RESUMO

Recently there have been a variety of methods to synthesize silver nanoparticles, among which the biosynthesis method is more noticeable due to features like being eco-friendly, simple, and cost-efficient. The present study aims for the green synthesis of silver nanoparticles from the extract of the three plants A. wilhelmsi, M. chamomilla, and C. longa; moreover, it pledges to measure the antibacterial activity against some variants causing a skin rash. The morphology and size of the synthesized silver nanoparticles were evaluated by UV.vis, XRD, SEM, and FTIR analyses. Then results showed a color alteration from light yellow to dark brown and the formation of silver nanoparticles. The absorption peak with the wavelength of approximately 450 nm resulting from the Spectrophotometry analysis confirmed the synthesis of silver nanoparticles. The presence of strong and wide peaks in FTIR indicated the presence of OH groups. The SEM results showed that most synthesized nanoparticles had a spherical angular structure and their size was about 10 to 20 nm. The highest inhibition power was demonstrated by silver nanoparticles synthesized from the extract combined from all three species against Gram-positive bacteria Staphylococcus aureus and Staphylococcus epidermidis (23 mm) which had a performance far more powerful than the extract. Thus, it can be understood that the nanoparticles synthesized from these three species can act as potential environment-friendly alternatives to inhibit some variations causing skin disorders; an issue that calls for further clinical studies.

Teucrium polium L. is used to treat many diseases like abdominal pains, indigestion, colds, and reproductive system diseases in Iranian folk medicine. This study was designed to investigate the yield, chemical profile of essential oil, and antibacterial and antifungal activity of this species. The flowering aerial parts of T. polium were collected from the Margh region of Kashan, Iran. The essential oil of the plant was extracted and separated using the Clevenger apparatus and analyzed using gas chromatography-mass spectrometry (GC-MS). The antimicrobial activity of the essential oil against a variety of standard microbial strains was investigated with the Agar well-diffusion method and determination of the lowest growth inhibitory and lethal concentration (MIC and MBC). The results showed that the yield of T. polium essential oil (TPEO) was 0.5778%. Based on the results of GC-MS, 76 compounds (99.3%) were identified in the TPEO, of which α-pinene (9.67%), ß-caryophyllene (8.07%), ß-pinene (5.04%), nerolidol (4.94%), and oleic acid (4.57%) were the dominant components. The results of antimicrobial tests showed that the TPEO on Staphylococcus aureus created the largest zone of inhibition (~ 14.29 mm). The dominant inhibitory activity of this essential oil was against Gram-negative bacteria Escherichia coli (~ 9.00 mm), which was almost similar to rifampin (~ 11 mm). It seems that the predominance of terpenoid and acidic compounds of the essential oil is one of the possible factors of the anti-bacterial activity of this essential oil. Therefore, the TPEO may be a promising and potential strategy to inhibit some bacterial strains.

Phlomis olivieri Benth. is an aromatic plant endemic to Iran belonging to the Lamiaceae family. It is used to treat pain, stomach ache and common cold in Iranian traditional medicine. P. olivieri also has valuable biological properties including antioxidant, antimicrobial and analgesic ones. This was the first study designed to assess the quality, quantity and antimicrobial activity of Phlomis olivieri Benth. essential oil (POEO). Samples were randomly collected from flowering twigs of this species in three locations between Azeran and Kamoo in Kashan, Iran at peak flowering in June 2019. Water distillation extraction was used to obtain the POEO the quantity of which was calculated by weight. Gas chromatography coupled to mass spectrometry (GC/MS) was also used for POEO qualitative analysis, which revealed its chemical compounds and their percentages. Antimicrobial activity of POEO was also determined using the agar well diffusion method. Minimum inhibitory concentration (MIC) and minimum bactericidal/fungicidal concentration (MBC/MFC) were also measured using the broth microdilution method. The results of the quantitative and qualitative analysis showed that the POEO yield was ~ 0.2292% and its main chemical compounds included the sesquiterpenes germacrene D (26.43%), ß-caryophyllene (20.72%), elixene (6.58%), ß-trans-farnesene (6.17%), ß-Cyclogermacrane (5.04%), germacrene B (4.73%), α-humulene (4.22%), and monoterpene α-pinene (3.22%). The agar diffusion method demonstrated the highest antimicrobial activity of POEO (MIC ~ 14.50 mm) was against the Gram-positive species Streptococcus pyogenes. The POEO also showed the strongest inhibitory and lethal activity against the gram-negative bacterial species Pseudomonas aeruginosa (MIC < 62.50 µg/mL) and S. paratyphi-A (MIC < 62.50 µg/mL and MBC = 125 µg/mL), and fungal species Candida albicans (MIC and MBC = 250 µg/mL) as compared to control-positive antibiotics. Therefore, POEO is a valuable natural alternative rich in sesquiterpenes with strong antimicrobial and antifungal activities against some fungal and bacterial strains. It can also be used in the pharmaceutical, food and cosmetic industries.

