OBJECTIVE: To evaluate the association between serum phosphorus concentration and the outcome of postpartum downer cows. ANIMALS: Postpartum downer dairy cows presented over a 22-year period. PROCEDURES: In this cross-sectional study (1994 to 2016), medical records of all postpartum downer cows presented to a referral large animal hospital were reviewed. The association between serum inorganic phosphorus concentration and survival was assessed using a multivariable logistic regression. RESULTS: 907 postpartum downer dairy cows were included and classified as hypophosphatemic (mild: > 2.25 to < 3.25 mg/dL, moderate: > 1.50 to ≤ 2.25 mg/dL, and severe: ≤ 1.50 mg/dL), normophosphatemic (≥ 3.25 and ≤ 8.76 mg/dL) or hyperphosphatemic (> 8.76 mg/dL). Hypophosphatemia was observed in 19.4% of the cows (n = 176). Of those, 54.5% (n = 96) were also hypocalcemic. Overall, 58.4% cows (n = 530) survived after hospitalization. Hypophosphatemia was not significantly associated with the outcome of postpartum downer cows (mild: OR = 1.0, 95% CI: 0.6 to 1.8; moderate: OR = 0.5, 95% CI: 0.2 to 1.1; severe: OR = 1.0, 95% CI: 0.4 to 2.4). CLINICAL RELEVANCE: Low serum phosphorus concentration was frequently observed with hypocalcemia and was not associated with the outcome of postpartum downer cows.

Paratuberculosis, a chronic disease affecting ruminant livestock, is caused by Mycobacterium avium subsp. paratuberculosis (MAP). It has direct and indirect economic costs, impacts animal welfare and arouses public health concerns. In a survey of 48 countries we found paratuberculosis to be very common in livestock. In about half the countries more than 20% of herds and flocks were infected with MAP. Most countries had large ruminant populations (millions), several types of farmed ruminants, multiple husbandry systems and tens of thousands of individual farms, creating challenges for disease control. In addition, numerous species of free-living wildlife were infected. Paratuberculosis was notifiable in most countries, but formal control programs were present in only 22 countries. Generally, these were the more highly developed countries with advanced veterinary services. Of the countries without a formal control program for paratuberculosis, 76% were in South and Central America, Asia and Africa while 20% were in Europe. Control programs were justified most commonly on animal health grounds, but protecting market access and public health were other factors. Prevalence reduction was the major objective in most countries, but Norway and Sweden aimed to eradicate the disease, so surveillance and response were their major objectives. Government funding was involved in about two thirds of countries, but operations tended to be funded by farmers and their organizations and not by government alone. The majority of countries (60%) had voluntary control programs. Generally, programs were supported by incentives for joining, financial compensation and/or penalties for non-participation. Performance indicators, structure, leadership, practices and tools used in control programs are also presented. Securing funding for long-term control activities was a widespread problem. Control programs were reported to be successful in 16 (73%) of the 22 countries. Recommendations are made for future control programs, including a primary goal of establishing an international code for paratuberculosis, leading to universal acknowledgment of the principles and methods of control in relation to endemic and transboundary disease. An holistic approach across all ruminant livestock industries and long-term commitment is required for control of paratuberculosis.

The non-neuronal, immunological effects of the cholinergic signaling are exerted on the system's scale of observation via the vagus nerve and on the cellular scale via α7 nicotinic acetylcholine receptor (nAChR) signaling in myeloid cells of the periphery or brain's microglia and astrocytes. The developmental effects of such multi-scale signaling can be conceived of as an example of psychoneuroimmunological (PNI) homeokinesis and, while reported in the literature, are not yet systematically well studied. To be better understood, the intricacy of the multi-scale interactions requires relevant preclinical animal models. Chronically instrumented non-anesthetized fetal sheep model comes with a strong track record of bench-to-bed translation and a large body of evidence for its strong resemblance to and relevance for human physiology on various scales of organization. Recently, there has been growing interest in pleiotropic effects of vagus nerve stimulation (VNS) on various organ systems such as innate immunity, metabolism, and emotion with implications for programming of PNI phenotype. Here we describe the procedures required to record and manipulate the vagus nerve activity in this large pregnant mammalian organism. Extending this in vivo model to in vitro, on the cellular scale, we present the method to manipulate the cholinergic signaling in ovine fetal microglia and astrocytes and analyze their responses on protein and RNA levels. Together these models can provide multi-scale-level mechanistic insights into the effects of cholinergic signaling on PNI phenotype.

The objective of this study was to determine if precolostral blood samples are useful to detect apparent fetal infections with bovine viral diarrhea (BVD) and infectious bovine rhinotracheitis (IBR) viruses. A convenience sample of 317 sera from 50 Canadian herds was used in the study. Antibody level was measured using 2 commercial IBR and BVD ELISA kits. Precolostral status of sera was confirmed on 304 samples using serum gamma-glutamyl transferase activity. Postcolostral serum samples yielded a higher proportion of positive results to IBR (OR = 86; 95% CI: 17.8 to 415.7) and BVD (OR = 199.3; 95% CI: 41.7 to 952.3) than did precolostral samples. All positive precolostral serum samples (n = 7 of 304) originated from calves born to vaccinated cows. Postcolostral positive serum samples (n = 11 of 13) originated mostly (60%) from calves born to non-vaccinated cows. Precolostral serum sampling can detect apparent fetal infections in a herd.


