Soil biogeochemical cycles are essential for regulating ecosystem functions and services. However, little knowledge has been revealed on microbe-driven biogeochemical processes and their coupling mechanisms in soil profiles. This study investigated the vertical distribution of soil functional composition and their contribution to carbon (C), nitrogen (N) and phosphorus (P) cycling in the humus horizons (A-horizons) and parent material horizons (C-horizons) in Udic and Ustic Isohumosols using shotgun sequencing. Results showed that the diversity and relative abundance of microbial functional genes was influenced by soil horizons and soil types. In A-horizons, the relative abundances of N mineralization and liable C decomposition genes were significantly greater, but the P cycle-related genes, recalcitrant C decomposition and denitrification genes were lower compared to C-horizons. While, Ustic Isohumosols had lower relative abundances of C decomposition genes but higher relative abundances of N mineralization and P cycling-related pathways compared to Udic Isohumosols. The network analysis revealed that C-horizons had more interactions and stronger stability of functional gene networks than in A-horizons. Importantly, our results provide new insights into the potential mechanisms for the coupling processes of soil biogeochemical cycles among C, N and P, which is mediated by specific microbial taxa. Soil pH and carbon quality index (CQI) were two sensitive indicators for regulating the relative abundances and the relationships of functional genes in biogeochemical cycles. This study contributes to a deeper understanding of the ecological functions of soil microorganisms, thus providing a theoretical basis for the exploration and utilization of soil microbial resources and the development of soil ecological control strategies.

BACKGROUND: Osteoarthritis (OA) is a common chronic age-related joint disease characterized primarily by inflammation of synovial membrane and degeneration of articular cartilage. Accumulating evidence has demonstrated that Danggui-Shaoyao-San (DSS) exerts significant anti-inflammatory effects, suggesting that it may play an important role in the treatment of knee osteoarthritis (KOA). METHODS: In the present study, DSS was prepared and analyzed by high-performance liquid chromatography (HPLC). Bioinformatics analyses were carried out to uncover the functions and possible molecular mechanisms by which DSS against KOA. Furthermore, the protective effects of DSS on lipopolysaccharide (LPS)-induced rat chondrocytes and cartilage degeneration in a rat OA model were investigated in vivo and in vitro. RESULTS: In total, 114 targets of DSS were identified, of which 60 candidate targets were related to KOA. The target enrichment analysis suggested that the NF-κB signaling pathway may be an effective mechanism of DSS. In vitro, we found that DSS significantly inhibited LPS-induced upregulation of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), interleukin-6 (IL-6), matrix metalloproteinase-3 (MMP3), and matrix metalloproteinase-13 (MMP13). Meanwhile, the degradation of collagen II was also reversed by DSS. Mechanistically, DSS dramatically suppressed LPS-induced activation of the nuclear factor kappa B (NF-κB) signaling pathway. In vivo, DSS treatment prevented cartilage degeneration in a rat OA model. CONCLUSIONS: DSS could ameliorate the progression of OA through suppressing the NF-κB signaling pathway. Our findings indicate that DSS may be a promising therapeutic approach for the treatment of KOA.

ETHNOPHARMACOLOGICAL RELEVANCE: Clinical research and basic scientific experiments have shown that modified Xiaoyaosan (MXYS) has antidepressant effects, whose system mechanism however has not been thoroughly characterized. AIM OF THE STUDY: This research was aimed at evaluating the treatment effects of MXYS on chronic unpredictable mild stress (CUMS)-induced depressive mice and exploring underlying mechanisms. MATERIALS AND METHODS: Whether MXYS has effects on depression was investigated via the depressive behaviors of mice, electron microscopy, real-time quantitative polymerase chain reaction (RT-qPCR), Western blot analysis, immunofluorescence (IF) staining and the stereotaxic injection of adeno-associated viruses (AAVs). In addition, network pharmacology was applied to predict relevant molecular targets and possible mechanisms and perform further in vivo validation. RESULTS: MXYS is effective in ameliorating the depression-like symptoms of CUMS mice. It can stimulate autophagosome formation, activate the expression of microtubule-associated protein 1 light chain 3 (LC3B), autophagy-related gene 5 (Atg5), Atg7 and neuron-specific nuclear protein (NeuN), and decrease the protein expression sequestosome 1 (SQSTM1/p62). The autophagy-upregulating effect of MXYS was weakened by silencing. The network pharmacology analysis revealed that mitogen-activated protein kinase 1 (MAPK1), MAPK3, serine/threonine-protein kinase (AKT1), proto-oncogene tyrosine-protein kinase (SRC), PI 3 kinase p85 alpha (PIK3R1), catenin (cadherin-associated protein) beta 1 (CTNNB1) and human thrombin activator 1 (HRAS) may be of importance to treat depression by MXYS. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis revealed that metabolic and autophagy pathways, pathways in cancer and MAPK, phosphoinositide 3-kinase (PI3K)-Akt and rhoptry-associated protein 1 (Rap1) signaling pathways are involved in the antidepressant effects of MXYS. As suggested by Western blot, the anti-depression mechanism of MXYS is possibly associated with the extracellular signal-regulated protein kinase (ERK)/P38 MAPK signaling pathway. CONCLUSION: The findings indicate the possible antidepressant effects of MXYS on CUMS mice via triggering autophagy to alleviate neuronal apoptosis and prompting autophagy, which may involve the ERK/P38 MAPK signaling pathway.

