There is currently a lack of high-quality research on the best dietary recommendations for patients with early glaucoma or at high risk for glaucoma. This meta-analysis aims to clarify the relationship between vitamin intake and glaucoma risk. Electronic databases, including PubMed, EMbase, ScienceDirect, Cochrane Database, Clinicaltrials.gov, and Google Scholar, were searched for publications indexed as of September 18, 2021. Data were expressed as odds ratios (ORs) with 95% confidence intervals (CIs). The I2 index was used to assess heterogeneity. We performed five meta-analyses of existing studies to summarize the evidence on the association between vitamin intake and glaucoma risk. The initial search identified 689 studies, eight of which (262,189 patients) met the eligibility criteria for the meta-analysis. The data showed that high-dose intake of vitamins A (OR=0.63, 95%CI [0.53, 0.76]) and B (OR=0.71, 95%CI [0.64, 0.80]) but not vitamins C (OR=0.69, 95%CI [0.48, 1.01]), D (OR=0.90, 95%CI [0.45, 1.83]), or E (OR=0.91, 95%CI [0.71, 1.16]) was associated with a low prevalence of glaucoma. The results of this study demonstrated that high-dose intake of vitamins A and B, but not vitamins C, D, or E, was associated with a low prevalence of glaucoma.

Objective: To clarify the clinical efficacy of Yiqi Huoxue recipe in the treatment of liver fibrosis of chronic viral hepatitis. Methods: An open, positive-drug, parallel-controlled study method was applied. A total of 207 cases of liver fibrosis with chronic hepatitis B and C diagnosed with liver biopsy and transient elastography were selected. According to the principle of syndrome differentiation in traditional Chinese medicine, self-made Yiqi Huoxue recipe (n = 127) and Fuzheng Huayu capsule (n = 80) were used for the treatment course of 24-48 weeks. Change score of TCM symptom, liver biochemistry, liver stiffness measurement (LSM), and noninvasive liver fibrosis index [aspartate transaminase to platelet ratio index (APRI), and fibrosis-4 score (FIB-4)] were compared between the two groups to evaluate the therapeutic effect of Yiqi Huoxue recipe on liver fibrosis. Results: Yiqi Huoxue recipe group and Fuzheng Huayu capsule group baseline LSM, APRI and FIB-4 was compared, and there was no statistically significant difference between them (P > 0.05). Yiqi Huoxue recipe and Fuzheng Huayu capsule received patients had improved symptom scores to a certain extent. Hepatic facies, discomfort over liver area, and soreness and weakness of waist and knees (P < 0.05) was significantly improved in Yiqi Huoxue recipe than Fuzheng Huayu capsule. Liver biochemical indicators (ALT, AST, GGT, ALP) had gradually relapsed with the extension of treatment duration and the normalization rate between the two groups after 24 to 48 weeks had reached 100% vs. 100%, 100% vs. 93.8%, 96.8% vs. 92.3% and 87.5% vs. 81.8%. After 12 weeks of treatment, APRI values ​​of both groups had significantly reduced, and after 48 weeks of treatment, LSM values of both groups had significantly improved. Moreover, Yiqi Huoxue recipe FIB-4 score was significantly improved after 48 weeks of treatment, and the difference was statistically significant compared to Fuzheng Huayu capsule group (P < 0.05). After treatment, LSM, APRI, and FIB-4 total effectiveness in the two groups were 80.0% vs. 63.6%, P = 0.046; 68.4% vs. 52.0%, P = 0.052; 68.4% vs. 62.0%, P = 0.437, respectively. LSM total effectiveness was significantly higher in Yiqi Huoxue recipe treated group than Fuzheng Huayu capsule group. Conclusion: Traditional Chinese medicine Yiqi Huoxue decoction can be used as an optimal treatment for liver fibrosis of chronic viral hepatitis.

