RESUMO

Mechanical allodynia (MA) represents one prevalent symptom of chronic pain. Previously we and others have identified spinal and brain circuits that transmit or modulate the initial establishment of MA. However, brain-derived descending pathways that control the laterality and duration of MA are still poorly understood. Here we report that the contralateral brain-to-spinal circuits, from Oprm1 neurons in the lateral parabrachial nucleus (lPBNOprm1), via Pdyn neurons in the dorsal medial regions of hypothalamus (dmHPdyn), to the spinal dorsal horn (SDH), act to prevent nerve injury from inducing contralateral MA and reduce the duration of bilateral MA induced by capsaicin. Ablating/silencing dmH-projecting lPBNOprm1 neurons or SDH-projecting dmHPdyn neurons, deleting Dyn peptide from dmH, or blocking spinal κ-opioid receptors all led to long-lasting bilateral MA. Conversely, activation of dmHPdyn neurons or their axonal terminals in SDH can suppress sustained bilateral MA induced by lPBN lesion.

RESUMO

Bulbus Fritillariae (BF) is a kind of herbal medicine widely used in many countries including China, Japan, Korea, and so on. Among the known BF medicinal materials, Bulbus Fritillariae cirrhosae (BF cirrhosae) was reported to have the best curative effect. Due to the limited resources of BF cirrhosae, a lot of adulterants have emerged in the market, impairing the market order, resource development, and above all, clinical efficacy. Here, a novel nucleic acid amplification technique, Recombinase Assisted Loop-mediated isothermal DNA Amplification (RALA), was used to establish a real-time fluorescence isothermal molecular authentication method for five commonly used BF drugs. Moreover, this RALA-based assay can also be developed as a colorimetric detection method for on-site detection. Both real-time fluorescence and visual methods could detect as low as 0.1% genuine targets in the mixed samples. In summary, we report an isothermal detection system for five kinds of BF circulating in the market, providing a new choice for the molecular identification of BF drugs and showing promise in the laboratory testing as well as field identification of other herbal medicines.

The AcOEt extract of Artemisia argyi-derived fungus Trichoderma koningiopsis QA-3 showed potent inhibitory activity against pathogenic bacteria. Fractionation of the extract resulted in the isolation of three new polyketides (1-3) and two new terpenoids (4 and 5), together with three known metabolites (6-8). Their chemical structures were analyzed by NMR spectra, ECD, HR-ESI-MS or HR-EI-MS, optical rotation, and X-ray crystallographic data, as well as by comparison with literature reports. In the antibacterial assays, 3-hydroxyharziandione (4) showed potent activity against human pathogen Escherichia coli with an MIC value of 0.5 µg/mL, while 6-(3-hydroxypent-1-en-1-yl)-2H-pyran-2-one exhibited strong activity against marine-derived aquatic pathogen Micrococcus luteus with an MIC value of 1.0 µg/mL.

In the search for antifungal lead compounds from natural resources, Notopterygium incisum, a medicine plant only distributed in China, showed antifungal potential against apple fruit pathogens. Based on the bioassay-guided isolation, chromatography fraction 6 of the ethyl acetate partition exhibited significant in vitro and in vivo antifungal activities against apple fruit pathogens. Furthermore, nine antifungal secondary metabolites, including five linear furocoumarins (1-5), two phenylethyl esters (6-7), one falcarindiol (8), and one sesquiterpenoid (9), were isolated and elucidated from fraction 6. Compound 5 is a new metabolite, and 9 isolated from the genus Notopterygium for the first time. The purified compounds (1-9) were firstly reported to exhibit antifungal activities against apple fruit pathogens of Colletotrichum gloeosporioides and Botryosphaeria dothidea with the MIC values ranging from 8 to 250 mg L-1, especially 8 of 16 and 8 mg L-1, respectively. Moreover, 8 could inhibit the spore germination and new sporulation of B. dothidea, as well as enhance the membrane permeabilization of B. dothidea spores. This was the first investigation for the antifungal components against apple fruit pathogens from Notopterygium incisum, which has great potential to be developed into bio-fungicides.