Woody plants possess great potential for phytoremediation of heavy metal-contaminated soil. A pot trial was conducted to study growth, physiological response, and Cd and Pb uptake and distribution in black locust (Robinia pseudoacacia L.), as well as the rhizosphere bacterial communities in Cd and Pb co-contaminated soil. The results showed that R. pseudoacacia L. had strong physiological regulation ability in response to Cd and Pb stress in contaminated soil. The total chlorophyll, malondialdehyde (MDA), soluble protein, and sulfhydryl contents, as well as antioxidant enzymes (superoxide dismutase, peroxidase, catalase) activities in R. pseudoacacia L. leaves under the 40 mg·kg-1 Cd and 1000 mg·kg-1 Pb co-contaminated soil were slightly altered. Cd uptake in R. pseudoacacia L. roots and stems increased, while the Pb content in the shoots of R. pseudoacacia L. under the combined Cd and Pb treatments decreased in relative to that in the single Pb treatments. The bacterial α-diversity indices (e.g., Sobs, Shannon, Simpson, Ace, and Chao) of R. pseudoacacia L. rhizosphere soil under Cd and Pb stress were changed slightly relative to the CK treatment. However, Cd and Pb stress could significantly (p < 0.05) alter the rhizosphere soil microbial communities. According to heat map and LEfSe (Linear discriminant analysis Effect Size) analysis, Bacillus, Sphingomonas, Terrabacter, Roseiflexaceae, Paenibacillus, and Myxococcaceae at the genus level were notably (p < 0.05) accumulated in the Cd- and/or Pb-contaminated soil. Furthermore, the MDA content was notably (p < 0.05) negatively correlated with the relative abundances of Isosphaeraceae, Gaiellales, and Gemmatimonas. The total biomass of R. pseudoacacia L. was positively (p < 0.05) correlated with the relative abundances of Xanthobacteraceae and Vicinamibacreraceae. Network analysis showed that Cd and Pb combined stress might enhance the modularization of bacterial networks in the R. pseudoacacia L. rhizosphere soil. Thus, the assembly of the soil bacterial communities in R. pseudoacacia L. rhizosphere may improve the tolerance of plants in response to Cd and/or Pb stress.

The oxide-zeolite (OXZEO) catalyst design concept has been demonstrated in an increasing number of studies as an alternative avenue for direct syngas conversion to light olefins. We report that face-centered cubic (FCC) MnGaOx -Spinel gives 40 % CO conversion, 81 % light olefins selectivity, and a 0.17 g gcat -1 h-1 space-time yield of light olefins in combination with SAPO-18. In comparison, solid solution MnGaOx (characterized by Mn-doped hexagonal close-packed (HCP) Ga2 O3 ) with a similar chemical composition gives a much inferior activity, i.e., the specific surface activity is one order of magnitude lower than the spinel oxide. Photoluminescence (PL), in situ Fourier-transform infrared (FT-IR), and density functional theory (DFT) calculations indicate that the superior activity of MnGaOx -Spinel can be attributed to its higher reducibility (higher concentration of oxygen vacancies) and the presence of coordinatively unsaturated Ga3+ sites, which facilitates the dissociation of the C-O bond via a more efficient ketene-acetate pathway to light olefins.