BACKGROUND: We conducted a single-arm phase II trial to evaluate the efficacy and adverse effects (AEs) of an anti-epidermal growth factor receptor monoclonal antibody, nimotuzumab, combined with cisplatin and 5-fluorouracil (PF) as first-line treatment in recurrent metastatic nasopharyngeal carcinoma after radical radiotherapy. METHODS: Patients who met the eligibility criteria were recruited from ten institutions (ClinicalTrials.gov; NCT01616849). A Simon optimal two-stage design was used to calculate the sample size. All patients received weekly nimotuzumab (200 mg) added to cisplatin (100 mg/m2 D1) and 5-fluorouracil (4 g/m2 continuous infusion D1-4) every 3-weekly for a maximum of six cycles. Primary end point was objective response rate (ORR). Secondary end points included disease control rate (DCR), progression-free survival (PFS), overall survival (OS) and AEs. RESULTS: A total of 35 patients were enrolled (13 in stage 1 and 22 in stage 2). Overall ORR and DCR were 71.4% (25/35) and 85.7% (30/35), respectively. Median PFS and OS were 7.0 (95% CI 5.8-8.2) months and 16.3 (95% CI 11.4-21.3) months, respectively. Unplanned exploratory analyses suggest that patients who received ≥2400 mg nimotuzumab and ≥4 cycles of PF had superior ORR, PFS and OS than those who did not (88.9% versus 12.5%, P < 0.001; 7.4 versus 2.7 months, P = 0.081; 17.0 versus 8.0 months, P = 0.202). Favourable subgroups included patients with lung metastasis [HROS 0.324 (95% CI 0.146-0.717), P = 0.008] and disease-free interval of >12 months [HROS 0.307 (95% CI 0.131-0.724), P = 0.004], but no difference was observed for metastatic burden. The only major grade 3/4 AE was leukopenia (62.9%). CONCLUSION: Combination nimotuzumab-PF chemotherapy demonstrates potential efficacy, and is well tolerated as first-line chemotherapy regimen in recurrent metastatic nasopharyngeal carcinoma.

OBJECTIVE: It has been well-established that microRNAs (miRNAs), a class of short non-coding RNA molecules, play an important role in the development of gastric cancer. In the present study, we focused on miR-105, a novel miRNA not previously linked to gastric cancer. PATIENTS AND METHODS: 36 paired surgically resected gastric cancer tissues and matched adjacent normal tissues were used to detect the expression of miR-105. AGS cells were used to overexpress or silence of miR-105 and to determine its effect on several tumorigenic properties. A cell proliferation enzyme-linked immunosorbent assay was used to analyze the incorporation of BrdU during DNA synthesis of AGS cells. Total cDNA from AGS cells was used to amplify the 3'-UTR of YY1 by PCR and luciferase activity was determined using the Dual-Luciferase Reporter Assay System RESULTS: We found that expression of miR-105 was reduced in gastric cancer tissues, compared with adjacent normal tissues, due to hypermethylation at its promoter region. Overexpression of miR-105 suppressed, whereas its inhibition promoted cell viability and proliferation. We further identified Yin Yang 1 (YY1) as a direct target of miR-105, by which miR-105 exerted its anti-proliferative role. Moreover, we found that DNMT3A was responsible for the down-regulation of miR-105 in gastric cancer cells. CONCLUSIONS: Our data demonstrate that miR-105 inhibits gastric cancer cell proliferation and progression, which might provide a therapeutical target for cancer therapy.

The Cdh23(erl/erl) mice are a novel mouse model for DFNB12 and are characterized by progressive hearing loss. In this study, erythropoietin (EPO) was given to the Cdh23(erl/erl) mice by intraperitoneal injection every other day from P7 for 7 weeks. Phosphate-buffered saline-treated or untreated Cdh23(erl/erl) mice were used as controls. Auditory-evoked brainstem response (ABR) thresholds and distortion product oto-acoustic emission (DPOAE) were measured in the mouse groups at the age of 4, 6 and 8 weeks. The results show that EPO can significantly decrease the ABR thresholds in the Cdh23(erl/erl) mice as compared with those of the untreated mice at stimulus frequencies of click, 8-, 16- and 32-kHz at three time points. Meanwhile, DPOAE amplitudes in the EPO-treated Cdh23(erl/erl) mouse group were significantly higher than those of the untreated groups at f2 frequency of 15383 Hz at the three time points. Furthermore, the mean percentage of outer hair cell loss at middle through basal turns of cochleae was significantly lower in EPO-treated Cdh23(erl/erl) mice than in the untreated mice (P<0.05). This is the first report that EPO acts as an otoprotectant in a DFNB12 mouse model with progressive hearing loss.

We report a novel mutation (erlong, erl) of the cadherin 23 (Cdh23) gene in a mouse model for DFNB12 characterized by progressive hearing loss beginning from postnatal day 27 (P27). Genetic and sequencing analysis revealed a 208 T >C transition causing an amino-acid substitution (70S-P). Caspase expression was upregulated in mutant inner ears. Hearing was preserved (up to 35-dB improvement) in pan-caspase inhibitor Z-VAD-FMK-treated mutants compared with untreated mutants (P<0.05). Outer hair cell (OHC) loss in the cochleae of Z-VAD-FMK-treated mutants was significantly reduced compared with those of untreated mice. Thus, the erl mutation can lead to hearing loss through apoptosis. This is the first genetic mouse model of hearing loss shown to respond to otoprotective drug therapy. The short interval from initial hearing loss to deafness (P27-P90) makes this model ideal for screening and validating otoprotective drugs.

