed OPN expression via the phosphorylation of c-Jun N-terminal protein kinase (JNK) signaling pathway. CONCLUSIONS: LBT improved CRS by inhibiting OPN expression through the JNK pathway. This study is the first to describe a cardioprotective effect of LBT and provides new insights into CRS drug discovery.

ETHNOPHARMACOLOGICAL RELEVANCE: Cardiorenal syndrome type 4 (CRS type 4), with high rates of morbidity and mortality, has become a social and economic problem worldwide over the last few decades. Zhen-Wu decoction, a traditional medicine used in East Asia, has been widely used in the treatment of cardiovascular disease and kidney disease, and has shown potential therapeutic effects for the clinical treatment of CRS type 4. However, the underlying mechanism has not been extensively explored. AIM OF THE STUDY: The purpose of this study was to investigate the effect and underlying mechanism of Zhen-Wu decoction on uremic cardiomyopathy, offering a potential target for clinical treatment of CRS type 4. MATERIALS AND METHODS: Five/six nephrectomized mice were utilized for experiments in vivo. The cardioprotective effects of Zhen-Wu decoction were evaluated by echocardiography and tissue staining. RNA-Seq data were used to investigate the potential pharmacological mechanism. The prediction of targets and active components was based on our previous strategy. Subsequently, the protective effect of the selected compound was verified in experiments in vitro. RESULTS: Zhen-Wu decoction alleviated cardiac dysfunction and endothelial injury in 5/6 nephrectomized mice, and the mechanism may involve the inflammatory process and oxidative stress. The activation of the Nrf2 signaling pathway was predicted to be a potential target of Zhen-Wu decoction in protecting endothelial cells. Through our machine learning strategy, we found that lactiflorin as an ingredient in Zhen-Wu decoction, alleviates IS-induced endothelial cell injury by blocking Keap1 and activating Nrf2. CONCLUSIONS: The present study demonstrated that Zhen-Wu decoction and lactiflorin could protect endothelial cells against oxidative stress in mice after nephrectomy by activating the Nrf2 signaling pathway.

Lake Hongze is an essential storage lake for the "Eastern Route of the South-North Water Diversion Project" and plays an important role in climate adjustment and flood prevention and control. To study the structural evolution of phytoplankton communities in Lake Hongze and their relationship with environmental factors and to understand the status of the ecosystem of Lake Hongze before the "ten-year fishing ban," monthly monitoring was carried out from 2015 to 2020. During the study period, total nitrogen showed a significant downward trend starting in 2017, the total phosphorus and chemical oxygen demand declined slightly, the water temperature had no obvious change trend, and the water depth and transparency increased from 2015 to 2018 and then declined significantly. A total of 102 genera of phytoplankton in eight phyla were identified, with a total of 310 species. The dominant phytoplankton phyla mainly included Chlorophyta and Bacillariophyta, followed by Cyanophyta and Pyrrophyta. The dominant genera were Scenedesmus, Aulacoseira, Cryptomonas, Cyclotella, Tetraedron, Microcystis, and Dolichospermum. The results of non-metric multidimensional scaling (NMDS) showed that the structural composition of the phytoplankton community differed significantly among years, seasons, and sampling areas, mainly due to the redistribution among dominant genera. NMDS analysis also indicated that variation in the phytoplankton community in Lake Hongze was mainly related to water temperature, total nitrogen, total phosphorus, water depth, and transparency. Total nitrogen, water depth, and phytoplankton community structure were significantly correlated with interannual succession, whereas water temperature, total nitrogen, and phytoplankton seasonal succession were also significantly correlated. Combined with the measures taken by the management department in recent years, the changes in the structure of the phytoplankton community may be related to the removal of the fence and other management and restoration measures in Lake Hongze.