The present study examined potential association between the daily intake and serum levels of copper (Cu), selenium (Se), and zinc (Zn) and the risk of osteoarthritis (OA) and rheumatoid arthritis (RA) using data from the National Health and Nutrition Examination Survey (NHANES). Daily intake and serum concentrations of Cu, Zn, and Se in 4200 adults from the 2011-2016 NHANES were examined and divided into normal, OA patients, and RA patients. The level of serum Cu was higher in OA and RA than in non-arthritis, while the levels of serum Se and Zn were no different in the three groups. Serum Se and Zn, but not Cu, concentrations were highly correlated with daily intake. Cu, Se, and Zn intake was independently associated with increased risk of OA, but not with RA. And there was a trend for higher odds of OA among participants in the higher Cu, Se, and Zn intake. Future large longitudinal studies are warranted to confirm these findings.

Under ongoing global change, whether grassland ecosystems can maintain their functions and services depends largely on their stability. However, how ecosystem stability responds to increasing phosphorus (P) inputs under nitrogen (N) loading remains unclear. We conducted a 7-year field experiment to examine the influence of elevated P inputs (ranging from 0 to 16 g P m-2 yr-1) on the temporal stability of aboveground net primary productivity (ANPP) under N addition of 5 g N·m-2·yr-1 in a desert steppe. We found that under N loading, P addition altered plant community composition but did not significantly affect ecosystem stability. Specifically, with the increase in the P addition rate, declines in the relative ANPP of legume could be compensated for by an increase in the relative ANPP of grass and forb species, yet community ANPP and diversity remained unchanged. Notably, the stability and asynchrony of dominant species tended to decrease with increasing P addition, and a significant decrease in legume stability was observed at high P rates (>8 g P m-2 yr-1). Moreover, P addition indirectly affected ecosystem stability by multiple pathways (e.g., species diversity, species asynchrony, dominant species asynchrony, and dominant species stability), as revealed by structural equation modeling results. Our results suggest that multiple mechanisms work concurrently in stabilizing the ecosystem stability of desert steppes and that increasing P inputs may not alter desert steppe ecosystem stability under future N-enriched scenarios. Our results will help improve the accuracy of vegetation dynamics assessments in arid ecosystems under future global change.

OBJECTIVE: To compare the clinical efficacy of scraping needling technique combined with western medication and simple western medication for neurogenic tinnitus of kidney essence deficiency. METHODS: A total of 68 patients with neurogenic tinnitus of kidney essence deficiency were randomly divided into an observation group and a control group, 34 cases in each group. In the control group, oral methylcobalamin tablets were given, 0.5 mg each time, 3 times a day; oral flunarizine hydrochloride capsules were given before bed, 5 mg each time, once a day, 4 weeks in total. On the basis of the treatment as the control group, scraping needling technique was applied at Tinghui (GB 2), Yifeng (TE 17), Yangchi (TE 4) on the affected side and Shenshu (BL 23), Lieque (LU 7), 5 min each acupoint, once a day, 5 times a week for 4 weeks. Before treatment, 2, 4 weeks into treatment and 4 weeks after treatment (follow-up), the tinnitus severity score, tinnitus visual analogue scale (VAS) score and pure tone average (PTA) were observed, and the clinical efficacy was evaluated in the two groups. RESULTS: The tinnitus severity scores, VAS scores and PTA of each time point after treatment in the two groups were lower than those before treatment (P<0.05), and those in the observation group were lower than the control group (P<0.05). The total effective rates of each time point after treatment in the observation group were 50.0% (17/34), 79.4% (27/34), 79.4% (27/34), which were higher than 26.5% (9/34), 64.7% (22/34), 61.8% (21/34) in the control group (P<0.05). CONCLUSION: Scraping needling technique combined with western medication could improve tinnitus severity, tinnitus volume and hearing in patients with neurogenic tinnitus of kidney essence deficiency, and its curative effect is better than simple western medication.

