RESUMO

Fatty acid spectrum of phospholipids and free cholesterol level in the myocardium, liver and erythrocytes were studied in dogs with the experimental lung-heart insufficiency (LHI). The content of polyunsaturated fatty acids of phospholipids in erythrocytes from peripheric blood and in free cholesterol level (2-3 times) in myocardium, liver and erythrocytes of dogs under investigated conditions decreased. It is shown that pharmacotherapy with alpha-adrenoblockers (thropaphen and prasozin) has led to the normalization of disturbances of the erythrocytic lipid complex and had caused inhibition of peroxidase reactions in the myocardium and liver of the dogs with LHI.

RESUMO

With the use of the erythrocyte osmotic and acidic resistance methods, the experiments on intact albino rats by applying a model of chronic non-specific lung diseases and those in vitro have shown that mildronate has a membrane-stabilizing action only when given in high doses (500 mg/kg) and concentrations (10(-3)-10(-4) M). When used in low doses (5, 25, 50, and 200 mg/kg) and concentrations (10(-5)-10(-7) M), in produces no positive effect.

In experiments on rabbits it was established that strophanthin (0.05 mg/kg) administered to animals with cardiac insufficiency due to a severe overloading of the heart function by occlusion of the diaphragmatic portion of the aorta by 4/5 of its diameter improved the contractile activity and relaxing properties of the left ventricle. This effect is however transient (up to 15-30 minutes) with the subsequent reduction of its characteristic indices, the development of symptoms of intoxication and not infrequently death of animals. alpha-tocopherol administered before the pathology reproduction (10 mg/kg, 3 days) increases the period of myocardial compensation and when given in combination with strophanthin significantly potentiates the cardiac effect of glycoside manifesting by an increase of the strength and rate of contraction and intensity of diastolic relaxation of the myocardium, prolongs the animal's life.

RESUMO

Experiments on intact dogs and animals with an early stage of experimental cardiopulmonary insufficiency due to chronic pneumonia were made to study the action of strophanthine on the hemodynamics of the left and right ventricles and pressure in the pulmonary artery. Administration of the glycoside to animals with cardiopulmonary insufficiency increased the contractility of the right ventricle and the intensity of diastolic relaxation of both ventricles. Strophanthine was revealed to exert the prevailing effect on the cardiodynamics of the right ventricle both in intact dogs and in animals with experimentally induced pathology. Strophanthine increased pressure in the pulmonary artery, promoting the overload of the right ventricle to a less degree than convallotoxin.

Experiments were made on anesthesized dogs with closed chest to examine the effect of the Soviet antihistaminic drug phencarol on the cardio- and hemodynamics energy metabolism and microcirculatory bed of the myocardium under focal immune injury to the heart. Preliminary administration of phencarol reduced the shifts in the cardio- and hemodynamics and energy metabolism in the myocardium, characteristic for immune injury. The affected area manifested a less marked rop of the systemic arterial pressure, myocardial contractility, the content of adenyl nucleotides, glycogen and phosphorylase activity. There was a considerable decrease in size of the affected area of the left ventricular myocardium.

RESUMO

The effect of convallotoxin on hemodynamics of the right and left ventricles and pressure in the pulmonary artery was studied in intact dogs and in surgical stenosis of the pulmonary trunk. In intact anesthetized dogs, convallotoxin produced changes in hemodynamics that were more pronounced in the right than in the left ventricle (decreased intraventricular pressure, increased dp/dtmax and dp/dtmin). Administration of convallotoxin ton dogs one month after pulmonary trunk stenosis without episodes of congestive heart failure did not improve the cardiohemodynamics.