Based on the data mining technology, the rules of acupoint selection and prescription were analyzed for impotence treated with acupuncture and moxibustion in ancient recorded in Zhonghua Yidian. By taking "yangwei" and "yinwei" as the searching terms, the database of Zhonghua Yidian (the 5th edition) were retrieved and the relevant information of impotence, such as prescription provision, acupoints and use frequency were extracted. Using the software, e.g. Microsoft Excel and Weka 3.8.4, the rules of acupoint selection and prescription for impotence treated with acupuncture and moxibustion in ancient were analyzed. Fifty five provisions of acupoint prescriptions were in compliance with the requirements and screened. Of them, there were 17 compound prescriptions and the rest were the single-point prescriptions, with 24 acupoints involved. Regarding the use frequency of acupoints in treatment of impotence, the top 5 acupoints were Yingu (KI 10), Ququan (LR 8), Qichong (ST 30), Taichong (LR 3) and Rangu (KI 2). The cluster analysis found that Yingu (KI 10), Ququan (LR 8)-Qichong (ST 30), Taichong (LR 3)-Rangu (KI 2)-Xingjian (LR 2), and Mingmen (GV 4)-Zhongfeng (LR 4)-Yuji (LU 10)-Yanggu (SI 5) were formed the group prescriptions respectively. Multilayer correlation analysis discovered that the commonly used meridians were the liver meridian of foot-jueyin, the kidney meridian of foot-shaoyin, the stomach meridian of foot-yangming and the conception vessel. The acupoints selected were generally on the lower extremities, the abdomen and the upper extremities. Regarding the special points, the five-shu points and the convergent points were mostly involved. By the analysis on compound prescriptions, 3 patterns of acupoint combination were discovered, the prescription by taking Yingu (KI 10), Rangu (KI 2) and Zhongfeng (LR 4) as the key points, the one by taking Shenshu (BL 23) and Yanggu (SI 5) as the key points and the relevant fixed combination of 4 acupoints, including Taichong (LR 3), Xingjian (LR 2), Neiting (ST 44) and Xiangu (ST 43). By the analysis on the compound prescriptions, 3 common meridian combinations were found, including the combination with the kidney meridian predominated, the relevant fixed combination with the liver meridian and the stomach meridian and the one with small intestine meridian and the lung meridian.

Eutrophication of shallow lakes in the middle and lower reaches of the Yangtze River has become an increasingly serious problem. In this study, we investigated the temporal and spatial variations in nutrient loads (nitrogen, N and phosphorus, P) in the Changdang Lake Catchment located to the northwest of Lake Taihu through field sampling and laboratory analysis in 2016-2017. The results show the severity of the N and P pollution in the Changdang Lake catchment. The mean river water concentrations of TN, NO3--N, NH4+-N, TP, Chla, and permanganate index are (3.70±0.76) mg ·L-1, (1.81±0.42) mg ·L-1, (1.03±0.61) mg ·L-1, (0.38±0.31) mg ·L-1, (25.74±37.00) µg ·L-1, and (6.35±0.81) mg ·L-1, respectively. N pollution in the river is more severe in winter and spring than in summer and autumn whereas P pollution in the river is worse in autumn and winter than in spring and summer. Spatially, the magnitude of river N and P pollution follows the order of northern > northwestern > southern > eastern part of the study area. The rivers are in a state of moderate to severe eutrophication. The mean lake water concentrations of TN, NO3--N, NH4+-N, TP, Chla, and permanganate index are (2.25±0.94) mg ·L-1, (0.98±0.47) mg ·L-1, (0.19±0.14) mg ·L-1, (0.11±0.03) mg ·L-1, (18.71±8.76) µg ·L-1, and (4.59±1.09) mg ·L-1, respectively. The water quality in Changdang Lake is categorized as worse than class Ⅲ for TN and TP concentrations, which show decreasing trends from the west to the east to the south of the lake. The lake is in a status of slight to moderate eutrophication. The lake water quality is affected by the combination of sewage discharge and non-point source pollutant losses. The inflow rivers including the Danjinlicao River, Tongji River, and Xuebu River are the dominant pollution sources for Changdang Lake. The Danjinlicao River transports 10-12 times the total N and P loads transported by Tongji and Xuebu rivers. Changes in land use and atmospheric deposition are the driving factors of the deterioration of water quality and eutrophication in the catchment.

As a water storage lake for the South-to-North Water Diversion Project, it is crucial to examine changes in aquatic ecosystem structures in Lake Luoma, Jiangsu province. Field sampling was carried out in Lake Luoma monthly from 2014 to 2018 to study the relationship between the phytoplankton community structure and environmental factors. During the studied period, total nitrogen, permanganate index, and electrical conductivity in water column gradually increased, whereas fluoride content declined. The pattern of total phosphorus and dissolved oxygen was not distinct. A total of 71 genera of phytoplankton were identified from 2014 to 2018, and the average monthly biomass variation ranged from 0.16 to 5.51 mg·L-1. Bacillariophyta and Chlorophyta were the dominant phyla in the four years, followed by Pyrrophyta and Cryptophyta. The dominant genera were Synedra sp., Chroomonas spp., Aulacoseira spp., Dinobryon sp., Scenedesmus spp. , Fragilaria spp., Mougeotia sp. , Ankistrodesmus sp. , and Euglena spp. The results showed that the phytoplankton community structure significantly changed in the four years, which was mainly ascribed to the redistribution of biomass. Specifically, in addition to the dominance of Bacillariophyta and Chlorophyta, the dominance of Pyrrophyta and Cyanophyta increased during the last two years. Non-metric multidimensional scaling analysis showed that variation of the phytoplankton community in Lake Luoma was mainly related to total nitrogen, fluoride, water temperature, total phosphorus, dissolved oxygen, pH, conductivity, and permanganate index, among which the total nitrogen, water temperature, and fluoride concentration dominated the phytoplankton community change after the generalized additive model test. Water temperature is the driving factor affecting seasonal changes of the phytoplankton community. Total nitrogen and fluoride concentrations are the driving factors affecting the interannual variation in the phytoplankton community. Our study indicated that in recent years, the implementation of the ban on sand mining and demolition of the enclosed aquaculture in Lake Luoma has affected the water environment, resulting in a significant succession of the phytoplankton community.