Mulberry (Morus alba L.) is a flowering tree traditionally used in Chinese herbal medicine. Mulberry leaf flavonoids (MLFs) have been reported to exert important anti-inflammatory and antioxidant properties. The purpose of this study was to select the MLF with the best anti-inflammatory and antioxidative activities from MLFs eluted by different ethanol concentrations (30%, 50%, and 75%) and explore its pharmacological properties. Three types of MLFs inhibited the production of nitric oxide (NO), prostaglandin E2 (PGE2), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and inflammatory cytokines in lipopolysaccharide (LPS)-induced RAW 264.7 cells. All MLFs boosted the antioxidative capacity by decreasing the reactive oxygen species (ROS) production and the scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals and improving the metal ion chelating activity and reducing power. The results revealed that the MLFs eluted by 30% ethanol exhibited the best anti-inflammatory and antioxidative activities. A nontargeted metabolomic analysis was used to analyze 24 types of differential flavonoids between the MLFs. Quercetin, kaempferol, and their derivatives in 30%MLF were more abundant than the other two MLFs. Furthermore, we evaluated the pharmacological activities of 30%MLF in dextran sodium sulfate (DSS)-induced ulcerative colitis (UC) mice. The 30%MLF could alleviate the clinical symptoms, reduce the secretion of inflammatory cytokines, and inhibit the activation of the inflammatory pathway in DSS-induced colitis mice. This study will provide valuable information for the development of MLFs eluted by 30% ethanol as a functional food.

Phosphorus (P) is an essential nutrient element for crop growth. The effects of P surplus or deficit on Cd absorption and transport in rice in Cd-polluted farmland is not clear. The effects of P deficiency and P sufficiency on Cd uptake, transport, and accumulation in rice under Cd stress were investigated by applying different levels of phosphorus (NaH2PO4) in a hydroponic experiment. The results showed that:① with the increase in ρ(P) (1.5-48.0 mg·L-1), the biomass in all parts of the rice plants had no obvious change, and the contents of photosynthetic pigment (chlorophyll a, chlorophyll b, and carotenoid) firstly ascended and then descended; high concentrations of P inhibited the synthesis of photosynthetic pigments. ② Under Cd stress, when the P was deficient (1.5-6.0 mg·L-1) or sufficient (12.0-48.0 mg·L-1), the Cd content in different parts of the rice increased with the increase in P addition level, and the maximum increase in Cd content in brown rice was 132.1% and 191.2%, respectively. ③ The P/Cd of brown rice showed a piecewise decreasing rule under P deficiency and P sufficiency, and the Cd content in brown rice was significantly negatively correlated with P/Cd (P<0.01). These results indicated that elevating phosphorus concentration when rice was under both the conditions of P deficiency and P sufficiency could promote the uptake and transport of Cd by rice roots under Cd stress, thus increasing the accumulation of Cd in aboveground parts and the risk of excessive Cd in rice.

Background: A national health education program in impoverished counties to promote health literacy among rural populations was released by the Chinese government in 2018. Under this nationwide campaign, an integrated health education program was implemented in Yunnan province, which included additional culturally sensitive educational components for the severely impoverished prefectures. Objective: This study examined the differential effects of the health education program models on health literacy outcomes among residents in poverty-stricken areas. Methods: A quasi-experimental design was applied with two arms that included surveys at baseline (in October 2019) and endline (in June 2021) to collect a range of individual-level health information, including the Chinese Resident Health Literacy Scale. The intervention group received the national health education program with the additional Yunnan specific program; the control group received only the national program. Respondents were recruited via a multi-stage stratified sampling, including 641 participants at baseline (261 from the intervention sites and 380 from the control sites) and 693 participants at endline (288 from the intervention sites and 405 from the control sites). Chi-square and logistic regression analyses were performed to examine the association between program intervention and health literacy outcomes. Results: The overall health literacy levels were low (1.87%) at baseline, and there was no statistically significant difference between two groups (1.92 vs. 1.84%, P = 1.000). A significant increase (from 1.87 to 11.11%, P < 0.001) in the health literacy level was observed at endline in both groups. The magnitude of increase was significantly greater in the intervention group relative to the control group (17.71 vs. 6.42%, P < 0.001). Adjusting for the confounding factors of individual and household characteristics, results from multivariate logistic regression revealed that the odds of having adequate health literacy among participants who received both the National Program and the Yunnan Program were 3.92 times higher than those who only received the National Program (95% CI: 2.10-7.33). Conclusion: The findings highlighted the importance of incorporating non-verbal visual aids and culturally-sensitive media tools in health literacy education to address healthy lifestyle and the living contexts of the populations in poverty-stricken areas.