This study investigated the effect of electroacupuncture pretreatment on the lipopolysaccharide (LPS)-induced inflammatory response and on acute kidney injury in adult male pathogen-free Wistar rats. Rats received electroacupuncture at the Zusanli (ST36) and Neiguan (PC6) acupoints, or electrical stimulation at sham points, for 30 min before stimulation with either 5 mg/kg LPS intravenously or normal saline. Plasma cytokines, plasma nitrite, renal inducible nitric oxide synthase (iNOS) and nuclear factor κB (NF-κB) activity were assessed 240 min after LPS or normal saline injection. Blood urea nitrogen (BUN), creatinine (Cr) and histopathological score for renal tubular damage were also measured. Electroacupuncture pretreatment significantly decreased LPS-induced plasma tumour necrosis factor-α and interleukin (IL)-1ß, increased plasma IL-10, and decreased plasma nitrite, renal iNOS and NF-κB activity. It also significantly decreased LPS-induced BUN, Cr and the renal histopathological score. These findings suggest that electroacupuncture pretreatment at the ST36 and PC6 acupoints attenuated the LPS-induced inflammatory response and mitigated acute kidney injury.

Qualitative analysis of the nonanthocyanin phenolics from 2 mulberry cultivars, da-10 and hongguo, was performed by the HPLC-DAD-ESI-MS/MS method. Six nonanthocyanin phenolics compounds were identified (procatechuic acid, chlorogenic acid, 4-caffeolyquinic acid, taxifolin, rutin, quercetin) and 3 others (3,5-diCQA, taxifolin-hexoside, kamepferol-hexoside) were tentatively identified. Quantification of nonanthocyanin phenolic was performed by HPLC-DAD, which revealed that rutin (111.38 and 90.79 microg/g FW) was the major nonanthocyanin phenolics in the 2 mulberry cultivars. Phenolic acids in cultivar da-10 (54.68 microg/g FW) were far more than in cultivar hongguo (14.93 microg/g FW). CQA and its isomers (40.02 microg/g FW) were the major phenolic acid in cultivar da-10. The nonanthocyanin phenolics in the 2 cultivars of mulberry had higher antiradical activities in superoxide anion and DPPH radical and antioxidant activity in beta-CLAMS assay than the Trolox. These results were relevant to not only the control of color stability and organoleptic characteristics of mulberry juice and wine but also the exploitation of the functional foods made from mulberry.

This study evaluated the developmental gene expression of lactoferrin (LF) and the effect of supplementary iron on gene expression of LF in the mammary gland of mice using semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) analysis. In experiment 1, a total of 12 female mice were used to determine the effect of different lactating stages on mRNA expression of LF. The Institute of Cancer Research mice were divided into 4 groups; each group of 3 mice was tested on d 1, 9, 17, and 25 of lactation. In experiment 2, 6 groups of mice (total of 24 female mice at d 12 after mating) were fed purified diets (without iron or supplement iron) and were assigned to 2 treatments (control and treatment). The experimental feeding period lasted 35 d. During the feeding experiment, 6 mice (3 animals in each group) were chosen on d 1, 9, 17, and 25 of lactation to determine the effect of iron on LF mRNA expression of mice at different stages of lactation. The results of experiment 1 showed that LF mRNA had strong expression on d 1 of lactation, decreased gradually on d 9 and 17 of lactation, and then increased again markedly on d 25 of lactation. These results imply that the expression of LF in the mammary gland at different lactating stages is consistent with the changes in LF concentrations in milk. Iron significantly increased LF mRNA expression on d 1 and 25 of lactation. Iron did not statistically increase LF gene expression on d 9 and 17 of lactation. These findings raised the possibility that iron supplementation may play a role in regulation of LF levels in vivo.

Poultry litter provides a rich source of nutrients for perennial forages, but the usual practice of surface-applying litter to pastures can degrade water quality by allowing nutrients to be transported from fields in surface runoff, while much of the NH4-N volatilizes. Incorporating litter into the soil can minimize such problems in tilled systems, but has not been used for perennial forage systems. In this study, we minimized disturbance of the crop, thatch, and soil structure by using a knifing technique to move litter into the root zone. Our objective was to determine effects of poultry litter incorporation on quantity and quality of runoff water. Field plots were constructed on a silt loam soil with well-established bermudagrass [Cynodon dactylon (L.) Pers.] and mixed grass forage. Each plot had 8 to 10% slopes, borders to isolate runoff, and a downslope trough with sampling pit. Poultry litter was applied (5.6 Mg ha(-1)) by one of three methods: surface-applied, incorporated, or surface-applied on soil-aeration cuts. There were six treatment replications and three controls (no litter). Nutrient concentrations and mass losses in runoff from incorporated litter were significantly lower (generally 80-95% less) than in runoff from surface-applied litter. By the second year of treatment, litter-incorporated soils had greater rain infiltration rates, water-holding capacities, and sediment retention than soils receiving surface-applied litter. Litter incorporation also showed a strong tendency to increase forage yield.