BACKGROUND: Sanhuangshu'ai decoction (SH), a traditional Chinese medicine (TCM) prescription, has been safely used to treat diarrhea, dysentery and other inflammatory diseases with little side effect and low cost for thousands of years. However, its mechanism remains elusive. This study was designed to investigate the anti-ulcerative colitis (UC) activity of SH and mechanism by detecting its anti-inflammatory, anti-oxidative, and intervention effects of intestinal flora with the dextran sodium sulfate (DSS)-induced colitis mice. METHODS: The DSS-induced colitis mice was orally administered SH for 1 week with 0.8 or 1.6 g kg-1 d-1 dosage. A clinical disease activity score was evaluated daily. The colonic tissues of the mice were collected and prepared to detect its anti-inflammatory, anti-oxidative, intervention effects of intestinal flora and hydrogen peroxide(H2O2) in vivo, cytotoxicity and ROS influencing effects in vitro. Histological colitis severity and expression of cytokines were also determined. RESULTS: Oral administration of SH significantly prevented the development of colitis. It reduced the expression of interleukin (IL)-1, IL-6, and tumor necrosis factor (TNF)-α in the colon. Moreover, SH administration alleviated the oxidative stress in the colon of DSS-induced colitis mice, evidenced by the decrease of myeloperoxidase (MPO) activity and malondialdehyde (MDA) level, and increase of ROS level. Furthermore, SH can prevent the decrease ofLactobacillus sp. and population abundance of intestinal flora caused by DSS. CONCLUSION: SH significantly ameliorates the symptoms of DSS-induced colitis mice and the potential mechanism of SH may involve in multiple kinds of metabolic pathway including the regulation of gut microbiota, inflammatory mediators and cytokines.

BACKGROUND: Huzhentongfeng (HZTF) is an extract from four Chinese medical herbs for treating gout. This study aims to evaluate its antigout activity and preliminary explore its mechanism in vivo and in vitro. METHODS: The rats were intragastrically administered with HZTF for 5 days and then injected 0.1 ml (10 mg) of MSU crystals to their joints for generating a gout model to analyze the paw volume and histopathology of joint synovial tissues of rats with different doses. We also investigated the antioxidant capacity of HZTF in vitro using indication including lipid peroxidation, DPPH·, and ABTS+ radical-scavenging capacity; besides, we used qRT-PCR to measure the effect of HZTF on interleukin (IL)-1ß, caspase-1, NLRP3, and NQO1 expression in hydrogen peroxide-stimulated RAW264.7 macrophages and IL-1ß, IL-6, and tumor necrosis factor (TNF)-α in MSU crystal-induced THP-1 monocytes. Confocal microscopy analysis was used to observe the dimerization of ASC adapter proteins. In addition, we also established quality standard of HZTF by using the high-performance liquid chromatography (HPLC) method. RESULTS: HZTF could significantly suppress the paw swelling and neutrophil infiltration induced by MSU intra-articular injection in rats compared with the control group. HZTF also showed inhibition effects of inflammatory cytokines (IL-1ß, IL-6, and TNF-α) secretion at 25.00 and 50.00 µg/ml in MSU-induced THP-1 cells but showed no effects of IL-1ß, IL-6, and TNF-α mRNA expression in MSU-induced THP-1 cells. Furthermore, confocal microscopy analysis showed that HZTF could prevent the oligomerization of ASC. Moreover, HZTF also showed effects in cell-free and cell-base tests of antioxidant capacity. CONCLUSION: The results prove that HZTF possessed the potential preventive effect against gout arthritis, and the effect may be attributed to its preventing effect on neutrophil infiltration and proinflammatory cytokines secretion such as IL-1ß, IL-6, and TNF-α which were caused by the activation of inflammasome.