ABSTRACT: The monoterpene glycoside paeoniflorin (PF) is the principal active constituent of the traditional Chinese herbal medicines, Radix Paeoniae Alba and Radix Paeoniae Rubra, which have been used for millennia to treat cardiovascular diseases (eg, hypertension, bleeding, and atherosclerosis) and neurological ailments (eg, headaches, vertigo, dementia, and pain). Recent evidence has revealed that PF exerts inhibitory effects on inflammation, fibrosis, and apoptosis by targeting several intracellular signaling cascades. In this review, we address the current knowledge about the pharmacokinetic properties of PF and its molecular mechanisms of action. We also present results from recent preclinical studies supporting the utility of PF for the treatment of pain, cerebral ischemic injury, and neurodegenerative diseases, such as Alzheimer's and Parkinson's diseases. Moreover, new evidence suggests a general protective role of PF in heart attack, diabetic kidney, and atherosclerosis. Mechanistically, PF exerts multiple anti-inflammatory actions by targeting toll-like receptor-mediated signaling in both parenchymal and immune cells (in particular, macrophages and dendritic cells). A better understanding of the molecular actions of PF may lead to the expansion of its therapeutic uses.

In this study, we investigated the effects of exogenous phosphorus on the accumulation and transportation of cadmium in rice plants through a hydroponic experiment. In the experiment, the rice variety was Huanghuazhan, P solution concentrations were 10.0-45.0 mg ·L-1 that was made using NaH2PO4, and Cd solution concentrations were 0.1 mg ·L-1 and 0.2 mg ·L-1. The results showed that: ① the biomass in all parts of rice plants and contents of photosynthetic pigment (chlorophyll a, chlorophyll b, and carotenoid) increased gradually with an increase in exogenous P. ② Content of Cd in rice stems, leaves, husk, and brown rice increased gradually with an increase in the amounts of exogenous P. The content of Cd in brown rice increased by 2.8%-22.8% and 40.9%-61.8% when treated with Cd concentrations of 0.1 mg ·L-1 and 0.2 mg ·L-1 in hydroponic solutions, respectively. ③ Cd accumulation in rice plants was accelerated due to the application of exogenous P. Cd accumulating amounts increased from 395.1 µg ·plant-1 to 542.6 µg ·plant-1 and 639.6 µg ·plant-1 to 1082.0 µg ·plant-1 when treated with Cd concentrations of 0.1 and 0.2 mg ·L-1 in hydroponic solutions, respectively. ④ With an increase in the applied amounts of exogenous P, the P/Cd quality ratio in rice roots increased, while those in rice stems, leaves, husks, and brown rice decreased; meanwhile, the Cd transfer coefficients from root to stem (TFroot-stem) and stem to leaf (TFstem-leaf) increased. This showed that there was a certain synergistic effect between P and Cd in the rice parts. Finally, the application of exogenous P promoted the transfer of Cd from the rice root to other rice tissues, resulting in a synergistic effect on Cd accumulation and transportation in various rice tissues and increased Cd contents in brown rice.

OBJECTIVE: To evaluate the analgesic effect of vertebral cancellous bone infiltration anaesthesia during percutaneous vertebroplasty (PVP). METHODS: Patients treated with vertebral cancellous bone infiltration anaesthesia (intervention group) or local anaesthesia alone (control group) during PVP at our institution during 2016-2018 were reviewed. The visual analogue scale (VAS) score before the operation, during establishment of the puncture channel, during pressure changes in the vertebral body (e.g., when removing or inserting pushers or needle cores), during bone cement injection, immediately after the operation, and at 2 h and 1 day postoperatively were compared between the groups. The patient's satisfaction with the operation was recorded and compared between groups. RESULTS: A total of 112 patients were enrolled (59 cases in the intervention group and 53 cases in the control group). There was no difference in the VAS score between the groups before the operation or during establishment of the intraoperative puncture channel (P > 0.05). The VAS score in the intervention group was significantly lower than that in the control group during pressure changes in the vertebral body (removal or insertion of puncture needle cores or pushers) and bone cement injection (P < 0.05). Immediately after the operation and at 2 h postoperatively, the pain in the intervention group was also significantly lower than that in the control group (P < 0.05), but there was no significant difference between the groups at 1 day postoperatively (P > 0.05). The patient satisfaction rate was 88% (52/59) in the intervention group and 67% (35/53) in the control group (P < 0.05). CONCLUSIONS: Vertebral cancellous bone infiltration anaesthesia may effectively relieve intraoperative pain and improve the surgical experience of patients without affecting the clinical effect of surgery.