BACKGROUND: Inducible nitric oxide synthase (iNOS) is expressed by leukocytes and smooth muscle cells in atherosclerotic lesions. To test whether NO produced by iNOS deficiency affects atherosclerosis, we studied apoE/iNOS-double knockout (dKO) and apoE-knockout (KO) control animals fed a "Western-type" diet. METHODS AND RESULTS: After 16 weeks of Western-type diet, the aortic lesion area in apoE/iNOS-dKO males and females was significantly reduced, by 22% and 21%, respectively, compared with apoE-KO males and females. This effect was more pronounced after 24 weeks of Western-type diet, after which lesion formation in male and female dKO mice was reduced by 38% and 40%, respectively. Plasma levels of lipoperoxides in apoE/iNOS-dKO mice (2.0+/-0.23 micromol/L) were significantly lower than in apoE-KO control animals (3.2+/-0.44 micromol/L; P=0.02). To test whether substrate deficiency plays a role in the proatherogenic actions of iNOS, we administered L-arginine to apoE-KO animals for 16 and 24 weeks. L-Arginine treatment did not affect lesion formation in apoE-KO animals fed a Western-type diet. CONCLUSIONS: Genetic deficiency of iNOS decreases diet-induced atherosclerosis and lowers plasma levels of lipoperoxides, a marker for oxidative stress, in apoE-KO animals. Reduction in iNOS-mediated oxidative stress could partly explain protection from lesion formation in dKO animals. L-Arginine supplementation did not change lesion area in apoE-KO mice, indicating that substrate deficiency is not a likely cause for iNOS-mediated injury in this model of atherosclerosis.

A reporter gene sequence was constructed within a eukaryotic expression vector. The altered plasmid contained 2 sequential estrogen response elements (ERE) coupled to a human phosphoglycerate kinase (PGK) promoter inserted upstream from a cDNA sequence encoding enhanced green fluorescent protein (GFP) with a 3'-polyadenylation signal. The plasmid was linearized and transfected into MCF-7 cells, a human breast cancer-derived line that expresses the estrogen receptor (ER). No selectable marker was present in the plasmid, requiring stably transfected cells to be selected by fluorescence-activated cell sorting based on GFP expression after the cells were treated with 10(-9) M 17beta-estradiol (E2). Stably transfected MCF-7 cells (MCF7-ERE) exhibited 2000-3000 times more fluorescence at 488 nm excitation and 512 nm emission than non-transfected cells. MCF7-ERE cells exhibited a linear increase in GFP expression induced over a range of 10(-12) M E2, a concentration giving 2 times the background expression, to maximal expression at 3 x 0(-10) M E2. From the maximal level, GFP expression plateaued, and then declined when E2 was increased to the highest concentration tested, 10(-7) M. 4-Hydroxytamoxifen (TFN-OH) treatment of cells produced a dose-dependent inhibition of E2-induced GFP expression, indicating the interaction of ER in the regulation of GFP gene expression. A series of estrogenic chemicals were evaluated for their capacity to induce GFP expression in MCF7-ERE cells, showing induced expression of GFP at concentrations 2-4 log units higher than the E2 concentration giving maximal GFP expression. The ERE-PGK-GFP reporter gene system is capable of rapid GFP expression in the presence of low concentrations of E2, and of quantifying estrogenicity of chemicals compared with a standard curve of the natural ligand, 17beta-estradiol.

Qingkailing Oral Tonic and Shuanghuanglian Oral Tonic are two kinds of complex formlae of Chinese traditional patent medicine, and both contain Scutellaria baicalensis Georgi. Baicalin is referred to the main active component with functions of dispelling wind-heat to reduce fever and detoxicating, and takes effect on uppering respiratory tract infection and tonsillities. In this work a high performance capillary electrophoresis (HPCE) method was established for separation and quantitative analysis of baicalin in prescriptions containing Scutellaria baicalensis Georgi, based on the mode of capillary zone electrophoresis. Electrophoretic conditions are as follows: uncoated fused silica capillary (34.8/39.5 cm x 50 microns i.d.), 40 mmol/L borax buffer(pH 9.0), applied voltage 17 kV, polarity anode (inlet) to cathode (outlet), temperature 25 degrees C, detection wavelength 285 nm, injection 68.95 kPa x s. Under the optimum conditions, baicalin was separated successfully from other components within 15 minutes. The relative peak area of baicalin increased linearly with the increase of its concentration in the range of 10-640 mg/L. The recoveries were (100.31 +/- 1.98)% for Qingkailing Oral Tonic and (100.60 +/- 2.36)% for Shuanghuanglian Oral Tonic. The analytical results demonstrate the method is simple, rapid and well reproducible, and can be used as a reliable tool for the quality control of Chinese traditional medicines containing Scutellaria baicalensis Georgi.