RESUMO

Qidong ( qi change) in Qidongzeyou ((), pulse change indicates the abnormal changes inside body) in the Book of Pulse (Maishu), the Bamboo Slips of Han Dynasty in Zhangjiashan is related to the classical thought on qi and the theory on meridians at early era. The connotation of qidong was explored in views of interdisciplinary perspectives, such as Chinese medicine, philology and qi philosophy in the ancient time. It refers to the pulse changes as mentioned in ancient medical books, indicating the abnormal changes in the body. Before the ancient medical canonization marked as sphygmology, the ancient medical scholar focused on qi monism. Based on the thinking mode as analogy and detecting the root from the phenomenon, it is believed that qidong refers to the imbalance of qi activity inside the body, manifested with the abnormal pulse change at relevant pulse region on the body surface. Accordingly, the disorders inside the body are detected by palpation.

Purpose: The prognosis for patients with refractory soft-tissue sarcoma (STS) is dismal. Anlotinib has previously shown antitumor activity on STS in preclinical and phase I studies.Patients and Methods: Patients 18 years and older, progressing after anthracycline-based chemotherapy, naïve from angiogenesis inhibitors, with at least one measurable lesion according to RECIST 1.1, were enrolled. The main subtypes eligible were undifferentiated pleomorphic sarcoma (UPS), liposarcoma (LPS), leiomyosarcoma (LMS), synovial sarcoma (SS), fibrosarcoma (FS), alveolar soft-part sarcoma (ASPS), and clear cell sarcoma (CCS). Participants were treated with anlotinib. The primary endpoint was progression-free rate at 12 weeks (PFR12 weeks).Results: A total of 166 patients were included in the final analysis. Overall, the PFR12 weeks was 68%, and objective response rate was 13% (95% confidence interval, 7.6%-18%). The median progression-free survival (PFS) and overall survival (OS) were 5.6 and 12 months, respectively. The PFR12 weeks, median PFS and OS were: 58%, 4.1 and 11 months for UPS (n = 19); 63%, 5.6 and 13 months for LPS (n = 13); 75%, 11 and 15 months for LMS (n = 26); 75%, 7.7 and 12 months for SS (n = 47); 81%, 5.6 and 12 months for FS (n = 18); 77%, 21 and not reached for ASPS (n = 13); 54%, 11 and 16 months for CCS (n = 7); and 44%, 2.8 and 8.8 months for other sarcoma (n = 23), respectively. The most common clinically significant grade 3 or higher adverse events were hypertension (4.8%), triglyceride elevation (3.6%), and pneumothorax (2.4%). No treatment-related death occurred.Conclusions: Anlotinib showed antitumor activity in several STS entities. The toxicity was manageable. Clin Cancer Res; 24(21); 5233-8. ©2018 AACR.

What's on the label is not what's in the bottle, from food products to herbal medicinal products (HMPs), economically-motivated biomaterials adulteration is a long-term problem affecting the food and drug industry. Accurate identification of the biomaterial ingredients in processed commodities is highly desirable. In this field, DNA-based techniques have proved to be powerful tools to overcome qualitative challenges. However, is it possible to quantify the weight of biological materials with PCR? Therefore, a basic scientific question needs to be answered: what's the relationship between DNA content and the mass of biological materials? Is DNA content directly proportional to the mass of biological materials as most of the researchers previously thought? In this study, we firstly found that there exists a linear relation between DNA contents and the weight of biomaterials indeed when the analytical practices are fully controlled. In this case, the mass of targeted biomaterials in the highly processed commercial products can also be calculated by quantifying the species-specific DNA through classic real-time PCR with a good reproducibility.

Dilated cardiomyopathy (DCM) is one of the main causes of heart failure and could increase death, hospitalization, and rehospitalization rate. The effect of conventional medicine treatment (CMT) is limited; meanwhile, the combination of CMT and Oral Chinese Herbal Medicine (OCHM) represents exciting adjunctive therapies. In this study, we ascertained the therapeutic effect of OCHM in combination with CMT for dilated cardiomyopathy by using meta-analysis methods for controlled clinical trials. We searched studies from five databases and extracted data from these studies. We also assessed the methodological quality of the included studies. We evaluated the following outcome measures to estimate the prognosis in patients with DCM: left ventricular ejection fraction (LVEF), left ventricular end-diastolic dimension (LVEDD), stroke volume (SV), brain natriuretic peptide (BNP), 6-minute walk test (6MWT), and overall efficacy. The result showed that OCHM combined with CMT for the improvement of therapeutic effect in DCM patients. However, the evidence remains weak due to the small sample size, high clinical heterogeneity, and poor methodological quality of the included trials. Further, large sample size and well-designed trials are needed.