An in-situ paddy experiment was conducted to study the remedying effects of a combined amendment(calcium carbonate+diatomite+ferric sulfate) on Cd bioavailability in soil and Cd accumulation in rice for spring and autumn rice planted in a certain Cd contaminated paddy soil in Beishan Town, Changsha, Hunan Province. The results showed that:Application of the combined amendment significantly increased soil pH and CEC values in both cultivated seasons, but decreased soil OM contents for the spring rice. During the experiment, the contents of TCLP-extractable Cd and CaCl2-extractable Cd in soil were reduced by 18.0%-33.0% and 5.4%-57.9%, respectively. The Cd contents in brown rice, husk, leave and root for the spring rice decreased by 29.6%-56.1%, 52.1%-54.0%, 18.1%-80.7% and 24.4%-41.6%, respectively, due to application of the combined amendment. There were significant differences in Cd transport capacity between spring rice and autumn rice, and the transportation of soil Cd in autumn rice was smoother than that in spring rice. For the autumn rice, the effects of the combined amendment were not significant(P>0.05), except that the Cd content in husk increased a little after applying 2 g·kg-1 of the combined amendment. Significant positive correlations were found between the Cd contents in brown rice and the contents of TCLP-extractable Cd or CaCl2-extractable Cd in soil. Application of the combined amendment effectively inhibited Cd uptake by rice root and reduced Cd content in brown rice for the spring rice; however, these remedying effects were significantly weakened for the autumn rice due to loss of the combined amendment with the movement of irrigation water in the spring season. Therefore, we suggest that supplement of the combined amendment is necessary between two rice cultivated seasons.

The research of plant ecological stoichiometry characteristics, nutrients distribution and their changes is of great significance to explain the response and adaptation of plants to environmental change. Leaves, root and soil from eight different abandoned years in Yanhe River basin were selected to study the content, characteristic ratio and distribution of carbon ( C) , nitrogen (N) , phosphorus (P), potassium (K). The results showed that the C, N, P, K contents of plant leaves were 444.21, 22.34, 1.49, 14.66 mg · g⁻¹ respectively, the C/N, C/P, C/K, N/P ratios of plant leaves were 21.86, 424.72, 39.82, 20.27 respectively; the C, N, P, K contents of root were 285.16, 5.79, 0.27, 6.07 mg · g⁻¹ respectively, the C/N, C/P, C/K, N/P ratios of root were .60. 56, 1019.33, 46.55, 21.36 respectively; the C, N, P, K contents of soil were 2.28, 0.18, 0.28, 4.33 mg · g⁻¹ respectively, the C/N, C/P, C/K, N/P ratios of soil were 16.43, 8.40, 0.54, 0.66 respectively. During the abandoned year of 1-35, C content of leaves increased, N content increased and then declined, P content declined overall, K content declined and then increased. The C/N, C/P, C/K, N/P ratios of plant leaves showed a rising trend overall. The changing pattern of root was different from that of leaves. Along with the increasing rehabilitation age, C and N contents of soil increased, P content changed as arc-sin function, K content changed as parabola, C/N decreased, C/P, C/K, N/P increased. With the increase of Abandoned Years, the ratio of C, P, K contents in leaves and root decreased, the ratio of C, N, P contents in leaves and soil decreased, the ratio of C, N contents in root and soil decreased. Corresponding relationship and its intension between different abandoned years and plant nutrient limit status and its allocation pattern were different.