To clarify the spatial distribution and pollution status of nutrients in the surface sediments from a macrophyte-dominated zone in Lake Taihu, total nitrogen (TN), total phosphorus (TP), alkali nitrogen (AN), available phosphorus (AP), and organic matter (OM) were measured at 60 sampling sites in Xukou Bay. A pollution evaluation and source apportionment analysis of these nutrients was also carried out. The average contents of TN, TP, AN, AP, and OM in the surface sediments of Xukou Bay were 1027.5, 423.2, 46.4, 15.3, and 17096.6 mg·kg-1, respectively, with the OM content being significantly higher in the northeast zone than in other areas of the bay. The average value of STN and STP was 1.75 and 0.91, respectively, indicating that TN and TP represent moderate and mild levels of pollution, respectively. The average comprehensive pollution index (FF) was 1.57, indicated that the surface sediment was moderately polluted. The observed organic pollution index indicated that the surface sediments in Xukou Bay were generally clean. Based on C/N values, organic matter in the surface sediments is derived from the bio-deposition of non-fibrous plants and phytoplankton. Based on correlation analysis, OM and TN in the surface sediments of Xukou Bay are derived from the same source, while TP is derived from an alternative source.

Ferroptosis is an iron-dependent cell death pathway that can eradicate certain apoptosis-insensitive cancer cells. The ferroptosis-inducing molecules are tailored lipid peroxides whose efficacy is compromised in hypoxic solid tumor and lack of tumor selectivity. It has been demonstrated that ascorbate (Asc) in pharmacological concentrations can selectively kill cancer cells via accumulating hydrogen peroxide (H2O2) only in tumor extracellular fluids. It was hypothesized that Asc-induced, selective enrichment of H2O2 in tumor coupled with Fe3+ codelivery could simultaneously address the above two problems via boosting the levels of hydroxyl radicals and oxygen in the tumor site to ease peroxidation initiation and propagation, respectively. The aim of this work was to synergize the action of Asc with lipid-coated calcium phosphate (CaP) hybrid nanocarrier that can concurrently load polar Fe3+ and nonpolar RSL3, a ferroptosis inducer with the mechanism of inhibiting lipid peroxide repair enzyme (GPX4). The hybrid nanocarriers showed accelerated cargo release at acidic conditions (pH 5.0). The combinational approach (Asc plus nanocarrier) produced significantly elevated levels of hydroxyl radicals, lipid peroxides, and depleted glutathione under hypoxia, which was accompanied with the strong cytotoxicity (IC50 = 1.2 ± 0.2 µM) in the model 4 T1 cells. In the 4 T1 tumor-bearing xenograft mouse model, the intravenous nanocarrier delivery plus intraperitoneal Asc administration resulted in a superior antitumor performance in terms of tumor suppression, which did not produce supplementary adverse effects to the healthy organs. This work provides a novel approach to enhance the potency of ferroptotic nanomedicine against solid tumors without inducing additional side effects.

Biodegradation is a significant part of pollutant integrated degradation, the process rate of which is represented by the biodegradation coefficient. To investigate the biodegradation law of typical pollutants in the plain rivers network located in the upstream of the Lake Taihu, experiments were conducted in site in September 2015, one order kinetics model was used to measure the biodegradation coefficients for permanganate index, ammonia, total nitrogen and total phosphorus, and influencing factors of the biodegradation coefficients were also analyzed. The results showed that the biodegradation coefficients for permanganate index, ammonia, total nitrogen and total phosphorus were 0.008 3-0.126 4 d⁻¹, 0.002 1-0.213 8 d⁻¹, 0.002 1-0.090 5 d⁻¹ and 0.011 0- 0.152 8 d⁻¹, respectively. The influencing factors of the biodegradation coefficients for permanganate index were permanganate index and pH; those for ammonia were ammonia concentration and pH; those for total nitrogen were inorganic nitrogen concentration, total dissolved solid concentration and nitrite concentration; and those for total phosphorus were background concentration and pH. The research results were of important guiding significance for pollutants removal and ecological restoration of the plain rivers network located in the unstream of the Lake Taihu.