Utilisation du serum précolostral pour le dépistage de la diarrhée virale bovine (BVD) et rhinotracheite infectieuse bovine (IBR) dans les troupeaux laitiers. L'objectif de cette étude était d'évaluer l'utilité du prélèvement de sérum précolostral de nouveaux nés pour détecter des infections foetales apparentes par IBR et BVD dans un troupeau. Un échantillonnage de convenance de 317 sérums, prélevés de 50 troupeaux canadiens, a été utilisé. Les niveaux d'anticorps des sérums ont été mesurés en utilisant 2 trousses ELISA (IBR et BVD). Le statut précolostral a été confirmé pour 304 échantillons par la mesure de l'activité sérique des gamma glutamyl transférases. Une plus grande proportion de résultats positifs à IBR (RC = 86; IC 95%: 17,8 à 415,7) et BVD (RC = 199,3; IC 95 %: 41,7 à 952,3) a été observée parmi les échantillons postcolostraux que parmi les précolostraux. Tous les échantillons précolostraux positifs (n = 7/304) provenaient de veaux nés de mères vaccinées. Les échantillons postcolostraux positifs (n = 11/13) étaient majoritairement (60 %) prélevés à partir de veaux nés de mères non vaccinées. Le prélèvement de sérum précolostal peut détecter des infections foetales apparentes dans les troupeaux.(Traduit par les auteurs).

RESUMO

This article describes hypokalemia syndrome. Lactating dairy cows seem to be at the highest risk, but younger animals may also develop the disease. At present, except for animals treated with repeated isoflupredone acetate administration, the exact determinants causing hypokalemia syndrome remain uncertain. Affected animals are anorexic, weak to recumbent, and most often show signs of gastrointestinal stasis. Treatment is directed toward supportive care and oral potassium supplementation.

Neonatal infections and sepsis occur most frequently in calves with failure of passive transfer. If the invading bacteria are not rapidly controlled, they can set up focal infections, such as in growth plates, joints, or meninges, or generalized sepsis may occur. If not successfully treated, sepsis can lead to a systemic inflammatory response, multiple organ dysfunction syndromes, septic shock, and death. Treatments are based on selecting an appropriate antimicrobial drug and dosage, supportive therapy, fluid therapy, nonsteroidal anti-inflammatory drugs, and plasma transfusion. Preventing the failure of passive transfer through good colostrum management is essential.

Epidemiological data, clinical signs, complementary examination findings, antimicrobial treatments, and outcome were reviewed in 15 calves diagnosed with otitis media at the Centre hospitalier universitaire vétérinaire de l'Université de Montréal between 1987 and 2002. Age at presentation ranged from 2 to 18 weeks. A purulent ear discharge and epiphora were seen in 8/12 and 6/15 cases, respectively. Neurological signs observed were head tilt (13), eyelid ptosis (7), paresis/paralysis of the pinna (8), ataxia (2), strabismus (2), and convulsions (1). Concurrent pneumonia was frequently diagnosed (n = 11). A Mycoplasma sp. was the principal pathogen isolated from ear discharge; 6 out of 6 samples submitted were positive for mycoplasma. Tympanic bullae radiographs were considered abnormal in 12 out of 13 cases. Cerebrospinal fluid analysis was considered abnormal in 2 out of 5 cases. The antibiotic most commonly used was enrofloxacin (n = 7). Average treatment duration was 19.6 days. Four out of 8 treated animals for which follow-up information was available completely recovered. These results suggest that M. bovis is a major pathogen of otitis media in dairy calves and effective antimicrobial therapy should be of long duration.

Risk factors associated with failure of passive transfer of immunity (FPT) were evaluated among newborn beef calves in Québec. Physical examination was performed on calves born of a normal calving and blood samples were collected for determination of health status and measurement of serum concentration of immunoglobulin (Ig) G1. Of 225 calves, from 45 herds, 19% showed FPT (serum IgG1 concentration < 10.0 g/L). Calves born in a stanchion-stall were more likely to show FPT (OR: 10.2). Calves bottle-fed colostrum were less at risk for FPT (OR: 0.06). Calf gender, month of birth, dam parity, and dam body condition score were not associated with FPT. No association was detected between FPT and health status. Special care should be given to calves born from cows in a stanchion-stall to ensure adequate colostrum intake. Failure of passive transfer of immunity should be considered with other risk factors when investigating morbidity.

A convenience sample of 234 colostral specimens, collected directly from the nursing bottle immediately prior to the first feeding, was studied. Samples originated from 6 farms and were collected over 24 months. Routine bacteriologic techniques were used to quantify the bacterial load of the colostrum, as well as to identify the bacteria. Overall, at least 1 microorganism was cultured from 221 colostral samples (94.4%). By using the upper tolerance level of 100,000 bacteria/mL, 84 samples (35.9%) were considered contaminated. Staphylococcus spp. (57.7%), gram-negative rods (47.9%), coliforms (44.0%), and Streptococcus uberis (20.5%) were among the most frequently isolated bacteria. The relative risk (RR) of contamination with more than 100,000 bacteria/mL was significantly greater in warm months [RR = 2.55, 95% confidence interval (CI): 1.63 to 4.02] than in cool months and in colostrum offered to male calves (RR = 1.55, 95% CI: 1.09 to 2.20). Bacterial load was also associated with the farm of origin (P < 0.0001). When assessing colostrum management, one should consider bacterial contamination. Multiple factors are likely associated with the degree of contamination, and farm-specific factors may be important. Further studies are necessary to evaluate the impact of bacterial contamination of colostrum on neonatal health.