BACKGROUND: One of the most important aromatic species of the Apiaceae family is Heracleum persicum Desf. ex Fisch., C.A.Mey. & Avé-Lall, which used as a spice and condiment in food. It is widely used in traditional Iranian medicine due to its anti-toxic properties. The aim of this study was to evaluate the essential oil of this plant in terms of content and ingredients, cytotoxicity and antimicrobial activity. METHODS: The fruit of H. persicum was collected in June 2019 from Maragheh region of Kashan, Iran. The essential oil was extracted by water distillation using Clevenger for 3 h. Identification analysis of H. persicum fruit essential oil (HPFEO) components was performed using gas chromatography-mass spectrometry (GC-MS). Evaluation of the effect of the HPFEO on the growth and proliferation of ovarian cancer cell line (OVCAR-3) was performed using MTT colorimetric method. Its antimicrobial activity was evaluated by agar diffusion method, minimum inhibitory concentration (MIC) and minimum bactericidal/fungal lethality concentration (MBC/MFC). RESULTS: The results of analysis of the HPFEO by GC-MS showed that there were 35 compounds with 99.54% relative content. Hexyl butyrate (35.24%), octyl 2-methylbutyrate (11.65%), octyl isobutyrate (9.23%), and octyl acetate (8.42%) were the predominant compounds. Examination of cell survival showed that the viability of cells decreased depending on the concentration of the HPFEO in 24 h and had value of IC50 ~ 12.08 µg/ml against OVCAR-3 cell line. The strongest inhibitory activity of the HPFEO was against Gram-negative bacteria Pseudomonas aeruginosa and Salmonella paratyphi-A serotype (MIC < 62.50 µg/mL). Also, the strong inhibitory and lethal activity of this essential oil against Candida albicans (MIC and MBC 250 µg/mL was one degree weaker compared to nystatin (MIC = 125 µg/mL). CONCLUSIONS: Thus, HPFEO, in addition to its traditional use, may have a strong and potential potential for the production of anti-proliferative and antimicrobial drugs.

Today, the lack of quality water supply has led to the tendency to use unconventional water to irrigate agricultural products. Considering the importance and application of essential oils of mint plants in various pharmaceutical, food, and health industries and also considering the approach of using unconventional waters in the cultivation of medicinal plants, the present study aimed to investigate and compare the chemical composition of essential oils of two species of Mentha spicata L. and Rosmarinus officinalis L. which was designed and implemented for the first time under the influence of different treatments of municipal and industrial wastewater. For this purpose, first R. officinalis cuttings and roots of M. spicata were prepared and after preparing and leveling the ground, in the spring of 2020, it was transferred to the planting site and planted in the form of creek and ridges. The treatments studied in this study included well water (WW), treated municipal wastewater (TMW), untreated municipal wastewater (UMW), treated industrial wastewater (TIW), and untreated industrial wastewater (UIW) in a randomized complete block design with four repeat runs. After watering the plants continuously for 3 months, the plant branches were collected and transferred to the laboratory for drying. After extracting the essential oil by water distillation (Clevenger) method, the analysis and identification of the compounds were performed by a chromatograph coupled with a mass spectrometer (GC/MS). The results showed that the highest and lowest yields of M. spicata belonged to the samples treated with UMW and WW, respectively. Also, R. officinalis essential oil irrigated with UMW and UIW had the highest and lowest yields, respectively. The number of essential oil compounds in of M. spicata was between 5 and 19 and in R. officinalis between 14 and 23 under different treatments. The results of the analysis of essential oil compounds showed that D-carvone (57.77-57.44%) and D-limonene (8.70-26.65%) for M. spicata and α-pinene (26.12-34.85%), 1,8-cineole (18.95-23.70%), and camphene (9.93-12.80%) for R. officinalis were predominant compounds in all studied treatments. The results show that UMW is a suitable and efficient treatment to have the best quantity of M. spicata essential oil and the best quality and quantity of R. officinalis essential oil.