Artificial vegetation restoration can induce variations in accumulation and distribution of soil carbon (C), nitrogen (N) and phosphorus (P). However, little is known about variations in soil C, N and P nutrient fraction stratification following artificial vegetation in Loess Plateau China. Based on the hypothesis that re-vegetated can improve soil quality and stratification ratios (SR) can be used as an indicator to evaluate soil quality. This study measured contents and storages of soil organic carbon (SOC), total nitrogen (TN), total phosphorus (TP) and their SRs in topsoil (0-20 cm) and subsoil (20-60 cm) in three 30-year re-vegetated lands that had been converted from arable land (Robinia pseudoacacia L., Caragana Korshinskii Kom. and abandoned cropland with low interferences and few management measures) and one slope cropland (SC) as a control for three soil profiles(0-20 cm, 20-40 cm and 40-60 cm) from June 2009 to June 2013. The results showed that the contents and storages of SOC, TN and TP in re-vegetated land were significantly higher than those in the SC in both topsoil and subsoil. The storages of SOC, TN and TP in the topsoil (0-20 cm) of the re-vegetated lands increased by 16.2%-26.4%, 12.7%-28.4% and 16.5%-20.9%, respectively, and increased by smaller but significant amounts in subsoil from 2009 to 2013. The SRs for SOC, TN and TP in the re-vegetated lands were mostly >2 (either for 0-20:20-40 cm or 0-20:40-60 cm) and greater than that in the SC. The SRs showed an increasing trend with increasing restoration age. The results also showed that the land use type and soil depth were the most influential factors for the SRs and storages, and the SRs of SOC and TN had significantly positive correlations with their storages. The SRs were concluded to be a good indicator for evaluating the soil quality, which can be significantly enhanced through vegetation restoration. Moreover, vegetation restoration can significantly enhance SOC, TN and TP accumulation in both topsoil and subsoil.

It was well known that physiological indices of submerged macrophytes could reflect change of water quality. The correlation between physiological indices of submerged macrophytes and change of water quality was studied under the cooperation of in-situ monitoring and lab analysis, combined with measuring Chlorophyll and free proline (PRO) contents as well as peroxidase (POD) activities in the leaves of Potamogeton wrightii Morong and Potamogeton crispus L. under different water quality and nutrition status. The results showed: ①there were significant spatial differences among water factors and the comprehensive eutrophication index (TLI) of distribution areas of Potamogeton wrightii Morong and Potamogeton crispus L. Mesotrophic water was more suitable for the growth of Potamogeton wrightii Morong, while Potamogeton crispus L. grew well in eutrophic water. ②there were significant spatial differences among physiological indices of Potamogeton wrightii Morong and Potamogeton crispus L. in Taihu Lake. Besides, there were significant relationships between Chlorophyll contents, POD activities of two species and TLI. ③water transparency as well as nitrogen and phosphorus nutrition were important factors leading to changes in Chlorophyll contents and POD activities of Potamogeton wrightii Morong and Potamogeton crispus L.. The findings from this study indicate that physiological properties of Potamogeton wrightii Morong and Potamogeton crispus L. have a very close correlation with nutrition status and physiochemical properties of water.

OBJECTIVE: To observe the effect of Chinese drugs for strengthening Pi, harmonizing Wei, and dispersing blood stasis (CDSPHWDBS) on the expression of gastric mucosal heat shock protein 70 (HSP70) in chronic atrophic gastritis (CAG) patients. METHODS: A total of 100 CAG patients were assigned to the control group and the treatment group by random digit table, 50 in each group. Patients in the control group took Folic Acid Tablet 10 mg each time, 3 times per day. Those in the treatment group took CDSPHWDBS, 100 mL each time, once per day. The treatment course was 6 months for all. Clinical symptoms and signs, endoscopic and histopathological changes were observed before and after treatment in the two groups. The expression of gastric mucosal HSP70 in CAG patients was determined using SP immunohistochemistry. Data were collected by HPIAS-1 000 pathological graphic analysis system, and its expression semi-quantitatively analyzed. RESULTS: The total effective rate of clinical Chinese medical symptoms and signs was 88. 0% (44/50 cases) in the treatment group and 56. 0% (28/50 cases) in the control group, with significant difference between the two groups (P <0. 01). The improvement rate of endoscopic manifestations such as congestion and edema, erosion, bile regurgitation, pale gastric mucosa, exposed blood vessels, particles proliferation in the treatment group were superior to those in the control group (P <0. 05). The total effective rate of atrophy was 80. 0% (40/50 cases) in the treatment group and 54. 0% (27/50 cases) in the control group, with significant difference between the two groups (P<0. 01). The effective rate of intestinal metaplasia was 75. 0% (12/16 cases) in the treatment group and 33.3% (5/15 cases) in the control group, with significant difference between the two groups (P < 0. 05). The optical density value of gastric mucosal HSP70 was significantly elevated in the two groups after treatment (both P <0. 05). It was higher in the treatment group than in the control group after treatment with significant difference (P <0. 01). CONCLUSION: CDSPHWDBS had obvious effect in treatment of CAG and could improve pathological changes of precancerous lesions possibly by promoting the expression of gastric mucosal HSP70 in CAG patients.