A high performance capillary electrophoresis method has been developed for separation and quantitative analysis of glibenclamide in Xiaotangling tablets. Electrophoretic conditions were as follows: micellar electrokinetic capillary chromatography (MECC) mode, trimethoprimum as internal standard, uncoated fused silica capillary (34.8 cm/39.5 cm(effective/total length), 50 microns i.d.), 25 mmol/L borax-30 mmol/L SDS(pH 9.0), applied voltage 17 kV, (+)-->(-), temperature 28 degrees C, detection wavelength 228 nm, pressure injection 68.95 kPa.s. Glibenclamide was separated successfully from other components within 14 minutes under the optimum conditions, and the relative peak area of glibenclamide increased linearly with the increase of its concentration within the range of 25 mg/L-275 mg/L. The recovery was (100.6 +/- 1.4)%. The method is simple, rapid and well reproducible, and can be used as a reliable tool for the quality control of Chinese traditional medicine containing glibenclamide.

Yinhuang Granule is a kind of complex formulae made up of extract of Flos lonicerae and that of Radix scutellariae, of the fuctions of dispelling wind-heat to reduce fever, detoxicating, relieving inflammation, promoting urination and tranquilizimg the mind and can be applied to upper respiratory tract infection and tonsillitis. Baicalin and chlorogenic acid are referred to main effective components of it. In this paper, the separation and determination of baicalin and chlorogenic acid in Yinhuang Granule by capillary electrophoresis is described. The experimental conditions are stated as follow: separation column: bare fused-sillica capillary (50 microns i.d., 370 microns o.d., total length 47 cm, 40 cm from the inlet to UV detector); electrolyte: 25 mmol/L borax, pH 8.5; detection wavelength: 310 nm; sampling: 17 kPa.s; running voltage: 25 kV, polarity from anode (inlet) to cathode (outlelt); temperature: 25 degrees C; internal standard: p-nitrobenzoic acid. The linear ranges of determination for baicalin and chlorogenic acid were 160-960 mg/L(r = 0.9993, RSD = 1.76%-2.33%) and 80-960 mg/L(r = 0.9989, RSD = 1.07%-2.51%), respectively. The recoveries of baicalin and chlorogenic acid were (102.09 + 1.74)% (RSD = 1.71%, n = 6) and (99.81 + 3.11)% (RSD = 3.12%, n = 6). The method is simple, fast and accurate. It can be used for the quality control of Yinhuang Granule and its related Chinese medicines.

Anti-mutagenic effects of some Chinese herbal medicine and green tea antagonizing cigarette tar was studied with unscheduled DNA synthesis (UDS) test in human peripheral lymphocytes in order to exploit the better use of Chinese herbal resources. Results showed that Scutellaria barbata, Hedyotis diffusa wildi, Xihuangwan, green tea and tea polyphenol all had anti-mutagenic effects, to some extent. And, 125 g/L of Scutellaria barbata, 125 g/L Hedyotis diffusa wildi, 325 g/L Xihuangwan, 78 g/L green tea and 25 g/L tea polyphenol could inhibit obviously the damage to DNA in lymphocytes caused by the total particle material (TPM) extracted from cigarette tar. If the Chinese herbal medicine and green tea were injected into cigarettes beforehand, extracted TPM (containing herbal medicine and green tea) also could protect DNA in lymphocytes from damage caused by cigarette tar, to even greater extent. This result is basically consistent with other anti-mutagenesis tests.

RESUMO

Effects of digital acupressure on the blood flow in the capillary arterioles of the cerebral pia mater covering the posterior sigmoidal gyrus in domestic dog (Canis familiaris) were observed. The speed of blood flow was found increased to 185% after continuous digital acupressure for 20 minutes. The increase was most prominent 15 minutes after the acupressure was completed; the effect subsided in 30 minutes. It was also found that in cases of low basal speed of flow before digital acupressure, the increase in the speed of blood flow after acupressure was more pronounced.
