RESUMO

Raffinose family oligosaccharides (RFOs) are very important for plant growth, development, and abiotic stress tolerance. Galactinol synthase (GolS) and raffinose synthase (RFS) are critical enzymes involved in RFO biosynthesis. However, the whole-genome identification and stress responses of their coding genes in potato remain unexplored. In this study, four StGolS and nine StRFS genes were identified and classified into three and five subgroups, respectively. Remarkably, a total of two StGolS and four StRFS genes in potato were identified to form collinear pairs with those in both Arabidopsis and tomato, respectively. Subsequent analysis revealed that StGolS4 exhibited significantly high expression levels in transport-related tissues, PEG-6000, and ABA treatments, with remarkable upregulation under salt stress. Additionally, StRFS5 showed similar responses to StGolS4, but StRFS4 and StRFS8 gene expression increased significantly under salt treatment and decreased in PEG-6000 and ABA treatments. Overall, these results lay a foundation for further research on the functional characteristics and molecular mechanisms of these two gene families in response to ABA, salt, and drought stresses, and provide a theoretical foundation and new gene resources for the abiotic-stress-tolerant breeding of potato.

BACKGROUND: The incidence of acute mastitis (AM) in lactating women has been increasing year by year. If there is no timely and appropriate treatment, AM may develop into mammary abscess and septicemia. This special situation has aroused social attention. Chinese massage has been widely used in the treatment of AM in recent years, but there is no systematic review of the effect of Chinese massage on AM. We plan to explore the efficacy and safety of Chinese massage in the treatment of AM. METHODS: We will use a computer to search the following 8 electronic databases (PubMed, Web of Science, Cochrane, Embase, Sinomed, China National Knowledge Infrastructure, Chongqing VIP Information, WanFang Data) on November 30, 2021. Randomized controlled trials (RCT) of Chinese massage therapy for AM were screened. Primary outcome measure: overall clinical response rate, breast pain score. Secondary outcome measures: milk secretion, temperature, mass size and time to resolution, White blood cell count, C-reactive protein, and the incidence of adverse reactions. According to the inclusion criteria and exclusion criteria, the included literature will be independently evaluated by two researchers using the RCT bias risk assessment tool in the Cochrane evaluation manual Handbook5.4, and meta-analysis will be performed by RevMan5.4 software. Funnel plots were used to analyze whether the study had publication bias. RESULTS: We will evaluate the clinical effect of Chinese massage therapy on AM based on RCTs. CONCLUSION: This study will provide evidence-based evidence for the effectiveness and safety of Chinese massage in the treatment of AM. PROTOCOL REGISTRATION NUMBER: INPLASY2021120019.

Aflatoxin B1 (AFB1), a mycotoxin found in food and feed, is immunotoxic to animals and poses significant threat to the food industry and animal production. The primary target of AFB1 is the liver. To overcome aflatoxin toxicity, probiotic-mediated detoxification has been proposed. In the present study, to investigate the protective effects and molecular mechanisms of Lactobacillus bulgaricus or Lactobacillus rhamnosus against liver inflammatory responses to AFB1, mice were administered with AFB1 (300 µg/kg) and/or Lactobacillus intragastrically for 8 weeks. AML12 cells were cultured and treated with AFB1, BAY 11-7082 (an NF-κB inhibitor), and different concentrations of L. bulgaricus or L. rhamnosus. The body weight, liver index, histopathological changes, biochemical indices, cytokines, cytotoxicity, and activation of the NF-κB signaling pathway were measured. AFB1 exposure caused changes in liver histopathology and biochemical functions, altered inflammatory response, and activated the NF-κB pathway. Supplementation of L. bulgaricus or L. rhamnosus significantly prevented AFB1-induced liver injury and alleviated histopathological changes and inflammatory response by decreasing NF-κB p65 expression. The results of in vitro experiments revealed that L.rhamnosus evidently protected against AFB1-induced inflammatory response and decreased NF-κB p65 expression when compared with L. bulgaricus. These findings indicated that AFB1 exposure can cause inflammatory response by inducing hepatic injury, and supplementation of L. bulgaricus or L. rhamnosus can produce significant protective effect against AFB1-induced liver damage and inflammatory response by regulating the activation of the NF-κB signaling pathway.

BACKGROUND: A laboratory-silo study was conducted to evaluate the fermentation quality, feed-nutritive value and aerobic stability of sweet sorghum silage with or without oil-extracted microalgae supplementation. Sweet sorghum was mixed with four microalgae levels (0%, 1%, 2% and 3% on a dry matter basis; control, M1, M2 and M3, respectively) and ensiled for 45 d. Further, the four experimental silages were subjected to an aerobic stability test lasting 7 d. RESULTS: All the silages except M3 silage had good fermentative characteristics with low pH and ammonia nitrogen concentrations, and high lactic acid concentrations and favorable microbial parameters. Meanwhile, oil-extracted microalgae supplementation improved the feed-nutritional value of sweet sorghum silage. Fibre (neutral detergent fibre, acid detergent fibre, acid detergent lignin and cellulose) concentrations decreased, while dry matter and crude protein levels markedly increased (P < 0.05). Compared with the control (69.7 h), treatments M2 and M3 improved the aerobic stability of sweet sorghum silage by 43.8% and more than 143% respectively, and decreased the clostridia spore counts during the stage of air exposure. CONCLUSION: Sweet sorghum silage produced with 2% oil-extracted microalgae addition was the most suitable for animal use due to the optimal balance of fermentation quality, feed-nutritional value and aerobic stability, which merits further in vivo studies using grazing ruminants. © 2018 Society of Chemical Industry.