The 44-item and 10-item Big Five Inventory (BFI) personality scales are widely used, but there is a lack of psychometric data for Chinese versions. Eight surveys (total N = 2,496, aged 18-82), assessed a Chinese-language BFI-44 and/or an independently translated Chinese-language BFI-10. Most BFI-44 items loaded strongly or predominantly on the expected dimension, and values of Cronbach's alpha ranged .698-.807. Test-retest coefficients ranged .694-.770 (BFI-44), and .515-.873 (BFI-10). The BFI-44 and BFI-10 showed good convergent and discriminant correlations, and expected associations with gender (females higher for agreeableness and neuroticism), and age (older age associated with more conscientiousness and agreeableness, and also less neuroticism and openness). Additionally, predicted correlations were found with chronotype (morningness positive with conscientiousness), mindfulness (negative with neuroticism, positive with conscientiousness), and mind wandering/daydreaming frequency (negative with conscientiousness, positive with neuroticism). Exploratory analysis found that the Self-discipline facet of conscientiousness positively correlated with morningness and mindfulness, and negatively correlated with mind wandering/daydreaming frequency. Furthermore, Self-discipline was found to be a mediator in the relationships between chronotype and mindfulness, and chronotype and mind wandering/daydreaming frequency. Overall, the results support the utility of the BFI-44 and BFI-10 for Chinese-language big five personality research.

The purpose of the present study is to confirm the protective effect of berberine (BBR) on gastrointestinal injury caused by acute heavy alcohol exposure, an effect that has not been reported previously. Our research details how BBR protects against gastrointestinal injuries from acute alcohol exposure using both in vivo and in vitro experiments. Acute high alcohol concentrations lead to obvious damage to the gastrointestinal mucosa, resulting in necrosis of the intestinal mucosa. Oral administration of BBR was able to significantly reduce this alcohol-induced damage, inhibit increases of alcohol-induced TNFα and IL-1ß expression in gastrointestinal mucosa as well as their upstream signals TLR2 and TLR4, and regulate cytokines that modulate tight junctions. Alcohol consumption is a popular human social behavior worldwide, and the present study reports a comprehensive mechanism by which BBR protects against gastrointestinal injuries from alcohol stress, providing people with a novel application of BBR.

The bamboo slips Fan yin (Anti-excessiveness) of the Western Han Dynasty, written in the form of conversation in 7-syllable prose-verse between hun (ethereal soul) and po (corporeal soul), with po as the host, and hun as the guest, reflecting the dualism of hun and po. In Huang di nei jing (Inner Canon of Yellow Emperor), appeared nearly at the same time, in which the hypothesis of all the five viscera embodies the spirit and its soul defined at the level of spirit-emotion, claiming that the physique and ethereal soul are born at the same time, and the spirit of Yang qi within the corporeal soul is the ethereal soul, which is different from the description in the Fan yin.

The aim of this study was to explore the molecular mechanisms of jolkinolide B (JB), which is extracted from the root of Euphorbia fischeriana Steud. In this study, we found that JB, a diterpenoid from the traditional Chinese medicinal herb, strongly inhibited the PI3K/Akt/mTOR signaling pathway. Furthermore, we evaluated the effects of JB on the proliferation and apoptosis of MCF-7 human breast cancer cells. Our results showed significant induction of apoptosis in MCF-7 cells incubated with JB. The viability of the MCF-7 cells was assessed by MTT assay. Flow cytometry was used to detect apoptosis and cell cycle analysis. Transmission electron microscopy (TEM) analysis was used to observe cell morphology. MCF-7 cells were subcutaneously inoculated into nude mice to study the in vivo antitumor effects of JB. The growth of MCF-7 cells was inhibited and arrested in the S phase by JB. The data showed significantly decreased tumor volume and weight in nude mice inoculated with MCF-7 cells. In addition, treatment with JB was able to induce downregulation of cyclinD1, cyclinE, mTOR, p-PI3K and p-Akt, and upregulation of PTEN and p-eIF4E. Collectively, JB-induced apoptosis of MCF-7 cells occurs through the PI3K/Akt/mTOR signaling pathway. Furthermore, the PI3K/Akt signaling cascade plays a role in the induction of apoptosis in JB-treated cells. These observations suggest that JB may have therapeutic applications in the treatment of cancer.

