RESUMO

Hyptis martiusii Benth. is an aromatic plant found in abundance in northeastern Brazil that is used in ethnomedicine to treat gastric disorders. The aim of this study was to elucidate the mechanisms of action involved in the gastroprotection of the essential oil of Hyptis martiusii (EOHM) and to evaluate its healing capacity. Wistar rats were exposed to different protocols and subsequently were treated with 1% Tween-80 aqueous solution (negative control), pantoprazole, carbenoxolone, N-acetylcysteine (depending on the specificity of each model) or EOHM. The antisecretory activity (basal or stimulated) was determined using the pyloric ligature method. The gastroprotective action of nitric oxide and sulphydryl groups (-SH groups), as well as the quantification of adherent mucus and the levels of malondialdehyde and -SH groups in gastric mucosa, were evaluated using ethanol-induced gastric ulcer model. The healing ability was evaluated using the acetic acid-induced gastric ulcer model and histological and immunohistochemical analysis (HE, PAS and PCNA). EOHM (400 mg/kg) reduced the volume and acidity of gastric secretion stimulated by histamine and pentagastrin. The gastroprotective effect of EOHM involves the participation of endogenous sulfhydryl groups. EOHM increased mucus production (54.8%), reduced levels of MDA (72.5%) and prevented the depletion of -SH groups (73.8%) in the gastric mucosa. The treatment with EOHM reduced in 70.3% the gastric lesion area, promoting significant regeneration of the gastric mucosa, as confirmed by histological analysis and analysis of proliferating cell nuclear antigen. The results show that gastroprotective effect of EOHM is mediated by cytoprotective and antioxidant mechanisms and by their antisecretory activity, and suggest that the essential oil of Hyptis martiusii is a promising candidate for the treatment of gastric ulcers.

Studies with human beings and animals have shown that dietary conjugated linoleic acid (CLA) promotes changes in body structure, markedly, body fat reduction. This research aimed to assess the effect of CLA (60 percent of conjugated linoleic acid methyl ester, at the proportion of 1:1 of the 9, 11: 10, 12 isomers) on changes in body weight, subcutaneous tissue thickness, body fat mass and concentration of serum lipids (triacylglycerols, total cholesterol, LDL and HDL) in overweight dogs. Dogs of different breeds weighting 16.85±7.13 assigned in a randomized assay. Two types of diets were offered for 120 days: control (-CLA) and test (+CLA, 0.3 percent). There was no difference between control and test groups considering body weight and serum levels of triacylglycerols, total cholesterol and LDL portion. There was a significant difference between the final and initial results for subcutaneous tissue thickness and body fat mass and for serum concentration of HDL in dogs supplemented with CLA. The addition of 0.3 percent of CLA in the diet of overweight dogs decreased the fat deposition on subcutaneous tissue and the development of body fat mass but increased blood levels of HDL.


Estudos em humanos e animais têm demonstrado que o uso do ácido linoléico conjugado (ALC) promove alterações na composição corporal, principalmente reduzindo a deposição de gordura. O objetivo deste trabalho foi avaliar as alterações sobre o peso corporal, tecido adiposo subcutâneo, gordura corporal e concentração sérica de lípideos (triacilglicerol, colesterol total, LDL e HDL) de cães com sobrepeso alimentados com ALC (60 por cento de ácido linoléico conjugado metil ester, na proporção de 1:1 de 9, 11: 10, 12 isômeros) na dieta. Duas dietas foram fornecidas por quatro meses, para 14 cães com sobrepeso (16,85±7,13) de diferentes raças: dietas controle (-ALC) e teste (+ALC, 0,3 por cento). O experimento seguiu delineamento inteiramente casualizado. Não houve diferença entre os grupos para peso corporal e nível sérico de triacilglicerol, colesterol total e LDL. No entanto, foram observadas diferenças significativas entre os períodos final e inicial para tecido adiposo subcutâneo, gordura corporal e nível sérico de HDL nos cães que receberam ALC. A suplementação de 0,3 por cento de ALC na dieta de cães com sobrepeso diminuiu a deposição de lipídios no tecido subcutâneo e a gordura corporal total, mas aumentou os níveis sanguíneos de HDL.