Studies have indicated that trichosanthin (TCS), a bioactive protein extracted and purified from the tuberous root of Trichosanthes kirilowii (a well­known traditional Chinese medicinal plant), produces antitumor effects on various types of cancer cells. However, the effects of TCS on glioma cells are poorly understood. The objective of this study was to investigate the antitumor effects of TCS on the U87 and U251 cell lines. The in vitro effects of TCS on these two cell lines were determined using a Cell Counting Kit­8 (CCK­8) assay, Annexin V­FITC staining, DAPI staining, Transwell assays, terminal deoxynucleotidyl transferase­mediated dUTP nick end­labeling (TUNEL) assays, 5,5',6,6'­tetrachloro­1,1',3,3'­tetraethyl­imidacarbocyanine iodide (JC­1) staining and western blotting, which was utilized to assess the expression of leucine­rich repeat­containing G protein­coupled receptor 5 (LGR5) and key proteins in the Wnt/ß­catenin signaling pathway. Our data indicated that TCS inhibited the proliferation of glioma cells in a dose­ and time­dependent manner and played a role in inhibiting glioma cell invasion and migration. Additional investigation revealed that the expression levels of LGR5 and of key proteins in the Wnt/ß­catenin signaling pathway were markedly decreased after TCS treatment. The results suggest that TCS may induce apoptosis in glioma cells by targeting LGR5 and repressing the Wnt/ß­catenin signaling pathway. In the future, in vivo experiments should be conducted to examine the potential use of this compound as a novel therapeutic agent for gliomas.

Fine roots (≤ 2 mm) play a major role in biogeochemical cycling in ecosystems. By the methods of soil cores and ingrowth soil cores, we studied the biomass and annual production of fine roots in 0-40 cm soil layers of four main vegetation types, i. e. , Robinia pseudoacacia plantation, deciduous shrubs, abandoned grassland, and Artemisia desertorum community in Loess Plateau, China. The spatial patterns of fine root biomass and production were negatively associated with latitudes. The fine root biomass in the 0-40 cm soil layer was in the order of deciduous shrubs (220 g · m(-2)), R. pseudoacacia plantation (163 g · m(-2)), abandoned grassland (162 g · m(-2)) and A. desertorum community (79 g · m(-2)). The proportion of ≤ 1 mm fine root biomass (74.1%) in the 0-40 cm soil layer of abandoned grassland was significantly higher than those in the other three vegetation types. The fine root biomass of the four vegetation types was mainly distributed in the 0-10 cm soil layer and decreased with soil depth. The proportion of fine root biomass (44.1%) in the 0-10 cm soil layer of abandoned grassland was significantly higher than those in other three vegetation types. The fine root productions of four vegetation types were in the order of abandoned grassland (315 g · m(-2) · a(-1)) > deciduous shrubs (249 g · m(-2) a(-1)) > R. pseudoacacia plantation (219 g · m(-2) · a(-1)) > A. desertorum community (115 g · m(-2) · a(-1)), and mainly concentrated in the 0-10 cm top soil layer and decreased with the soil depth. The proportion of the annual production (40.4%) in the 0-10 cm soil layer was the highest in abandoned grassland. Fine roots of abandoned grassland turned over faster than those from the other three vegetation types.

OBJECTIVE: To study the effect of taspine hydrochloride (TA/HCl) on skin wound healing in rats and its mechanism. METHODS: Bilateral round wounds were made on the backs of SD rats. The effect of TA/HCl on the skin wound was evaluated through determining closure time and contracting ability of the skin wound, observing histopathological characteristics and measuring contents of hydroxyproline (Hyp) and protein in the wound tissue. RESULTS: The closure time of the skin wounds was significantly shorter in the TA/HCl-treated groups than that in the model group. The percentages of wound contraction were higher in the TA/HCl-treated groups than that in the dimethyl sulfoxide (DMSO) control group of the same group (P<0.05 or P<0.01) on the 3rd to 14th days after wounding. The content of the protein in the wound tissue in the TA/HCl-treated group (2 mg/ml) was higher than that in the model group (P<0.05) on the 3rd to 7th days after wounding, and it arrived at the peak on the 7th day and gradually decreased to the normal level in skin tissue on the 14th to 21st days after wounding. The contents of Hyp in the wound tissues in the TA/HCl-treated groups were higher than that in the model group (P<0.05 or P<0.015) on the 3rd to 21st days after wounding, and they arrived at the peak on the 14th day and at the normal level in skin tissue on the 21st day. Histopathological test results showed that TA/HCl could promote the formation of newly born capillaries in the early period of the wound healing. CONCLUSION: TA/HCl has the ability of promoting skin wound healing in rats, and it can also accelerate the growth of newly born capillaries and raise the production of protein and collagen in wound tissue.