OBJECTIVE: To evaluate the sensitization effect of polysorbate 80 from different factories on Beagle dogs. METHOD: Beagles dogs were randomly divided into 5 groups, 3 in each group, received respectively the intravenous infusion of polysorbate 80 made by four factories in the concentration of 0.5%, with the constant infusing speed of 5 mL x min(-1) and volume of 10 mL x kg(-1). Changes were observed before infusion and in the 24 h after infusion, time of symptom appearance and disappearance was recorded, and the grade of response was determined. Moreover, blood pressure and heart rates were tested by the machine of Bp-98E, blood samples of animals were collected before infusion and at 10 min after ending infusion for measuring histamine content in plasma using ELISA. Then the sensitization effect was comprehensively estimated by combined consideration of the responding grade and histamine level. RESULT: Typical symptoms of anaphylactoid reactions were found in sample 3 group, atypical symptoms were found in other polysorbate 80 groups. Different degrees of heart rate speeding and blood pressure downing were found in polysorbate 80 groups. No over 1-fold increase of plasma histamine level was found in all groups. The atypical anaphylactoid reactions and blood pressure of sample 2 group was lighter than other polysorbate 80 groups. Estimation showed that the sample 3 induced the suspicious anaphylactoid reactions, other test solutions did not induce the typical anaphylactoid reactions on Beagle dogs. CONCLUSION: For allergies and other special populations, there is still a certain risk to applicate polysorbate 80 in the concentration of 0.5%. Production process of polysorbate 80 have a certain influence on allergenicity, refined polysorbate 80 increase the security, but further reasearchs are needed to confirmed.

From April 14 to October 10, 2009, the dynamic changes of Robinia pseudoacacia sap flow in the hilly-gully region of Loess Plateau were measured with thermal dissipation probe. Meantime, the photosynthetic active radiation (PAR), air temperature, relative air humidity, water vapor pressure deficit (VPD), wind speed, and rainfall were monitored. The diurnal variation of the sap flow velocity (SFV) displayed a single-peak curve, but the pattern varied among different months. In April, the sap flow started to increase at about 12:00, and reached the peak at about 18:00. From May to August, the sap flow started to increase at 5:30-7:30, and reached the peak at about 15:00. In September and October, the sap flow started to increase at about 8:00, and reached the peak at 11:30-13:00. The monthly average SFV was the highest in August and the lowest in April, with the value being 0.002610 cm x s(-1) and 0.000549 cm x s(-1), respectively. During the monitoring period, the sap flow velocity was significantly correlated with PAR, air temperature, VPD, wind speed, and air relative humidity, and the correlation coefficients declined in the order of air temperature > VPD > PAR > relative humidity > wind speed. The sap flow velocity could be estimated by the linear equation with variables PAR and VPD, and the regression coefficients were highly significant.

OBJECTIVE: To prepare and characterize Herba pogostemonis, Rhizoma atractylodis and Radix aucklandiae compound essential oil-beta-cyclodextrin complex. METHODS: The essential oil-p3-cyclodextrin complex was prepared by colloidal mill method. TLC, XRD, DSC and TGA analysis were applied to characterize it. RESULTS: The complex inclusion rate of essential oil-beta-CD complex was 82.4%, and the complex recovery was 80.7%. The formation of essential oil-beta-CD complex was proved by the results of TLC, XRD, DSC, TGA analysis. CONCLUSION: The stability of compound essential oil is improved by the preparation of oil beta-cyclodextrin complex.