This study investigated the acute effects of green tea extract (GTE) and its polyphenol constituents, (-)-epigallocatechin-3-gallate (EGCG) and (-)-epicatechin (EC), on basal and stimulated testosterone production by rat Leydig cells in vitro. Leydig cells purified in a Percoll gradient were incubated for 3 h with GTE, EGCG or EC and the testosterone precursor androstenedione, in the presence or absence of either protein kinase A (PKA) or protein kinase C (PKC) activators. The reversibility of the effect was studied by pretreating cells for 15 min with GTE or EGCG, allowing them to recover for 1 h and challenging them for 2 h with human chorionic gonadotropin (hCG), luteinizing hormone releasing hormone (LHRH), 22(R)-hydroxycholesterol or androstenedione. GTE and EGCG, but not EC, inhibited both basal and kinase-stimulated testosterone production. Under the pretreatment conditions, the inhibitory effect of the higher concentration of GTE/EGCG on hCG/LHRH-stimulated or 22(R)-hydroxycholesterol-induced testosterone production was maintained, whereas androstenedione-supported testosterone production returned to control levels. At the lower concentration of GTE/EGCG, the inhibitory effect of these polyphenols on 22(R)-hydroxycholesterol-supported testosterone production was reversed. The inhibitory effects of GTE may be explained by the action of its principal component, EGCG, and the presence of a gallate group in its structure seems important for its high efficacy in inhibiting testosterone production. The mechanisms underlying the effects of GTE and EGCG involve the inhibition of the PKA/PKC signalling pathways, as well as the inhibition of P450 side-chain cleavage enzyme and 17beta-hydroxysteroid dehydrogenase function.

Toxocara vitulorum, a nematode parasite in the small intestine of cattle and water buffaloes, causes high morbidity and mortality of 1-3 months old buffalo calves. This research evaluated the specific perieneteric antigens (Pe) reactivity of anti-T. vitulorum-Pe antibody (Tv-Pe-Ab) in both immune sera and colostrum from buffalo cows immediately post-partum from buffalo cows. The presence of Tv-Pe-Ab in sera of buffalo newborn calves was also examined at 1 day before and after suckling the colostrum as well as in sera from naturally infected calves at the beginning and peak of the maximum infection and then again during the period of rejection and post-rejection of the parasite. Pe antigens were characterized for Tv-Pe-Ab by SDS-PAGE and Western blot (WB). The SDS-PAGE showed that Pe contained nine protein bands (11, 14, 31, 38, 58, 76, 88, 112 and 165 kDa). All Pe bands were recognized by Tv-Pe-Ab in sera and colostrum of buffalo cows. Only the serum antibodies of buffalo calves at 1 day of age after suckling the colostrum and during the beginning of T. vitulorum infection recognized Pe antigen's nine bands. In contrast, serum antibodies from 1-day-old buffalo calves, taken before suckling colostrum, did not react with any protein band. In suckling calves, which reached peak egg output, rejection and post-rejection stages of the infection, serum Tv-Pe-Ab reactivity with lower molecular weight protein bands (11-76 kDa) was lost and only reactivity with the Pe protein bands of higher molecular weight (88, 112 and 165 kDa) remained.

Toxacara vitulorum é um nematódeo que acomete principalmente bezerros búfalos na faixa etária de um a três meses de vida, causando grande morbidade ou mortalidade quando não tratados. A pesquisa objetivou a obtenção do antígeno extrato larval solúvel bruto(Ex) de larvas infectantes de T. vitulorum, bem como a separação das frações antigênicas pelo SDS-PAGE e pelo "Western blotting" (WB), utilizando-se soros imunes e colostros de búfalos naturalmente infectados com T. vitulorum. O acompanhamento do quadro parasitário dos bezerros búfalos.também foi realizado. Pôde-se verificar que o antígeno revelou 11 (11, 13, 16,22,25, 32, 43, 53, 68, 82 e 96 kDa) bandas protéicas. Quando analisadas pelo WB, cinco dessas bandas (43, 53, 68, 82 e 96 kDa) foram reconhecidas pelos anticorpos presentes nas amostras de soros e de colostros das búfalas e de soros dos bezerros búfalos com um de vida após mamarem o colostro. No entanto, somente as bandas de 53 a 96 kDa que foram as mais evidentes persistiram nos grupos de bezerros búfalos que se encontravam tanto no pico da infecção como no período de expulsão ou pós-expulsão dos helmintos adultos do intestino. As bandas antigênicas de 68 e de 92 kDa, por serem as mais proeminentes podem ser consideradas componentes para futuras vacinas contra T. vitulorum em búfalos.
