OBJECTIVE: To observe the effect of electroacupuncture (EA) at "Zusanli"(ST36) on the phosphorylated tau levels in pancreas and hippocampus of type 2 diabetes mellitus (T2DM) ratsï¼so as to explore the underlying mechanism of EA in diabetic demention rats. METHODS: Forty-eight male Sprague-Dawley rats were randomly divided into control, model and EA groups, with 16 rats in each group. The T2DM model was established by 6 weeks of high-fat, high-sugar diet as well as intrape-ritoneal injection of streptozocin (STZ) solution (35 mg/kg). After that, EA (2 Hz, 0.1 mA) was applied to unilateral "Zusanli"(ST36) for 30 min, once a day, 6 times a week for 4 weeks. The survival rate was recorded every week, and the fasting blood glucose (FBG) was detected on the 1st, 6th and 11th week. The level of serum insulin (INS) was measured by using ELISA. The morphological structure of pancreas islet was observed by H.E. staining. The expressions of phosphorylated tau at the sites of Ser 396 (pS396) and Thr 231 (pT231), total tau (Tau5), phosphorylated glycogen synthase kinase-3ß (pGSK-3ß) and total glycogen synthase kinase-3ß (GSK-3ß) in pancreas and hippocampus were detected by Western blot. The expression and distribution of pS396 and pT231 in pancreas and hippocampus were assayed with immunohistochemistry. RESULTS: Compared with the control group, the survival rate presented a significant decline, the contents of FBG and INS were obviously higher(P<0.01), and the structure of the pancreas islet appeared shrunken, obscure and disordered in the model group. Furthermore, the levels of pS396, pT231 in pancreas and hippocampus were obviously higher in the model group(P<0.01)ï¼while the level of pGSK-3ß in pancreas and hippocampus was significantly lower in the model group(P<0.01). In comparison with the model group, the survival rate of EA group was higher. Following 4 weeks' interventions, the enhanced levels of tau phosphorylation and GSK-3ß activity in pancreas and hippocampus were partly reversed in the EA group compared to the model group(P<0.05ï¼P<0.01). CONCLUSION: EA at ST36 can reduce the level of tau phosphorylation via regulating the activity of GSK-3ß in the pancreas and hippocampus of T2DM rats, which may be related with the effect of EA on the brain function in T2DM rats.

To further study and fully exploit the medicinal plant Sophora alopecuroides, the molecular markers related with the phenotypic traits of alkaloid content in S. alopecuroides should be detected. In this study, SSR molecular markers were used to analyze the genetic diversity and genetic structure of 23 S. alopecuroides populations, in combination with the association analysis between molecular markers and the alkaloid contents. The results showed that P, H, I, G_(st) and N_m values were 40.10%, 0.335 3, 0.504 5, 0.433 7 and 0.625 9 respectively, in 23 S. alopecuroides populations. This indicated that there was less gene exchange and higher genetic differentiation among different S. alopecuroides populations. The results of SSR unweighted pair-group method with arithmetic means(UPGMA) cluster showed that the S. alopecuroides populations relationship from Xinjiang was far from the populations of other regions, but the populations of S. alopecuroides from Gansu, Inner Mongolia and Qinghai were closely relevant to those from Ningxia. The 23 populations were further divided into 2 genetic subpopulations by the population structure analysis. Through association analysis, a total of 26 loci in 13 SSR markers were found to be significantly associated(P<0.005)with the content of MA, OMA, SC and OSC, and the rate of explanation on the phenotype variance of related markers ranged from 36.45% to 77.93%. Among the locus, 1 each were related with MA and OSC content at interpretation rate reached as high as 50% with high threshold(P<0.000 1). These results could provide support for the discovery of important genes in the alkaloid biosynthetic and metabolic pathway of S. alopecuroides.