The goal of this study is to evaluate the chemical compounds, the anti-bacterial/fungal activity, and the cytotoxicity of the essential oil of three species of lamiaceae in Iran. After the extraction of the essential oil implementing the hydrodistillation method, the analysis and identification of the compounds were carried out with a chromatograph coupled with a mass spectrometer. For the evaluation of the anti-bacterial/fungal activity of the essential oils, the measurement of the diameter of inhibition halo, the minimum inhibitory concentration (MIC), bactericidal and fungicidal concentrations (MBC/MFC) were utilized; and for the evaluation of the cytotoxic activity of the essential oils, the 3-(4,5-dimethylthiazolyl)-2,5-diphenyltetrazolium bromide (MTT) method was used. The results show that the dominant compounds in the Perovskia abrotanoides Kar essential oil were camphor (21.68%), 1,8-cineole (14.26%), and α-pinene (7.23%); moreover, the dominant compounds in the Salvia reuteriana Boiss. Essential oil were benzyl benzoate (27.10%), linalool (13.27%), and sclareol (7.75%); in addition, the dominant compounds in the Ziziphora clinopodioides subsp. rigida (Boiss.) Rech.f. were cyclofenchene (25.29%), pulegone (14.14%), and menthol (7.70%). The largest halo diameter of inhibition halo (~ 22 mm) was against Streptococcus pyogenes and the strongest inhibiting and killing activity was against Candida albicans (MIC and MFC = 125 µg/mL) shown by the S. reuteriana essential oil which, respectively, matched the control antibiotics rifampin and nystatin. The analysis of the MTT test results showed that the Z. clinopodioides subsp. rigida essential oil (with IC50 value of ~ 144.2500) had the strongest cytotoxic activity against human ovarian cancer cells (OVCAR-3). On the whole, the results show that the essential oil of the Lamiaceae family plants is a source for various compounds with potential biological activities which can serve as a possible alternative to produce herbal medicine which are effective on some microorganisms and cancer cell lines.

Damask rose (Rosa damascena Mill.) is an aromatic medicinal plant rich in bioactive compounds with high value in the food, pharmaceutical and cosmetic industries. Knowledge of the factors affecting the quantitative and qualitative properties of the compounds in its essential oil (EO) and the bioactivity of this EO is important in optimizing Damask rose cultivation and production. This research studied, for the first time, the effects of irrigation water and soil chemical and physical characteristics on the EO yield of this important commercial species and on it chemical composition and antimicrobial activity. The results showed the significant effect of crop cultivation site on yield, chemical composition and inhibition zone diameter (IZD) at the 1% significance level. The highest EO yield (~0.0266%), which belonged to the Noushabad site (EON), resulted from the increased soil electrical conductivity (EC) and the higher sand, gypsum and lime contents and irrigation water salinity. Analysis of the chemical composition of the EOs showed that their main compounds at all three crop sites were citronellol, nonadecane, heneicosane and geraniol. The EO obtained from the Yazdel site (EOY) had the highest contents of citronellol and geraniol (~29.05% and ~6.85%) that were directly correlated with soil potassium and phosphorus contents and inversely correlated with soil acidity and EC and its lime, nitrogen, and organic carbon contents. Antimicrobial assays indicated that the EO extracted from the Sefidshahr site (EOS), which had the largest inhibition zone diameter (~14.67 mm) for Aspergillus brasiliensis (IZD~14.67 mm) and the lowest MIC (~31.25 µg/mL) for Staphylococcus aureus and Pseudomonas aeruginosa, exhibited efficacy similar to that of rifampin, probably due to the dominance of the alkanes in it. The EOY and EOS also exhibited the strongest inhibitory and lethal activity against Candida albicans (MIC and MBC <15.63 µg/mL for EOY and MIC and MBC = 62.5 µg/mL for EOS), which were six and four times stronger than those of nystatin, respectively. Therefore, the selected EOs can act as a potentially promising strategy for fighting microbial strains.