OBJECTIVE: To explore the significance of application of Dannang Recipe No. 2 (DNR2), a Chinese herbal preparation, in the perioperative period of laparoscopic cholecystectomy (LC). METHODS: Three hundred and sixty patients with LC were randomly assigned to two groups, 180 in each group. The treatment group was treated with DNR2 in the perioperative period, one dose of the recipe composed of aucklandia root 10 g, red peony root 15 g, giant knotweed rhizome 10 g, scutellaria root 10 g, honeysuckle flower 15 g, forsythia fruit 10 g, rhubarb 9 g, immature bitter orange 10 g, magnolia bark 10 g, peach kernel 10 g, red sage root 20 g and licorice root 4 g, which was boiled with water and taken one dose per day, starting from the previous night of operation. The control group was treated by antibiotic with Ceftizoxime sodium 2.0 g or Levofloxacin 200 mg via intravenous dripping once 0.5 h before operation preventively and 1-3 days after operation according to patients' condition. The indexes, including recovery time of borborygmus, gas elimination and defecation, post-operation body temperature, days needing fluid transfusion, hospitalization time, incidence of infectious complication, as well as the white blood cell counting (WBC) and serum C-reactive protein (CRP) before and after operation, were analyzed and compared between groups. RESULTS: The recovery time of borborygmus, gas elimination and defecation were shorter in the treatment group as compared with that in the control group (10.42 +/- 4.38 h vs. 17.11 +/- 6.25 h, 15.60 +/- 5.03 h vs. 32.74 +/- 9.43 h and 38.81 +/- 9.87 h vs. 56.09 +/- 11.00 h, respectively) and all the other indexes were better in the treatment group than those in the control group, showing significant difference (P < 0.01 or P < 0.05), except the incidence of postoperative infectious complication, it was similar in the two groups. CONCLUSION: Application of DNR2 in perioperative stage of laparoscopic cholecystectomy can effectively promote the recovery of postoperative gastrointestinal motility and suppress the occurrence of acute inflammation.

Water is a key factor influencing the yield and quality of crops. Plants mainly adapt to water deficits by biochemical changes and osmotic adjustment (OA). Research on drought tolerance of field crops has been done intensively, but there is little work to be done in medical plants. Traditional Chinese medicine (TCM) has a long history of several thousand years. TCM is playing an important role in daily life in China and applied widely in clinical experience on the globe. More and more wild medical plants are cultivated and introduced. It is known that ecological and environmental conditions are vital to cultivation and efficient component accumulation of medical plants. This study is concerned about biochemical changes of three genotypes of Radix Astragali during water deficient periods and we evaluated the relative ability of their drought tolerance on the above basis. We analyzed the effect of soil water deficits on antioxidant enzymes activity and osmoregulation substances in R. Astragali leaves of three genotypes collected on day 0, 5, 10, 15, 20 and 25 after onset of water deprivation. Under water deficient conditions, biochemical changes include protecting enzyme system, for instance superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD). Osmoregulation matters include proline (Pro) and soluble sugar. Antioxidant enzyme activities and Pro, and soluble sugar content correlated between water deficient degree and time course. Antioxidant enzyme activities increased before 20 days, then decreased at the end of experiment. Proline content increased gradually, and soluble sugar content reached the highest on day 20. The order of the ability of drought tolerance in three genotypes of R. Astragali is Mongolia>Wild>Hebei by using index of drought tolerance. The research results are instructive for cultivation and introduction of R. Astragali under different conditions of water status.

OBJECTIVE: To observe the effects of Relinqing granules (powder of Polygonum capitatum extract) on the bacterial pyelonephritis model in rats. METHOD: The rat bacterial pyelonephritis model was induced by injecting the escherichia coli ATCC-25922 into kidney parenchyma. The rats were divided ramdamly into Relinqing groups(52.32, 26.16 g x kg(-1)), norflorin group (0.03 g x kg(-1)), model group and normal control group, and were given experimental drugs by gastrogavage. The contents of leucocytes (WBC), occult bloo (BLD), glucose (GLU), protein (PRO), ketones, bilirubin and urobilinagen in urine were determined. RESULT: As compared with the model group, Relinqing granules 6.0 g x kg(-1) (crude drug 52.32 g x kg(-1)) could decrease significantly the contents of WBC and BLD in urine and, however, had no markedly effects on the other biochemical parameters of urine. CONCLUSION: Relinqqing granule has significant effects of decreasing urine WBC and BLD on the bacterial pyolonephritis in rats.
