RESUMO

Nanotechnology has many potential applications in cancer treatment. For example, nano-drug delivery systems (NDDS) with high bioavailability, biodegradability, and biocompatibility have been developed, in order to increase the therapeutic effects of anticancer drugs. Among these NDDS, high-performance hydroxyapatite (HA) nanoparticles are rapidly advancing in the targeted cancer treatment due to their numerous benefits. Curcumin is an herbal metabolite that acts as a chemical inhibitor through the inhibition of tumor cells and the progression of many cancers. However, the poor bioavailability of curcumin is the most important challenge in using this substance. In this study, HA nanoparticles coated by chitosan were used as a pH-sensitive biopolymer to improve the efficiency and bioavailability of curcumin. For this purpose, HA nanoparticles were first synthesized by the sol-gel method. Then, a layer of chitosan was coated on it, and the curcumin drug was encapsulated in the nanocarrier, under controlled conditions. Techniques such as scanning electron microscopy (SEM), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR) were used to characterize the nanocarriers. In the second part, nano-drugs prepared by various bioassays were examined. For this purpose, the rate of cytotoxicity by the methyl-thiazol-tetrazolium (MTT) assay and the rate of apoptosis induction by the acridine orange and ethidium bromide (AO/EB) staining method on the brain carcinoma U87MG cell line were investigated.

ABSTRACT The present study was designed to evaluate the in vivo effect of Allium sativum (garlic) hydroalcoholic extract on wound healing in rats. For this purpose, 72 mature Wistar rats were divided into four groups (n=18/each) to receive no treatment, placebo, Cicalfate(r), or 2% Allium sativum (AS) extract, administered topically to the wound area, for 21 days. Following the experimental period, tissue samples were dissected out and underwent to histopathological analyses. Fibroblasts, fibrocytes, mast cells, intra-cytoplasmic carbohydrate ratio, neovascularization, collagen deposition, and re-epithelialization were analyzed in all groups. Animals in the treated groups showed significant enhancement in fibroblast, fibrocyte, and mast-cell distribution. Significantly higher neovascularization was observed on day 3 after wound induction in AS-treated animals versus those in the placebo, Cicalfate, and untreated groups (P<0.05). A dose-dependent, significantly higher intra-cytoplasmic carbohydrate storage was observed in treated animals. Our data show that AS promotes wound healing due to its preliminary impact on mast-cell distribution, which enhanced collagen synthesis and upregulated angiogenesis, and shortened the healing process by enhancing the intra-cytoplasmic carbohydrate ratio.

Tendon repair is still one of the challenges for rehabilitation. Various treatments for tendon injuries have been used in recent decade. This study was established to investigate the effects of low-level laser therapy (LLLT), platelet-rich plasma (PRP) treatment alone, and using combined method on the healing of Achilles tendon in rabbits. Seventy-two healthy mature male white New Zealand rabbits were divided randomly into four groups of 18 animals each: control: partial tenotomy with no treatment, only 1 mL normal saline was injected on days 1, 8, and 15 at the site of splitting; PRP: partial tenotomy with PRP treatment on days 1, 8, and 15 at the site of splitting; LLLT: partial tenotomy with LLLT (K30 hand-held probe, AZOR, Technica, Russia, 650 nm, 30 mW, surface area = 1 cm(2), 60 S/cm(2), energy density = 1.8 J/cm(2)) for 15 consecutive days; LLLT + PRP: partial tenotomy with LLLT + PRP. At the end of trial, the rabbits were euthanatized and tendon specimens were harvested and were submitted for histopathological evaluation, hydroxyproline levels, and biomechanical measurement. The Tukey post hoc test was performed. The results for these parameters showed that PRP or LLLT alone has significant advantages over untreated animals (P < 0.05). Furthermore, it was found that the combined treatment with PRP and LLLT is even more efficient. There was no significant difference (P > 0.05) between the two groups of LLLT and PRP. However, the treatments combining PRP and LLLT showed significant results in comparison of PRP or LLLT alone (P < 0.05). Our results demonstrate that the healing time of injured tendon decreases by using the two therapies combined.