ETHNOPHARMACOLOGICAL RELEVANCE: Dracocephalum kotschyi is a medicinal plant widely used in traditional medicine to treat pain, fever, inflammation, and seizures. AIM OF THE STUDY: Due to the importance of this plant and the well-known antibacterial activity of essential oils, the aim of the present study was to investigate the composition of essential oil and evaluate the antimicrobial activity of its main active compounds. MATERIALS AND METHODS: In order to test its possible application at industrial level the oil was extract from the cultivated and wild plants. The epigean parts were collected in June 2018 from the same region of Daran (Isfahan, Iran). The extraction of essential oil was carried out using a Clevenger apparatus. The composition of the essential oil was assayed by using a gas chromatography/mass spectroscopy apparatus (GC/MS). RESULTS: Results showed that the predominant compounds of essential oil of cultivated plants were α-pinene (13.66%), (E)-citral (12.89%), neral (11.25%), methyl geranate (8.66%), limonene (8.33%), campholenal (6.22%) and geraniol (5.69%), while those found in naturally grown plants were two main compounds: cyclohexylallene (52.63%) and limonene (35.88%). The antimicrobial properties of the plant were determined against 12 strains of microorganism by evaluating inhibition halo, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The highest inhibition halo for both oils from cultivated and wild plants was obtained against Aspergillus brasiliensis. The MIC value against Gram-positive Bacillus subtilis was 31.25 µg/ml, it was the lowest value provided by the essential oil obtained from the cultivated sample, the MIC was significantly lower than that obtained by treating the same strain with Rifampin. On the other hand, Candida albicans had the highest sensitivity (MIC value of 31.25 µg/ml) for the essential oil obtained from wild plants as the inhibitory concentration was lower than that obtained treating the yeast with Nystatin. CONCLUSIONS: Therefore, according to the results of the present study, the use of the essential oil obtained from D. kotschyi can be used to protect food and to treat microbial infections.

The majority of essential oils obtained from vascular plants have been demonstrated to be effective in treating fungal and bacterial infections. Among others, Salvia hydrangea is an endemic half-shrub belonging to the Lamiaceae family that has been widely used from ancient times in Iranian traditional medicine. The aim of this study was to compare the composition and antimicrobial properties of essential oils obtained from leaves or flowers of this plant, collected from the Daran region of Iran during June 2018. The oils were obtained using Clevenger apparatus, their composition was evaluated by means of gas chromatography/mass spectrometry (GC/MS) and the antimicrobial properties were assayed by measuring inhibition halos, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The yield of leaf oil was ~ 0.25% and that of flower oil was ~ 0.28%. Oil composition was affected by the part of the plants used: the most abundant bioactives contained in leaf essential oil were (+)-spathulenol (16.07%), 1,8-cineole (13.96%), trans-caryophyllene (9.58%), ß-pinene (8.91%) and ß-eudesmol (5.33%) and those in flower essential oil were caryophyllene oxide (35.47%), 1,8-cineole (9.54%), trans-caryophyllene (6.36%), ß-eudesmol (4.11%), caryophyllenol-II (3.46%) and camphor (3.33%). Both the oils showed a significant inhibitory and lethal effect on the Gram-negative bacteria Pseudomonas aeruginosa (MIC ~ 16 µg/mL), Shigella dysenteriae and Klebsiella pneumoniae (MIC ~ 62 µg/mL). Therefore, the essential oils obtained from both leaves and flowers of S. hydrangea may have potential application as bactericidal agents against some bacteria.