Objective: To observe the clinical efficacy of heat-sensitive moxibustion for adjuvant treatment of depression in Parkinson disease and explore its mechanism. Methods: A total of 80 patients with Parkinson disease coupled with depression were randomized into an observation group and a control group, with 40 cases in each group. The control group was treated with levodopa and benserazide hydrochloride tablets and paroxetine tablets, while the observation group was treated with heat-sensitive moxibustion on the basis of the medications in the control group. The treatment course was 2 months. The Hamilton depression scale-17 (HAMD-17), unified Parkinson's disease rating scale (UPDRS) and Parkinson's disease quality of life questionnaire-39 (PDQ-39) were scored before and after the treatment, and the efficacy was evaluated after treatment. Levels of patients' serum dopamine (DA), 5-hydroxytryptamine (5-HT), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were detected before and after the treatment. Results: After treatment, the total effective rate of the observation group was higher than that of the control group (P<0.05). The HAMD-17 scores in the two groups decreased significantly after treatment (both P<0.05), and the score in the observation group was obviously lower than that in the control group (P<0.05). The component scores and total scores of UPDRS in both groups decreased significantly (all P<0.05), and the scores in the observation group were lower than those in the control group (all P<0.05). The score of PDQ-39 in the observation group decreased significantly (P<0.05), and was lower than that in the control group (P<0.05). After treatment, the serum DA and 5-HT levels in the observation group increased significantly (both P<0.05) and the TNF-α and IL-6 levels decreased significantly (both P<0.05), which were statistically different from those in the control group (all P<0.05). Conclusion: Heat-sensitive moxibustion has certain auxiliary effect in treating depression in Parkinson disease, significantly improving clinical symptoms and the quality of life, which may be related to the up-regulation of DA and 5-HT levels and down-regulation of TNF-α and IL-6 levels.

Objective By analyzing and summarizing the acupoint application rules in the treatment of dry eye syndrome (DES) with acupuncture, to provide references and evidences for acupuncture prescriptions.Method Clinical articles published during the recent 10 years on the treatment of DES with acupuncture were collected to summarize and assort the frequency, meridian affiliation and attribution of the commonly-used acupoints.Result Jingming (BL 1) had the highest frequency, and the selected acupoints, predominantly the periocular points, were majorly from the Bladder Meridian of Foot Taiyang.Conclusion In the treatment of DES with acupuncture, periocular acupoints such as Jingming (BL 2), Cuanzhu (BL 2), Taiyang (EX-HN 5) and Sizhukong (TE 23) were mainly selected, and the selection of acupoints should follow the along-meridian and syndrome-differentiation principles.

Cell apoptosis induced by Angiotensin II (Ang II) has a critical role in the development of cardiovascular diseases. The aim of the present study was to investigate whether sanguinarine (SAN), a drug which was proved to have antioxidant, antiproliferative and immune enhancing effects, can abolish cell apoptosis induced by Ang II. In the present study, H9c2 cardiac cells were stimulated with 10 µM Ang II with or without SAN. The level of intracellular reactive oxygen species (ROS) generation was assessed using dichlorodihydrofluorescein diacetate, and changes of the mitochondrial membrane potential (MMP) were assessed using JC1 staining. Furthermore, mRNA expression of NOX2 was determined by reverse transcription quantitative polymerase chain reaction, and apoptosis was detected by Annexin V/propidium iodide staining and flow cytometry. The expression of Bcell lymphoma 2 (Bcl2), Bcl2associated X protein (Bax) as well as cleaved (c)caspase 3 and 9 were detected by western blot analysis, and the activity of caspase 3 and 9 was detected using an ELISA. The results of the present study showed that NOX2 expression and ROS generation induced by Ang II were inhibited by SAN, and the Ang 2induced MMP loss was also ameliorated. Furthermore, Ang IIinduced H9c2 cardiac cell apoptosis as well as ccaspase 3 and 9 levels were significantly reduced by SAN. Investigation of the possible pathway involved in the antiapoptotic effect of SAN showed that the expression of Bcl2 was decreased, while that of Bax was increased following stimulation with Ang II, which was reversed following treatment with SAN. In addition, Ang II enhanced the activity of caspase 9 and cleaved downstream caspases such as caspase3, initiating the caspase cascade, while pretreatment of H9c2 cardiac cells with SAN blocked these effects. In conclusion, the findings of the present study indicated that SAN inhibits the apoptosis of H9c2 cardiac cells induced by Ang II, most likely via restoring ROSmediated decreases of the MMP.

Transmembrane transports of four kinds of lipophilic organic chemicals (LOCs) on suspending multilamellar liposomes (SML) and Escherichia coli (E. coli) were investigated, where both anthracene and phenanthrene were accorded to the lipid-water partition law and Sudan I and III to the Langmuir isothermal adsorption. Less than half of phenanthrene is transported into E. coli, where more than 60% are located in the cytoplasm. About 60% of anthracene entered the E. coli where only 10% was released into the cytoplasm. The partition coefficients of phenanthrene and anthracene partitioning from the extracellular liquid into membrane are 502 and 1190L/kg but their inverse partition coefficients are only 0.180 and 0.018kg/L. Over 60% of Sudan I and less than 40% of Sudan III accumulated on E. coli where most of them remained on the membrane. The transmembrane impedance effect (TMIE) is proposed for evaluating the cell-transport of polar LOCs.