This work aimed at developing a mouthwash based on liposomes loading Citrus limon var. pompia essential oil or citral to treat oropharyngeal diseases. Vesicles were prepared by dispersing phosphatidylcholine and pompia essential oil or citral at increasing amounts (12, 25 and 50 mg/mL) in water. Transparent vesicle dispersions were obtained by direct sonication avoiding the use of organic solvents. Cryogenic transmission electron microscopy (cryo-TEM) confirmed the formation of unilamellar, spherical and regularly shaped vesicles. Essential oil and citral loaded liposomes were small in size (~110 and ~100 nm, respectively) and negatively charged. Liposomes, especially those loading citral, were highly stable as their physico-chemical properties did not change during storage. The formulations were highly biocompatible against keratinocytes, were able to counteract the damages induced in cells by using hydrogen peroxide, and able to increase the rate of skin repair. In addition, liposomes loading citral at higher concentrations inhibited the proliferation of cariogenic bacterium.

OBJECTIVE: Dracocephalum kotschyi Boiss is one of eight species of Dracocephalum, an endemic species of Iran, and grows in areas such as Isfahan, Yasuj, Mazandaran, and Tabriz. The present study was designed to analyze the antioxidant, phenol and flavonoids contents of this plant under different conditions of cultivation in a completely randomized, factorial design. MATERIALS AND METHODS: Shrubs of different ages (two, three, and six) in cultivated rangeland collected from three randomly plant at the end of May 2018 simultaneously with the flowering time. Antioxidant activity of methanolic extract of plant samples were evaluated with DPPH method. Total phenolic compounds and the total content of flavonoids were measured using the Folin Sioukhlu and aluminum chloride methods respectively. RESULTS: The result showed that there was a significant effect of planting time, plant organs and interaction of time and organ on the total antioxidant capacity and total phenol and flavonoids contents. The highest antioxidant activity, total phenol, and total flavonoid have belonged to the flower of six age plants. The highest antioxidant activity, total phenol, and total flavonoid have belonged to the flower of six age. The highest level of antioxidant activity with IC50 15.8489 ±â€¯0.001 µg/ml belonged to the flower of the two-year-old plant, which has a stronger antioxidant activity than the BHT standard with 19.95 µg/ml. CONCLUSION: But in general, it can be said that the six-year-old plant with the highest antioxidant capacity, phenolic, and flavonoid compounds is the best age and mainly flower organ is preferred.

Scrophularia striata, commonly known as figwort, is one of the most important medicinal plants that mainly grows in cold regions of the Zagros Mountains (West of Iran). Although the chemical composition of this plant species has not yet been explored, people living in Ilam province (W Iran) have used it for many years to treat different illnesses. The present study aims to analyze the effect of some ecological factors on the antioxidant potential and the amount of phenol present in this plant species, using a random factorial design with two factors (elevation and region) and three replicates. The fruits of the plant were gathered from three different elevations. They were collected from three regions of the Ilam province (Badreh, Dareshahr, and Dehloran) in June 2016, when the fruits appear. Moreover, to analyze different soil chemical and physical features, soil samples were gathered from a depth of 0.5 m under the shrubs. The antioxidant action of the methanol extract from the plant samples and the total amount of phenol compounds were measured using 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and the Folin-Ciocalteu method, respectively. The results showed that the effects of site and elevation, and the interaction between these factors, on the antioxidant potential and total phenol amount were significant with a probability of error of 1%. The maximum extract efficiency (19.37 ± 3.07%), antioxidant potential (126.5656 ± 0.96 µg/mL), and total amount of phenol (55.7689 ± 3.17 µg/mL) were obtained from Dareshahr at an elevation of 600 m above mean sea level. The minimum amount of total phenol (24.6544 ± 3.21 µg/ml) was recorded at the lowest elevation of Badreh, at which phosphorus, potassium, organic carbon, organic material, nitrogen, acidity, lime, and silt were present at the lowest amount. However, the antioxidant activity and total amount of phenol had a strong direct correlation in the two districts of Dareshahr and Badreh, but were reversely and strongly correlated in Dehloran. Therefore, it can be stated that Scrophularia striata has the potential for antioxidant activity, however, the complexity of the effect of ecological factors on one hand, and the emergence of different chemical processes in the plant under such effects on the other hand, has led to the synthesis of different compounds with antioxidant potential in the plant in different regions.
